Deregulation of Aspartokinase by Single Nucleotide Exchange Leads to Global Flux Rearrangement in the Central Metabolism of Corynebacterium glutamicum

  • The wild-type Corynebacterium glutamicum ATIC 13032 and Corynebacterium glutamicum ATTC 13032 lysC S301Y, exhibiting a deregulated aspartokinase, were compared concerning growth, lysine production, and intracellular carbon fluxes. Both strains differ by only one single nucleotide over the whole genome. In comparison to the wild-type, the mutant showed significant production of lysine with a molar yield of 0.087 mol (mol glucose$^{-1}$) whereas the biomass yield was reduced. The deregulation of aspartokinase further led to a global rearrangement of carbon flux throughout the whole central metabolism. This involved an increased flux through the pentose phosphate pathway (PPP) and an increased flux through anaplerosis. Because of this, the mutant revealed an enhanced supply of NADPH and oxaloacetate required for lysine biosynthesis. Additionally, the lumped flux through phosphoenolpyruvate carboxykinase and malic enzyme, withdrawing oxaloacetate back to the glycolysis and therefore detrimental for lysine production, was increased. The reason for this might be a contribution of malic enzyme to NADPH supply in the mutant in the mutant. The observed complex changes are remarkable, because they are due to the minimum genetic modification possible, the exchange of only one single nucleotide.

High Temperature Reaction Behaviors of Oxide Materials with Carbon for Refractory Application (내화물 응용을 위한 산화물 재료들과 탄소와의 고온 반응거동)

  • High temperature reaction behaviors of various oxide materials (such as bauxite, pyrophyllite, mullite and fused silica powders) used in the refractory materials for tap-hole plugging of blast furnace were investigated with varying temperature in the carbon surrounding. Kinetics of carbothermal reduction of $SiO_2$ for forming SiC with high corrosion resistance were strongly dependent on it's crystalline phase. SiC generation yield increased with increasing catalyst amount in oxide regardless of generated SiO gas amount at temperature of $<1500^{\circ}C$. However, in case of fused silica over $1500^{\circ}C$, SiC generation yield was dominantly influenced by SiO amount without catalyst effect. Bauxite showed the most effective carbothermal reduction reaction, since bauxite have a large amount of catalyst and well-dispersed $SiO_2$ phase in oxide matrix.

Effects of Heat Treatment on the Micro-structures and the Mechanical Properties of 0.002% Boron-added Low Carbon Steel (0.002% 보론첨가 저탄소강의 미세조직 및 기계적 성질에 미치는 열처리의 영향)

  • The effect of heat treatment on the micro-structures and the mechanical properties of 0.002% boron added low carbon steel was investigated. The tensile strength reached the peak at about $880-890^{\circ}C$ with the rising quenching temperature and then the hardness decreased sharply, but the tensile strength hardly decreased. The tensile and yield strength decreased and the total elongation increased with a rising tempering temperature, but the tensile and yield strength sharply fell and the total elongation prominently increased from above a $400-450^{\circ}C$ tempering temperature. Tempered martensite embrittlement (TME) was observed at tempering condition of $350-400^{\circ}C$. In the condition of quenching at $890^{\circ}C$ and tempering at $350^{\circ}C$, the boron precipitates were observed as Fe-C-B and BN together. The hardness decreased in proportion to the tempering temperature untill $350^{\circ}C$ and dropped sharply above $400^{\circ}C$ regardless of the quenching temperature.

  • The purpose of this research are to investigate experimentally flexural strengthening effects and flexural behaviour of RC beams strengthened by carbon fiber sheet(CFS) with/without superimposed pre-load. Test parameters of experiment are tension reinforcement ratio(0.85, 1.32, 1.91%) and pre-load(80% of yield capacity of unstrengthened beams). The structural behaviour of strengthened beams are compared with in terms of yield load and ultimate load, load-deflection relation, ductility, strengthened efficiency. From the test results, it were shown that ultimate capacity and flexural failure behaviour of RC beams strengthened by CFS changed by initial stresses between original beams and bonded CFS.

Steam Reforming of Toluene Over Ni/Coal Ash Catalysts: Effect of Coal Ash Composition

  • The development of a low cost catalyst with high performance and small amount of carbon deposition on catalyst from toluene steam reforming were investigated by using coal ash as a support material. Ni-loaded coal ash catalyst showed similar catalytic activity for toluene steam reforming compared with the Ni/Al2O3. At 800 ℃, the toluene conversion was 77% for Ni/TAL, 68% for Ni/KPU and 78% for Ni/Al2O3. Ni/TAL showed similar toluene conversion to Ni/Al2O3. However, Ni/KPU produced higher hydrogen yield at relatively lower toluene conversion. Ni/KPU catalyst showed a remarkable ability of suppressing the carbon deposition. The difference in coke deposition and hydrogen yield is due to the composition of KPU ash (Ca and Fe) which increase coke resistance and water gas shift reaction. This study suggests that coal ash catalysts have great potential for the application in the steam reforming of biomass tar.

  • Recently, hot rolled galvanized steel is widely used in automotive parts. As the paradigm of the automotive market has changed from fossil fuel vehicle to electric vehicle, the automotive industry needs more high-strength steels to reduce weights of automobiles. However, because high-strength steel contains high solute carbon, it is expected to have a risk of stretcher-strain on the surface due to dislocation trapping by solute [C] and [N]. Generally, galvanized steel is supposed to pass through a furnace around the temperature of Zinc pot to increase material temperature. Otherwise, the inhibition layer could not be formed. However, solute carbon and nitrogen are volatile enough to move around the furnace temperature. Moreover, the ratio of ferrite phase and precipitated Fe3C can be variable, resulting in yield point elongation related to the stretcher strain. Furthermore, the quality of the galvanized surface can be affected by a high temperature of the furnace. Although a relatively hot rolled galvanizing line furnace has a lower temperature than an annealing line furnace, it can affect various quality aspects. In other words, this paper aims to determine how these phenomena appear concerning furnace temperature.

Long-term Application Effects of Soil Amendments on Yield and Soil Properties in Paddy (논토양에서 토양개량제 장기연용에 따른 벼의 생육 및 토양특성 평가)

  • This research evaluated the long-term application effects of different soil amendments on yield, dissolved organic carbon, nitrogen and soil organic carbon stock in rice paddy. The experiment consisted of four different fertilizations; Inorganic fertilization (NPK), NPK+Lime (NPKL), NPK+Silicate (NPKS), NPK+Compost (NPKC). There was no significant difference in rice yield between the treatment groups in 1995, but the rice yields in the NPKL and NPKC treatments in 2019 increased by 4.3% and 14.3% compared to NPK. In terms of soil properties, the pH of NPKS(6.7) and NPKL(6.4) in 2019 increased the most compared to the soil pH before experiment(5.2). The organic matter(OM) content from NPKC treatment increased upto 34 and 27 g kg-1 in year of 1995 and 2019, respectively, compared to before the test. In NPKS and NPKL treatment, labile carbon and nitrogen content, used as a soil quality indicator, increased by 1.1-1.9 times over the control. From these result, it is suggested that type and application rate of soil amendment should be determined based on the soil analysis before cultivation for sustainable agricultural environment and productivity.

Comparative Study on The Composition of Essential Oil by Supercritical Carbon Dioxide Extraction and Hydro-distillation from Chamaecyparis obtusa Leaves (편백 잎에서 추출한 정유와 초임계 이산화탄소 추출물의 성분 비교분석)

  • This study was focused on the comparison of the variations in the yield and chemical composition of Chamaecyparis obtusa leaf oil obtained under different pressure conditions of the supercritical carbon dioxide extraction (SCE), and by hydro-distillation. SCE was carried out varying the pressure in the range of 100~400 bar at $40^{\circ}C$. The chemical composition of C. obtusa leaf oils was determined by gas chromatography-mass spectroscopy (GC-MS) analysis. The maximum yield of 4.4% (relative to the initial mass of oven dry mass) was obtained in the extraction under 300 bar pressure, which was higher than that of the hydro-distillation method (1.9%). The contents of sesquiterpenes in the extracts obtained by the SCE were higher than those of the essential oils of C. obtusa by the hydro-distillation. The sesquiterpenes in the SCE extracts made up approximately 39%~46% of the total, followed by monoterpenes, diterpene, and lignan. The contents of each constituent in the supercritical carbon dioxide extracts were varied on the extraction pressure. Therefore, these results showed that the extraction condition of SCE had significant effect on the yield of C. obtusa oils and its chemical composition.

Change of Growth and Yield Characters in Rice at Different Transplanting Densities under Agrivoltaics (영농형 태양광 발전 시스템 하부 벼 재배 시 재식밀도별 생육 및 수량변화)

  • Recently, interest in renewable energy development has been increasing to promote carbon-neutral policies. Agrivoltaic is a solar power generation facility with the potential to aid in meeting carbon-neutral policies. It has the advantage of generating electricity while farming takes place, but it also has the disadvantage of reducing crop yield and cultivation safety. We analyzed the rice yield, quality, and stem growth characteristics according to different transplanting densities under agrivoltaics. Under agrivoltaics, the number of rice panicles was reduced by the shading effect, but the reduction was lower under 60 hills than under 80 and 100 hills. Brown rice perfect ratio was increased under 60 hills under agrivoltaics. Brown rice yield did not differ significantly between 60 and 80 hills under agrivoltaics. However, stem dry weight by unit(mg/cm) in each internode showed highest under 60 hills compared to 80 and 100 hills under agrivoltaics. Therefore, 60 hill density was considered appropriate to ensure cultivation safety and yield when cultivated rice under agrivoltaics.

Effect of Martensite Fraction on the Tensile Properties of Dual-phase Steels Containing Micro-alloying Elements (미량합금 원소가 첨가된 2상 조직강의 인장 특성에 미치는 마르텐사이트 분율의 영향)

  • In this study dual-phase steels with different ferrite grain size and martensite fraction were fabricated by varying micro-alloying elements and intercritical anneling temperatures, and then the tensile properties were investigated in terms of yield and tensile strengths, elongation, and yield ratio. The addition of micro-alloying elements reduced ferrite grain size, and the increased intercritial transformation tempeature increased the martensite fracton. The tensile test results showed that yield and tensile strengths of all the steel specimens increased with increasing the martensite fraction. However, the elongation and yield ratio were differently changed according to variations in the morphology and carbon content of martensite, ferrite grain size, and precipitates resulting from the addition of micro-alloying elements and intercritical annealing.
