미생물 생장 예측모델과 반응표면분석법을 이용한 Vibrio parahaemolyticus의 신속 증균배지 개발 (Development of a Rapid Enrichment Broth for Vibrio parahaemolyticus Using a Predictive Model of Microbial Growth with Response Surface Analysis)

  • 본 논문은 적은 농도로 존재하는 Vibrio parahaemolyticus를 6시간이내에 106 CFU/mL까지 신속하게 배양가능한 배지인 Rapid Enrichment Broth for V. parahaemolyticus(REB-V)를 개발하여 V. parahaemolyticus를 신속하게 검출할 수 있도록 하였다. Modified Gompertz model과 RSM를 활용하여 V. parahaemolyticus를 신속하게 증균할 수 있도록 첨가 성분을 최적화하였다. 그 결과 2%(w/v) NaCl BPW에 D-(+)-mannose 0.3 g/L, L-valine 0.2 g/L, magnesium sulfate 0.2 g/L를 첨가하였을 때 V. parahaemolyticus의 증균량이 최대였다. REB-V의 배양조건을 pH 7.84와 37℃으로 최적화하였으며, 2%(w/v) NaCl BPW와 비교하여 REB-V의 증균 효율을 확인하기 위해 7시간 증균을 통해 증균량을 평가하였다. 또한 매시간마다 배양액에서 DNA를 추출하였고, 추출한 DNA로 real-time PCR을 수행하여 REB-V의 분자진단법 적용가능성을 확인하였다. 그 결과 7시간 동안 2%(w/v) NaCl BPW에서 V. parahaemolyticus는 5.452±0.151 Log CFU/mL까지 증균되었고, REB-V에서는 7.831±0.323 Log CFU/mL의 V. parahaemolyticus가 증균되었다. 최종적으로 REB-V에서 6시간 이내에 106 CFU/mL 이상으로 증균된 것을 확인하였고, REB-V의 증균속도가 2%(w/v) NaCl BPW보다 빠르고 같은 시간 내의 증균량이 2%(w/v) NaCl BPW보다 많았음을 통해 REB-V의 증균효율이 우수한 것으로 확인되었다.

  • 양식 농어에 있어서 연속 채혈 스트레스는 혈장내 glucose 농도의 뚜렷한 증가 및 말초 혈액내 식세포의 chemiluminescent 반응을 유의적으로 감소시켰다. 실험어는 연속적인 스트레스에 대해 축적적인 반응을 나타내었으며, 혈장내 glucose의 농도는 스트레스 부과 시간간격에 의존하는 것으로 나타났다. 혈장내 glucose의 농도와 chemiluminescent 반응간에는 통계적으로 유의적인 (P<0.05) 음의 상관관계를 나타내었다.

The Novel SCN- Ion-selective Electrode Based on the 1-Benzyl-3-(4-nitrophenyl) thio-urea Ionophore

  • A potentiometric sensor based on the 1-benzyl-3-(4-nitrophenyl) thio-urea was synthesized and tested as an ionophore in PVC based membrane sensor towards $SCN^-$ ions. This membrane exhibits a linear stable response over a wide concentration range ($1.0{\times}10^{-5}$ to $1.0{\times}10^{-2}M$) with a slope of -59.2 mV/dec., a detection limit of ${\log}[SCN^-]=-5.05$, and a selectivity coefficient for thiocyanate against perchlorate anion of ${\log}K^{pot}_{SCN^-j}=-0.133$. The selectivity series of the membrane is as follows: $SCN^-$ > $ClO_4{^-}$ > $I^-$ > $NO_3{^-}$ > $HSO_3{^-}$ > $Cl^-$ > $HSO_4{^-}$ > $F^-$ > $CH_3COO^-$ > $HCO_3{^-}$ > $Br^-$ > $H_2PO_4{^-}$ > $SO{_3}^{2-}$ > $SO{_4}^{2-}$ > $CO{_3}^{2-}$. The proposed electrode showed good selectivity and a good response for the $SCN^-$ ion over a wide variety of other anions in pH 6.0 buffer solutions and has a fast response time of about < 5s. The influences of the membrane by pH, ionophore, and plasticizer were studied.

Optimisation of Calcium Alginate and Microbial Transglutaminase Systems to form a Porcine Myofibrillar Protein Gel

  • The aim of this study was to model and optimize the calcium alginate (CA) and microbial transglutaminase (TG) systems to form a cold-set myofibrillar protein (MP) gel containing 0.1 M or 0.3 M NaCl using a response surface methodology. The gel strengths of cold-set and heat-induced MP gels, and cooking yields were measured. All measured parameters showed determination coefficients ($R^2$) above 0.7 without a lack-of-fit. The CA system had the best results with component ratios of 1.0:0.3:1.0 corresponding to sodium alginate, calcium carbonate and glucono-$\delta$-lactone, respectively, and was favourable at 0.1 M NaCl. In contrast, the TG system only had an effect on cold-set MP gelation at 0.3 M salt, and the optimal ratio of TG to sodium caseinate was 0.6:0.5. By combining the two systems at 0.3 M NaCl, an acceptable cold-set MP gel with an improved texture and high cooking yield could be formed. Therefore, these results indicated that the functionality of the cold-set MP gel could be enhanced by combining these two optimized gelling system.

Comparison of Membrane Currents in Xenopus Oocytes in Response to Injection of Calcium Influx Factor (CIF) and Depletion of Intracellular Calcium Stores

  • The depletion of intracellular calcium stores by thapsigargin treatment evoked extracellular calcium-dependent membrane currents in Xenopus laevis oocytes. These currents have been compared to those evoked by microinjection of a calcium influx factor (CIF) purified from Jurkat T lymphocytes. The membrane currents elicited by thapsigargin treatment (peak current, $163{\pm}60$ nA) or CIF injection (peak current, $897{\pm}188$ nA) were both dependent on calcium entry, based on their eradication by the removal of extracellular calcium. The currents were, in both cases, attributed primarily to well-characterized $Ca^{2+}-dependent$ $Cl^-$ currents, based on their similar reversal potentials (-24 mV vs. -28 mV) and their inhibition by niflumic acid (a $Cl^-$ channel blocker). Currents induced by either thapsigargin treatment or CIF injection exhibited an identical pattern of inhibitory sensitivity to a panel of lanthanides, suggesting that thapsigargin treatment or CIF injection evoked $Cl^-$ currents by stimulating calcium influx through pharmacologically identical calcium channels. These results indicate that CIF acts on the same calcium entry pathway activated by the depletion of calcium stores and most lanthanides are novel pharmacological tools for the study of calcium entry in Xenopus oocytes.

죽염의 일반약리작용 (General Pharmacology of Bamboo Salt)

  • This study has been carried out to investigate general pharmacological action of bamboo salt (jukyom) in terms of effects on central nervous system and cardiovascular system in experimental animals. After bamboo salt, crude salt or reagent-grade NaCl were orally administered into male ICR mice with dose of 2.0 g/kg, general behavioural syndromes such as body weight and locomotor activity, spontaneous motor activity, pento-barbital-induced sleeping time, muscle incoordination, electroshock-induced convulsion, body temperature and writhing response caused by 0.6% acetic acid solution were observed. Bamboo salt had no influences in these indices for examinition of effect on central nervous system. Additionally, conscious male Sprague Dawley rats fastened overnight won ere treated with bamboo salt, crude salt or reagent-grade NaCl (2.0 g/kg, p.o.) to examine the effect of these salts cardiovascular system. Systolic, median and diastolic food pressure and heart rate were dertemined using tail cuff indirect method. Treatment with Hydralazine (50 mg/kg, p.o) as a positive control produced the decreases in systolic, median and diastolic blood treasure and an increase in heart rate. whereas no changes were observed in bamboo salt, crude salt and reagent-grade NaCl treated groups. These results strongly suggest that bamboo salt may have no effects on general pharmacology of central nervous systems and cardio-vascular systems.

Characterization of the Aroma of Salt-fermented Anchovy Sauce Using Solid Phase Microextraction-Gas Chromatography-Olfactometry Based on Sample Dilution Analysis

  • Aroma-active compounds were evaluated from salt-fermented anchovy sauce by solid phase microextraction-gas chromatography-olfactometry (SPME-GC-O) based on sample dilution analysis (SDA). SPME extract from carboxen/polydimethylsiloxane (CAR/PDMS) fiber was the most similar to the original odor of salt-fermented anchovy sauce used for this experiment, followed by divinylbenzene/CAR/PDMS (DVB/CAR/PDMS) fiber. Because salt-fermented anchovy sauce contains 23% NaCl, NaCl concentration of diluent was considered when salt-fermented anchovy sauce was serially diluted. Linear relationship between GC response and sample concentration was observed when diluted with 23% NaCl solution, whereas not observed when diluted with deodorized distilled water. Eleven and 16 aroma-active compounds were detected by SPME-GC-O based on SDA using CAR/PDMS and DVB/CAR/PDMS fibers, respectively. Butanoic acid and 3-methyl butanoic acid showed the highest ${\log}_2SD$ factors for CAR/PDMS and DVB/CAR/PDMS fibers. Dimethyl trisulfide, methional, trimethyl amine, 1-penten-3-ol, and acetic acid were also detected as potent aroma-active compounds.

Role of Calcium in the Osmoregulation under Salt Stress in Dunaliella salina

  • Involvement of calcium in signal transduction of salt stress was investigated in 1.7 M NaCl adapted Dunaliella salina, extremely halotolerant, unicellular green alga. When hyperosmotic (3.4 M NaCl) or Hypoosmotic (0.8 M NaCl) stress was treated, extracellular calcium was influxed in or intracellular calcium effluxed from D. salina, respectively, and these fluxes were proportional to the degree of stress. This might indicate indirectly that the change of calcium level occurred within the cells. In addition, the change of calcium flux was ahead of glycerol synthesis which has been known as the physiological response to salt stress. Osmoregulation was affected byextracellular calcium concentration, and increase of glycerol content as an osmoticum was inhibited about 50% by treatment of TFP and W-7 known as calmodulin specific inhibitors. Furthermore, in the case of the hyperosmotic stressed cells, the amount of 21 kD and 39 kD protein appeared to be calcium binding protein were increased. Among these, the 39 kD protein was detected only in the hyperosmotic stressed cells. The results obtained in the present work suggest that the possibility of calcium as a second messenger in the transduction of salt stress signal exists in the osmoregulation system of D. salina.

Influence of Intermittent Lighting on Broiler Performance, Incidence of Tibial Dyschondroplasia, Tonic Immobility, Some Blood Parameters and Antibody Production

  • The aim of this study was to determine the effect of two lighting programs (continuous lighting (CL) 24L:0D and intermittent lighting (IL) 1L:3D) on the broiler performance, carcass traits, incidence of tibial dyschondroplasia (TD), relative asymmetry (RA), duration of induced tonic immobility (TI), heterophils-lymphocytes ratio (H/L), serum glucose, cholesterol and triglyceride levels. The chicks were randomly divided into 2 treatment groups consisting of 100 chicks per treatment, continuous lighting (CL) 24L:0D or intermittent lighting (IL) 1L:3D. Each treatment consists of 5 replicates of 20 chicks. The experimental period was 6 weeks. Use of IL decreased feed to gain ratio, improved immune response and reduced fearful. Body weight, carcass traits, TD and stress parameters (organ weights, RA, H/L, glucose, cholesterol and triglyceride levels) were not significant in different lighting groups. As a result IL was beneficial for producers and chickens than CL.

Inhibition of ethylene biosynthesis enhances embryogenesis of cultured microspores of Brassica napus

  • Procedures that induce microspore embryogenesis have been described for a range of Brassica species, but embryo yield remains low for a number of genotypes. We have carried out experiments with the microspores from a weakly responsive line of B. napus to determine the culture conditions that optimize their in vitro embryogenesis by treating them with effectors of ethylene synthesis and action. The results revealed that isolated microspores subjected to an initial heat stress in a medium supplemented with inhibitors of ethylene synthesis such as AVG and $CoCl_2$ exhibited significantly increased embryo yields. This suggested that regulatory effects are exerted by the ethylene produced by the isolated microspores on the early processes of gametogenesis. As a consequence, treatment of microspores with SAM, an ethylene synthesis precursor, or with the ethylene-releasing agent ethephon, led to decreases in embryo yield. A special response to ethylene during the early stages of microspore development was finally shown to occur through experiments where isolated microspores were treated for increasing periods of time with $CoCl_2$. Collectively, our data demonstrated that the induction of embryogenesis induced by heat stress can be enhanced by inhibitors of ethylene biosynthesis.
