The Stimultaneous Determination of Phenolic Compounds by GC and GC/MS

  • To develop a simple, rapid and simultaneous analytical method of phenolic compounds using gas chromatography (GC) and gas chromatography/mass spectrophometer (GC/Ms), this experiment was carried out to search the retention times of capillary columns and the characteristics of fragment ions in electron impact mass spectra. Most of trimethylsilyl derivatives and underivatized phenolic compounds were separated very well on three kinds of capillary columns(HP-1), Ultra-2 and HP-35). Quantitiative determination of phenolic compounds was achieved by internal standards (p-hydroxybenzoic acid iopropyl ester, p-hydroxybenzoic acid ethyl ester). Calibration plts were linear in the investigated range, and the limits of detection were about 5 ng at split mode method. When analyzed by three columns, theseparation times were fairly constant on two nonpolar columns, but a few compounds showed slightly different separation order by the itnermediate polar HP-35 column. The important characteristic patterns of TMS derivatives of phenolic compounds on the EI/MS spectrra appeared at the base peak of [M-15]+ ion and presented at high abundance in most TMS derivatives of phenoloc compounds. [M]+, [M-CH3-COO]+, [M-Si(CH3)4]+ and [M-Si(CH3)4 -CH3]+ also observed in mass spectra of these compounds . Although several compounds have the same retention times on GC column, it might be possible to identify these compounds by the different patternsof mass frgement ions. The TMS derivatives, thus , provide additional information for identification of phenolic compounds in biological systems.

Sex determination by radiographic localization of the inferior alveolar canal using cone-beam computed tomography in an Egyptian population

  • Purpose: The purpose of this study was to evaluate possible differences in the location of the inferior alveolar canal in male and female Egyptians. Materials and Methods: This cross-sectional retrospective study involved the evaluation of 210 CBCT scans of Egyptian individuals (18-70 years old). The inferior alveolar canal was localized by measuring 8 linear dimensions: 2 for the vertical localization of the mental foramen (superior and inferior to the mental foramen), 4 at the first molar bifurcation for the vertical and horizontal localization of the inferior alveolar canal (superior, inferior, buccal, and lingual to the inferior alveolar canal), and 2 for the horizontal localization of the mandibular foramen (anterior and posterior to the mandibular foramen). The measurements were statistically analyzed via comparative analysis, stepwise logistic regression, and receiver operating characteristic (ROC) curve analysis. Results: Six of the 8 measured distances differed to a statistically significant extent between the sexes. Regression analysis suggested a logistic function with a concordance index of 84%. The diagnostic accuracy capabilities of the linear measurements as sex predictors were calculated using ROC analysis, and the 6 best predictors for sex determination were selected and ranked from highest to lowest predictive power. Moreover, combining these 6 predictors increased the predictive power to 84%. Conclusion: The location of the inferior alveolar canal in the Egyptian population varies significantly by sex; accordingly, this anatomic landmark could be used as a reliable indicator of sexual dimorphism.

Determination of the Strouhal number based on the aerodynamic behavior of rectangular cylinders

  • The Strouhal number is an important nondimensional number which is explanatory of aerodynamic instability phenomena. It takes on the different characteristic constant value depending upon the cross-sectional shape of the body being enveloped by the flow. A number of investigations into this subject, especially on the drag test, surface pressure test and hot-wire test, have been carried out under the fixed state of the body in the past. However, almost no investigations concerning the determination of the St on wind-induced vibration of the body have been reported in the past even though the aerodynamic behavior of the body is very important because the construction of wind-sensitive structures is recently on the sharp increase. Based on a series of wind tunnel tests, this paper addresses a new method to determine the Strouhal number of rectangular cylinder in the uniform flow. The central idea of the proposed method is that the Strouhal number can be obtained directly by the aerodynamic behaviors of the body through wind-induced vibration test. The validity of proposed method is evaluated by comparing with the results obtained by previous studies in three B/Ds at attack angle $0^{\circ}$ and a square cylinder with various attack angles. The values and trends of the proposed Strouhal numbers are in good agreements with values of previous studies. And also, the Strouhal numbers of B/D=1.5 and 2.0 with various attack angles are obtained by the proposed method and verified by other method. This proposed method is as good as any other previous methods to obtain the Strouhal number.

A Study on the Simultaneous Determination of Residual Zeranol, Zearalenone and Their Metabolites in Beef by Gas Chromatography/Mass Spectrometry (Gas Chromatography/Mass Spectrometry에 의한 우육 중의 잔류 Zeranol, Zearalenone 및 그 대사산물들의 동시 분석법에 대한 연구)

  • A Simultaneous determination method was improved for the determination and confirmation of zeranol, zearalenone, as well as their isomers and metabolites, in beef. The analytes were extracted from tissue by CH3CN, hydrolyzed enzymatically(for glucuronide conjugates), cleaned up by a strong basic anion exchange resin combined with a liquid/liquid partitioning, derivatized using MSTFA and confirmed, quantified by GC/MS/SIM with a internal standard, zearalane. The results were as follows : (1) all the estrogens were separated on the GC/MS chromatogram under the extraction method and the chromatographic conditions improved, the retention times of zearalane-TMS2, zearalanone-TMS2, zearalenone-TMS2, zeranol-TMS3, taleranol-TMS3, and $\alpha$-zearalenol-TMS3, $\beta$-zearalenol-TMS3, were 18.49, 19.44, 19.63, 19.71, 19.79 and 19.99, 20.08 minutes, respectively. (2) The calibration curves of residual zeranol, zearalenone and their metabolites showed constantly linear(r=0.99) in the range of 5~20 ng. The minimum detection concentration of residual zeranol, zearalenone and their metabolites was 1 ppb. (3) The total average recovery of residual zeranol, zearalenone and their metabolites from spiked beef was 60.2%(CV=29.7%) at the 1 ppb and 63.5%(CV=26.5) at the 2 ppb, 72.9%(CV=18.2%) at the 4 ppb. (4) The preservation method for 6 estrogens was improved for the fast running time(21 min) and MSTFA was utilized for derivatizing 6 estrogens for improvement of recovery, for good resolution, for characteristic mass spectra unlike Jose's method and Tina's method. The utilization of zearalane as internal standard showed good quantification result for zeranol, zearalenone, as well as their isomers and metabolites, in beef.

Determination of isoquinoline alkaloids by UPLC-ESI-Q-TOF MS: Application to Chelidonium majus L.

  • In this study, we set up an analytical method that can be used for rapid and accurate determination of representative isoquinoline alkaloids in medicinal plants using UPLC-ESI-Q-TOF MS (ultra pressure liquid chromatography-electrospray ionization-quadrupole-time-of-flight mass spectrometry). The compounds were eluted on a C18 column with 0.1 % formic acid and acetonitrile, and separated with good resolution within 13 min. Each of the separated components was characterized by precursor ions (generated by ESI-Q-TOF) and fragment ions (produced by collision-induced dissociation, CID), which were used as a reliable database. We also performed method validation: analytes showed excellent linearity ($R^2$, 0.9971-0.9996), LOD (5-25 ng/mL), LOQ (17-82 ng/mL), accuracy (91.6-97.4 %) as well as intra- and inter-day precisions (RSD, 1.8-3.2 %). In the analysis of Chelidonium majus L., magnoflorine, coptisine, sanguinarine, berberine and palmatine were detected by matching retention times and characteristic fragment ion patterns of reference standards. We also confirmed that, among the quantified components, coptisine was present in the highest quantity. Furthermore, alkaloid profiling was carried out by analyzing the fragment ion patterns corresponding to peaks of unknown components. In this manner, protopine, chelidonine, stylopine, dihydroberberine, canadine, and nitidine were tentatively identified. We also proposed the molecular structure of the fragment ions that appear in the mass spectrum. Therefore, we concluded that our suggested method for the determination of major isoquinoline alkaloids by UPLC-Q-TOF can be useful not only for quality control, but also for rapid and accurate investigation of phytochemical constituents of medicinal plants.

Sex Determination of Scolopendra Subspinipes through Morphological Characteristic Analysis (형태적 특성 분석을 통한 왕지네(Scolopendra subspinipes) 암·수판별)

  • The centipede is predatory arthropod that form head and numerous segments. It is known that there are about 3,000 species worldwide and 44 species (four species and seven families) in Korea. Among them, Scolopendra subspinipes called "Wang-ji-ne" has been known as an important medicinal resource and its value is expected to increase in the future. In this study, the Scolopendra subspinipes in Korea was classified as Chilopoda through morphological analysis of forcipule and venom gland, and a method for sex determination was established. These results are expected to be used as basic data for artificial breeding of Scolopendra subspinipes in the future, and through this, it is expected that it will greatly contribute to the expansion of the Scolopendra subspinipes market as medicinal resources.

Comparison of Temperature Distribution in Agar Phantom and Gel Bolus Phantom by Radiofrequency Hyperthermia

  • The usefulness of Gel Bolus phantom was investigated by comparing the temperature distribution characteristic of the agar phantom produced to investigate the dose distribution characteristic of radiofrequency hyperthermia device with that of the Gel Bolus phantom under conditions similar to those of an agar phantom that can continuously carry out temperature measurement. The temperatures of the agar phantom and the Gel Bolus phantom were raised to $36.5{\pm}3^{\circ}C$ and a temperature sensing was inserted at depths of 5, 10, and 15 cm from the phantom central axis. The temperature increase rate and the coefficient of determination were analyzed while applying output powers of 100 W and 150 W, respectively, at intervals of 1 min for 60 min under conditions where the indoor temperature was in the range $24.5{\sim}27.5^{\circ}C$, humidity was 35~40%, internal cooling temperature of the electrode was $20^{\circ}C$, size of the upper electrode was 250 mm, and the size of the lower electrode was 250 mm. The coefficients of determination of 150 W output power at the depth point of 5 cm from the central axis of the phantom were analyzed to be 0.9946 and 0.9926 in the agar and Gel Bolus phantoms, respectively; moreover, the temperature change equation of the agar and Gel Bolus phantoms with time can be expressed as follows in the state the phantom temperature is raised to $36^{\circ}C:Y(G)$ is equation of Gel Bolus phantoms (in 5 cm depth) applying output power of 150 W. Y(G)=0.157X+36. It can be seen that if the temperature is measured in this case, the Gel Bolus phantom value can be converted to the measured value of the agar phantom. As a result of comparing the temperature distribution characteristics of the agar phantom of a human-body-equivalent material with those of the Gel Bolus phantom that can be continuously used, the usefulness of Gel Bolus phantom was exhibited.

Movement Analyzing of Soil Moisture at a Hillslope Scale Considering Spatial-Temporal and Seasonal Characteristics (시공간적, 계절적 특성을 고려한 사면에서의 토양수분의 거동파악)

  • In order to analyze movement of soil moisture, Time Domain Reflectometry(TDR) with multiplex system has been installed at the Bumreunsa hillslope of Sulmachun Watershed to configure spatial-temporal variation pattern considering seasonal characteristic. An intensive surveying was performed to build a refined digital elevation model(DEM) and flow determination algorithms with inverse surveying have been applied to establish an efficient soil moisture monitoring system. Soil moisture data were collected through an intensive and long term monitoring 380 hrs in November of 2003 and 1037 hrs in May and June of 2004. Soil moisture data shows corresponding variation characteristics of soil moisture on the up slope, buffer, main channel zones of the hillslope which were classified from terrain analysis. Inferences and limitations of measured soil moisture data were discussed in conjunction with flow characteristic through terrain analysis.

  • A technique to measure the adsorption characteristic of surface area and pore size of a ceramic thin film is proposed, and its performance is examine. The thin film is fabricated directly on the resonator surface to measure the adsorption capacity of the film as it is, and using carbon dioxide makes the measurement easy. The results indicates that the measured surface area is satisfactory, while the pore size has some error. It is suggested that readily available carbon dioxide can be used to determine adsorption capacity of thin film at room temperature.

VPI Varnishing Technology Effects on Frequency Characteristics of an Air Core Inductor Used in LISN Circuit Application

  • The functional characteristic of LISN circuit, which is used for measurements of conductive noise in mains power line, is basically related to frequency characteristics of passive elements like inductors used in the circuit as well as the frequency response of inductors is highly related to the resins used in the varnishing process. The significant problem in determination of an inductor's frequency characteristic is the intrinsic resistance, inductance and parasitic capacitance. In this triplet, the parasitic capacitance is the major limiting factor of inductor's frequency range. This capacitance depends on inductor design parameters and materials filling the spaces of coil like resin and its coherency after curing process. In this paper, two similar inductors were designed and built. The first inductor was not varnished while the second one was varnished with VPI technology. VPI, or Vacuum, Pressure, Impregnation technology is one of the most reliable methods performing good insulating conditions for electrical circuits and windings based on resins. The measured results show that implying varnishing technology does not significantly affect the frequency response. However, due to mechanical solidity aspects and improved environmental protection, it is better to varnish the inductors.
