The allelopathic effects of volatile substances from Chamaecyparis obtusa (S. et Z.) Endl. were examined on the germination and seedling growth of some plant species, and on the population growth of some microorganisms. The germination and seedling growth of the receptor plants were suppressed more severely by leaf and fruit essential oils than by those of other parts. Colonial growth of fungi was severely inhibited by essential oils extracted from leaves and fruits. The development of root hairs of the receptor plants was also severely inhibited by the essential oils. The cortical cells at the root tips of Lactuca sativa L. treated with essential oils showed contraction of the cytoplasm, resulting in plasma membranes becoming detached from the cell walls and the cells metamorphosing irregularly. Accumulation of lipid granules inside the contracted cytoplasm and degeneration of mitochondrial cristae were also observed.
Objectives : The present study was designed to investigate effects of mixed extracts (9:1, v/v) of Hwangryunhaedoktang, Chamaecyparis obtusa essential oil. We evaluated the antimicrobial and antioxidant activity by manufacturing mixed extracts as the materials for functional medicinal herb cosmetics. Methods : We performed antimicrobial were tested microbes (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Propionibacterium acnes) by disc diffusion method and measure clear zone. Antioxidant activities were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. Results : Antimicrobial activities of mixed extracts against S. epidermideis, P. acnes, S. aureus, E. coli, and Candida. A were $10.9{\pm}3.0mm$, $16.9{\pm}2.0mm$, $9.2{\pm}2.0mm$, $10.3{\pm}1.0mm$, $11.8{\pm}3.0mm$ respectively. The Chamaecyparis obtusa essential oil had the highest antimicrobial activities against S. epidermideis, P. acnes, S. aureus, E. coli, and Candia. A and clear zone of microbes ware $16.7{\pm}3.0mm$, $28.8{\pm}0.2mm$, $15.9{\pm}2.0mm$, $11.5{\pm}1.0mm$, $16.3{\pm}3.0mm$. Hwangryunhaedoktang extract showed antimicrobial activity but only P. acnes and S. aureus The antioxidant activities of the mixed extracts were tested through the evaluation of DPPH radical scavenging activity. The 100 % mixed extracts were found to have 90 % DPPH radical scavenging activity. The mixed extracts was presented similar antioxidant activities compared with that of ascorbic acid. As a result, A mixture extract is expected to have antimicrobial effects and free radical scavenging activity was found. Conclusions : Accordingly, It can be concluded that mixed extracts has the potential to cosmetic material.
The essential oils were extracted from the needles of several softwood tree species, Pinus densiflora, Pinus koraiensis, Chamaecyparis obtusa and Chamaecyparis pisifera to investigate their biological activities. The antifungal activity by hyphal growth inhibition method was determined by three wood rot fungi, one mouldous fungi, and three tree pathogenic fungi and the antibacterial activity by paper disc method and turbidiometric method by three gram positive bacteria and one gram negative bacteria, Escherichia coli, were also tested. Antifungal activity was the best in the essential oil from C. obtusa, and the following was resulted in order of P. densiflora and P. koraiensis. The essential oil from P. densiflora showed higher antifungal activity against wood rot fungi than that from P. koraiensis while the result was contrary against tree pathogenic fungi. However, the antibacterial activity was the best in the essential oil from P. densiflora, and the following was in order of C. obtusa and P. koraiensis. From these results, the variation of biological activity seemed to be due to the chemical composition of essential oils.

The present study was conducted to evaluate the application of Chamaecyparis obtusa and to investigate potential utilization of essential oil from C. obtusa as plant-based medicine. The antifungal activity of essential oil from leaves and twigs of C. obtusa (Sieb. Et Zucc) was determined and the major components of active fractions against Microsporum canis (KCTC6591) and Trichophyton mentagrophytes (KCTC6077) were identified by GC/MS analysis. In treatment of essential oil from C. obtusa, the strain M. canis was more resistant than the other, T. mentagrophytes. In the agar diffusion assay, essential oil from C. obtusa inhibited hyphal growth of M. canis and T. mentagrophytes at the concentration of more than 5,000 ppm. The zones named B and C in the TLC assay of essential oil from C. obtusa showed antifungal activities. Among four sub-fractions of n-hexane extract from B and C zones, named as B-1, B-2, C-1 and C-2, the C-2 showed the highest antifungal activity. Instrumental GC/MS analysis for sub-fractions showed that a major component of C-1 was ${\alpha}$-terpineol as terpene alcohol, while C-2 contained sesquiterpenes such as elemol, cedrol and eudesmol.

Chamaecyparis obtusa (C. obtusa) belongs to Cupressaceae and grows wild in southern area of Korea. C. obtusa has been used for furnitures because of the superior quality of the lumber and remaining branches and leaves were used for extraction of essential oil (C. obtusa oil). C. obtusa oil has numerous health effects, including anti-inflammation, antibacterial, deodorization, sedative effect. This study was performed to evaluate the effects of C. obtusa oil on anti-wrinkles, skin-barrier and moisturizing. First, DPPH and ABTS assay were conducted to evaluate antioxidant effect and C. obtusa oil showed significance scavenging activity on both radicals. And elastase activity was examined for anti-wrinkle effect. At the result, elastase activity was significantly reduced by C. obtusa oil. Next, real-time PCR was performed to evaluate gene expression. At the results, MMP-1 mRNA expression was significantly reduced by C. obtusa oil in HDF cells. In addition, filaggrin and HAS-2 mRNA expression significantly increased by C. obtusa oil in HaCaT cells. Taken together, these results indicate that C. obtusa oil has potential as a cosmetic material for anti-aging by enhancing anti-wrinkle, skin-barrier and moisturizing effects.

The essential oil from Chamaecyparis obtusa was investigated for biological activities in anti-oxidative, anti-inflammation and antibacterial method, respectively. The Growth inhibitory effect of C. obtusa oil on the bacteria was evaluated with MIC (minimum inhibitory concentration), $IC_{50}\;(50\%$ inhibitory concentration), and paper disc method. Two kinds of gram positive strains and two kinds of gram negative strains were used in this study. Gram positive strains were B. subtilis and S. aureus. and Gram negative strains were E. coli and P. aeruginosa. Gram positive strains showed much more intensive effect than gram negative strains. Anti-oxidative effect was investigated with DPPH (1,1-diphenyl-2-picrylhidrazyl) in methanol based and $IC_{50}\;was\;0.78\%.$ Our results suggest that the essential oil from Chamaecyparis obtusa has effects on anti-bacterial, anti- oxidative and anti-inflammation in in vitro and in uiuo. Then this material could be expect synergic effect with other candidated extracts and oils.

This study was carried out to investigate the potential promise of Chamaecyparis obtusa oil as a natural antioxidant. C. obtusa oil and its fractions were subjected to screening for their antioxidant activities by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and ammonium thiocyanate method. In the first case, $IC_{50}$ value of the C. obtusa oil was determined as $40{\mu}{\ell}/m{\ell}$. At $0.5{\mu}{\ell}/m{\ell}$ concentration level, free radical scavenging effect of fraction G determined as 66.94% was the highest among the fractions of C. obtusa oil. In the ammonium thiocyanate method, essential oil of C. obtusa and its fraction C, D, and E showed activities of 72.0%, 71.2%, 71.9% and 71.1%, respectively. Fraction G, most active fraction, was mainly consisted of $\alpha$-terpineol, elemol, widdrol and $\alpha$-cadinol.
The compositions of essential oils isolated from the leaves and fruits of Chamaecyparis obtusa (Sieb. et Zucc). Endl. and Chamaecyparis pisifera (Sieb. et Zucc.) Endl. were analyzed through GC and GC-MS. The oil yields were 0.83% (as fresh weight) and 1.36% in the leaves and the fruits of C. obtusa, and were 0.92% and 1.28% in those of C. pisifera, respectively. More than 90 components were identified, including high contents of monoterpenoids and sesquiterpenoids. Contents of monoteipenoids in the leaf and fruit oils of C. pisifera were higher than in those of C. obutsa. The major constituents in the leaf oil of C. obtusa were sabinene (11.81% as determined through GC peak area), limonene (7.73%), bornyl acetate (6.92%), $borneol+{\alpha}-teirineol$ (15.67%), and elemol (12.82%), and those in the fruit oil were myrcene (8.12%), ${\gamma}-terpinene$(5.91%), p-cymene(7.62%), $borneol+{\alpha}-terpineol$(6.53%) and ${\beta}-caryophyllene$ (23.74%). The major constituents in the leaf oil of C. pisifera were ${\alpha}-pinene$(32.34%), ${\delta}-3-carene$(25.28%), myrcene(11.72%), and bornyl acetate (8.77%), and those in the fruit oil were ${\alpha}-pinene$ (29.38%), ${\delta}-3-carene$(30.27%), myrcene(15.05%), and limonene(8.10%).
Antimicrobial activity of the essential oil extracted from plants Artemisia princeps var. orientalis, Thuja orientalis and Chamaecyparis obtusa were tested against selected pathogenic bacteria and fungi of fish. At the concentrations above 500ppm, ingibitory effect of the oil of A. princeps var. orientalis was at its peak against Aeromonas hydrophila ATCC 14715, A. hydrophila CF-2, A. salmonicida ATCC 14174 and A. salmonicida EL-1 but the bacteria Edwardsiella tarda KBF-1, Vibrio anguillarum ATCC19264, V. ordalii ATCC33509 and Streptococcus sp. were insensitive. Likewise, the oil extract of T. orientalis showed the highest inhibitory activity against V. ordalii ATCC33509, E. tarda ECK-1, and E. tarda KBF-1 at 300ppm; however the activity was highest at 500ppm or A. hydrophila ATCC14715, A. hydrophila CF-2, A. salomonicida ATCC14174, A. salmonicida EL-1 and Streptococcus sp. SF-1. With increasing dose of C. obtusa oil, the inhibitory activity became more and more effective against A. hydrophila CF-2, A. salomonicida ATCC14174, E. tarda ECK-1 and Streptococcus sp. SF-1, but A. hydrophila ATCC14174, A. salmonicida EL-1, E. tarda KBF-1, V. anguillarum ATCC19264, V. ardalii ATCC33509 and gram positive bacteria (Streptococcus sp.) were somewhat resistant. A. princeps var. orientalis, T. orientalis and C. obtusa were also tested against Saprolegnia sp. at the oil concentrations of 10, 100, 500, 1,000, 1,500 and 2,000ppm. The inhibitory effect of the oil on the inhibit the mycelial growth of Saprolegnia sp. at 10ppm and completely inhibited at over 500ppm.
