Comparison Between Phylogenetic Relationships Based on 18S rDNA Sequences and Growth by Salinity of Chlorella-like Species (Chlorophyta)

  • This study was carried out to understand the correlation between phylogenetic relationships based on 18S rDNA sequences and growth by salinity of Chlorella-like species. The 18S rDNA sequences of 71 Chlorella-like species which were mainly collected from Korean waters were analyzed. The 18S rDNA sequences of Chlorella-like species were divided into three groups (group A, B and C) and group B was further divided into three subgroups (subgroup B-1, B-2 and B-3). Thirty-seven Chlorella-like species in group A grew well at high salinity (32 psu) but the other groups grew well in freshwater. The sequence identities of the species in group A and B were 97.2-99.5%, but those of 6 species in group C ("Chlorella" saccharophila), which contained group I intron sequences region were 75.0-75.4%. Two representative species of each group were cultured at different salinities (0, 16 and 32 psu) to examine the correlation between the molecular phylogenetic groups and the phenotypic characteristics on cell growth and size by different salinities. The size of cell cultured at different salinities varied according to the species of each molecular phylogenetic group. The size of "Chlorella" saccharophila in group C was bigger and more obviously elliptical rather than that of the other Chlorella-like species. Considering the results on molecular and phenotypic characteristics, the group A and B belonged to Chlorellaceae, but group C was distinctly different from them.

독도의 해조상 및 군집구조 (Marine Algal Flora and Community Structure in Dokdo, East Sea, Korea)

  • We investigated species composition and variation of benthic marine algae between 2003 and 2006 at Dokdo on the East Sea. The total number of benthic algae species identified was 96, which included 15 Chlorophyta, 30 Phaeophyta, 50 Rhodophyta and 1 Spermatophyta species. The representative species in this study were Ulva pertusa, Caulerpa okamurae, Codium adhaerens, C. frgile, Undaria pinnatifida, Ecklonia cava, Eisenia bicyclis, Dictyopteris undulata, Padina arborescens, Sargassum horneri, Hildenbrandtia rubra, Lithophyllum okamurae, Amphiroa dilatata, Corallina officinalis, C. pilulifera, Prionitis cornea, Grateloupia elliptica, Plocamium telgairiae, Chondrus ocellatus, Chondracanthus intermedia, Chondria crassicaulis, Polysiphonia morrowii, Melobesioidean algae and Phyllospadix sp. Wet weight biomass according to various depths ranged between 1,094.8 to $2,595.3\;g\;m^{-2}$ during the study period. Mean biomass at the investigated sites was greater in the 15m depth range than in the 5 and 10m depths. Vertical distribution was characterized by Ulva intestinalis, U. linza, Endarachne binghamiae, Bangia atropurpurea, Gloiopeltis furcata and Chondria crassicaulis at intertidal zone, Amphiroa dilatata, Corallina pilulifera, Gelidium amansii, Eisenia bicyclis and Ecklonia cava at 2m depth, Ecklonia cava, Eisenia bicyclis, Padina arborescens, Undaria pinnatifida and Sargasum horneri at 5m depth, Ecklonia cava, Sargassum horneri and Padina arborescens at 10m depth, Ecklonia cava, Myagropsis myagroides, Padina arborescens and Sargassum horneri at 15m depth. The R/P, C/P and (R+C)/P value were 1.67, 0.50 and 2.17, respectively.

Life History and Systematic Studies of Pseudothrix borealis gen. et sp. nov. (=North Pacific Capsosiphon groenlandicus, Ulotrichaceae, Chlorophyta)

  • We cultured a tubular marine green alga, originally identified as Capsosiphon groenlandicus (J. Agardh) K.L. Vinogradova, from Amaknak Island, Alaska. The alga had an alternation of heteromorphic generations in which tubular monoecious fronds produced quadriflagellate zoospores and/or biflagellate isogametes. The gametes fused to produce cysts or Codiolum-like zygotes with long, tortuous stalks. Cysts and codiola produced 8-16 aplanospores, which germinated in situ to yield upright fronds. Fronds arising from both aplanospores and zoospores displayed a distinctive development in which non-septate colorless rhizoids from the base of the initially uniseriate, Ulothrix-like filament were transformed into septate uniseriate Ulothrix-like photosynthetic filaments. These transformed filaments then developed new basal non-septate rhizoids. This pattern of rhizoids becoming filaments, which then produced new rhizoids, was repeated to yield a tuft of up to 50 fronds. Periclinal and longitudinal divisions occurred in each filament, starting basally, until the mature tubular thallus was achieved. Pyrenoid ultrastructure revealed several short inward extensions of chloroplast lamellae, each of which was surrounded by pyrenoglobuli. Analysis of ribosomal SSU and ITS sequences placed this alga in the family Ulotrichaceae, order Ulotrichales, together with but as a distinct species from North Atlantic Capsosiphon groenlandicus. Analysis of a partial ITS sequence from authentic Capsosiphon fulvescens, the current name of the type of the genus Capsosiphon, indicated that neither our material nor C. groenlandicus belongs in that genus, and we propose a new genus, Pseudothrix, to accommodate both species. We propose P. borealis for the North Pacific entity formerly called C. groenlandicus and make the new combination P. groenlandica for the Atlantic species.

Effects of future climate conditions on photosynthesis and biochemical component of Ulva pertusa (Chlorophyta)

  • Ulva pertusa, a common bloom-forming green alga, was used as a model system to examine the effects of elevated carbon dioxide (CO2) and temperature on growth and photosynthetic performance. To do this, U. pertusa was grown under four temperature and CO2 conditions; ambient CO2 (400 μatm) and temperature (16℃) (i.e., present), elevated temperature only (19℃) (ET; i.e., warming), elevated CO2 only (1,000 μatm) (EC; i.e., acidification), and elevated temperature and CO2 (ET and EC; i.e., greenhouse), and its steady state photosynthetic performance evaluated. Maximum gross photosynthetic rates (GPmax) were highest under EC conditions and lowest under ET conditions. Further, ET conditions resulted in decreased rate of dark respiration (Rd), but growth of U. pertusa was higher under ET conditions than under ambient temperature conditions. In order to evaluate external carbonic anhydrase (eCA) activity, photosynthesis was measured at 70 μmol photons m−2 s−1 in the presence or absence of the eCA inhibitor acetazolamide (AZ), which inhibited photosynthetic rates in all treatments, indicating eCA activity. However, while AZ reduced U. pertusa photosynthesis in all treatments, this reduction was lower under ambient CO2 conditions (both present and warming) compared to EC conditions (both acidification and greenhouse). Moreover, Chlorophyll a and glucose contents in U. pertusa tissues declined under ET conditions (both warming and greenhouse) in conjunction with reduced GPmax and Rd. Overall, our results indicate that the interaction of EC and ET would offset each other’s impacts on photosynthesis and biochemical composition as related to carbon balance of U. pertusa.

Growth responses of Chondrus ocellatus Holmes (Gigartinales, Rhodophyta) to two endophytes, Mikrosyphar zosterae Kuckuck (Ectocarpales, Ochrophyta) and Ulvella ramosa (N. L. Gardner) R. Nielsen (Ulvales, Chlorophyta) in culture

  • To examine the effects of two endophytic algae, Mikrosyphar zosterae (brown alga) and Ulvella ramosa (green alga), on the host Chondrus ocellatus (red alga), culture experiments were conducted. Four treatments were made: endophyte-free (Chondrus only), endophyte-M (Chondrus + Mikrosyphar), endophyte-U (Chondrus + Ulvella), and endophytes-M U (Chondrus + Mikrosyphar + Ulvella). After 3 weeks, the relative growth rates (RGRs) of frond lengths and the number of newly formed bladelets were examined. M. zosterae formed wart-like dots on C. ocellatus fronds, whereas U. ramosa made dark spots. The RGRs of frond lengths of C. ocellatus were significantly greater in the endophyte-free and endophyte-M treatment groups than in the endophyte-U and endophytes-M U treatment groups, indicating that the growth of host C. ocellatus was inhibited more by the green endophyte U. ramosa than the brown endophyte M. zosterae. The number of newly produced bladelets was greater in the endophyte-U and endophytes-M U groups than in the endophyte-free and endophyte-M treatment groups. These results indicate that the two endophytes inhibit growth of the host C. ocellatus. The negative effects of U. ramosa on C. ocellatus growth were more severe than those caused by M. zosterae. Furthermore, U. ramosa destroyed the apical meristems of C. ocellatus, whereas M. zosterae did not. On the other hand, C. ocellatus showed compensatory growth in the form of lateral branch production as U. ramosa attacked its apical meristems.

진해만 해역의 조간대에 서식하는 해조류의 군집 구조 (Marine Benthic Algal Community at Intertidal Zone in Jinhae Bay)

  • 진해만 조간대에서 서식하는 해조류 군집 구조를 파악하기 위하여 2007년에 8개 정점을 대상으로 계절별로 조사하였다. 조사기간 동안 총 45종의 해조류가 서식하였으며, 이 중 녹조류 4종, 갈조류 10종, 그리고 홍조류가 29종으로 나타났다. 우점종은 Ulva pertusa, U. conglobata, Sargassum horneri, Gelidium divaricatum, Gracilaria textorii, Polysiphonia morrowii으로 나타났다. 출현종수는 계절 및 정점에 따라 변하였다. 겨울에 가장 많은 42종이 출현하였으며, 여름에는 33종으로 가장 적었다. 정점별로는 정점 1,7,8에서 가장 많았으며, 정점 2와 3에서 가장 낮게 나타났나. 해조류 군집의 공간적 분포를 파악하기 위한 MDS 분석 결과에서는 해조류의 종조성 및 현존량이 각 정점의 물리해양학적인 특징 및 지형적인 차이에 의한 것으로 나타났나.

The phytohormone abscisic acid increases triacylglycerol content in the green microalga Chlorella saccharophila (Chlorophyta)

  • Microalgae are currently a very promising source of biomass and triacylglycerol (TAG) for biofuels. In a previous study, we identified Chlorella saccharophila as a suitable source of oil for biodiesel production because it showed high biomass and lipid content with an appropriate fatty acid methyl esters profile. To improve the TAG accumulation in C. saccharophila, in this study we evaluated the effect of abscisic acid (ABA) addition on cell concentration, lipid content and TAG production in this microalga. First, we evaluated the effects of four ABA concentrations (1, 4, 10, and 20 μM) added at the beginning of a single-stage cultivation strategy, and found that all concentrations tested significantly increased cell concentration and TAG content in C. saccharophila. We then evaluated the addition of 1 μM ABA during the second stage of a two-stage cultivation strategy and compared it with a nitrogen deficiency treatment (ND) and a combination of ND and ABA (ND + ABA). Although ABA alone significantly increased lipid and TAG contents compared with the control, ND showed significantly higher TAG content, and ND + ABA showed the highest TAG content. When comparing the results of both strategies, we found a superior response in terms of TAG accumulation with the addition of 1 μM ABA at the beginning of a single-stage cultivation system. This strategy is a simple and effective way to improve the TAG content in C. saccharophila and probably other microalgae as a feedstock for biodiesel production.

녹조류 청각의 생식방법을 이용한 인공종묘생산 (Artificial Seed Production Using the Reproduction Methods in Codium fragile (Chlorophyta))

  • Codium fragile (Suringar) Hariot, an edible green alga is farmed in Korea by natural blooming zygotes attachment. Experiments were conducted to reveal the conditions for artificial seed production of C. fragile by sexual and asexual reproduction. Growth was compared between zygotes attachment (sexual reproduction) and isolated utricles with medullary filaments (asexual reproduction). Zygotes and isolated utricles with medullary filaments were cultured under different light conditions (10, 20, 40, 60 and $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and temperatures (5, 10, 15, 20 and $25^{\circ}C$) under 16:8LD. Maximum growth of zygote was $261.3{\pm}21.0\;{\mu}m$ under $15^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ after 13 days culture. Maximum regeneration of isolated medullary filament was $8.1{\pm}1.7\;mm$ per one isolated utricle under $20^{\circ}C$ and $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ after 15 days culture. After intermediate culture during two months in the field, morphogenesis occurred in both sexual and asexual reproduction, and growth of young thalli was not significantly different (p>0.05) between the both reproduction methods. Even though seed production of C. fragile is possible in both sexual and asexual reproduction, the mass artificial seed production of asexual reproduction is much more effective than that of sexual reproduction that is too much affected by maturity.

녹조류 참홑파래 (Monostromu nitidum)의 생태와 초기생장 (Ecology and Early Growth of Monostroma nitidum (Ulvales, Chlorophyta) in Korea)

  • This study investigated the ecology and growth of Monostroma nitidum Wittrock in both its natural habitat and the laboratory. The maximum length, width, and weight of M. nitidum in March were 9.0$\pm$4.7 cm, 9.6$\pm$3.6 cm, and 1.52$\pm$1.13 g, respectively. Yellowish-green or yellowish-brown reproductive thalli began to appear in January, and over 80% of the thalli matured by March. The male and female spores were ca. 6 $\mu$m long, and elongate and ovoid in shape. The spores had two flagella and one-eye spot, and tended to swim toward light. Maximum number of spores released from matured thalli was 236 cells/mL after 70 minutes at a light intensity of 100 $\mu$mol/m$^2$/s. The zygote diameter ranged from 3.4-6.0 $\mu$m (mean 4.2 $$m) and increased to 69.8 $\mu$m 14 weeks after culture. The mass release of zoospores was observed from thalli in the dark (3 to 12 days), after 30 min under dry conditions in the shade, at 25$^{\circ}C$, and a light intensity of 100 $\mu$mol/m$^2$/s. The maximum number of zoospores released was 109.8 cells/mL after 60 min of induction. M. nitidum fronds on the net increased to 6.8-7.2 cm in length, and 6.6-8.9 cm in width during the winter.

한국 연안 해조류 생물량의 연간 변동 양상: 경상남도 욕지도 지역 (Temporal Variation of Seaweed Biomass in Korean Coasts: Yokjido, Gyeongnam Province)

  • The seasonal variation of marine algal biomass and community were studied with nondestructive sampling and quadrat method during May 2006 to April 2008 in the intertidal and subtidal zone at Yokjido. As result, total 120 species, 13 Chlorophyta, 34 Phaeophyta, 71 Rhodophyta and 2 Spermatophyta were identified in this study. The major species which contributed significantly to the total biomass were Ulva pertusa, Undaria pinnatifida, Ecklonia stolonifera, Sargassum horneri, S. serratifolium, Gelidium amansii and Prionitis cornea. The mean biomass was 235.5 g wet wt m$^{-2}$ in intertidal, 1,038.9 g wet wt m$^{-2}$ in 1 m, 1,013.1 g wet wt m$^{-2}$ in 5 m, and 19.7 g wet wt m$^{-2}$ in 10 m depths of mean sea level. Seasonal biomass change in intertidal zone, the peak season was in spring, while the lowest was in autumn. The vertical distribution of marine vegetation was characterized by Ulva pertusa - Gelidium divaricatum - Gloiopeltis furcata - Sargassum thunbergii - Hizikia fusiformis at intertidal zone, and Caulerpa okamurae - Gelidium amansii - Chondrus ocellatus - Ecklonia stolonifera - Sargassum horneri - Gracilaria textorii at subtidal zone. These result indicates that the marine algal species and biomass of 1 m and 5 m depths in subtidal zone are greater than intertidal zone, and Ecklonia stolonifera and Sargassum spp. are the dominant species of Yokjido.
