The purpose of this study was the discrimination of different tobacco types by the E-Nose/tongue and the analysis of what human sensory attributes are correlated with e-instrument's sensors. Samples were made from five groups of Korean domestic tobacco leaves, aged burley and not aged, aged flue-cured and not aged and blending types of the four. Instrumental tests were conducted to discriminate characteristics among different tobacco samples by the E-Nose and the E-Tongue. Sensory attributes of tobacco tastes were impact, irritation, bitterness, hay-like, tobacco taste, smoke volume, smoke pungent and mouth cleanness. STATISTICA software was used to analyze correlation between the human sensory data and the raw data of e-instruments. Discrimination analysis can be achieved using principal components analysis (PCA) and discriminant factorial analysis(DFA). As a result, impact, bitterness, irritation, smoke volume and smoke pungent of human sensory attributes were correlated with data from the several clustered E-Nose sensors(p < 0.10). And bitterness, irritation, and smoke pungent of human sensory attributes were correlated with data from the E-Tongue sensors(p < 0.10). PCA plot by the E-Nose shows that aged tobacco and not aged were discriminated and DFA plot shows that three groups(aged burley, not aged burley and flue-cured) were discriminated. PCA plot by the E- Tongue shows that flue-cured tobacco was separated from burley. Our results indicated that the e-instruments are sensitive enough to distinguish among tobacco types and their several sensors are reacted to the human sensory attributes.
In order to obtain the basic information for the most successful breeding of Burley tobacco, these studies were performed on the correlations among agronomic characters and on the heterosis and combining ability in $F_1$ generation. Heterosis ratio was highly variable in different $F_1$ combinations. Positive heterosis was observed in all characters except days to flowering and yield showed the highest heterosis ratio. As the results of analysis of dialled table, the general combining ability (GCA) in all characters was shown to be significant . All characters except the number of leaves in the specific combining ability (SCA) were significant, Variety, L8, contributed negative GCA for all characters. Burley 21, ky 10 and ky 14 contributed negative GCA for leaf width, quality and yield, respectively. Hybrid of ky 10 x ky 14 showed negative SCA and hybrids of ky I0x L8 and ky 14 x L8 showed positive SCA for all characters. There were significant correlations between yield and number of leaves, leaf length, leaf width or leaf area. Correlation between yield and days to flowering was non- significant .
This experiment was conducted to investigate varietal responses to cool temperature (18℃) and short photoperiod (8 hours a day) in a controlled room of phytotron. Five burley tobacco varieties (Nicotiana tabacum L.), Awelius (very early flowering type), Burley 21 and KB 103 (medium), Banket Al and MB 1 (slightly late) and TC 681 (late) were compared in number of leaves per plant and day to flower in different treatment periods (10, 15, 20, 25 and 30 days). The days to flower decreased significantly than the control (in the phytoron with 14hr photoperiods and a minimum temperature 20'c) when the treatment periods were over 15 days for the varieties Aurelius, Burley 21 and KB 103, and 20 days for TC 681, and 30 days for Banket Al, respectively. And also the number of leaves decreased significantly than that of the control when the periods of treatment were over 10 days for Burley 21 and Aurelius, 15 days form 103, and 20 days for TC 681 and Banket Al, and 30 days for MB 1, respectively. The decreasing ratio in the number of leaves per plant under cool temperature and short photoperiod treatments was the lowest in Banket Al and MB 1. To obtain the optimum condition for analysis of 10 alkaloids in tobacco leaves, such as nicotine, nornicotine, anatabine, anabasine, myosmine, cotinine, 2,3'-dipyridyl, $\beta-nicotyrine,\;\beta-nornicotyrine\;and\;\beta-formylnornicotin$, 5 types of extraction method were investigated by GC-FID and GC/MS. The optimum condition of alkaloid extraction was achieved by using methanol:dichloromethane(1:3, v/v) after NaOH treatment. The use of mass selective detector (MSD) provided unambiguous nicotine related alkaloid analysis. Alkaloids in various tobacco leaves were extracted with the optimum extraction condition and quantified by GC/MS/SIM mode. Compared with concentrations of alkaloids among the various tobacco leaves, the concentration of alkaloids was generally in the order burley > flue-cured > oriental tobacco. In flue-cured tobacco leaves, the order of concentration of alkaloids was nicotine > anatabine > nornicotine > $\beta-nicotyrine\;>\;\beta-formylnornicotine\; >\;myosmine\;>\;2,3'-dipyridyl\;>\;cotinine\;>\;anabasine\;>\;\beta-nornicotyrine$. However, in the case of burley and oriental tobacco leaves, the concentration of nornicotine was higher than that of anatabine.
The effect of mineral oil treatment in Burley 21 tobacco field on the control of potato virus Y(PVY-VN) mostly transmitted by green peach apid(Myzus persicae Sulzer) in nature was studied and the virus infection in some plants including potato, pepper, bramble, radish, etc near the tobacco fields as a virus infection source was tested by capillary tube precipitatioin test with PVY-antibody and bioassay in Xanthi-nc tobacco. The main source of PVY-VN infection in tobacco field in korea was potato(ca. 40% of test plants infected). Pepper and bramble were also infected by PVY-VN. The control level of PVY-VN infection by treatment of 0.75% liquid mineral oil with 3 % nonionic emulsifier to the plants was 84.8 % in case of the artificial transfection with a infected apterous aphid in laboratory. However, the reduction of PVY-VN disease severity in tobacco fields treated with mineral oil at late June was only 35.5%. These results suggest that mineral oil treatment is not so effective for the protection of aphid-born virus(PVY - VN) infection in tobacco fields.
Burley leaf tobacco extracts has been heated at loot for 2 hours and changes in non-volatile fructosazines and volatile components were investigated. Major changes for the heat treatment with corn syrup of burley leaf tobacco extracts were as follows, increases in the contents of 2,5-deoxyfructosazine and 2,5-fructosazine that is produce for the heating reaction of sugar and ammonia, production of pyraEine compounds, such as 2,6-dimethyl pyrazine,2,4-dimethyl pyrazine, ethrnyl pyrazine, methylethyl pyraxine, trimethyl pyrazine, 2-ethenyl-5-methyl pyrazine, 2-acetyl parazine and 2-acetyl-3-methyl pyrazine, increases in the content of furfuryl alcohol derived from sugar degrad ation, production of 2,5-dimethyl-4-hydroxy-3(2H)-furanone and 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one derived from thermal degradation of Amadori compounds.
This study was conducted to obtain the basic information on the burley tobacco (Nicotiana tabacum L.) curing methods. The effects of the three kinds of curing methods on the dry weight losses during curing, chemical contents and physical properties of cured leaves, and organoleptic and smoking qualities were observed. The dry weight losses during curing was the highest in the stalk curing, and the lowest in the conventional priming. The longer the curing periods was, the more the loss of dry weight. The percent of the excessive dried leaves was high and the price per kilogram was low in the conventional priming. The chemical contents and the physical properties of the cured leaves in the conventional priming were similar to those of yellowing or browning stage of curing. But, the chemical contents and the physical properties of the cured leaves in the stalk curing were comparable to those of the air curing. The smoking quality was the best in the stalk curing.
The nitrogen contents of some forms in Burley tobacco leaf cultivated in Yaesan, Mokpo and Namwon district were investigated. The rate of each form in total nitrogen contents were as follows ; Protein form nitrogen 30~33 % Nitrate form nitrogen 10~123% Alkaloid form nitrogen 8~16 % Ammonia form nitrogen 6~9% Amide form nitrogen 2~ 3% Other form nitrogen 26~44 % The order of nitrate form nitrogen content on the nitrogen of each tobacco cultivated in three area was Yaesan > Namwon > Mokpo, but that of alkaloid form nitrogen was reverse order of nitrate form nitrogen. As for Quality ( grade ), the orders of alkaloid and ammonia form nitrogen content on total nitrogen were H5> 3> 1> L 1> 3> 5, but that of nitrate form nitrogen was reversed.
The incitant of soft rot on burley tobacco leaves in the curing vinylhouse was identified as Erwinia carotovra subsp. carotovora on the basis of its physiological characters. The bacterium yew best at $25^{\circ}C$ and $30^{\circ}C$, but no growth was detectable at $40^{\circ}C$ in the nutrient broth for 24 hours period. Burley tobacco leaves inoculated with the bacteria (Ecc) produced typical soft rot lesions when the incubation temperature was 25 to $30^{\circ}C$ and the relative humidity was more than 8075, however, the lesion development was suppressed when the temperature was $40^{\circ}C$ and the relative humidity was below 80%. Significant negative correlation was found between hanging space in the curing vinylhouse and the incidence of soft rot on the tobacco leaves harvested in a rainy day regardless of streptomycin treatment.
Potato virus Y (PVY) distribution, areas where the virus occurred, and incidence, Percentage of plants infected, on burley tobacco in Korea was surveyed in 1982. Most of the fields Investigated were infected with PVY. The virus incidence was 12.5%. District)union and incidence generally were sporadic, but Onyang, Hongseung and Iksan area virus incidence was higher than that of other areas. For strain identification, approximately 95% was nonnecrotic (PVY-VB) and 5olo necrotic strain (PVY-VN) .
