Identification of Aster Yellows Phytoplasma in Dendranthema grandiflorum

  • Phytoplasmas were identified from two chrysanthemum (Dendranthema grandiflorum) plants showing different symptoms ; one with stusting, rosette, and excessive branching (Ph-ch1), and the other with stunting and chlorosis (Ph-ch2). Electron microscopy of midrib of the plants with the symptoms revealed that numerous phytoplasmas were localized in the phloem cells. The disease was transmitted from infected plants to healthy ones by grafting. Phytoplasma-specific DNA was detected in polymerase chain reaction (PCR) analysis with template DNA extracted from the leaves of Ph-ch1 and Ph-ch2, both of which yielded a same DNA band corresponding to 1.5 kb. Using a specific primer pair (R16F1/R1) synthesized based on aster yellows (AY) phytoplasma, a DNA fragment of 1.1 kb was amplified by PCR. Endonuclease restriction patterns of the 1.1 kb PCR products from Ph-ch1 and Ph-ch2, which were dgeste with each of the restriction endonucleases Sau3A, Hha, Alu and Rsa, were same as those of AY phytoplasma from periwinkle. This suggests that the chrysanthemum plants (Ph-ch1 and Ph-ch2) be infected with a phytoplasma belonging to AY phytoplasma.

Volumetric Behaviour of Binary Liquid Mixtures at a Temperature of 303.15 K

  • Excess molar volumes (Vm E ) of binary liquid mixtures: xC6H5CH3 + (1-x1)CH3CN or + (1-x1)C6H5NO2, or + (1-x1)C2H5NO2 have been determined as a function of mole fraction of C6H5CH3 (x) at a temperature of 303.15 K over a entire range of composition. The densities of the binary liquid mixtures were determined by pycnometrically. The VmE values of the mixtures have been found to be negative over the whole composition in order of C6H5CH3 + C6H5NO2, < C6H5CH3 + CH3CN, and < C6H5CH3 + C2H5NO2. The negative magnitude of VmE suggests the presence of intermolecular interaction in the three binary liquid mixtures.

Response Characteristics of Thick Film Sensors Using Nano ZnO:Ni for Hydrocarbon Gas (나노 ZnO:Ni를 이용한 후막 가스센서의 탄화수소계 가스에 대한 감응특성)

  • The effects of a Ni coating on the sensing properties of nano ZnO:Ni based gas sensors were studied for $CH_4$ and $CH_3CH_2CH_3$ gases. Nano ZnO sensing materials were prepared by the hydrothermal reaction method. The Ni coatings on the nano ZnO surface were deposited by the hydrolysis of zinc chloride with $NH_4OH$. The weight % of Ni coating on the ZnO surface ranged from 0 to 10 %. The nano ZnO:Ni gas sensors were fabricated by a screen printing method on alumina substrates. The structural and morphological properties of the nano ZnO : Ni sensing materials were investigated by XRD, EDS, and SEM. The XRD patterns showed that nano ZnO : Ni powders with a wurtzite structure were grown with (1 0 0), (0 0 2), and (1 0 1) dominant peaks. The particle size of nano ZnO powders was about 250 nm. The sensitivity of nano ZnO:Ni based sensors for 5 ppm $CH_4$ gas and $CH_3CH_2CH_3$ gas was measured at room temperature by comparing the resistance in air with that in target gases. The highest sensitivity of the ZnO:Ni sensor to $CH_4$ gas and $CH_3CH_2CH_3$ gas was observed at Ni 4 wt%. The response and recovery times of 4 wt% Ni coated ZnO:Ni gas sensors were 14 s and 15 s, respectively.

The Patterns of CH4 and N2O fluxes from used Litter Stockpile from Korean Native Cattle (Hanwoo) (사용한 한우 깔짚에서 배출되는 CH4 및 N2O의 배출 특성)

  • This study was conducted to measure methane ($CH_4$) and nitrous oxide ($N_2O$) emissions from the 6 month old litter stockpile used for korean native cattle (Hanwoo) from August 3, 2007 to October 4, 2007. Daily mean $CH_4$ emissions was peaked to 273.013 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}1.047{\mu}g\;m^{-2}\;s^{-1}$) on first day and then gradually decreased to 2.309 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.061{\mu}g\;m^{-2}\;s^{-1}$) at the end of this experiment. Daily mean $N_2O$ emissions was as little as 0.269 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.018{\mu}g\;m^{-2}\;s^{-1}$) on first day, but exponentially increased up to 3.569 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.454{\mu}g\;m^{-2}\;s^{-1}$) on 43rd day and then slowly decreased to 1.888 ${\mu}g\;m^{-2}\;s^{-1}$ (SE : ${\pm}0.012{\mu}g\;m^{-2}\;s^{-1}$) at the end of this experiment. Carbon dioxide equivalent ($CO_2$-eq), calculated by global warming potentials of $CH_4$ or $N_2O$, of $CH_4$ on first day occupied approximately 99% of sum of $CO_2$-eq of $CH_4$ and $N_2O$. Methane emissions decreased and $N_2O$ emissions increased so that $CO_2$-eq ratio of $CH_4$ to $N_2O$ was 50:50 on 34th day. The effect of $N_2O$ on the ratio was increase thereafter. The ratio of daily mean $CH_4$ and $N_2O$ emissions to daily error of the mean was calculated to find daily fluctuation of $CH_4$ and $N_2O$ emissions. The ratio of $CH_4$ was less than 1.0% till 11th day but increased to 10.9% on 57th day. The ratio of $N_2O$ (0.4%~51.0%) was higher than that of $CH_4$, showing high in early stage and then gradually decrease, which was different from the pattern of $CH_4$. The ratio of daily mean emissions to daily error of the mean was little in case of active $CH_4$ or $N_2O$ generation period, which would be caused by the temporal and spatial heterogeneity of composting process. Hence more air supply on early stage to decrease $CH_4$ generation and proper turning to reduce spatial heterogeneity are needed to mitigate greenhouse gas emissions.

  • The electrochemical behavior of N-methyl-2-amino-l-cyclopentene-l-dithiocarboxylic acid $(N-CH_3 acdc) and 2-amino-l-cyclopentene-l-dithiomethyl ester (S-CH_3 acdc) in DMF have been investigated by the use of polarography, cyclic voltammetry and coulometry. The dimer of N-CH_3 acdc is further oxidized at +0.98 V via 2-electron process to produce free sulfur atom and cyclization product. The ring formation between two dithio group occurs along with the elimination of one sulfur atom. The elimination of sulfur atom occurs via two electron oxidation process at + 0.98 V vs. Ag/AgCl electrode. However, the cyclization does not occur in the S-CH_3$ acdc.

Carboxy-terminus truncations of Bacillus licheniformis SK-1 CHI72 with distinct substrate specificity

  • Bacillus licheniformis SK-1 naturally produces chitinase 72 (CHI72) with two truncation derivatives at the C-terminus, one with deletion of the chitin binding domain (ChBD), and the other with deletions of both fibronectin type III domain (FnIIID) and ChBD. We constructed deletions mutants of CHI72 with deletion of ChBD (CHI72${\Delta}$ChBD) and deletions of both FnIIID and ChBD (CHI72${\Delta}$FnIIID${\Delta}$ChBD), and studied their activity on soluble, amorphous and crystalline substrates. Interestingly, when equivalent amount of specific activity of each enzyme on soluble substrate was used, the product yield from CHI72-${\Delta}$ChBD and CHI72${\Delta}$FnIIID${\Delta}$ChBD on colloidal chitin was 2.5 and 1.6 fold higher than CHI72, respectively. In contrast, the product yield from CHI72${\Delta}$ChBD and CHI72${\Delta}$FnIIID-${\Delta}$ChBD on ${\beta}$-chitin reduced to 0.7 and 0.5 fold of CHI72, respectively. These results suggest that CHI72 can modulate its substrate specificities through truncations of the functional domains at the C-terminus, producing a mixture of enzymes with elevated efficiency of hydrolysis.

Synthesis and Characterization of Various Di-N-Functionalized Tetraaza Macrocyclic Copper(II) Complexes

  • Two copper(II) complexes, [CuL3](ClO4)2 bearing one N-CH2CH2CONH2 group as well as one N-CH2CH2CN group and [CuL4](ClO4)2 bearing two N-CH2CH2CONH2 groups, have been prepared by the selective hydrolysis of [CuL2](ClO4)2 (L2 = C-meso-1,8-bis(cyanoethyl)-5,5,7,12,12,14-hexamethyl-1,4,8,11-tetraazacyclotetradecane). The complex [CuL5](ClO4)2 bearing one N-CH2CH2C(=NH)OCH3 and one N-CH2CH2CN groups has been prepared as the major product from the reaction of [CuL2](ClO4)2 with methanol in the presence of triethylamine. In acidic aqueous solution, the N-CH2CH2C(=NH)OCH3 group of [CuL5](ClO4)2 undergoes hydrolysis to yield [CuL6](ClO4)2 bearing both N-CH2CH2COOCH3 and N-CH2CH2CN groups. The crystal structure of [CuL5](ClO4)2 shows that the complex has a slightly distorted square-pyramidal coordination polyhedron with an apical Cu-N (N-CH2CH2C(=NH)OCH3 group) bond. The apical Cu-N bond distance (2.269(3) A) is ca. 0.06 A longer than the apical Cu-O (N-CH2CH2CONH2 group) bond of [CuL4](ClO4)2. The pendant amide group of [CuL3](ClO4)2 is involved in coordination. The carboxylic ester group of [CuL6](ClO4)2 is also coordinated to the metal ion in various solvents but is removed from the coordination sphere in the solid state.

  • Methanolysis rate constants were determined for $CH_3O(CO)Cl,\;CH_3S(CO)Cl\;and\;CH_3S(CS)Cl\;in\;CH_3OH-CH_3CN$ mixtures. Results show that the rates are not predominantly influenced by the bulk solvent properties but are partly influenced by specific electrophilic solvation.Polarity of the solvent is not a dominant factor but it nevertheless plays a role in charge stabilization of the $S_N1$ like transition state. The methanolysis proceeds through $S_N1$ mechanism for $CH_3S(CS)Cl$ for which both specific solvation of leaving group by methanol and charge stabilization by a high dielectric medium are important, while for $CH_3O(CO)Cl\;methanolysis occurs\;via\;S_N2$ mechanism in which both of the solvent effects are unimportant.

  • The Wittig-oxy-Cope rearrangements of deprotonated diallyl ether, I, $CH_2={\bar{C}}H-CH-O-CH_2-CH=CH_2$, have been investigated theoretically by the AM1 method. A two step mechanism forming a Wittig product ion, II, $(CH_2=CH)$ $(CH_2=CH-CH_2)$ $CHO^-$, through a radical-pair intermediate was found to provide the most favored reaction pathway in the Wittig rearrangement. The subsequent oxy-Cope rearrangement from species II also involves a two step mechanism through a biradicaloid intermediate. The intramolecular proton transfer in I (to form $CH_2=CH-CH_2-O-{\bar{C}}H-CH=CH_2$) is a higher activation energy barrier process compared to the Wittig and oxy-Cope rearrangements and is considered to be insignificant. These results are in good agreement with the condensed-phase as well as gas-phase experimental results.

Photodissociation of Methane at Lyman Alpha (121.6 nm)

  • Laser induced fluorescence studies of hydrogen atom using four wave mixing technique are reported for the photodissociation of CH4 and its isotopomers at Lya (121.6 nm). The source of dissociating and probe radiation is one and the same (delay time??20 nsec). The average translational energy of ejected hydrogen atoms (50 Kcal/mol) reveals that CH4 + hn??CH3 + H(2S) and CH4 + hn??CH2(a1A1) + H2(1Sg) are the main dissociation processes. The absolute quantum yield for CH4 and CD4 are the same, FH(CH4) = FD(CD4) = 0.31 0.05. If one divides the experimental H/D ratios from the isotopomers CH3D, CH2D2, CHD3 by the isotopic H/D ratios, a value 2 is obtained in all three cases. Overall, the heavier D atoms are more likely than the H atoms to remain attached to the carbon atom.
