Studies on the Components of Pholiota squarrosa(II) (비늘버섯의 성분(成分)에 관한 연구(硏究)(제 2 보))

  • To investigate antitumor components in Korean higher fungi, the carpophores of Pholiota squarrosa belonging to the family Strophariaceae were collected and extracted with hot water. A protein-bound polysaccharide fraction was obtained by adding ethanol to the extract and by dialyzing through Visking tube. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. It showed an inhibition ratio of 78.7% at the dose of 20mg/kg/day. The tumor in two of the ten mice was completely regressed. The chemical analysis of the antitumor fraction by Anthrone and Lowry-Folin methods showed that it consisted of 42% polysaccharide and 55% protein. The enzyme fraction of the carpophores showed no proteolytic activity on casein.

  • The dried carpophores of Sarcodon aspratus were examined for mineral elements and a protein-polysacccharide. Calcium, iron, zinc, magnesium, manganese, copper, lead, cadmium and mercury were detected in that order of contents by atomic absorption spectrometry and automatic mercury analysis. The protein-polysaccharide fraction extracted from the carpophore was found to contain 31.5% protein and 52.8% polysaccharide. The fraction showed no antitumor activity against sarcoma 180 implanted in mice.

Antimicrobial Activity of Elfvingia applanata extract alone and in Combination with Some Antibiotics (Elfvingia applanata 엑스가 수종 항생물질의 항균력에 미치는 병용효과)

  • As part of our search for less toxic antimicrobial agents from natural resources, the carpophores of Elfvingia applanata$(P_{ers}.)K_{ARST}.$ was extracted with hot water. EA, the aqueous extract from the carpophores of E. applanata, was lyophilized and a dark brownish powder was obtained. Antimicrobial activity of EA was tested in vitro against Gram positive and Gram negative bacteria by serial broth dilution method, and the antimicrobial activity was expressed by minimal inhibitory concentration(MIC). Among fourteen species of bacteria tested, the antimicrobial activity of EA was the most potent against Proteus vulgaris showing MIC of 1.250 mg/ml. To investigate the effect of antimicrobial combinations of EA with four kinds of antibiotics(ampicillin, cefazolin, oxytetracycline and chloramphenicol), the fractional inhibitory concentration index(FICI) was determined by checkerboard assay for each strain. The antimicrobial combinations of EA with four kinds of antibiotics resulted in synergism in four instances, but no antagonism was observed. Four instances of synergism were observed when EA was combined with ampicillin against Micrococcus luteus, with cefazolin against Bacillus subtilis, with cefazolin against Micrococcus luteus and with oxytetracycline against Staphylococcus aureus.

Immunoactivities of the Protein-Polysaccharides of the Tips of the Growing Carpophores of Ganoderma lucidum (영지버섯(Ganoderma lucidum) 생장점으로부터 분리한 단백다당류의 면역활성)

  • Two protein-polysaccharide fractions, GLA and GLB, respectively, were prepared from the pileus of the fully grown carpophores and the tips of the growing carpophores of Ganod erma lucidum. At a dose of 100mg/kg/day ip, GLA and GLB inhibited the growth of sarcoma 180 solid tumor in ICR mice by 56.3% and 81.8%, respectively. In a flow cytometric (FCM) analysis, GLA and GLB enhanced the formation of lymphoblasts of BALB/c, splenic leukocytes at a concentration of 100 ${\mu}$g/ml, by 38.3% and 61, 3%, respectively. When ip injected into ICR mice, GLB exerted anti-leukopenic effect against cyclophospamide (75mg/kg, ip) in that the leukocyte counts of the peripheral blood of the normal and the cyclophosphamide-treated mice. respectively. was (11.1 ${\pm}$ 3.8) ${\times}$ 10$^3$ and (4.0 ${\pm}$ 1.8) ${\times}$ 10$^3$, while the GLB-cyclophosphamide treated mice showed a leukocyte count of (10.8 ${\pm}$ 5.1) ${\times}$ 10$^3$ CELLS/${\mu}$l. These results suggest that GLB is a promising candidate for an effective, cancer immunotherapeutic agent.

  • The carpophores of Phallus impudicus collected at Gal-mae-ri in Gyeong-gi Province were subjected to chemical analysis for sterol constituents. The dried carpophores were homogenized and extracted with chloroform-methanol(2 : 1). After saponification of the dried extract, unsaponified components were obtained by ether extraction of the reaction mixture. A sterol fraction was separated by preparative TLC and from this fraction, ergosterol was identified by gas­chromatographic analysis with authentic sterols.

  • Antimicrobial activity of GL (the aqueous extract from the carpophores of Ganoderma lucidum ($F_RK_{KARST}$) was tested in vitro aginst Gram positive and Gram negative bacteria by serial broth dilution method, and the antimicrobial activity was expressed by minimal inhibitory concentration (MIC). Among fifteeen species of bacteria tested, the natimicrobial activity of GL was of antimicrobial combinations of GL with four kinds of antibiotics (ampicilin, cefazolin, oxytet-racycline and chloramphenicol), the fractional inhibitory concentraction index (FICI) was determined by checkerboard assy for each stain. The antimicrobial combinations of GL with four antibiotics resulted in additive effect in most instances, synergism in two instances, and antagonism in two instances. Synergism was obversed when GL was combined with cefazolin against Bacillus subtilis and Klebsiella oxytoca.

  • To find antitumor components, a protein-bound polysaccharide fraction was obtained by extracting with hot water and precipitating with ethanol from the carpophores and the cultured mycelia of Laccaria laccata. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. The component extracted from the carpophores showed 75% and 65% tumor inhibition ratios in the doses of 20 mg and 50 mg/kg/day, respectively. The protein bound polysaccharide fraction of the cultured mycelia of L. laccata also showed 57.8% tumor inhibition ratio in the dose of 20 mg/kg/day. The chemical analysis of the protein-bound polysaccharide fraction showed that it contained a polysaccharide and a protein. The hydrolysates of the polysaccharide moiety contained six and one unknown monosaccharides. Fourteen amino acids were identified in the hydrolysates of the protein moiety.

  • To find toxic components in Korean higher fungi, the carpophores of Naematoloma fasciculare which had caused several cases of lethal intoxication were examined for toxicity. The components of high molecular weight were separated by ethanol precipitation and dialysis from the aqueous extract of the carpophores. After the components were freeze-dried, a brown powder was obtained. When a dose of 60mg/kg of this macromolecular fraction was intraperitoneally injected into mice, the mice began to die in six days and a half of them died within seven days. This toxic component was named nematoxin after the genus name of the mushroom.

  • To investigate the acute toxicity of Elfvingia applanata$(P_{ers})K_{ARST}$, it was extracted with hot water. EA, the aqueous extract from the carpophores of E. applanata, was lyophilized and a dark brownish powder was obtained. The acute toxicity of EA was investigated in ICR mice of both sexes. Five groups of mice were administered orally and intraperitoneally with 375, 750, 1,500, 3,000, 6,000 mg/kg(p.o.) and 312.5, 625, 1,250, 2,500, 5,000 mg/kg (i.p.) of EA Abnormal clinical signs related to EA were not observed and no deaths occurred. $LD_{50}$ values administered orally and intraperitoneally were evaluated to be over 6,000 mg/kg and over 5,000 mg/kg, respectively. Gross finding of necropsy revealed no evidence of specific toxicity related to EA. There were no significant changes in body weights, serum biochemical values and histopathological changes compared with control group.

  • To investigate constituents of Strobilomyces floccopus (Fr.) Karst. and Coprinus comatus (Fr.) S. F. Gray, free and total amino acids of the two mushrooms were quantitatively analyzed by G. L. C. and an amino acid analyzer. Free amino acids were extracted from both mushrooms with ethanol. Fourtenn free amino acids were detected from the ethanol extract of S. floccopus and fifteen free amino acids from C. comatus by G. L. C. And the dry carphopores of both mushrooms were hydrolyzed with hydrochloric acid and then the total protein amino acids were analyzed by A. A. A. Seventeen total amino acids were detected from each acid-hydrolysate of S. floccopus and C. comatus. Lipids were extracted from the carpophores of S. floccopus and saponified with alcoholic potassium hydroxide. The isolated sterols were subjected to G. L. C. and two sterols were detected. The isolated free fatty acids were methylated with diazomethane and subjected to column chromatography and G. L. C. Eleven saturated and nine unsaturated free fatty acids were detected from the carpophores of S. floccopus. The presence of these nutrient components shows that the two mushrooms can be utilized as edible ones.
