가토의 신장 기능에 미치는 측뇌실내 Bethanidine의 영향 (Influence of Intraventricular Bethanidine on the Renal Function of the Rabbit)

  • Bethanidine was administered into the lateral ventricle of the rabbit brain for the investigation of the effect on the renal function in doses ranging from 0.1 to 1.0mg/kg. In a dose of 0.1 mg/kg, bethanidine did not exhibit significant changes on the renal function of the rabbit, on the other hand, in the doses of 0.3 and 1.0mg/kg bethanidine elicited the reduction of renal plasma flow and glomerular filtration rate with a marked antidiuresis, at the same time bethanidine produced the decrement of urinary sodium and potassium excretion. After intravenous pretreatment of phentolamine, intraventricular bethanidine in a dose of 0.3mg/kg did not produced the antidiuresis and the decrement of urinary sodium and potassium excretion, wherease renal plasma flow and glomerular filtration rate reduced as before of phentolamine pretreatment although the durations of their reduction were shortened. These observations suggest that bethanidine induces the antidiuresis through the centrally mediated mechanism which interposed other factors in addition to sympathetic stimulation affected by phentolamine, alpha adrenergic blocking agent.

Phototransduction and Visual Cycle in the Ascidian Tadpole Larva

  • Ascidians are lower chordates, and their tadpole-like larvae share a basic body plan with vertebrates. To study photoreceptive systems in ascidians, we have isolated and characterized cDNA clones for three opsins, five G protein ${\alpha}$ subunits (G${\alpha}$), catalytic and regulatory subunits of cGMP phosphodiesterase (PDE), and arrestin from the ascidian Ciona intestinalis tadpole larva. Ci-opsin1 and Ci-opsin2 are vertebrate-type opsins, while Ci-opsin3 is a retinal photoisomerase similar to retinochrome and mammalian RGR. Both Ci-opsin1 and arrestin are specifically localized in the photoreceptor cells of the ocellus, whereas Ci -opsin2 is not expressed in the photoreceptors, but is co-localized in another population of neurons in the brain with PDE (Ci-PDE9 and Ci-PDE$\delta$). Ci-opsin3 is present in the entire region of the brain. Though five different cDNAs encoding Ga have been cloned, no transducin-type G protein has been found yet. Interestingly, one of G${\alpha}$i isoform is conspicuously expressed in the entire region of the brain. The Ci-opsin3 gene expression was observed in a broad area of the brain vesicle as well as in the visceral ganglion. Genes encoding ascidian homologs of CRALBP and ${\beta}$-CD, whose function is required for the mammalian visual cycle, are co-expressed with Ci-opsin3 in the brain vesicle and visceral ganglion. Localization of Ci-opsin3, CRALBP, and ${\beta}$-CD in a broad area of the brain suggests that the brain of the ascidian larva has a visual cycle system similar to that of the vertebrate RPE. Based on these data, we discuss the evolution of vertebrate visual systems.

  • We previously isolated the OsCBT gene, which encodes a calmodulin (CaM)-binding protein, from a rice expression library constructed from fungal elicitor-treated rice suspension cells. In order to understand the function of OsCBT in rice, we isolated and characterized a T-DNA insertion mutant allele named oscbt-1. The oscbt-1 mutant exhibits reduced levels of OsCBT transcripts and no significant morphological changes compared to wild-type plant although the growth of the mutant is stunted. However, oscbt-1 mutants showed significant resistance to two major rice pathogens. The growth of the rice blast fungus Magnaporthe grisea, as well as the bacterial pathogen Xanthomonas oryzae pv. oryzae was significantly suppressed in oscbt-1 plants. Histochemical analysis indicated that the hypersensitive-response was induced in the oscbt-1 mutant in response to compatible strains of fungal pathogens. OsCBT expression was induced upon challenge with fungal elicitor. We also observed significant increase in the level of pathogenesis-related genes in the oscbt-1 mutant even under pathogen-free condition. Taken together, the results support an idea that OsCBT might act as a negative regulator on plant defense.

Altered Expression of RANKL/OPG after Alendronate Administration in the Developing Teeth of Postnatal Rats

  • The working mechanism of bisphosphonate on bone cells is unclear despite its powerful inhibitory activity on bone resorption. The differentiation and activation of osteoclasts are essential for bone resorption and are controlled by the stimulatory RANKL and inhibitory OPG molecules. Teeth exhibit a range of movement patterns during their eruption to establish their form and function, which inevitably accompanies peripheral bone resorption. Hence, the mandible, which contains the teeth during their eruption processes, is a good model for revealing the inhibitory mechanism of bisphosphonate upon bone resorption. In the present study, RANKL and OPG expression were examined immunohistochemically in the mandible of rats with developing teeth after alendronate administration (2.5 mg/kg). The preeruptive mandibular first molars at postnatal days 3 to 10 showed the developing stages from bell to crown. No morphological changes in tooth formation were observed after alendronate administration. The number of osteoclasts in the alveolar bone around the developing teeth decreased markedly at postnatal days 3, 7 and 10 compared with the control group. RANKL induced strong positive immunohistochemical reactions in the dental follicles and stromal cells around the mandibular first molar. In particular, many osteoclasts with strongly positive reactions to RANKL appeared above the developing mandibular first molars at postnatal days 3 and 10. Immunohistochemical reactions with RANKL after alendronate administration were weaker than the control groups. However, the immunohistochemical reactivity to OPG was stronger after alendronate administration, at postnatal days 3 and 10. These results suggest that alendronate may decrease bone resorption by regulating the RANKL/OPG pathway in the process of osteoclast formation, resulting in a delay in tooth eruption.

유산소운동이 여자 아동의 뇌세포 생성 관련인자에 미치는 영향 (Effect of Aerobic Exercise on Factors Relative to the Brain Nerve Growth in Girls)

  • 운동은 신경의 생성, 적응, 보호 기능을 통해 뇌 기능 및 생성에 긍정적인 영향을 미치며, 유산소운동은 뇌신경 성장인자(GH, IGF-1, estrogen, serotonin)를 변화시키는 것으로 보고되었다. 본 연구는 여자 아동의 뇌기능 발달 및 뇌세포 생성 유도인자의 유산소운동 효과를 알아보고자 유산소운동 프로그램에 따른 뇌신경 성장인자(GH, IGF-1, estrogen, serotonin)의 변화를 분석하여 다음과 같은 결론을 얻었다. 반복측정 분산분석에 의해 운동군과 대조군의 평균을 비교한 결과 유산소운동은 집단간 유의한 차이를 보이며 체중, BMI, 체지방량, 체지방률을 감소 시켰으며, 제지방률을 증가시켰다. 또한 뇌신경 성장인자인 serotonin을 감소시켰으며, GH와 IGF-1을 증가시켰다. 본 연구결과 유산소운동 프로그램이 아동의 체중, BMI, 체지방, 체지방률 감소와 제지방률 증가에 효과적이다. 또한 serotonin이 운동 후 감소된 결과를 볼 때 유산소운동 프로그램이 아동의 중추피로 감소에 효과적이며, GH, IGF-1의 운동 후 증가된 결과를 볼 때 유산소운동이 아동의 뇌기능 발달 및 뇌세포 생성 유도에 긍정적인 프로그램임을 알 수 있다.

허혈성 뇌졸중을 위한 뇌 자기공명영상의 의미적 특징 기반 템플릿 중심 의료 영상 매핑 기법 (Brain MRI Template-Driven Medical Images Mapping Method Based on Semantic Features for Ischemic Stroke)

  • 허혈성 뇌졸중은 뇌혈관의 혈전이나 색전에 의해 뇌 혈류가 감소하게 되어 뇌 조직이 기능을 못하는 질환으로, 질환의 특성상 뇌혈관의 폐색 여부를 확인하는 것이 중요하기 때문에 질환의 진단에 있어서 의료 영상이 필수적으로 활용된다. 그 중에서도 뇌 자기공명영상은 뇌의 구조적인 정보들을 얻을 수 있어 질환을 진단하는데 그 지표로 널리 활용되고 있다. 하지만 허혈성 뇌졸중과 같은 응급 질환의 경우 빠른 진단과 처치에 도움이 될 수 있는 지능적인 시스템이 요구됨에 비해, 기존의 의료 영상 저장 시스템으로는 신속하고 직관적인 영상 정보 제공이 어렵다. 즉, 기존의 시스템은 피상적인 메타 데이터를 이용하여 의료 영상을 관리하고 있어 의료 영상에 내재된 주요 의미적 정보를 고려하지 못하고 있다. 따라서 본 논문에서는 뇌 자기공명영상이 내포하고 있는 주요 의미적인 정보인 뇌의 해부학적 구조와 같은 영상 정보를 제공할 수 있도록 하는 템플릿 중심의 영상 매핑 기법을 제안하고자 한다. 제안하는 기법은 방대한 양의 영상을 대표할 수 있는 대표 영상(템플릿)을 선정하여 의미적 특징과 대표 영상(템플릿) 사이의 대응성을 정립하고, 전문가(의사)에 의해서만 분석될 수 있는 영상 사이의 의미적 연관성을 표면화 시켜 의미 기반의 영상 관리를 가능케 한다.

Transcallosal Ipsilateral Motor Pathway from the Unaffected Hemisphere in a Patient with Traumatic Brain Injury

  • Purpose: We presented with a patient with traumatic brain injury who showed a transcallosal ipsilateral motor pathway from unaffected motor cortex to affected extremities, as evaluated by diffusion tensor tractography (DTT). Methods: One patient and six age-matched normal subjects were enrolled. A 42-year-old left hemiparetic male who suffered from brain injury by motor vehicle accident 9 years ago showed a leukomalactic lesion in the right corona radiata and parieto-temporal lobe. His left extremities were completely paralyzed initially, but recovered slowly over 2 years. At the time of the evaluation, he was able to grasp and release an object, and to walk with spastic gait pattern. DTT was performed using 1.5 T with a Synergy-L Sensitivity Encoding head coil. DTT was obtained with termination criteria of FA <0.2 and an angle change > $45^{\circ}c$. Results: The motor tracts of the unaffected (left) hemisphere of the patient and control subjects originated from the motor cortex and descended along the known corticospinal tract without any transcallosal tract. By contrast, the tract of the affected (right) hemisphere originated from the left premotor cortex, descended through the left corona radiata, and then crossed the mid-portion of the corpus callosum. The tract then descended through the known corticospinal tract pathway to the right medulla. Conclusion: We conclude that the transcallosal ipsilateral motor pathway from the unaffected hemisphere appeared to contribute to the motor recovery in this patient.

약물치료 중인 주의력결핍 과잉행동장애 아동 청소년에서 작업기억력 훈련이 임상증상과 신경심리소견에 미치는 부가적 효과 (Augmentative Effects of Working Memory Training on Clinical Symptoms and Neuropsychology in Medicated Children and Adolescents with Attention-Deficit Hyperactivity Disorder)

  • Objectives: Executive dysfunction including working memory deficit has been suggested to be one of the major neuropsychological etiologies of attention-deficit hyperactivity disorder (ADHD). The purpose of this study was to investigate the augmentative effects of Cogmed working memory training on the symptoms and neurocognitive functions in medicated children and adolescents with ADHD. Methods: Twenty-five children with ADHD, aged 7 to 19 years, taking ADHD medication participated in this study. The participants were trained for 5 weeks with a commercially available and computerized working memory program ($Cogmed^{(R)}$) without any changes to their medication. The Korean version of the ADHD Rating Scale, Clinical Global Impression Scale, and Comprehensive Attention Test were administered before training and 4 weeks and 7 months after training, respectively. Results: After completing the training, the clinical symptoms and function, rated by the parents and clinician, were improved. In addition, the level of commission errors was significantly reduced in the selective attention (visual/auditory) task, sustained attention to response task, and flanker task. The untrained visuospatial short-term memory and working memory were also improved. These effects were still observed 7 months after the training. Conclusion: Cogmed working memory training can be a promising training option for the additional improvement of the symptoms and deficits in working memory and response inhibition in medicated children with ADHD.

인간 태아의 뇌로부터 만들어진 cDNA library에서 내생 레트로바이러스 HERV-W LTR의 클로닝 및 분자계통분류 (Molecular Cloning and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in cDNA Library of Human Fetal Brain)

  • Long terminal repeats(LTRs) of the human endogenous retrovirus(HERV) heve been found to be coexpresed with genes located nearby. It has been suggested that the LTR elements have contributed to the genetic variation of human genome connected to various diseases. Recently, HERV-W family was identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using cHNA library derived from human fetal brain, we performed PCR amplification and identified seven new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity(98∼99%) with HERV-W (AF072500). A phylogentic tree obtained by the neighbor-joining method revealed that seven new HERV-W LTR elements(FB-1, 2, 4, 8, 9, 10, 12) were closely related to the AX000960, AF072504, and AF072506 from Gen Bank database. Our data suggest that several copy numbers of the HERV-W LTR elements are expressed in human feta brain and may contribute to an understanding of biological function connected to neuropsychiatric diseases.

Effects of Rice Bran Extracts Fermented with Lactobacillus plantarum on Neuroprotection and Cognitive Improvement in a Rat Model of Ischemic Brain Injury

  • This work aimed to study whether rice bran extract fermented with Lactobacillus plantarum (LW) promotes functional recovery and reduces cognitive impairment after ischemic brain injury. Ischemic brain injury was induced by middle cerebral artery occlusion (MCAO) in rats. Four groups were studied, namely the (1) sham, (2) vehicle, (3) donepezil, and (4) LW groups. Animals were injected with LW once a day for 7 days after middle cerebral artery occlusion. LW group showed significantly improved neurological function as compared to the vehicle group, as well as enhanced learning and memory in the Morris water maze. The LW group showed the greatest functional recovery. Moreover, the LW group showed an enhanced more survival cells anti-apoptotic effect in the cortex and neural cell densities in the hippocampal DG and CA1. In addition, this group showed enhanced expression of neurotrophic factors, antioxidant genes, and the acetylcholine receptor gene, as well as synaptophysin (SYP), Fox-3 (NeuN), doublecortin (DCX), and choline acetyltransferase (ChAT) proteins. Our findings indicate that LW treatment showed the largest effects in functional recovery and cognitive improvement after ischemic brain injury through stimulation of the acetylcholine receptor, antioxidant genes, neurotrophic factors, and expression of NeuN, SYP, DCX, and ChAT.
