Associations of physical activity with gut microbiota in pre-adolescent children

  • [Purpose] To determine whether physical activity (PA), primarily the recommended 60 minutes of moderate-to-vigorous PA, is associated with gut bacterial microbiota in 10-year-old children. [Methods] The Block Physical Activity Screener, which provides minutes/day PA variables, was used to determine whether the child met the PA recommendations. 16S rRNA sequencing was performed on stool samples from the children to profile the composition of their gut bacterial microbiota. Differences in alpha diversity metrics (richness, Pielou's evenness, and Faith's phylogenetic diversity) by PA were determined using linear regression, whereas beta diversity (unweighted and weighted UniFrac) relationships were assessed using PERMANOVA. Taxon relative abundance differentials were determined using DESeq2. [Results] The analytic sample included 321 children with both PA and 16S rRNA sequencing data (mean age [SD] =10.2 [0.8] years; 54.2% male; 62.9% African American), where 189 (58.9%) met the PA recommendations. After adjusting for covariates, meeting the PA recommendations as well as minutes/day PA variables were not significantly associated with gut richness, evenness, or diversity (p ≥ 0.19). However, meeting the PA recommendations (weighted UniFrac R2 = 0.014, p = 0.001) was significantly associated with distinct gut bacterial composition. These compositional differences were partly characterized by increased abundance of Megamonas and Anaerovorax as well as specific Christensenellaceae_R-7_group taxa in children with higher PA. [Conclusion] Children who met the recommendations of PA had altered gut microbiota compositions. Whether this translates to a reduced risk of obesity or associated metabolic diseases is still unclear.

  • Purpose: The purpose of this study was to classify mandibular molar furcation involvement (FI) in periapical radiographs using a deep learning algorithm. Materials and Methods: Full mouth series taken at East Carolina University School of Dental Medicine from 2011-2023 were screened. Diagnostic-quality mandibular premolar and molar periapical radiographs with healthy or FI mandibular molars were included. The radiographs were cropped into individual molar images, annotated as "healthy" or "FI," and divided into training, validation, and testing datasets. The images were preprocessed by PyTorch transformations. ResNet-18, a convolutional neural network model, was refined using the PyTorch deep learning framework for the specific imaging classification task. CrossEntropyLoss and the AdamW optimizer were employed for loss function training and optimizing the learning rate, respectively. The images were loaded by PyTorch DataLoader for efficiency. The performance of ResNet-18 algorithm was evaluated with multiple metrics, including training and validation losses, confusion matrix, accuracy, sensitivity, specificity, the receiver operating characteristic (ROC) curve, and the area under the ROC curve. Results: After adequate training, ResNet-18 classified healthy vs. FI molars in the testing set with an accuracy of 96.47%, indicating its suitability for image classification. Conclusion: The deep learning algorithm developed in this study was shown to be promising for classifying mandibular molar FI. It could serve as a valuable supplemental tool for detecting and managing periodontal diseases.

  • In order to obtain basic information on soil inprovement and management of two reclaimed paddy soils, Munpo and Pori series, this study was carried out by investigating the change of physico-chemical and soil mechanical properties. 1. In the Munpo series, the contents of salt in the soil and ground water before paddy rice transplanting at 6 years after reclamation were 0.60 and 1.84%, respectively, and the safty cultivation of paddy rice was possible after 20 years, while that of the Pori series was after 12 years. 2. In the Munpo series, the change of the soil particle size according to the ages after reclamation showed negative correlation with sandy contents, but clay contents had positive correlation both in the surface and subsurface soils. The pori series showed reverse tendency. 3. Between the N-value and Atterberg limits and the ages showed negative correlation in both Munpo and Pori series, but clay-activity showed positive. Arid the cole values showed positive correlation in the Munpo series, but negative in the Pori series. 4. Cone penetrating resistance in the surface of the Munpo series after 6 years of reclamation was 62.9, after 20 years 24.5 and $16.7kg/cm^2$ in 15cm of soil depth at depth of 22.5cm after 27 years. In the Pori series, it was $3kg/cm^2$ at 15-20cm of soil depth after 12 years and it used after being creation of hard pan layer after 23 years. 5. Change of the clay minerals by the different year after accomplishment of tidal reclamation did not show. The major clay minerals were illite and kaolinite, and accessory clay minerals were montmorillonite and vermiculite in both Munpo and Pori series.

Antimicrobial Activity of the Aerial Part (Leaf and Stem) Extracts of Cnidium officinale Makino, a Korean Medicinal Herb (천궁(Cnidium officinale Makino) 지상부(잎과 줄기) 추출물의 항균활성)

  • This study was carried out to investigate the usefulness of the aerial part of Cnidium officinale Makino as a bioactive material source. The aerial part(leaf and stem) of Cnidium officinale Makino was extracted with three kinds of solvents and determined their antimicrobial activities against several bacteria and yeast strains using the paper disc method and the microtiter dilution method. The extracts of the Cnidium offocinale aerial part exhibited the broad spectrum of antibacterial activity against Gram (+) and Gram (-) bacteria, including food-borne pathogens such as Listeria monocytogenes, Salmonella typhimurium, and Staphylococcus aureus. The extracts of Cnidium officinale also showed antifungal activity against Saccharomyces cerevisiae. The ethyl acetate extracts completely inhibited the growth of Staphylococcus aureus and Pseudomonas aerogenes, and moderately inhibited the growth of Escherichia coli and Enterobacter cloacae at the concentration of 0.5 mg/mL. However, water extract of Cnidium officinale exhibited lower antimicrobial activity than ethyl acetate and methanol extracts. The inhibitory effect of the ethyl acetate extract of Cnidium officinale Makino was not destroyed by heating at $100^{\circ}C$ for 30 min or at $121^{\circ}C$... for 15 min. These results suggest that the aerial part of Cnidium officinale Makino could be a useful source for a natural antimicrobial material.

Comparison Study of Extraction Properties of Solids, Protein and Color Pigments of Several Soybean Varieties (콩 품종에 따른 고형분, 단백질, 색소의 추출 특성의 비교)

  • The solid and protein yields and extraction properties of color pigments were compared for 7 varieties of soybeans during soaking in water at $4-100^{\circ}C$. The varieties investigated were Paldal, Danyeob, Jangbaek, Baegun, Jangyeob and 2 cultivars of Local 1 and Local 2. The Hunter values showed that Jangbaek was the highest in 'L' value while other varieties except Local 1 and Local 2 were comparatively high in 'L' value. Local 1 and Local 2 were low in 'b' value. The yields of solid and protein during water extraction showed that most of solids and proteins were recovered with three consecutive extractions. The cumulated yields were 73.2 % for solid and 83.2 % for protein. Extraction of color pigments of seed coats in $4-100^{\circ}C$ water showed that the extraction rate was very much dependent on extraction time and temperature. A linear relationship of A=aT+b was obtained for equilibrated absorbance(A) and extraction temperature(T). The activation energy calculated from initial extraction rate of cole. pigments and temperature had two different values of low($4-60^{\circ}C$) and high($60-100^{\circ}C$) temperature range.

Effecs of Soyangin Indongdeungjikolpitang on the Complication of Diabetes (소양인(小陽人) 인동등지골피탕(忍冬藤地骨皮湯)이 중소합병증(中消合倂症)에 미치는 영향(影響))

  • The purpose of this research was to investigate the effects of Indongdeungjikolpitang water extract(IJTE) on the complication of diabetes. IJTE did not affect the level of blood glucose in alloxan- or streptozotocin-induced hyperglycemic mice, but inhibited the motility of gastrointestine. IJTE inhibited the writhing syndrome induced by acetic acid, the permeability of evans blue into peritoneal cavity induced by acetic acid, the paw edema induced by histamine, and the formation of cotton pellet granuloma. IJTE increased the cell viability of thymocytes and splenocytes. IJTE decreased the release of ${\gamma}-interferone$(${\gamma}-IFN$) and interleukin-2(IL-2), but did not affect the release of interleukin-4(IL-4) from murine thymocytes. IJTE increased the release of IL-4 and decreased the release of tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) and $interleukin-1{\beta}$($IL-1{\beta}$), but did not affect of ${\gamma}-IFN$ and IL-2 from murine splenocytes. IJTE decreased the release of $TNF-{\alpha}$ and $IL-1{\beta}$ from murine peritoneal macrophages. IJTE decreased the production of niric oxide(NO) from murine peritioneal macrophages and increased the phagocytic activity of murine peritoneal macrophages. These results suggest that IJTE has an anti-inflammatory action via the inhibition of $TNF-{\alpha}$, $IL-1{\beta}$ and NO production from immune cells.

EFFECTS OF CONDENSED PHOSPHATES ON THE DENATURATION OF ALASKA POLLACK MUSCLE DURING REFREEZING AN COLE STORAGE (재동결 명태육의 냉동변성에 미치는 축합인산염처리의 효과에 대하여)

  • The present study was ,conducted to evaluate the effects of condensed phosphates on the refeezing damage of Alaska pollack muscle. The fillets were dipped in such solution as 5 and $10\%$ sodium polyphosphate, 1 and $5\%$ mixture of sodium polyphosphate and sodium pyrophosphate (1:1, w/w) for 1 and 5 minutes, respectively, before refreezing. And fillets were frozen at $27^{\circ}C\~28^{\circ}C$ and stored for 15 days at $-18^{\circ}\~-20^{\circ}$. The degree of denaturation was estimated by determining amounts of drip relased, content of total solids, nitrogen, and DNA in the drip an cooking-weight-loss. Phosphorus absorbed in the muscle was also determined. Phosphorus absorbed in the fillets treated with loft solution of sodium polyphosphate for 5 minutes amounted to 101 mg/100g muscle as $P_2O_5$. The absorption was dependent on tile concentration of treating solution rather than on the dipping time. The increase of phosphorus absorption seemed to affect to reduction of drip. Among the treating conditions, $10\%$, 5 minutes and $10\%$ 1 minute with sodium polyphosphate appeared most effective ones on drip reduction. The effect of $5\%$, minutes with the mixture of sodium polyphosphate and sodium pyrophosphate did not show so benefitable effect in refrozen fillets. As a tendency total solids, nitrogen, and DNA in tile drip varied proportionally to the amount of drip released. And the content of DNA was lower than the amount. Treatment, at higher the concentration and longer the dipping time, resulted in the lower cooking-weight-loss and the better quality on organoleptic test of thawed fillets.

Effects of Supplemental Humic Substances on Egg Production and Quality in Laying Hens (Humic Substances의 급여가 산란계의 산란율과 난 특성에 미치는 영향)

  • The effects of dietary humic substances (HS) on egg Production and egg Quality were studied using 252(55-wk old) ISA brown laying hens. laying were divided into 21 groups of 12 hens each and seven groups (experimental units) were assigned to 1) CON (basal diet), 2) HS5 (basal diet 4- 5% humic substances) or 3) HS10 (basal diet +10% humic substances) in a completely randomized block design. Hens had free access to diets and water fur 6 wk. Egg Production and egg quality were monitored over the 6-wk Period. Results showed that 10% dietary HS decreased egg Production and yolk diameter (P<0.05) compared to CON. Egg weight and yolk cole. were improved (P<0.05) in HS10 compared to CON. Egg shell breaking strength was increased significantly (P<0.05) when hens were fed HS5 diet compared to the others. There were no effects of treatments on egg shell thickness, yolk index, albumen height and Haugh nit. The results suggest that the dietary supplementation of HS at 5% or 10% decreases egg Production, but HS at 5% can increase egg shell breaking strength. Hens fed 10% HS could increase egg weight and yolk color and decrease yolk diameter.

Effect of Prunus mume Extract on the Sensory Quality and Shelf Life of Cooked Rice (매실(Prunus mume)추출물이 쌀밥의 관능적 특성 및 저장성에 미치는 효과)

  • This study was investigated the effect of Prunus mume water extract (PW) and alcohol extract (PA) to the sensory properties, cole. value and shelf-life characteristics (pH, titratable acidity and total bacteria count) of cooked rice. Sensory attributes such as taste, color and flavor were significantly higher in cooked rices prepared with 500 ppm of PA than cooked rices prepared with 1000 ppm of PA and PW (p<0.05), but there was no significant difference among samples in texture and overall acceptability. Redness, yellowness and total color difference ($\Delta$E) were higher when PA and PW were added. When cooked rice was putrefied, the value of pH, titratable acidity and total bacteria count were about 5.8, 0.1% and 10$\^$8/ cfu/g, respectively. The shelf life of cooked rice was prolonged to 1∼2 day by adding 500 and 1000 ppm of PA as compared with control group. The shelf life of cooked rice was prolonged to 2∼3 day by adding 500 and 1000 ppm of PW as compared with control group.ith control group.

Physiological Characteristics of Anticoagulant Fractions from Eugenia caryophyllata (정향으로부터 추출한 항응고활성 획분의 기능적 특성)

  • The alkali extraction of anticoagulant from a clove revealed 3-to 6-fold more effective than hot-water extraction. The highest anticoagulant activity was found with 0.1 N NaOH at 7$0^{\circ}C$. The anticoagulant fractions from a clove, EC-2B and EC-2C were separated by alkali extraction, ethanol precipitation, cetavlon treatment, and ultrafiltration. The anticoagulant activities of these two fractions were, respectively, 6.57 and 8.63 times higher than those extracted with hot-water. As of the sensory evaluation, boiled pork added with EC-2B fraction revealed similar sensory acceptability to raw clove material, while EC-2C fraction had low sensory acceptability due to a mild chemical odor. Antibacterial characteristics against pathogenic microorganism of both fractions were confirmed in the control strain. The inhibitory effect of growth by EC-2B was noticed above 0.016% in S. aureus. Also, EC-2C showed the inhibitory effect at 0.004% in both E. cole and S. aureus control strains.
