Evaluation of the Agreement between Immunodot Assays and Intradermal Skin Testing or Favrot Diagnostic Criteria in Canine Atopic Dermatitis

  • This study was undertaken to identify differences between atopic and non-atopic dogs in three rapid screening immunodot assays as well as the ability of the assays to predict the results of intradermal skin testing (IDST) or Favrot diagnostic criteria (FDC). Twenty-nine dogs diagnosed with canine atopic dermatitis (CAD) were selected as the atopic group. Twenty-five dogs without CAD were included as the non-atopic group. Three types of immunodot assays were conducted on all serum samples from both groups: Allercept E-screen 2nd generation (ES2G), Canine Allergic Tendency Reference Test (ALERT), and Asan Easy Test Canine IgE (AETC). IDST, which included 39 allergens, and immunodot assays were performed concurrently in 13 dogs from the atopic group and compared. While there were no significant differences in positivity between the two groups in the evaluation of ALERT (P = 0.435) and AETC (P = 0.313), positivity in ES2G testing was significantly higher in the non-atopic group than the atopic group (P = 0.038). The ES2G, ALERT, and AETC results showed fair (${\kappa}=0.235$), slight (${\kappa}=0.133$), and slight (${\kappa}=0.014$) accordance with IDST, respectively. The outcomes of ES2G, ALERT, and AETC indicated poor (${\kappa}=-0.211$), slight (${\kappa}=0.106$), and slight (${\kappa}=0.087$) agreement with FDC. In conclusion, rapid screening immunodot assays were not useful for the diagnosis of CAD. These assays may provide a supplementary method for predicting the results of IDST in atopic dogs.

Efficacy of 10-Injection Induced Modified Rush Immunotherapy in Dogs with Atopic Dermatitis

  • Modified rush immunotherapy (IT), by combination of rush IT and conventional IT, provides a faster method to reach maintenance dose, leading to higher patient adherence when compared with conventional IT, decreasing systemic adverse reactions when compared with a standard rush IT. Ten atopic dogs of this study include fulfillment of Favrot's criteria. Offending allergens were identified by the use of IDST. During the induction period, the dogs were received a total of 10 injections. Five injections were administrated every 30 minutes in a day with gradually increasing amounts and concentrations of allergens, and the last 5 injections were administered every 3 days. The efficacy of 10-injection induced mRIT was assessed using the canine atopic dermatitis extent and severity index (CADESI). During maintenance period, reduction rate from baseline scores varied between 3.2% and 60.9% and the after 6 months of therapy for CADESI-03 score in 6 of the 10 dogs. Adverse reactions were not observed in these dogs during induction period by mRIT with 10 injections. Based on these results, our modified rush IT protocol is considered to be a useful protocol to treat canine atopic dermatitis.

Efficacy of Modified Rush Allergen-Specific Immunotherapy on Canine Atopic Dermatitis

  • Modified rush ASIT protocol has been performed to identify the ideal schedule that allows the dose considered effective to be reached in the shortest possible time with the fewest adverse effects. Ten atopic dogs of this study includes fulfillment of Favrot's criteria. Offending allergens were identified by the use of IDST. During the induction period, the dogs were received a total of 15 injections. Ten injections were administrated every 30 minutes in a day with gradually increasing amounts and concentrations of allergens, and the last 5 injections were administered every 3 days. Disease severity was quantified by using the canine atopic dermatitis extent and severity index (CADESI). During induction period, reduction rate from baseline scores varied between 1% and 67% and the improvement of ${\geq}50%$ was recorded after induction period of therapy for CADESI-03 score in 6 of the 10 dogs. This study of ten dogs with atopic dermatitis provide evidence for the efficacy and safety of modified rush ASIT for clinical improvement.

개의 아토피성 피부염에서 분리한 Staphylococcus intermedius 균의 세균단백질의 면역반응 (Immune Response of Bacterial Proteins of Staphylococcus intermedius from Canine Atopic Dermatitis)

  • 개의 아토피성 피부염의 주된 세균성 감염은 Staphylococcus intermedius에 의해 발생한다. 본 연구의 목적은 아토피성 피부염을 가지고 있는 환견의 혈청과 정상견의 혈청을 이용하여 체액성 면역반응을 유발하는 주된 세균단백질을 규명하는데 있다. 건국대학교 수의과대학 부속동물병원에 내원한 아토피성 피부염 및 표재성 세균성 농피증을 앓고 있는 환견의 혈청 및 정상견의 혈청을 분리하여 본 실험에 사용하였으며 아토피성 피부염을 가지고 있는 환견에서 S. intermedius 균을 분리하였다. Brain heart infusion 액체배지 조건 및 $37^{\circ}C$ 배양조건에서 호기상태로 증균시켰으며 증균후 포도상구균을 PBS완충액에 부유시킨후 원심분리하고 최종적으로 lysostaphin을 이용하여 세균단백질을 분리하였다. 수거한 세균단백질은 SDS-PAGE을 이용하여 단백질을 전기영동하였으며 영동후 nitrocellulose membrane을 이용하여 단백질을 이동시켰다. Western blot은 anti-dog-IgG, 아토피성 피부염의 혈청 및 정상혈청을 이용하였다. 결론적으로 아토피성 피부염의 혈청을 이용해 확인한 S. intermedius의 주요 세균단백질은 18, 31, 75, 및 110 kDa이였다. 현재의 연구결과로 볼 때 아토피성 피부염에 감염된 대부분의 환견은 S. intermedius의 세균단백질에 대한 체액성 면역반응이 유발된다고 생각된다.

개의 아토피성 피부염의 피부증상 호발부위의 비만세포분포조사 (Mast Cell Distribution at Predilection Sites of Atopic Dermatitis in Normal Canine Skin)

  • 개의 아토피성 피부염의 피부증상이 호발하는 8부위에서 비만세포의 분포를 조사 비교하였다. 피부증상을 보이지 않는 10두의 개의 8부위의 피부조직을 채취하여 비만세포에서 특이적인 acidfied toluidine blue 염색을 하여 현미 경하에서 계수비교하였다. 비만세포수는 표재진피에서 각각의 부위간에 유의한 변화가 관찰되었으며 (p<0.0001) 표재진피가 심재진피에 비해 유의하게 높았다 (p<0.05). 호발 부위중 비만세포가 가장 높은 부위는 내측이개와 지간으로 각각 $74.88{\pm}17.93\;per\;mm^2$, $28.33{\pm}6.24\;per\;mm^2$이었다. 이 부위는 다른 부위에 비해 비만세포가 $280{\%}$ 높게 분포하였다. 따라서 개의 아토피성 피부염의 호발증상인 귀와 발의 소양증의 발생은 비만세포의 수가 다른 부위에 비해 높은 것이 하나의 요인으로 사료된다. 그러나 비만세포의 수가 낮게 나타난 다른 호발부위에 대해서는 다른 요인이 관계하는 것으로 사료되며 이에 관한 연구가 필요하다.

두 마리 개의 피부염에 있어서 Geotrichum candidum의 존재 (Occurrence of Geotrichum candidum in Two Cases of Canine Dermatitis)

  • 다양한 피부병을 갖고 있는 52두의 개에 대해 Geotrichum candidum의 감염율을 조사하였다 피부가검물은 15% KOH용액으로 케라틴을 용해한 후 현미경으로 직접 검사하였다. 또한 Sabouraud dextrose agar, chloramphenicol을 함유한 Sabouraud dextrose agar 및 brain heart infusion agar에 피부가검물을 접종하여 $25^{\circ}C와\;37^{\circ}C$에서 배양하여 원인균을 배양하고, 배양물을 현미경으로 검사하였다. 52두 중 2두의 가검물에 대한 직접 현미경 검사에서 직사각형 내지 원기둥형의 분절포자와 격벽이 있는 균사가 관찰되어 Geotrichum 감염이 제시되었다. 이 가검물의 배양물에서 균사와 $4-8\;\mu\textrm{m}$의 포자가 관찰되었다. 또한 이 진균은 분아포자를 생성하지 않고, 요소의 이용과 맥아당의 동화작용을 나타내지 않았다. 이 두 마리리의 피부염은 2% gentian violet또는 2% miconazole 의 국소적용에 임상적인 차도를 나타내었다 Geotrichum candidum은 사물기생성 진균이지만 개에 있어서 피부염의 원인이 될 수 있기 때문에 피부염 감별진단에 포함시켜야 할 것임을 제시하였다.

Comparison of Intradermal Skin Test and Multiple Allergen Simultaneous Test Results in Canine Atopic Dermatitis

  • Intradermal skin test (IDST) is generally considered a useful tool in identifying causal allergens in canine atopic dermatitis. Currently, multiple allergen simultaneous test (MAST), an in vitro testing method for allergen-specific immunoglobulin E, is being used as an alternative method. However, there are no reports comparing the IDST and MAST results in the same dogs. This study compared the results of both tests to evaluate the agreement and correlation between them. The sensitivity, specificity, and accuracy of the MAST were 76.2%, 64%, and 66.7%, respectively. Moderate positive predicted value (PPV, 50-75%) or high sensitivity (80-100%) were identified for indoor allergens, such as cat epithelia, house dust, and house dust mites. In contrast, high negative predicted value (NPV, 93.3-100%) and specificity (60-100%) were observed for environmental allergens and fungi. Although the agreement between IDST and MAST for all allergens was fair (κ = 0.301), that for each allergen was poor (κ < 0.01), except for birch (κ = 0.158). Spearman's rank correlation analysis revealed a low correlation between the MAST and IDST results (ρ = 0.308, p = 0.001). As compared to the IDST results, the MAST results did not identify the causative allergens sufficiently. IDST may not be performed for environmental allergens and fungi with high NPV and specificity if the MAST result is negative, but it may have to be performed for indoor allergens with moderate PPV and high sensitivity when the MAST result is positive.

개의 아토피성 피부염에 대한 cyclosporine의 경구 요법 (Application of oral cyclosporine in a dog with atopic dermatitis)

  • A 7.1 kg, seven-year old, castrated male, Shih-Tzu with severe pruritus, chronic otitis externa and Malassezia infection was referred to Veterinary Medical Teaching Hospital of Chungnam National University. In local animal hospital, steroid therapy was used to treat uncontrollable pruritus, but the clinical signs were recurrent when steroid therapy was discontinued. On physical examination, generalized alopecia, erythema, papules, severe crust and diffuse lichenification were presented. Tape strip test of skin lesions revealed cocci and Malassezia infections. Based on the result of history, clinical signs and examination described above, canine atopic dermatitis with secondary superficial pyoderma and Malassezia dermatitis was diagnosed. Oral challenge with cyclosporine and antibiotics had good results in clinical signs. Clinical sign scores were evaluated by investigator with CADESI at 2weeks, 4weeks, 6weeks, 8weeks and 10weeks after cyclosporine administration. And in the result of comparing of allergen-specific IgE value, the level of allergen-specific IgE to general causative allergen after 10 weeks of cyclosporine therapy was higher than that before cyclosporine therapy.

오소리의 개 디스템퍼 감염증 (Canine Distemper Virus Infection in Badgers)

  • Two dead and two terminally sick badgers with signs of lacrimation and nasal discharge were submitted to the Pathology Division of the National Veterinary Research Institute for necropsy. The major gross findings included chronic dermatitis and pneumonia. histologically intracytoplasmic and intranuclear acidophilic inclusion bodies consistent with Canine Distemper (CD) virus particles in lung kidney urinary bladder skin foot pad stomach and small intestine. Additionally there were diffuse bronchointerstitial pneumonia hyperkeratosis of foot pads and focal non-suppurative encephalitis. Canine distemper infection in these badgers was further confirmed by immuofluorescent technique which demonstrated CD virus-specific antigens in lung and kidney sections.
