Yuklinzu Aqueous Extracts Ameliorate Experimental Climacterium Symptoms Induced by Ovariectomy in Mouse (난소적출로 유발된 갱년기장애 마우스 모델에서 육인주(毓麟珠) 열수 추출물의 증상 개선 효과)

  • Objectives: The object of this study was to observe the complex anti-climacterium potentials of Yuklinzu aqueous extracts (YLZ), using bilateral ovariectomy (OVX) female ddY mice similar to women postmenopausal symptoms, as including cardiovascular diseases, obesity, hyperlipidemia, osteoporosis and hepatic steatosis. Methods: In order to evaluate anti-climacterium effects of YLZ, six groups of mice were used; sham control, OVX control, 17β-estradiol, YLZ 500, 250 and 125 mg/kg treated groups. Since 28 days after bilateral OVX surgery, YLZ were administered orally for 84 days, once a day. And then we evaluated anti-climacterium effects divided into five categories; estrogenic effects, anti-obesity, hypolipidemic effects, hepatoprotective effects and anti-osteoporotic effects. The results of YLZ were compared with 17β-estradiol 0.03 ㎍/head/day subcutaneous treated OVX mice. Results: As a result of OVX, obvious changes related to the estrogen-deficient menopausal symptoms - obesity, hyperlipidemia, hepatic steatosis and osteoporosis were displayed in mice. However, these menopausal symptoms induced by OVX were significantly inhibited by 84 days of consecutive treatment of 17β-estradiol, YLZ 500, 250 and 125 mg/kg, respectively. Especially, YLZ showed obvious dose-dependent inhibitory activities on the OVX-induced climacterium changes in mice, and YLZ 500 mg/kg showed comparable inhibitory effects against menopausal symptoms in comparison with those of 17β-estradiol 0.03 ㎍/head/day subcutaneous treatment. Conclusions: The results suggest that oral administration of YLZ 500, 250 and 125 mg/kg has obvious dose-dependent favorable anti-climacterium effects in OVX mice. Especially, YLZ 500 mg/kg showed comparable inhibitory effects against menopausal symptoms in comparison with those of 17β-estradiol 0.03 ㎍/head/day subcutaneous treatment.

The anti-climacterium effects of red clover dry extracts combined with pomegranate concentration powder in ovariectomized rats

  • Objective : In this study, the addition of dried pomegranate concentrate powder (PCP) was affected the anti-climacterium activity of red clover dry extracts (RC) in ovariectomized (OVX) rats. Materials and methods : After bilateral OVX surgery, RC 40 mg/kg, PCP 20 mg/kg and RC:PCP 2:1 mixture (g/g) 120, 60 and 30 mg/kg (of body weight) were orally administered, once a day for 84 days, and then the changes on the serum estradiol levels, abdominal fat pad and uterus weights were observed for estrogenic effects. In addition, liver weights, serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels were also evaluated for hepatoprotective effects, and serum total cholesterol (TC), low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride (TG) levels were monitored for hypolipidemic effects. Results : As a result of OVX, the estrogen-deficient climacterium symptoms, increments of abdominal fat pad weights, serum AST, ALT, TC, LDL and TG levels with decrease of uterus and liver weights, serum estradiol levels, were demonstrated. However, these estrogen-deficient climacterium symptoms induced by bilateral OVX in rats were significantly inhibited by continuous oral treatment of RC 40 mg/kg, PCP 20 mg/kg and RC:PCP 2:1 mixture (g/g) 120, 60 and 30 mg/kg, respectively. Conclusion : The results suggested that RC:PCP 2:1 mixtures synergistically increased the anti-climacterium effects of RC in OVX rats. It, therefore, is expected that RC:PCP 2:1 mixture will be promising as a new potent protective agents for relieving the climacterium symptoms.

Aqueous Extracts of Jibaekjihwang-tang Ameliorate Ovariectomy-induced Climacterium Symptoms in Mouse (난소적출 마우스에서 지백지황탕 열수 추출물의 갱년기 장애 개선 효과 평가)

  • Objectives: The purpose of this study was to evaluate the anti-climacterium effects of Jibaekjihwang-tang (JBJHT), especially on estrogenic, anti-obesity, hypolipidemic, hepatoprotective against fatty liver and anti-osteoporotic effects by Ovariectomy (OVX) mice. Methods: In order to evaluate anti-climacterium effects of JBJHT, we used bilateral OVX female ddY mice. In this study, six groups were used; sham control, OVX control, estradiol, JBJHT 500, 250 and 125 mg/kg treated groups. Since 28 days after OVX surgery, JBJHT extracts were orally treated, and $17{\beta}$-estradiol $0.03{\mu}g/head$ were subcutaneously injected for 84 days, once a day. And then, we observed anti-climacterium effects classified into five categories; estrogenic, anti-obesity, hypolipidemic, hepatoprotective against fatty liver and anti-osteoporotic effects. The results were compared with $17{\beta}$-estradiol $0.03{\mu}g/head$/day subcutaneous treated OVX mice. Results: OVX control mice showed noticeable hypertrophic changes of adipocytes in abdominal fat pads, fatty liver, uterine atrophic changes, decreases of bone strength were also observed in OVX control. However, these estrogen-deficient climacterium symptoms were significantly and dose-dependently inhibited by JBJHT 500, 250 and 125 mg/kg treatment. Moreover, JBJHT 500 mg/kg showed comparable inhibitory effects as compared to those of estradiol $0.03{\mu}g/head$/day subcutaneous treatment. Conclusions: The results suggest that oral administration of JBJHT 500, 250 and 125 mg/kg has clear dose-dependent anti-climacterium effects in OVX mice.

Anti-climacterium Effects of Gagamguibiondam-tang in Ovariectomized Rats (난소적출로 유발된 랫트 갱년기 장애에 대한 가감귀비온담탕의 생리활성 효과 평가)

  • Purpose: The object of this study was to observe the anti-climacterium activity of Gagamguibiondam-tang (GGOT) on ovariectomized (OVX) rats, a well-documented rodent models resembles with women postmenopausal climacterium symptoms, as including cardiovascular diseases, obesity, hyperlipidemia, osteoporosis, organ steatosis and mental disorders. Methods: In this study, anti-climacteric effects were evaluated separated into three categories; 1) anti-obese, 2) anti-uterine atrophy and 3) anti-osteoporotic effects. Five groups were used (8 rats in each group); sham control, OVX control, GGOT 500, 250 and 125 mg/kg administered groups. Twenty-eight days after bilateral OVX surgery, GGOT were orally administered, once a day for 84 days, and then the changes on the body weight and gain during experimental periods, serum estradiol levels, abdominal fat pad and uterus weights with histopathology of abdominal fat pads (total thickness and mean adipocyte diameters) and uterus (total, epithelial and mucosal thickness, percentages of uterine gland regions) for anti-obese and estrogenic effects. In addition, femur, tibia and fourth or fifth lumbar vertebrae (L4 or L5) wet, dry and ash weights, mineral density (BMD), bone strength (failure load), serum osteocalcin and bone specific alkaline phosphatase (bALP) contents, histological and histomorphometrical analyses - bone mass and structure with bone resorption, were monitored for anti-osteoporosis activity. Results: As a result of OVX, noticeable increases of body weight and gains, food and water consumption, weights of abdominal fat pad deposited in dorsal abdominal cavity, serum osteocalcin levels were demonstrated in this experiment with decrease of uterus, femur, tibia and L5 weights, serum bALP and estradiol levels. In addition, marked hypertrophic changes of adipocytes located in deposited abdominal fat pads, uterine disused atrophic changes, decreases of bone mass and structures of femur, tibia and L4 were also observed in OVX control rats with dramatic increases of bone resorption markers, the Ocn and OS/BS at histopathological and histomorphometrical analysis in this study as compared with sham-operated control rats, suggesting the estrogen-deficient climacterium symptoms - obese and osteoporosis were induced by OVX, respectively. However, these estrogen-deficient climacterium symptoms induced by bilateral OVX in rats were significantly inhibited by 84 days of continuous oral treatment of GGOT 500, 250 and 125 mg/kg, respectively. Especially, GGOT 500, 250 and 125 mg/kg showed clear dose-dependent inhibitory activities on the OVX-induced climacterium signs. Conclusion: The results suggest that oral administration of GGOT 500, 250 and 125 mg/kg has clear dose-dependent favorable anti-climacterium effects - estrogenic, anti-obese and anti-osteoporotic activities in OVX rats in this experiment.

Development of a Genistein-enriched Doenjang Using Corn $\beta$-Glucosidase

  • Genistein, one of the isoflavones in doenjang, is generally known to prevent various cancers, osteoporosis, climacterium, and menopause symptoms, and has better bioavailability and healthful physiological effects than its glucoside, genistin. In both traditional and commercial doenjangs, genistein content ranged from 370 to 1,510 mg/kg, however, significant amounts of genistin also existed at the level of 190 to 350 mg/kg. After treating with corn $\beta$-glucosidase, over 84% of genistin in doenjang was converted to genistein. However, physiochemical characteristics such as pH, viscosity, 2-thiobarbituric acid (TBA) value, and color were not changed significantly after corn $\beta$-glucosidase treatments. Therefore, this study shows that the improved doenjang with the increased genistein content can be produced using corn $\beta$-glucosidase.

  • The climacterium is that phase in the aging precess during which a woman passes from the reproductive to the non-reporductive stage. The signals, such as hot flashes, vaso-motoric disturbances, perspiration, stiff shoulders, emotinal symptoms, are refered to as climacteric disturbances. Treatment of climacteric symptoms centers around estrogen replacement and transfuilizers, but there are many problems to be solved to use these hormones/drugs as far as dossage, duration and complications are concerned. The care of women during the climacteric years should provide relief of distressing symptoms with as high a degree of safety as possible. From this view point, we used red ginseng powder to those patients with high menopausal index successfully. We studied its mechanism of action and proved that red ginseng improved the micro-circulation system via improvement of erythrocyte reformability which enhanced sex steroidgensis consequently.

Depression and Health Promoting Behavior Depending on the Climacteric Symptoms of Middle-aged Male Workers (중년 직장 남성의 갱년기 증상에 따른 우울과 건강증진행위)

  • Purpose: This study is the narrative investigating research to comprehend the climacteric symptoms, depression and health promoting behavior of the male workers and to develop the nursing intervention. Methods: The research was done from August to September 2011, and the subjects were the middle-aged males between 40 and 64 working in capital cities of Chungcheongnamdo. Among the 220 questionnaires, 189 were retrieved and used for the analysis. The data analysis was done by $x^2$-test, and t-test using the SPSS/WIN 19.0 program. Results: The 70.9% of the subjects showed male climacterium and among them, those in their 40s showed high rate to the change in the body and emotions whereas those in their 50s showed high rate to the sexual function-related symptoms. Also, having the climacteric symptoms showed significant changes in depression and health promoting behavior. Conclusion: To promote health of the middle-aged male workers and the nursing intervention for those who show depression, verifying whether these males have climacteric symptoms is necessary.

The Literatual Study of Kamisoyo-san on Menopausal Disorder (갱년기장애에 활용된 가미소요산(加味逍遙散)의 문헌적 고찰)

  • Purpose: The purpose of this study is to discuss the usage of Kamisoyo-san on menopausal disorder by literatual research. Methods: 33 papers and 47 books about Kamisoyo-san and menopausal disorder were used to develop this article. Results: According to the papers and the books, Soyo-san was first appeared in Taepyungheminhwajegookbang. It was modified to Kamisoyo-san and it has been used to treat menopausal disorder for more than several hundreds of years. There is much evidence of the causes, mechanisms, and symptoms of menopausal disorder corresponding with Kamisoyo-san's indications. Currently, scientific research supports that Kamisoyo-san has many effects on the symptoms of menopausal disorder. Conclusions: Kamisoyo-san is effective on menopausal disorder. We suggest that Kamisoyo-san should be given an insurance code for the treatment of menopausal disorder for the benefits to numerous patients suffering from the disorder.

  • I got the following result from the bibliographic study about the meaning of Hwa(火) and Hwa- byung(火病) at the concept and the aspect of pathological and etiological factors.The results are as follows.1. The pathological characteristics of Hwa(火) mean the inflammed state, the excited mental condition and perinervous hyperfunction caused by exopathy produced by any of the six extermal etiological factors(外感六淫) and Hwa caused by interior injury(內傷火熱).2. Hwa caused by the fire emotions(五志之火); anger, gladness, contemplation, sadress, fear by menta complications brings to Hwa caused by the five viscera(五臟之火) and Hwa caused by the stagnation of the liver - energy(肝鬱之火) is the rpresentive Hwa caused by the five viscera and the six hollow organs(贓腑之火).3. A symptom caused by a pathogenic Hwa(火證) is understood in a pathological sense involved an inflammed disease and is considered as a pathological state of excess syndrome(實證) caused by exterior Hwa( 外火).4. The pathologic process of Hwa-byung(火病) as the mental disease is based upon our people's emotion; Han(恨) to be pathologized without being overcome and can be considered as the process of Hwa caused by long-term(鬱久而火化).5. Hwa-byung(火病) means a psychosomatic disorder resulting from stress reaction caused by personal relations in the climacterium.Form the above mentioned study, the meaning of Hwa(火) and Hwa-byung(火病) is set theoretically and it also needs to be maintained reserching and studying about Hwa-byung(火病) consistantly.

A Systematic Review of Non-pharmacological Intervention for Depression in Korean Middle-aged Women (한국 중년여성의 비약물적 우울중재연구: 체계적 문헌고찰)

  • The purpose of this study was to review the literature and examine the effects on non-pharmacological depression intervention for middle-aged women in Korea. This systematic review analyzed the literature from 2006 to September, 2015. The existing literature was searched in the electronic databases in RISS, KISS and DBPia using the following key words: middle-aged women and depression, menopause and depression, climacterium and depression. Two hundred eighty studies were found through the database. According to PICOTS-SD, twelve studies were included for analysis. Among the twelve studies, four studies were conducted in the nursing field. Six studies were randomized controlled trials. Aerobic exercise, laughter therapy, meditation, aroma massage, counseling and art therapy were performed as non-pharmacological interventions for depression in Korean middle-aged women. The length of each session ranged from 20 to 120 minutes. The intervention period varied from 2 to 24 weeks, and the total number of the interventions ranged from 8 to 72 times. They influenced not only depression, but also physical aspects that are associated with obesity and psychosocial variables, such as anxiety, quality of life, life satisfaction, etc.
