Efficacy and Safety of Codonopsis lanceolata (S. et Z.) Trautv. Extract on the Improvement of the Hypersensitivity Reaction in Allergic Rhinitis Patients (알레르기 비염 질환자를 대상으로 한 더덕(Codonopsis lanceolata (S. et Z.) Trautv.) 추출물의 과민면역반응 개선에 대한 유효성 및 안전성 평가)

  • Objectives : This clinical trial is performed to investigate the effect of improving the hypersensitivity reaction and the safety of powdered ethanol extract Codonopsis lanceolata (S. et Z.) Trautv. for allergic patients. Methods : This study was randomized, double blinded, placebo-controlled clinical trial. 60 allergic rhinitis subjects were randomly assigned to Treatment group(n=30) and Control group(n=30). They were received the test substance or placebo in their $1^{st}$ and $2^{nd}$ visit, and took it once a day, 1 powder bag per 1 time, for 8 weeks. The freeze dried powder of Codonopsis lanceolata (S. et Z.) Trautv. was mixed with 30% ethanol and the filtrate was collected thrice with constant stirring of the mixture. The placebo was prepared in the same form as the test substance by replacing with corn starch. Total Nasal Symptom Score(TNSS) questionnaires and laboratory tests were performed at the screening and last visit to evaluate the efficacy. And to assess safety, vital signs and adverse events were confirmed at every visit. Statistical analysis about the result was performed using SAS 9.4. Results : In treatment group, there was a significant decrease in B cell after taking the test substance(p<0.005). And there was a statistically significant difference between groups(p<0.005). Changes in nasal symptoms were not statistically significant between groups, and both groups showed statistically significant differences before and after the test(p<0.0001). Also there were no adverse events associated with the clinical trial product. Conclusions : This clinical trial showed that Codonopsis lanceolata (S. et Z.) Trautv. extract was helpful in improving the hypersensitivity reaction in allergic patients without significant side effects.

Changes of Crude Components and Essential Oil Content by Shading Treatment in Codonopsis lanceolata Trautv (차광처리에 따른 더덕의 조성분과 정유성분 변화)

  • This experiment was conducted to increase essential oil and aromatic contents on roots of Codonopsis lanceolata Trautv with different shading treatments. Vine length was prolonged by shading treatment compared with non-shading. Fresh root weight in 75% shading treatment was highest with 13g per plant. Crude components such as crude protein, fat and fiber contents decreased, while crude ash increased as shading degree increased. Forty-eight volatile aromatic compounds in the root were identified by GC/MS. Major aromatic compounds were 1-hexanol, cis-3-hexanol, and trans-2-hexanol. In particular, trans-2-hexanol was highest in the 75% shading treatment attaining the 160.32 % area. Conclusively, it was suggested that the 75% shading treatment was to some extent effective to improve the content of essential oil and aromatics in the roots of Codonopsis lanceolata Trautv.

Discrimination and Genetic Relationship of Adenophorae triphylla(Thunb) A.DC. var. japonica Hara and Codonopsis lanceolata Trauty using RAPD analysis (RAPD분석에 의한 잔대와 더덕의 유연관계 비교 및 감별)

  • Dried parts of the two species are difficult to distinguish morphologically, thus Codonopsis radix has been sold instead of Adenophorae radix in herbal medicine market. Therefore, this study was conducted to develop the genetic marker through the examination of the phylogenetic relationships between two Adenophora triphylla(Thunb.) A. DC. var. japonica Hara, two Adenophora radiatifolia Nakai, five Codonopsis lanceolata(Sieb. et Zucc)Trautv. using RAPD analysis. Fifty decarmer oligonucleotide primers were screened for the RAPD analysis, and four primers generated distinct RAPD markers specific to Adenophorae radix and Codonopsis radix. Based on the RAPD patterns, the genetic relationships between three herbal medicine were analyzed by UPGMA method. As a result, Adenophorae radix and Codonopsis radix were classified into two major subgroups on the basis of the genetic similarity coefficient. The specific RAPD patterns generated by the selected primers were reproducible from dried materials. Furthermore, the specific RAPD patterns were produced from the mixture of dried roots of A. triphylla and C. lanceolata. These results prone the usefulness of the RAPD analysis for the discrimination of pure materials from the mixtures of A. triphylla and C. lanceolata.

Effects of Additional Fertilization after Cultivating Green Manure Crops on the Growth and Saponin Content of Codonopsis lanceolata Trautv. (풋거름작물 재배 후 추비처리가 더덕의 생육 및 사포닌 함량에 미치는 영향)

  • Background: This study was conducted to investigate the effects of fertilization with the application of mixed organic matter after cultivating green manure crops on the growth and saponin content of Codonopsis lanceolata Trautv. Methods and Results: Five type of cultivation of green manure crops were done: hairy vetch as a single seedling crop (HV), hairy vetch and rye as a mixture of seedling (HV/R), additional fertilization with mixed organic matter after cultivation of a seedling mixture (HV/R/MO), chemical fertilizer (CF), and a non-treated control (NT). The total nitrogen content was the highest in the HV/R/MO treatment, followed by the HV, CF, HV/R and NT treatments, respectively. Nitrogen fixation was higher in the single seedling treatment with hairy vetch (HV) than in the treatment with a mixture of hairy vetch and rye seedling (HV/R). Moreover, the growth of C. lanceolata was greatly increased by the application of additional fertilizer after treatment with the mixture of hairy vetch and rye seedlings. The HV/R/MO treatment produced the highest total lancemaside content among the treatments, followed by HV, HV/R, CF, and NT, respectively. Conclusions: It was verified that C. lanceolata experienced superior growth with the application of mixed organic matter as additional fertilizer after the cultivation of green manure crops, and the content of its major saponins, lancemasides were also increased by this treatment.

Isolation and Characterization of Thioredoxin cDNA from Codonopsis lanceolata (S. et Z.) Trautv

  • A thoredoxin (CTRX) gene was cloned and characterized from a full length cDNA library prepared from taproot of three-year old Codonopsis lanceolata. A CTRX was 666 nucleotides long and has an open reading frame of 372 bp with 124 amino acid residues (pI = 4.92). The deduced amino acid sequence of the CTRX matched to the previously reported plant thioredoxin h genes. The deduced amino acid sequence of CTRX exhibited the similarity of 33-67% among previously registered thioredoxin genes. The expression of CTRX in leaves of Codonopsis lanceolata was increased by wounding and 1 mM $H_2O_2$, but decreased by 0.1 mM cadmium.

  • A cytosolic ascorbate peroxidase, hydrogen peroxide-scavenging enzyme, was characterized from Codonopsis lanceolata. The cytosolic ascorbate peroxidase cDNA (CAPX) was 983 nucleotides long and possess an open reading frame of 753 bp with 251 amino acids (MW 27.9 kDa) with pI 5.61. The deduced amino acid sequence of CAPX shows high homology to other known cytosolic APXs ($78{\sim}83%$), but the CAPX was clustered independently from compared ten plant APXs. The CAPX gene was highly expressed in leaf and stem tissues, but not in root. When Codonopsis leaves cut using scalpel were soaked in 1 mM hydorgen peroxide, the expression of CAPX gene was suppressed.

Simultaneous Quantitative Analysis of Natural Complex Extract of Codonopsis lanceolata (Siebold & Zucc.) Trautv., Platycodon grandiflorum A. De Candolle, Glycyrrhiza uralensis Fischer (더덕, 길경, 감초 복합추출물에 대한 지표성분 동시분석 및 함량평가)

  • Fine dust has emerged as seriously hazardrous particles, which can cause respiratory and cardiovascular diseases. Thus, we have developed a natural complex extract consisted of Codonopsis lanceolata (Siebold & Zucc.) Trautv., Platycodon grandiflorum A. De Candolle, and Glycyrrhiza uralensis Fischer in the ratio of 3 : 5 : 2, which have a beneficial effect on respiratory system. In this study, simultaneous quantitative analysis of marker compounds, lobetyolin, platycodin D, and glycyrrhizic acid in the natural complex extract was developed and validated with high performance liquid chromatography-photodiode array detector. The marker compounds were shown in a large linearity with a correlation coefficient (R2) of 1.000. The limit of detection (LOD) of lobetyolin, platycodin D and glycyrrhizic acid were 5.40 ㎍/mL, 3.94 ㎍/mL and 5.86 ㎍/mL, respectively. The limit of quantification (LOQ) of lobetyolin, platycodin D and glycyrrhizic acid were 16.36 ㎍/mL, 11.94 ㎍/mL and 17.75 ㎍/mL, respectively. The content analysis revealed that the amounts of lobetyolin, platycodin D, and glycyrrhizic acid were 0.039±0.013 mg/g, 0.337±0.048 mg/g, and 1.171±0.003 mg/g, respectively, in the natural complex extract. Therefore, these results could be used as basic data for the standardization and quality control of the natural complex extract.

Molecular Cloning and Characterization of Type 2 Metallothionein cDNA from Codonopsis lanceolata (S. et Z.) Trautv

  • A class I type 2 metallothionein (CMet2) cDNA from taproot of Codonopsis lanceolata was isolated and characterized. A CMet2 cDNA was 572 nucleotides long and had an open reading frame of 234 bp with a deduced amino acid sequence of 78 residues (pI = 4.99). The deduced amino acid sequence of CMet2 matched to the previously reported type 2 metallothionein-like protein genes and showed 74% identity with that of G. max (BAD18377) and C. arietinum (CAA65009). Expression of CMet2 by the RT-PCR was increased at 1 hr after cadmium and hydrogen peroxide treatment, respectively.
