Expression of the Brother of the Regulator of Imprinted Sites Gene in the Sputum of Patients with Lung Cancer

  • Background: Brother of the regulator of imprinted sites (BORIS) is a putative new oncogene that is classified as a cancer germline gene; however, its role in the development of cancer is unclear. This study investigated the expression of BORIS in lung cancer and its clinical implications. Methods: The expression of BORIS messenger ribonucleic acid (mRNA) in the sputum of 100 patients with lung cancer (50 with squamous cell carcinoma, 36 with adenocarcinoma, and 14 with small-cell carcinoma) was evaluated by reverse transcription polymerase chain reaction. Results: The overall expression rate of BORIS in patients with lung cancer was 36.0%: 19 of 50 squamous cell carcinomas (38.0%), 13 of 36 adenocarcinomas (36.1%), and 4 of 14 (28.6%) small-cell carcinomas. There was no significant difference in the BORIS expression according to age, gender, or histologic type. However, the mRNA expression of BORIS was significantly related to the pathologic cancer stage (p=0.004) and lymph node metastasis (p=0.001). The expression of the melanoma antigen gene family A1-6 was not associated with the expression of BORIS. Conclusion: Our results suggest that the expression of BORIS might be a negative prognostic factor in lung cancers and implicate BORIS as a molecular target for immunotherapy.

익수영진고가미방의 지황박, 복령피 배합과 발효에 따른 제제가 면역활성에 미치는 영향 (The effect of prescriptions prepared by adding medicinal herbs(Rehmannia glutinosa Residue, Poria cocos Bark) and fermenting herbal materials based on formulas Iksuyoungjingo on immunological activity)

  • Objectives : This study was carried out to enhance the activity of Iksuyoungjingo, which has the tonify Qi, nourish Yin effect of oriental medicine, and to study the effect of the prescriptions on the immunological activities. Methods : Immunosuppression was induced by methotrexate (MTX) 2 mg / kg, the experimental group was divided into IYGgami-I, IYGgami-I-F, IYGgami-II and IYGgami-II-F groups. Each prescription was administered with drinking water for 20 days, and body weight was measured every 5 days during this period. Leukocyte, $TNF-{\alpha}$, IL-2, IL-6, IgE, spleen weight and body weight were measured Results : In the changes of $TNF-{\alpha}$, IL-2 and IL-6 as pro-inflammatory elements, all of the experimental groups showed a significant increase compared to the control group. In the IgE changes, the IYGgami-I-F, IYGgami-II and IYGgami-II-F groups showed a significant decrease compared to the control group. In the changes of spleen weight, the IYGgami-II-F group showed a significant increase compared to the control group. In the changes of WBC and lymphocytes, the IYGgami-I-F group showed a significant increase compared to the control group. Conclusions : From the above results, it can be observed that the efficacy against immunity is exerted in all of the preparations, and it was confirmed that the efficacy was maintained constant even when utilizing the Rehmannia glutinosa Residue and Poria cocos Bark, and that a more beneficial effect can be exerted in the effectiveness when the fermentation is carried out.

랫드에서 혈액지질 감소에 관한 오메가 6와 3 지방산 비율의 생화학적 메카니즘 (Biochemical mechanism of the ratio of omega 6 to 3 fatty acid on blood lipid reduction in rats)

  • 본 연구는 차세대 랫드에서 혈액 지질 감소에 관한 식이 내 오메가 6와 3 지방산 비율의 생화학적 메카니즘을 조사하였다. 실험처리군은 오메가 6:오메가 3비율 0 (대조구), 1:1, 8:1, 19:1로 구분하였다. 혈액 중성지방, 총콜레스테롤, 저밀도지질단백질(LDL-C), aspartate aminotransferase (AST), alanine aminotransferase (ALT), glucose의 수준은 오메가 6:오메가 3 비율 1:1 그룹에서 가장 낮았다. 혈액 고밀도지질단백질(HDL-C), 인지질의 수준은 오메가 6:오메가 3 비율 1:1 그룹이 가장 높았다. HMG-CoA reductase activity는 전체 저리구 가운데 오메가 6:오메가 3 비율 1:1, 8:1 그룹에서 가장 강하게 억제되었으나 분을 통한 스테롤의 배설량은 촉진되었다. 혈액 오메가 6:오메가 3 비율은 섭취한 식이 내 오메가 6:오메가 3 비율 증가에 따른 농도 의존적으로 감소하였다. 본 연구결과는 부모 세대로부터 차세대 랫드에 이르기까지 오메가 6:오메가 3 비율이 8:1 이하로 낮아진 식이를 섭취하면 영양소 대사작용 활성화 메카니즘에 의한 혈액 중 유해지질 감소, 간 기능 유지 및 성장을 자극한다는 새로운 사실을 나타낸다.

Cysteine 첨가로 배양된 소 수정란의 발달과 동결성 효과 (Developmental and survivability according to cryopreservation of in vitro produced bovine embryos cultured by addition of Antioxident cysteine)

  • The aim of the present study was to assess the embryo development and survivability of post-thawed bovine embryos produced in vitro by addition of cysteine. The rates of metaphase II formation were not differed significantly among three groups(TCM199 73.8%, TCM199 with 0.3% cysteine 76.9%, TCM199 with 0.5% cysteine 83.8%, respectively). No difference of cleavage rate(70.6~74.6%) was seen among three culture medium(TCM199 70.6%, CR1aa 71.3%, SOF 74.6%) with 0.5M cysteine. however, Significantly(P<0.05) higher development rate into blastocyst stage by 0.5M cysteine addition was obtained in SOF medium(35.6%) than in TCM199(27.6%) or CR1aa(26.6%), however no significant differences in the cleavage rates were among three culture medium. After frozen the blastocysts cultured with 0.5M cysteine, The re-expansion rates were 61.3%~86.4% among groups, and hatching rates were 26.3%~46.9% among groups, the rates of re-expansion and hatching were significantly(P<0.05) higher in SOF medium(86.4% and 46.9%) than those in TCM199(61.3% and 26.3%) and CR1aa medium(87.1 and 44.4%). After thawing, the blastocyst re-expansion rate was significantly(P<0.05) higher in in vivo (87.1%) and in vitro (70.3%) embryos. In conclusion, our results demonstrate that supplementation of IVM and IVC medium with 0.5M cysteine improved the quality of in vitro production embryo and post- thawed embryo. Future studies comparing these media systems in well-designed trials should be performed.

배양 조건에 따른 한우 수정란의 발달과 초자화 동결 후 수정란의 생존성 비교 (Effect of Culture Condition on Hanwoo Embryonic Developments and Their Sunrival after Vitrification)

  • We investigated the cleavage rate and blastocyst yield for each culture condition to enhance tolerance of cryo-preservation of bovine IVF embryo with relatively lower cryo-tolerance compared to in vivo embryo. The cleavage rate and blastocysts yield for CR1aa, IVMD, IVD, CR1aa+10% FBS were 73.2, 69.3, 72.8, 68.5% and 44.1, 30.8, 33.3, 48.0%, respectively. The values did not differ among each treatments without serum. For embryo vitrification, In vivo and In vitro blastocysts were exposed to VS1(10% glycerin, 0.1 M glucose, 0.1 M sucrose, PEG 1%) for 5 min, and VS2 (10% glycerin, 10% EG, 0.2 M glucose, 0.2 M sucrose, PEG 2%) for 5 min and then VS3 (10% glycerin, 30% EG, 0.3 M glucose, 0.3 M sucrose, PEG 3%) for 1 min. The exposed embryos were then loaded into the 0.25 ml plastic straws and then plunged into liquid nitrogen. The straws were held for period of 1 to 2 weeks before thawing. In embryo viability, no differences in blastocyst re-expansion rates were found between in vivo and in vitro embryos. whereas expansion-BL rates was significantly higher for in vivo-derived embryos (72.7%) when compared to in vitro-derived embryos (51.4%), respectively (P<0.05). In conclusion, our results indicate that combined use of CRIaa culture medium with vitrification might enhance tolerance of cryopreservation for bovine IVF embryo production.

다배란 처리에 따른 한우 체내 수정란 생산 효율과 수정란이식 (Effect of In Vivo Embryo Production and Embryo Transfer Following Superovulation in Hanwoo)

  • In vivo embryo produced from Hanwoo donor cows were collected and transferred to Hanwoo recipients. Cows, at random stages of the estrous cycle, received Progesterone Releasing Intravaginal Device (CIDR-plus, InterAg, New Zealand) together with injection of 1 mg estradiol benzoate and 50 mg progesterone, and gonadotropin treatment began 4 day later. For superovulation, a total of 28 mg FSH was intramuscularly injected twice a day in the way of decreasing doses 4 day (5, 5, 4, 4, 3, 3, 2 and 2 mg). Twenty one Hanwoo donor cows were flushed on day 7 of estrus cycle with same FSH and artificial insemination by the same technicians. Embryos were recovered 7 days after the second insemination by flushing the uterus with Embryo Collection Medium. The results obtained were as follows: The rates of transferable embryos were 50.3%, and 78 fresh embryos at morulae and blastocysts stage were transferred into Hanwoo recipients on day 7 of estrus cycle. The pregnancy rates were first embryo transfer 55.6%, 2nd 62.9% and 3rd 57.9%, respectively. In conclusion, These results suggest that CIDR-based superovulation protocol may be effectively used for production of superior Hanwoo embryos. Also, since it seems the condition of recipient cows greatly affect pregnancy rate, it is very important to evaluate recipient for effective cattle production.

한우 체내.외 유래 소 수정란의 생존성 비교 (Comparison of Viability on In Vivo and In Vitro-derived Bovine Embryos in Korean Native Cattle (Hanwoo))

  • 포유동물 수정란의 동결보존기술은 최근 기후 변화에 따른 생물종 다양성을 보존하기 위해서 중요하게 여겨지는 연구 분야이다. 따라서 멸실 위험에 처한 동물의 개량과 증식, 보존과 복원 및 생명공학의 분야에 이르기까지 응용 기술은 다양하게 이용되어진다. 본 연구에서는 한우 수정란의 동결 후 생존성 향상을 위해서 동결 방법에 따른 체내 외수정한의 내동성을 조사하였다. 완만동결에 따른 체내 외수정란의 동결 융해 후 수정란의 재확장률은 89.6%와 81.5% 그리고 72시간 배양하였을 때 부화된 수정란은 76.9%와 43.4%, 생존수정란의 총세포수는 136${\pm}$3.6개와 107${\pm}$3.8개의 결과를 보여 동결 융해 후 생존율에서는 차이를 보이지 않았으나, 수정란의 부화율과 세포수 조사에서는 체내수정란이 유의적으로(p<0.05) 높은 결과를 보였다. 체내수정란의 완만동결 후 배반포 회복률은 91.3% 그리고 12시간 이상 배양하였을 때 확장배반포까지의 발달률은 71.4%의 결과를 보였으며, 초자화 동경에서는 85.7%와 75.0%의 결과를 보였다. 결과적으로 본 연구에서 수행된 체내 외 수정란의 동결기술을 이용한다면 수정란이식 현장에서도 효과적인 활용이 가능할 것으로 사료된다.

국산원료(國産原料)를 활용한 복합분(複合粉) 및 제품개발(製品開發)에 관한 연구(硏究) 제4보 보리 및 고구마복합분(複合粉)을 이용한 면류(麵類)의 Texture 특성(特性) (Development of Composite Flours and Their Products Utilizing Domestic Raw Materials IV. Textural Characteristics of Noodles made of Composite Flours Based on Barley and Sweet Potato)

  • 밀가루 대체원료(代替原料)로써 쌀보리가루 또는 고구마가루를 활용한 복합분(複合粉)으로 국수를 만들고 그들의 품질(品質)을 조사하여 다음과 같은 결과를 얻었다. 1) 면대(麵帶) 및 건면(乾麵)생산에 있어서 밀가루에 대한 보리가루 대체율(代替率)은 100%까지, 고구마가루 대체율(代替率)은 70%까지 가능하였으며 품질개량제(品質改良劑)로서 glyceryl monostearate 및 sodium polycrylate의 첨가는 이들 제품의 texture 특성(特性)을 향상시켰다. 2) 면대(麵帶)와 건면(乾麵)의 texture 특성치(特性値)인 견고성(堅固性), 응집성(凝集性), gumminess는 보리가루나 고구마가루의 배합에 의하여 떨어졌으나 첨가제의 효과가 뚜렷하였고 면대(麵帶)는 견고성(堅固性) 6.2 이상, gumminess 430 이상, 건면(乾麵)은 견고성(堅固性) 6.8 이상이 필요하였다. 3) 조리면(調理麵)에 있어서 밀가루 대체(代替)와 첨가제의 사용은 texture 특성(特性)을 떨어뜨리는 경향이 있었으며 조리특성(調理特性)으로 보아 보리가루는 60%까지, 고구마가루는 40%까지 대체(代替)할 수 있었다. 4) 조리면(調理麵)의 색깔, 맛, texture에 관한 관능(官能)시험에서 20% 대체복합분(代替複合粉)은 밀가루와 5% 수준에서 유의차(有意差)가 없었으며 품질저하(品質低下)는 주로 착색(着色)에 기인(起因)하였다.

인공 온천수가 퇴행성 슬관절염에 미치는 효과 (The Effects of Artificial Balneotherapy on Osteoarthritis)

  • The purpose of this study was to evaluate effects of artificial balenotherapy on the with degenerative osteoarthritis were selected as the subject do this study among the patients with degenerative osteoarthritis having pains in their knees by the criteria of America Rheumatoid Association. And the randomly selected ten patients out of the twenty were classified as an artificial balenotherapy group and the other the Patients were in a control group. Ten individuals with degenerative osteoarthritis patients in the artificial balenotherapy group Participated in artificial balenotherapy Program four weeks from March 1st, 2003 to March 31st, 2003 in Daegu 00 hospital. Ten individuals with degenerative osteoarthritis in the control group did not receive Artificial Balenotherapy. The conclusions were as follows: 1. After 4 weeks of therapy, the score of WBC(p < 0.01), RBC(p < 0.01), HTC(p < 0.01) in the artificial blalenotherapy group were significantly decreased, compared with their scores in pre-intervention. 2.In the balenotherapy group, significantly increase $Na^{+}$ ion(P < 0.05), decrease K ion(p < 0.01), unchanged Cl ion scores were found, compared with their scores in pro-intervention. 3. The result of the substudy about the effects of artificial balenotherapy on the substances that have relations with the function of liver were following that artificial balenotherapy group, increased total bilirubin(P < 0.01), unchanged total Protein, Albumin, total cholesterol, GOT, GPT, ALP were found, compared with their score in pre-intervention. 4. The result of the substudy about the effects of artificial balenotherapy on the substances that have relations with the function of kidney were following that in the artificial balenotherapy group, reduced BUN(P < 0.05), and increased creatine were found. 5. The result of the substudy about effects of artificial balenotherapy on the knee pain visual analog score, knee's functional score, ADL score were following that in the artificial balenotherapy group, decreased VAS score(P < 0.05), increased ADL score(p < 0.01), unchanged knee's functional score were founf, compared with their scores of the pre-intervention. In conclusion, the result of this study suggest that artificial balenotherapy improved ADL score and reduced knee pain visual analog score of the patients with degenerative osteoarthritis. The result proposed that appropriate use of artificial balenotherapy improves the degenerative osteoarthritis

객체지향 분석-함성 부호화를 위한 효율적 움직임 파라미터 추정 알고리듬 (Efficient Algorithms for Motion Parameter Estimation in Object-Oriented Analysis-Synthesis Coding)

  • 객체지향 분석-합성 부호화는 일련의 영상들을 여러 개의 동 객체로 분할한 후 각 객체의 움직임을 추정하고 보상한다. 그것은 각 객체에 있는 움직임 정보를 추정하기 위해 변환 파라미터 기법을 적용하는데 이때 변환 파라미터 기법은 그레디언트 연산자를 사용하기 때문에 매우 복잡한 계산이 요구된다. 본 논문의 목적은 객체지향 분석-합성 부호화에서 계층적 구조를 사용한 효율적인 변환파라미터 기법을 개발하는 것이다. 이러한 목표를 달성하기 위해 본 논문은 계층적 구조를 사용한 하이브리드 변환파라미터 추정 방법과 적응형 변환 파라미터 방법의 두 가지 알고리듬을 제안한다. 전자는 파라미터 검증 방법을 사용하는데 원 영상을 1/4로 축소한 저해상도 영상에서 파라미터 검증 처리 방법에 의해 6-파라미터 또는 8-파라미터로 추정한다. 후자는 동일한 계층적 방법을 적용한 다음 변환 파라미터를 적응적으로 추정하기 위해 temporal co-occurrence 행렬에 기반 한 움직임 량을 측정하는 움직임 판단기준을 사용한다. 이러한 방법은 고속이며, 병렬처리 기법을 사용할 경우 쉽게 하드웨어로 구현할 수 있는 이점이 있다. 이론 분석 및 모의시험 결과 제안한 방법이 기존 방법에 비해 약 1/4 정도로 월등한 계산량 감축을 얻을 수 있었으며, 아울러 제안한 방법들에 의해 복원된 신호대 잡음비는 6-파라미터와 8-파라미터 추정 방법에 의해 복원된 결과들 사이에 있음을 보여 준다.
