The Effects of KR-10876, a new Quinolone Antimicrobial Agent, on the Central Nervous System

  • To evaluate KR-10876, a new fluoroquinolone antibacterial agent, its effects on the central nervous system(CNS) were investigated in mmice as part of phamacological study, and the results were compared with those for ciprofloxacin and ofloxacin, two prototypes of quinolone antiabctrial agents. All the parameters indicative of CNS function and acute toxicity were measured by close observation of the animals at regular time intervals after oral treatment of test compounds. KR-10876 did not have any effect on the parameters measured at lower does (100, 300 mg/kg, p.o.), it caused ptosis, suppressed spontaneous locomotor activity, hypothemia, and prolonged hexobarbital-induced sleeping time. KR-10876 also had a slight effect on motor coordination only at high dose. Simialr to ciprofloxacin, KR-10876 did not protect mice from pentylenetetrazol-strychnine-, and electroshock-inducedl convulsions at doses tested. These findings demonstrate that KR-10876 affects CNS functions only at high doses. The rank order for effects is ofloxacin$\le$KR-10876>ciprofloxacin.

  • Cold Neutron Source(CNS) facility comprises moderator circulation system, helium cooling system, neutron guide and auxiliary sistems. To increase the amount of cold neutron, the thermal neutron should pass cold moderator at cryogenic temperature. As cold moderator in Hanaro, the liquid hydrogen or liquid deuterium will be used and the temperature in operation will be used and the temperature in operation will be maintained to be $250^{\circ}C$ below zero. To maintain the moderator at this cryogenic temperature. He refrigerator is used to cool it down in thermosiphon having natural circulation. As a part of the conceptual design of Hanaro CNS, study on the characteristics of moderators, design of moderator chanmber and cooling method were done through the collaboration of Korea Atomic Energy Research Institute and Petersburg Nuclear Physics Institute. During the collaboration, a program for the design of moderator cooling system design concept through the parametric study using this program. In the parametric study, the effect of the moderator type on the design parameters was investigated. Also, the requirements on the performance test for the cooling system, which will be made before the basic design, were investigated.

Central nervous system activity of the methanol extracts of Caesalpinia bonducella and Bauhinia racemosa (Caesalpinaceae) in experimental animal model

  • The aim of the present study is to investigate central nervous system (CNS) activity of the methanol extracts of leaves of Caesalpinia bonducella (MECB) and stem bark of Bauhinia racemosa (MEBR) (Caesalpinaceae) in Swiss albino mice and Wistar albino rats. General behavior, exploratory behavior, muscle relaxant activity and phenobarbitone sodium-induced sleeping time were studied. The results revealed that the methanol extracts of leaves of Caesalpinia bonducella at 100 - 200 mg/kg and stem bark of Bauhinia racemosa 100 - 200 mg/kg caused a significant reduction in the spontaneous activity (general behavioral profile), remarkable decrease in exploratory behavioral pattern (Y-maze and head dip test), a reduction in muscle relaxant activity (rotarod and traction tests), and also significantly potentiated phenobarbitone sodium-induced sleeping time. The results suggest that MECB and MEBR exhibit CNS depressant activity in tested animal models.

  • The authors studied 5 cases of idiopathic CNS hypersomnia who visited Division of Sleep Studies, Seoul National University Hospital in 1995. Detailed medical history was taken and nocturnal polysomnography(NPSG), multiple sleep latency test(MSLT) and human leukocyte antigen(HLA) typing were performed. Neither cataplexy nor hypnagogic hallucination was reported in all cases and in NPSGs, there were tendencies of increased sleep period time and decreased slow wave sleep time. In MSLT, all the subjects showed average sleep latencies less than 8 minutes without sleep-onset rapid eye movement period(SOREMP). In HLA typing, some correlation between idiopathic CNS hypersomnia and HLA DR4 was observed. In contrast to previous reports, overall treatment response with methylphenidate was remarkable. Therefore, the authors suggest that patients suspected of idiopathic CNS hypersomnia be actively evaluated and treated with rather optimistic perspective.

Prevalence of Low Back Symptom and Impact of Job Stress among Working Women as Clinical Nurses in University Hospitals

  • Purpose: This study was aimed to investigate the impact of job stress on low back symptoms among Clinical nurses (CNs) in university hospital. Methods: A total of 322 CNs employed in a hospital in Seoul were interviewed by a well-trained interviewer using the structured questionnaire. Data collected for this study includes demographics, social and work characteristics, low back symptoms, and job stress. To test the impact of job stress on low back symptoms, we used multiple logistic regression analysis. Results: The prevalence of low back symptoms was 25.8% in this study. Low back symptoms differed significantly by factors, such as physical work burden, past history of injury and work duration. Also low back symptoms differed significantly by organizational system among independent variables of job stress. In a multiple logistic regression analysis, the odds ratio of organizational system to low back symptoms was 2.07 after an adjustment. Conclusion: This study showed that organizational system among job stress was a significant variable in explaining low back symptoms. We suggest the improvement of organizational system category as a preventive strategy for low back pain in CNs working in hospital.

Promotion of Remyelination by Sulfasalazine in a Transgenic Zebrafish Model of Demyelination

  • Most of the axons in the vertebrate nervous system are surrounded by a lipid-rich membrane called myelin, which promotes rapid conduction of nerve impulses and protects the axon from being damaged. Multiple sclerosis (MS) is a chronic demyelinating disease of the CNS characterized by infiltration of immune cells and progressive damage to myelin and axons. One potential way to treat MS is to enhance the endogenous remyelination process, but at present there are no available treatments to promote remyelination in patients with demyelinating diseases. Sulfasalazine is an anti-inflammatory and immune-modulating drug that is used in rheumatology and inflammatory bowel disease. Its anti-inflammatory and immunomodulatory properties prompted us to test the ability of sulfasalazine to promote remyelination. In this study, we found that sulfasalazine promotes remyelination in the CNS of a transgenic zebrafish model of NTR/MTZ-induced demyelination. We also found that sulfasalazine treatment reduced the number of macrophages/microglia in the CNS of demyelinated zebrafish larvae, suggesting that the acceleration of remyelination is mediated by the immunomodulatory function of sulfasalazine. Our data suggest that temporal modulation of the immune response by sulfasalazine can be used to overcome MS by enhancing myelin repair and remyelination in the CNS.

A study on effective disinfection methods of medical ultrasound probe resident floras (의료용 초음파 프로브 상제균의 효과적인 소독법에 관한 연구)

  • In the probe of a medical ultrasound device, three parts were selected randomly by the examiner and the bacteria in the probe were detected by the blood examiners. In addition, the degree of death of the pathogenic bacteria after each disinfection of the detected pathogens, disinfecting ethanol, and disinfecting tissue of the detected pathogens were analyzed quantitatively. The following was detected: S Aureus (32.3 %), Bacillus spp. (26.5 %), Micrococcus spp. (21.5 %), and CNS (20 %). With the conventional probe, S. aureus (26.2 %), a playback curve (24.2 %), and a micron (19.5 %), Micrococcus spp. (15.5 %), and CNS (14.6 %) were observed. In the fan probe, S. aureus (24.7 %), Enterococcus (24.7 %), Enterococcus (17.7 %), and CNS (13.8 %) were detected. The disinfection of the three pathogens detected revealed sterilization of most of the pathogens, and most cases contained at least 91.3 % of the total sterilizing effect (P>0.05). In addition, for the disinfection of Propolis extract and disinfecting tissue, the disinfection effect was lower than that of disinfecting ethanol, but the difference was not statistically significant (P>0.05). The results revealed bacteria on most of the ultrasound probes. Antiseptic disinfection of surgical instruments using an extract of propolis works with results similar to those of ethanol. A blood test along with disinfection can help prevent infection if an ultrasound probe is applied to food.

GBAS Flight Testing and Performance Assessment using Flight Inspection Aircraft at Gimpo International Airport (비행검사용 항공기를 이용한 김포국제공항 GBAS 비행시험 및 성능평가)

  • Ground Based Augmentation System(GBAS) is a system that offers an aircraft within 23 NM radius from the airport precision positioning service and precision approach service using the concept of Differential Global Positioning System(DGPS). After GBAS ground equipment installing at the airport, functionalities and performances of GBAS should be verified through the GBAS ground and flight testing. This paper describes the methods and results for GBAS flight test using the flight inspection aircraft at Gimpo International Airport. From the test results, we confirmed that the VDB data was received without misleading within the VDB coverage of Gimpo International Airport, and VDB field strength, protection level, and course alignment accuracy met the evaluation's criteria.

A Study on Learning Experiences and Self-Confidence of Core Nursing Skills in Nursing Practicum among Final Year Nursing Students (졸업학년 간호학생의 핵심기본간호술 학습경험과 수행자신감 조사연구: 실습교과를 중심으로)

  • Purpose: This study was done to identify learning experiences and self-confidence and to analyze nursing students' self-confidence according to learning experiences for core nursing skills (CNS). Method: Participants were 502 final year nursing students. Data were collected using a structured questionnaire and analyzed using descriptive statistics and t-test. Results: Over 60% of participants had practiced 15 items of the 24 CNS in the basic nursing lab (BNL). In clinical practice (CP), they had practiced five items but experienced only one item in a simulation lab (SL). Items with the highest confidence level were vital signs (4.69) followed by blood sugar test (4.60), pulse oximeter (4.38), and oral medication (4.12). Items with the lowest confidence level were blood transfusion (2.17) followed by enema (2.64) and indwelling catheterization (2.67). The group with CNS experience in the practice curriculum was generally more confident than the group with no experience. Self-confidences in some skills was significantly different depending on availability of SL, credits for BNL and CP, and presence of preceptors. Conclusion: Results indicate a need to develop practice education strategies such as changing the CP to practice-centered learning with preceptors and including well-designed SL to increase confidence of nursing students.

Shear Load-Transfer Function of Rock-Socketed Drilled Shafts Considering Borehole Roughness (굴착면 거칠기를 고려한 암반 근입 현장타설말뚝의 주면 하중전이함수 제안)

  • Shear load transfer characteristics of rock-socketed drilled shafts were analyzed. The constant normal stiffness (CNS) direct shear tests were performed to identify the major influencing factors of shaft resistance, i.e., unconfined compressive strength, borehole roughness, normal stiffness, initial confining stress, and material properties. Based on the CNS tests, shear load transfer function of drilled shafts in rocks is proposed using borehole roughness and the geological strength index (GSI), which indicates discontinuity and surface condition of rock mass in Hoek-Brown criterion (1997). The proposed load-transfer function was verified by the load test results of seven rock-socketed drilled test shafts subjected to axial loads. Through comparisons of the results of load tests, it is found that the load-transfer function by the present study is in good agreement with the general trend observed by in situ measurements, and thus represents a significant improvement in the prediction of load transfer of drilled shafts.
