Analysis of Environmental Factors Related to Seasonal Variation of Bacteria and Heterotrophic Nanoflagellate in Kyeonggi Bay, Korea (경기만에서 박테리아와 종속영양편모류의 계절변화에 미치는 환경요인 분석)

  • From June 2007 to May 2008, seasonal variations of bacterial abundance and heterotrophic nanoflagellate (HNF), together with environmental factors, were investigated at weekly and monthly intervals in Kyeonggi Bay. During the study period, the water temperature and salinity varied from $1.9^{\circ}C{\sim}29.0^{\circ}C$ and 31~35.1 psu, respectively. The concentration of ammonia, nitrate+nitrite, phosphate, and silicate ranged from 0.01 to $3.22{\mu}M$, 2.03 to $15.34{\mu}M$, 0.06 to $1.82{\mu}M$, and 0.03 to $18.3{\mu}M$, respectively. The annual average concentration of Chl. a varied from $0.86{\mu}g\;L^{-1}$ to $37.70{\mu}g\;L^{-1}$; the concentration was twice as much at the surface than at the deeper layers. The abundance of bacteria and HNF ranged from $0.29{\times}10^6$ to $7.62{\times}10^6cells\;mL^{-1}$ and $1.00{\times}10^2$ to $1.26{\times}10^3cells\;mL^{-1}$, respectively. In particular, there were significant correlations between bacteria and HNF abundance (p<0.05), and then the high abundance of HNF was frequently observed with an increase of bacterial abundance in summer (p<0.001). Our results therefore indicate that bacterial abundance in the bay was mainly controlled by resources supplied as organic and inorganic substances from Lake Shihwa due to the daily water exchange after dike construction. Also, the bacterial abundance was significantly controlled by HNF grazing pressure (top-down) in the warm seasons, i.e. excluding winter, in the Kyeonggi Bay.

Characteristics of Aquatic Environment and Algal Bloom in a Small-scaled Agricultural Reservoir (Jundae Reservoir) (소규모 농업용 전대저수지의 수환경 변화와 조류발생 특성)

  • This study was conducted to identify the relationship between environmental factors and algal bloom, and provide information for efficient management based on the results of monitoring the environmental parameters and algal diversity in the Jundai reservoir from March 2011 to October 2013. Little change in the weather conditions was observed during the study period except for a slight decrease in rainfall. Concentration of TN and TP in the reservoir exceeded water quality standards for agriculture and significant correlation between algal growth and environmental factors was observed. Phytoplankton in Jundai reservoir included 6 classes, 40 genus, 62 species, and the phytoplankton abundance was in the range of $1.3{\times}10^4{\sim}2.8{\times}10^6$ cells $mL^{-1}$. The annual average of phytoplankton abundance and Chl-a gradually decreased as TN and TP concentrations decreased. Overall Anabaena sp., Oscillatoria sp., and Microcystis sp. were the dominant species in Jundai reservoir. As the water temperature increased, the dominant species were Anabaena sp., Microcystis sp. and Oscillatoria sp., in that order. Anabaena sp. was dominant from spring to early summer with increase in water temperature and pollutant concentrations, and high correlation with environmental factors was observed. Microcystis sp. was dominant depending on changes in the nutrient levels. In the case of Oscillatoria sp., there was no significant correlation between phytoplankton biomess and Chl-a. However, efficient management of water environment and practical control of algal bloom in small scale reservoir polluted by livestock and farm irrigation should be achieved by identification of the relationship between algal growth and environmental factors.

The Characteristics on the Spatio-temporal Distributions of Phytoplankton Communities in Deukryang Bay, Southwestern Korea (득량만 식물플랑크톤 군집의 시.공간적 분포특성.)

  • The observations on the spatio-temporal distribution and seasonal fluctuations of phytoplankton community were carried out in Deukryang Bay of the Korean Southwestern Sea from June 1992 to April 1993. A total of 75 species of phytoplankton belonged to 47 genera was identified. In Deukryang Bay seasonal succession in dominant species; P. alata, G. flaccida, S. costatum, L. danicus and N. longissima in summer, St. palmeriana, Ch. curvisetus and B. paxillifera in autunm, S. costatum, Ch. curvisetus, E. zodiacus and Pn. pungens in winter, and As. glacialis, As. kariana, N. pelagica, Th. nitzschioides and S. costatum in spring, were very marked, that is to say, the communities structure of phytoplankton in Deukryang Bay appeared to be various species composition and it was occupied with diatoms all the year round. Phytoplankton standing crops fluctuated with an annual mean of $1.4{\times}10^5 cells/1 between the lowest value of 2.6{\times}10^3 cells/1 in July and the highest value of 1.0{\times}10^6 cells/1$ by S. costatum in January. Densities of the phytoplankton cell number by the samples of Deukryang Bay ranged from $2.6{\times}10^3cells/1 to 1.2{\times}10^5 cells/1 with the mean value of 3.6{\times}10^4cells/1 in summer, from 6.0{\times}10^3cells/1 to 2.6{\times}10^5 cells/1 with mean of 1.5{\times}10^5 cells/1 in autumn, from 1.3{\times}10^4cells/1 to 1.0{\times}10^6 cells/1 with mean 3.5{times}10^5 cells/1 in winter, and from 4.8{\times}10^3cells/1 to 6.0{\times}10^5 cells/1 with mean of 1.6{\times}10^5 cells/1$ in autumn. That is to say, phytoplankton standing crops was large in low temperature seasons, on the other hand small in high temperature seasons. Chlorophyll $\alpha$ concentration fluctuated between 0.l9 $\mu$g/l and 12.3 $\mu$g/l in March. in Deukryang Bay seasonal flucturation in chi-$\alpha$ concentration was not marked. Especially, chl-$\alpha$ concentration in the water around Deukryang Island located in the middle part of Deukryang Bay showed patchy distributions with a very high concentration. And chl-$\alpha$ concentration was high during a year. Therefore, phytoplankton production in Deukryang Bay could be very high year-round.

  • Mutagenicity of recombinant human erythropoietin (rhEPO) was examined in the reverse mutation test on bacteria, in the chromosomal aberration test on cultured mammalian cells and in the micronucleus test on mice. The reverse mutation test was performed by a plate incorporation method with or wothout a metabolic activation system (59 Mix) using Salmonella typhimurium strain TA100, TA1535, TA98 and TA 1537. The rhEPO did not significantly increase revertant colonies in any of the test strains under any conditions at dose levels ranging from 1000 H/ml to 62.5 lu/plate, compared with the vehicle control. In the chromosomal aberration test using cultured Chinese Hamster Lung (CHL) cells, the number of aberrant cells was not increased in the presence or absence of 59 Mix at concentrations of 1000 lU/ml to 250 lU/ml, compared with the vehicle control. In the micronucleus test, male ICR mice were given rhEPO intraperitoneally at a dose level of 25000, 12500 and 6250 lU/kg. The incidence of bone marrow micronucleated polychromatic erythrocytes was not different from that of the vehicle control. From these results, rhEPO is considered to be non-mutagenic under the present test conditions.

Water Quality Modeling of Youngju Dam Reservoir by HSPF, EFDC and WASP (HSPF, EFDC 및 WASP에 의한 영주다목적댐 저수지의 수질예측)

  • This study was carried out to investigate the effect of EFDC hydrodynamic result on the WASP7.3 water quality modeling result in accordance with the change of number of grid for the dam reservoir to be constructed. The simulated flow and BOD, T-N and T-P loads by the HSPF watershed model was used for boundary conditions and the hydrodynamic modeling results was linked with WASP model to predict future water quality after dam construction. The scenarios for EFDC modeling were composed of Scenario 1(141 grid cells) and Scenario 2(568 grid cells). The results of Scenario 2 showed that BOD, T-N, T-P and Chl-a concentrations were decreased 0.073mg/L(8.5%), 0.032mg/L(2.6%), 0.003mg/L(6.8%), 0.644mg/L(4.2%) compared with those of Scenario 1, respectively. As number of grid cell increased, water quality concentrations were decreased and also it caused the longer running time. Therefore, this study suggests that the consideration of the geometry of water body is more important than the number of grid cells for the prediction of water quality of a dam reservoir in EIA.

Mutagenicity of DA-5018, a Non-narcotic Analgesic Agent (비 마약성 진통제 DA-5018의 변이원성 연구)

  • DA-5018, a non-narcotic analgesic agent, was examined for mutagenicity in the reverse mutation test on bacteria, chromosomal aberration test on cultured mammalian cells and micronucleus test on mice. The reverse mutation test was performed by a plate incorporation method with or without a metabolic activation system(S9 mix) using Salmonella typhimurium strain TA100, TA1535, TA98 and TA1537. DA-5018 did not significantly increase revertant colonies in any of the test strains under any conditions at concentrations ranging from 0.0049 to 1.25 mg/plate, compared with the vehicle control. In the chromosomal aberration test using cultured Chinese Hamster Lung(CHL) cells, DA-5018 did not increase the number of aberrant cells in the presence or absence of S9 mix at concentrations of 0.016 mM/plate to 0.25 mM/plate, compared with the vehicle control. In the micronucleus test, male ICR mice were given DA-5018 intraperitoneally at a dose level of 0.55, 1.10 and 2.20 mg/kg. The incidence of bone marrow micronucleated polychromatic erythrocytes in the DA-5018 treated mice was not significantly different from that of the vehicle control. These results indicate that DA-5018 does not have mutagenic potential under the present test conditions.

The incidence of bone marrow micronucleated polychromatic erythrocytes in the DA-3002 treated mice did not differ from that of the vehicle control. These results indicate that DA-3002 doesn't have mutagenic potential under the present test conditions.

Evaluation of Genotoxicity of SU-Eohyeol Pharmacopuncture Using an In Vitro Chromosome Aberration Test in Chinese Hamster Lung Cell

  • Objectives: This study was conducted to evaluate the safety of SU-Eohyeol pharmacopuncture (SUEP) by assessing its potential to cause chromosomal abnormalities in Chinese hamster lung cells (CHL/IC). Methods: A dose-curve was conducted to determine the highest dose of SUEP. Doses of 10, 5, 2.5, 1.25, 0.625, and 0.313% were used, and no cytotoxicity or SUEP precipitation was observed. SUEP doses of 10, 5, and 2.5%, with positive and negative controls, were used in a chromosome aberration test. Results: In this study, the frequency of abnormal chromosomal cells in the SUEP group did not show a statistically significant difference from that of the negative control group in short-term treatments with and without metabolic activation and the continuous treatment without metabolic activation. Compared with the negative control group, the positive control group had a significantly higher frequency of cells with structural chromosomal abnormalities. This test's results satisfied all conditions for determining the results. Conclusion: SUEP did not induce chromosomal aberrations under the conditions of this study. Other toxicity evaluations, safety studies in humans, and various clinical trials are required to evaluate the safety and efficacy of SUEP.

Monthly HPLC Measurements of Pigments from an Intertidal Sediment of Geunso Bay Highlighting Variations of Biomass, Community Composition and Photo-physiology of Microphytobenthos (HPLC를 이용한 근소만 조간대 퇴적물내의 저서미세조류 현존량, 군집 및 광생리의 월 변화 분석)

  • In this study, the surveys were carried out from October (2016) to October (2017) along the tidal flat of Geunso Bay, Taean Peninsula of the western edge of Korea. The sampling trips were carried out for a total of 16 times, once or twice a month. In order to investigate the monthly variation of the microphytobenthos (MPB) biomass, community composition and photo-physiology were analyzed by HPLC (High performance liquid chromatography). The total chlorophyll a (TChl a) concentrations used as an indicator of biomass of MPB in the upper 1 cm sediment layer ranged from 40.4 to $218.9mg\;m^{-2}$ throughout the sampling period. TChl a concentrations showed the maximum level on $24^{th}$ of February and remained high throughout March after which it started to declined. The biomass of MPB showed high values in winter and low values in summer. The monthly variations of Phaeophorbide a concentrations suggested that the low grazing intensity of the predator in the winter may have partly attributed to the MPB winter blooming. As a result of monthly variations of the MPB community composition using the major marker pigments, the concentrations of fucoxanthin, the marker pigment of benthic diatoms, were the highest throughout the year. The concentrations of most of the marker pigments except for chlorophyll b (chlorophytes) and peridinin (dinoflagellates) increased in winter. However, the concentrations of fucoxanthin increased the highest, and the relative ratios of the major marker pigments to TChl a except fucoxanthin decreased during the same period. The vertical distribution of Chl a and oxygen concentrations in the sediments using a fluorometer and an oxygen micro-optode Chl a concentrations decreased with oxygen concentrations with increasing depth of the sediment layers. Moreover, this tendency became more apparent in winter. The Chl a was uniformly vertical down to 12 mm from May to July, but the oxygen concentration distribution in May decreased sharply below 1 mm. The increase in phaeophorbide a concentration observed at this time is likely to be caused by increased oxygen consumption of zoobenthic grazing activities. This could be presumed that MPB cells are transported downward by bioturbation of zoobenthos. The relative ratios (DT/(DD+DT)) obtained with diadinoxanthin (DD) and diatoxanthin (DT), which are often used as indicators of photo-adaptation of MPB, decreased from October to March and increased in May. This indicated that there were monthly differences in activity of Xanthophyll cycle as well.

Genotoxicity of Environment-friendly Organic Materials of Plant Origin in the Micronucleus Test Using Chinese Hamster Lung Cells (Chinese Hamster Lung Cell의 소핵시험을 이용한 식물추출물 유기농업자재의 유전독성평가)

  • BACKGROUND: Azadirachta Indica extract(AIE) and Sophorae radix extract(SRE) are widely used as environment-friendly organic materials of plant origin in South Korea. METHODS AND RESULTS: In this study, the in vitro micronucleus(vitMN) tests of two samples of AIE and SRE were conducted to evaluate their genotoxicity using the Chinese hamster lung(CHL) cell. This study was composed of two parts; cytochalasin B(cyto B) test and non-cyto B test. Mitomycin C and colchicine were used as positive controls. As a result, the incidence of micronucleus(MN) in all AIE and SRE treated groups increased in dose-dependent manner, but were less than 2.2% in 1,000 binucleated cells. In addition, there were no significant increases of MN incidence in all AIE and SRE treated groups, compared with the negative control group. CONCLUSION: Therefore, we suggest that AIE samples and SRE samples used in this study may have no genotoxicity in the in vitro micronucleus test using the CHL cells. In our previous study, we reported that AIE and SRE did not cause genotoxicity in Ames test. According to the genotoxicity battery system, we concluded that AIE and SRE used in this study have no genotoxic effects to humans.
