Morphological Variations, Genetic Diversity and Pathogenicity of Colletotrichum species Causing Grape Ripe Rot in Korea

  • Ripe rot was frequently observed on fruits, leaves and stems of grape growing in eight locations in Korea from 2004 to 2006. All 30 isolates of Colletotrichum sp. were obtained from lesions of the ripe rot on grape plants. Out of the isolates, 19 isolates were identified as Colletotrichum acutatum and the others as Colletotrichum gloeosporioides based on morphological and cultural characteristics. Inter and intra specific variations of the Colletotrichum spp. isolates were investigated using RAPD and sequences of rDNA ITS and $\beta$-tubulin-2. Isolates of C. acutatum and C. gloeosporioides were distinctly differentiated by molecular analyses. Phylogenetic trees of ITS and$\beta$-tubulin-2 showed that Korean isolates of C. acutatum were clustered into groups A2 and A3 among the eight global groups. A2 included non-chromogenic isolates and A3 chromogenic ones. Both C. acutatum and C. gloeosporioides isolates were tested for pathogenicity to grape leaves. All isolates tested induced lesions on the leaves of grape by artificial inoculation. There was no difference in pathogenicity between C. acutatum and C. gloeosporioides isolates. This is the first report that C. acutatum except C. gloeosporioides causes grape ripe rot in Korea.

Inheritance of Anthracnose Resistance in a New Genetic Resource, Capsicum baccatum PI594137

  • Pepper fruit anthracnose caused by Colletotrichum species is an economically important disease that causes serious yield loss and quality deterioration in many Asian countries including South Korea and Taiwan. Recently, Capsicum baccatum PI594137 was found to exhibit broad-spectrum resistance to Colletotrichum acutatum. The inheritance of anthracnose resistance to C. acutatum was analyzed in an intraspecific population derived from a cross between C. baccatum Golden-aji and PI594137. Detached mature green fruits were inoculated using the microinjection method. The disease response was evaluated as the disease incidence and the overall lesion diameter at 7 days after inoculation(DAI). The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_s$ populations significantly fit a 3:1 Mendelian model. This result indicates that the resistance of PI594137 to C. acutatum is controlled by a single dominant gene.

Safflower (Carthamus tinctorius L.) Anthracnose Casued by Colletotrichum acutatum in Korea (Colletotrichum acutatum에 관한 의한 탄저병)

  • A destructive anthracnose of safflower has severely occurred at Kyeongsangnam-do Agricultural Research and Extention Services in 1998. Incidence of the disease surveyed at 12 fields in Chinju, Sanchong and Hamyang was ranged fro 55.7 to 85.4%. Small irregular yellowish to brown spots appeared on all parts of the plant at initial infection stage and the spots enlarged or united into larger irregular to cylindrical black lesions with hollows. Above portions of infected stems were often blighted and pink colored conidial masses were colonized on the lesions. The causal fungus consistently isolated from the infected tissues were identified as Colletotrichum acutatum by following characteristics. The fungus grew well on PDA at 15~3$0^{\circ}C$ developing apricot to salmon color. Conidia were single-celled, colorless, fusiform and bisectioned and measured as 8~18$\times$3~5 ${\mu}{\textrm}{m}$. Brownish appressoria were cylindrical or ovate and sized as 5~10$\times$4~8 ${\mu}{\textrm}{m}$. Optimum temperature for growth and germination of conidia was recorded at $25^{\circ}C$ and at least 6 hours were required for infection to the host plants. Over 90% conidia were germinated and all plants were infected by artificial inoculation by 24 hours at $25^{\circ}C$. This is the first report of safflower anthracnose caused by C. acutatum in Korea.

  • The strains of Colletotrichum gloeosporioides, C. coccodes, C. acutatum isolated from black raspberry were pathogenic to apple and strawberry after dropping inoculation, but showed weak pathogenicity in hot-pepper and tomato. The anthracnose pathogens of C. gloeosporioides, C. orbiculare, C. acutatum isolated from apple, hot-pepper and pumpkin showed pathogenicity in black raspberry. Moreover, the anthracnose pathogens isolated from apple caused disease symptoms in non-wounded inoculation.

Response to Fungicides of Colletotrichum Species Isolated from Infected Tissues of Several Crops (다양한 작물로부터 분리한 탄저병균(Colletotrichum spp.)의 살균제에 대한 반응)

  • This study has been conducted to investigate the responses of various isolates of four Colletotrichum species such as C. gloeosporioides, C acutatum, C. coccodes, and C. dematium isolated from infected tissues of several crops to fungicides such as carbendazim, carbendazim+diethofencarb, four protective fungicides, and three ergosterol biosynthesis-inhibiting (EBI) fungicides. All the isolates of C. acutatum showed $EC_{50}$ values in a range of 0.001-3.040 ${\mu}g/ml$ against carbandazim, a benzimidazole fungicide. As for the response to carbendazim, the isolates of C. gloeosporioides obtained from pepper, apple, and strawberry were clearly divided into two groups, resistant or sensitive isolates. All the resistant isolates showed $EC_{50}$ values above 1000 ${\mu}g/ml$, whereas the sensitive isolates had lower $EC_{50}$ values than 0.550 ${\mu}g/ml$. The isolates of C. gloeosporioides exhibited a negative cross resistance between carbendazim and diethofencarb (a N-phenylcarbamate fungicide), but isolates of C. acutatum did not. Toward carbendazim, C. coccodes and C. dematium isolates showed a similar response to C. acutatum isolates and the sensitive isolates of C. gioeosporioides, respectively. As for response of Protective fungicides, all the isolates of C. acutatum showed a more resistant reaction than all the isolates of C. gloeosporioides. However, there was no difference among 4 species of Colletotrichum against EBI fungicides.

Anthracnose of Salvia plebeia Caused by Colletotrichum acutatum in Korea (Colletotrichum acutatum에 의한 배암차즈기 탄저병)

  • Anthracnose of Salvia plebeia caused by Colletotrichum acutatum was observed in the exhibition field of Hamyang-gun, Gyeongsangnam-do Agricultural Research and Extension Services, in Korea. The disease mainly occurred on the leaves, petioles and stems of whole plant. The typical symptoms were water-soaked lesions on stems and petioles, rotted stems and withering then eventually died. The color of colonies of the pathogenic fungus grown on potato dextrose agar was pinkish gray. Conidia were fusiform in shape and was $8{\sim}17{\times}3{\sim}4\;{\mu}m$ in size. Appressoria were pale to dark brown in color, clavate in shape and the sizes were $8{\sim}11{\times}4{\sim}6\;{\mu}m$. Optimum temperature for mycelial growth was $25^{\circ}C$. On the basis of mycological characteristics and pathogenicity test on host plants, the fungus was identified as Colletotrichum acutatum J. H. Simmonds. This is the first report on the anthracnose of S. plebeia caused by C. acutatum in Korea.

Anthracnose of Pomegranate Caused by Colletotrichum acutatum in Korea (Colletotrichum acutatum에 의한 석류나무 탄저병)

  • Anthracnose of pomegranate (Punica granatum) caused by Colletotrichum acutatum was observed in the exhibition field at Hamyang-gun, Gyeongsangnam-do Agricultural Research and Extension Services, and in farmer's orchard at Gaheo-myon, Hapcheon-gun, Gyeongnam province, Korea. The pathogen generally attacks on the fruits and induces dark lesion and rotting. The infected fruits were mostly dropped or mummified. The color of colonies of the pathogenic fungus grown on potato dextrose agar was pinkish gray. Conidia were fusiform in shape and was $8{\sim}16{\times}3{\sim}4{\mu}m$ in size. Appressoria were pale to dark brown in color, clavate in shape and was $8{\sim}12{\times}4{\sim}6{\mu}m$ in size. Optimum temperature for mycelial growth was $25^{\circ}C$. Pathogenicity of the causal organism was proved according to Koch's postulate. On the basis of mycological characteristics and pathogenicity test on fruit, the fungus was identified as Colletotrichum acutatum. This is the first report on the anthracnose of pomegranate caused by C. acutatum in Korea.

Agrobacterium tumefaciens-mediated Transformation in Colletotrichum falcatum and C. acutatum

  • Agrobacterum tumefaciens-mediated transformation (ATMT) is becoming an effective system as an insertional mutagenesis tool in filamentous fungi. We developed and optimized ATMT for two Colletotrichum species, C. falcatum and C. acutatum, which are the causal agents of sugarcane red rot and pepper anthracnose, respectively. A. tumefaciens strain SK1044, carrying a hygromycin phosphotransferase gene (hph) and a green fluorescent protein (GFP) gene, was used to transform the conidia of these two Colletotrichum species. Transformation efficiency was correlated with co-cultivation time and bacterial cell concentration and was higher in C. falcatum than in C. acutatum. Southern blot analysis indicated that about 65% of the transformants had a single copy of the T-DNA in both C. falcatum and C. acutatum and that T-DNA integrated randomly in both fungal genomes. T-DNA insertions were identified in transformants through thermal asymmetrical interlaced PCR (TAIL-PCR) followed by sequencing. Our results suggested that ATMT can be used as a molecular tool to identify and characterize pathogenicity-related genes in these two economically important Colletotrichum species.

Studies on the Seed Transmission of Colletotrichum spp. in Red Pepper (Capsicum annuum) (고추 탄저병균(炭疽病菌)의 종자전염(種子傳染)에 관(關)한 연구(硏究))

  • Colletotrichum acutatum, C. coccodes, C. dematium and C. gloeosporioides were detected in seed samples of red pepper (Capsicum annuum). C. dematium and C. gloeosporioides were the predominant species, maximum seed infection of the species in some samples were 84% and 28%, respectively. C. acutatum and C. coccodes were recorded only in low percentages of 1-2. The blotter method proved more suitable for detecting Colletotricum spp. than the deep freezing blotter or agar plate methods. Plating of seed components showed that C. dematium and C. gloeosporioides were recovered more frequently from seed coat, and decreasing amounts of infection were observed in the endosperm and cotyledon. Seed-borne C. dematium and C. gloeosporioides caused seed rot, damping-off, seedling blight and brown discoloration of cotyledon and hypocotyl when infected seeds were sown in agar of test tube or in soil. Inoculation experiments showed that C. acutatum was pathogenic to red fruit of red pepper and C. coccodes was highly pathogenic to red fruit and weakly pathogenic to leaf of the plant. C. dematium was highly pathogenic to leaf and green fruit and C. gloeosporioides was pathogenic to not only leaf but also green and red fruits. Host range of the four seed-borne species of Colletotrichum was also investigated.

Species identification and pathogenicity study of Colletotrichum isolates isolated from red-pepper and Chinese matrimony vine

  • This study reports the identification of species of Colletotrichum strains originating from red-pepper and Chinese matrimony vine in Cheongyang. Ninteen isolates of red-pepper and 26 Coiletotrichum isolates of Chinese matrimony vine were compared with 5 isolates of strawberry representing C. gloeosporioides, by use of morphological and cultural criteria. Twenty three isolates among 26 isolates from Chinese matrimony vine were identified as C. acutatum, characterized by the low growth rates and the low sensitivity to carbendazim and diethofencarb. Also, all the isolates of red-pepper were identified as C. acutatum, showing the same characteristics as those of Chinese matrimony vine. Three and five isolates from Chinese matrimony vine and strawberry, respectively, were identified as C. gloeosporioides, characterized by the high growth rates and the high seneitivity to carbendazim and diethofencarb. There were differences in colony color and pathogenicity between Chinese matrimony vine isolates and red-pepper isolates of C. autatum. The isolates of C. acutatum from Chinese matrimony vine producing orange colored colonies with abundant spores showed the strong pathogenicity to Chinese matrimony vine, although they could not infect fruits of red-pepper by the wound inoculation. However, red-pepper isolates of C. acutatum producing gray colonies showed the strong pathogenicity to Chinese matrimony vine as well as red-pepper. Furthermore, comparative study on PCR amplification of ITS regions of rDNA was carried out using a number of Colletotrichum isolates. A species-specific primer could be used for the identification of C. acutatum from red-pepper and Chinese matrimony vine.
