Studies on the Identification of the Anthocyanins in Plum (자두 Anthocyanin의 구조확인(構造確認)에 관(關)한 연구(硏究))

  • The anthocyanins in Santa rosa plum (widely growing var. in Korea) were extracted with 0.5% methanolic HCl and purified with Amberlite IRC-50 cation exchange resin. The individual pigments were separated by paper chromatography. The two pigments were identified as cyanidin-3-monoglucoside and cyanidin-3-xyloglucoside based on the following evidence: Rf values in various solvents, partical acid hydrolysis, stepwise hydrolysis, sugar moiety, alkaline degradation, and absorption spectra in the visible region. Cyanicin-3-monoglucoside was the abundant pigment of the plum.

Protein Adsorption on Ion Exchange Resin: Estimation of Equilibrium Isotherm Parameters from Batch Kinetic Data

  • The simple Langmuir isotherm is frequently employed to describe the equilibrium behavior of protein adsorption on a wide variety of adsorbents. The two adjustable parameters of the Langmuir isotherm - the saturation capacity, or $q_m$, and the dissociation constant, $K_d$ - are usually estimated by fitting the isotherm equation to the equilibrium data acquired from batch equilibration experiments. In this study, we have evaluated the possibility of estimating $q_m$ and $K_d$ for the adsorption of bovine serum albumin to a cation exchanger using batch kinetic data. A rate model predicated on the kinetic form of the Langmuir isotherm, with three adjustable parameters ($q_m,\;K_d$, and a rate constant), was fitted to a single kinetic profile. The value of $q_m$ determined as the result of this approach was quantitatively consistent with the $q_m$ value derived from the traditional batch equilibrium data. However, the $K_d$ value could not be retrieved from the kinetic profile, as the model fit proved insensitive to this parameter. Sensitivity analysis provided significant insight into the identifiability of the three model parameters.

  • Formation of the complexes of barium, strontium and calcium ions with dibasic organic acid ions in dilute solution was studied at room temperature, utilizing the equilibrium exchange technique which involved the uses of radioactive alkaline earth metal ions and cation excbange resin. The organic acids used in this study were succinic and tartaric acids, and the solvents used were water, 20 % acetone-water and 20 % ethanol-water. The pH of the solutions was controlled to 7.2∼7.4, and the ionic strength of the solutions was kept at approximately 0.1. The experimental results indicated that the alkaline earths formed one-to-one complexes in solution with the dibasic acids examined, and that the relative stabilities of the complexes increased in the order: $Ba^{++}; succinic

A New Streptothricin Family Antibiotic Producing Streptomyces Spp. Snus 8810-111 ; Characterization of The Producing Organisms, Fermentation, Isolation, and Structure Elucidation of Antibioitics

  • A new streptothricin family antibiotic producing Streptomyces spp. SNUS 8810-111 was isolated from a soil sample. Study of its morphological and physiological characters indicated that the antibiotic producing organism was a Streptomyces spp. Taxonomical studies suggested that the organism might belong to the genus streptomyces gougeroti. The organism produced antibiotics most in calcium carbonate-tryptic soy broth. The active principles were recovered from the broth with a cation exchange resin and eluted from the resin with HCI. Cellulose column chromatography gave two active principles.$^1H-^1H$ Homo-COSY study on the first compound revealed four structural components. Total hydrolysis of the antibiotic with HCI allowed isolation of $\beta-lysine$. From these data the antibiotic was found to be streptothricin D. The other compound showed one additional signal in the .$^1H$NMR and the $^{13}C$ NMR spectra. The signal was from a methyl group attached to a nitrogen atom. Comparison of the NMR signals with those of streptothricin D suggested that the compound was N-methyl-streptothricin D which was a new compound in the family of streptothricin antibiotics.

An Improved Method for the Determination of Scandium by Neutron Activation Analysis (스칸듐定量을 위한 改良된 放射化分析法)

  • A rapid and simple method is described here for the determination of scandium in monazite by neutron activation analysis. The sample is irradiated for 20 hours at the neutron flux of $10^{12}$ thermal neutrons/$cm^2$/sec in the TRIGA MARK Ⅱ reactor, after which the sample is decomposed by fusion with concentrated sulfuric acid. The scandium-46 together with scandium carrier are separated from the irradiated sample by precipitating with ammonia, and are extracted by solvent extraction of the thiocyanate complex into ether. The induced radioactivity is measured by gamma scintillation spectrometry using the Multichannel Pulse Height Analyzer connected with 2"${\times}$2" NaI(Tl). The chemical yield is determined gravimetrically by precipitating scandium with mandelic acid. In order to check the efficiency of scandium separation and the errors from interfering activities of the other elements, scandium was separated by the cation exchange resin column, and the results from both samples were compared each other, which showed that the chemical procedure used in this work was as selective as the ion-exchange method with respect to scandium separation. The scandium contents in Korean monazite were found to be about 12 p. p. m.

  • U(IV) ion, the valance state of uranium presumed at in a deep-depth disposal of a spent fuel, was prepared and separated from U(VI) ion. In order to prepare U(IV) ion, tests were performed by adding several reducing agents into a uranyl solution or by dissolution of uranium oxide in a mixed acid added with a reducing agent. The valance states of the uranium in the prepared solutions were identified by separating two ions with a Dowex AG 50W-X8 cation exchange resins and measuring the solutions using a laser-induced fluorescence spectroscopy. However, U(IV) and U(VI) were not separated by a Lichroprep Si60 exchange resin in the same separation condition of Pu(IV) and Pu(VI).

  • Copolymerization of the 4-vinylpyridine with vinylacetate and divinylbenzene initiated by azobis-isobutyronitrile was carried out in DMF in presence $BaCl_2$ at $98^{\circ}C$. Ion exchange res in, poly 4-vinylpyridine-vinylsulfonic acid-divinylbenzene was prepared by sulfonation of 4-vinylpyridine-vinylacetatp-divinylbenzene with concentrated sulfuric acid. The compositions of each synthetic resin were identified by means of ir adsorption spectroscopy. Anion and cation capacities of 4-vinylpyridine-vinylsulfonic acid-divinylbenzene ion exchanger were 2.5meq/g and 4.8meq/g, respectively. Adsorption of Cd(II) and Cu(II) ions have showed larger quantity in alkalie media. A study also was made of the influence of alcohol on the distribution coefficient of Cd(II) and Cu(II) ions between the synthetic ion exchanger, and solution containing hydrochloric acid, various alcohols and water.   • The quantitative separations of a mixture containing equal amounts of each cation such as Mn(Ⅱ), Cr(Ⅲ), V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ), Co(Ⅱ), and Fe(Ⅲ) are carried out by the elution through $35cm{\times}3.14cm^2$ column of cation exchange resin, $Dowex 50w{\times}12$. The eluents are a mixture of 0.6 M sodium chloride and 0.1 M sodium tartrate (pH = 2.00 and 4.50) for Fe(Ⅲ), V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ) and Co(Ⅱ), and a mixture of 3 M sodium chloride and 0.1 M sodium tartrate (pH = 4.50) or a mixture of 0.7 M sodium chloride and 0.5 M sodium oxalate (pH = 4.50 and 5.00) for Mn(Ⅱ) and Cr(Ⅲ). The subsidiary cations in a standard iron mixture such as V(Ⅴ), Cu(Ⅱ), Ni(Ⅱ), Mn(Ⅱ) and Cr(Ⅲ) are separated together from the large amount of Fe(Ⅲ) through $15cm{\times}3.14cm^2$ column of the resin, $Dowex 1{\times}8$, by elution with the eluent of 4.0 M hydrochloric acid. A small amount of Fe(Ⅲ), however, is eluted together with Cu(Ⅱ). V(Ⅴ), Ni(Ⅱ), Mn(Ⅱ) and Cr(Ⅲ) eluted together are separated quantitatively through $10cm{\times}3.14cm^2$ column of the resin,$Dowex 50w{\times}12$. Cu (Ⅱ) and a small amount of Fe(Ⅲ) are separated quantitatively through $10cm{\times}3.14cm^2$ column of the resin, $Dowex 50w{\times}12$, by the elution with a mixture of 0.6 M sodium chloride and 0.1 M sodium tartrate (pH = 2.00 and 4.50) as an eluent. By the conditions obtained in the separations of the standard iron mixture, Fe(Ⅲ) and all of the subsidiary cations in steel are quantitatively separated.

The Investigation of a Novel Indicator System for Trace Determination and Speciation of Selenium in Natural Water Samples by Kinetic Spectrophotometric Detection

  • A novel catalytic kinetic method is proposed for the determination of Se(IV), Se(VI) and total inorganic selenium in water based on the catalytic effect of Se(IV) on the reduction of bromate by p-nitrophenylhydrazine at pH 3.0. The generated bromine, $Br_2$ or $Cl_2$ plus $Br_2$ in 0.1 M NaCl (or NaBr) environment efficiently decolorized Calmagite and the reaction was monitored spectrophotometrically at 523 nm as a function of time. In this indicator reaction, bromide acted as an activator for the catalysis of selenium (IV) and a reducing agent for selenium (VI) at pH 3.0, which allowed the determination of total selenium. The fixed time method was adopted for the determination and speciation of inorganic selenium. Under the optimum conditions, the calibration graph are linear in the range 1 - 35 ${\mu}gL^{-1}$ of Se(IV) for the fixed time method at $25^{\circ}C$. The detection limit based on statistical $3S_{blank}$/m-criterion was 0.215 ${\mu}gL^{-1}$ for the fixed time method (7 min). All of the variables that affect the sensitivity at 523 nm were investigated, and the optimum conditions were established. The interference effect of various cations and anions on the Se (IV) determination was also studied. The selectivity of the selenium determination was greatly improved with the use of the strongly cation exchange resin such as Amberlite IR120 plus. The proposed kinetic method was validated statistically and through recovery studies in natural water samples. The RSDs for ten replicate measurements of 5, 15 and 25 ${\mu}gL^{-1}$ of Se(IV) and Se(VI) was changed between 2.1 - 4.85%. Analyses of a certified standard reference material (NIST SRM 1643e) for selenium using the fixed-time method showed that the proposed kinetic method has good accuracy. Se(IV), Se(VI) and total inorganic selenium in environmental water samples have been successfully determined by this method after selective reduction of Se(VI) to Se(IV).

  • Ethylenediamine-triacetatocobalt (III) complexes with an ammine, an ethylene-diamine, and a trimethylenediamine as the unidentate ligand were prepared, and were isolated as only one isomer for each case by the Dowex 50W-X8, cation exchange resin in $H^+$ form. The geometrical isomer of these complexes have been assigned cis-equatorial form in the three possible geometrical isomers from the elemental analysis, pH titration, IR, NMR, and electronic absorption spectrum. It was found that $[CoN_3O_3]$ system of the meridional form with multidentate ligand have the first absorption band of the largely splitting pattern, and that the diamines (ethylenediamine, trimethylenediamine) have coordinated to the central cobalt (III) ion as a unidentate ligand.
