Fractionation and Electrophoretic Patterns of Rice Proteins (쌀단백질의 분획 및 전기영동)

  • The composition of four rice protein groups is greatly affected by the extraction conditions. The extraction amounts of albumins and glubulines primarily depended on the temperature rather than the method of extraction. The total amount of glutelins, the major components of rice storage proteins, could be extracted by a successive extraction processes, extraction with 0.5% SDS-0.1M borate buffer(pH 8.3) followed by extraction with 0.5% SDS-0.6% ${\beta}-mercaptoethanol-0.1M$ borate buffer(pH 8.3). The extracted amounts of glutelin with these solvents were 54.1 and 45% respectively. The further purification of SDS soluble glutelins was achieved by Sephadex G-150 gel column chromatography. The molecular weight of the components in four protein groups has been estimated by SDS-polyacrylamide gel electrophoresis with or without ${\beta}-mercaptoethanol.$ The comparison of albumins and globulins by starch gel electrophoresis at pH 3.1 permitted us to identify seven rice varieties. However, at pH 8.95, the specific bands for Japonica type rice varieties were observed.

  • In this research, isothermal conduction calorimetry analysis has been conducted to investigate the reactivity of alkali activated slag binders. In order to secure the reactivity and workability of alkali activated slag binders, experiences with various types and concentrations of alkali activators were performed. Isothermal conduction calorimetry were measured with different alkali activators and mass ratio of $SO_3$ to binders as variables, and sodium tripolyphosphate ($Na_2P_3O_{10}$) and hydrated sodium borate ($Na_2B_4O_710H_2O$) were used to control setting time. As a results, alkali activated slag binders required alkali activators with 4 to 5 percent of concentration to accelerate the formation of calcium silicate hydrate(C-S-H) by alkali-activation, and overall heat generation rate delayed as accumulated heat decreased due to the high $SO_3$ contents. Moreover, the use of hydrated sodium borate as setting retarder causes elongated setting time due to delaying heat generation, so it can be considered that setting retarder played an important role in delaying total heat generation rate.

Analysis on Chemical Ingredients with Anti-microbial Activity in Water-based Metalworking Fluids

  • This study was conducted to estimate if the level of several chemical ingredients including alkanolamines or ethanolamines (EA) examined in the specific synthetic metalworking fluid (MWF) ‘A’ can cause anti-microbial activity and health effect. Three water-based MWF products (‘A’, ‘B’, and ‘C’) were studied every week for two months. Chemical ingredients such as formaldehyde, boron, EA, and copper were examined. In the sump where MWF ‘A’ was used, not only the total level of EA, monoethanolamine(MEA), diethanolamine(DEA) and triethanolamine(TEA), but also boron level were significantly higher than those of the other MWFs. ANOVA statistical tests indicated that levels of pH, alkalinity, boron, MEA, DEA and TEA in MWF ‘A’ were significantly higher than those in other MWF types. Correlation tests also found that levels of pH, alkalinity, boron, MEA, DEA and TEA in MWF ‘A’ are significantly correlated. We suggested the assumptions that excessive concentrations of EA, and borate at a high pH level, may cause anti-microbial resistance synergically. To demonstrate this assumption, additional study is needed to examine the relationship between the levels of microbes and excessive concentrations of EA, and borate at a high pH level.

  • Among various CE separation methods, micellar electrokinetic capillary chromatography(MECC) method using sodium dodecylsulfate(SDS) provides rapid and accurate separation of organic explosive constituents with easy. The running buffer was composed with 2.5 mM borate and 25mM SDS(pH 8.5). Addition of 1M urea and 10% organic modifiers (acetonitrile, methanol and ethanol) improves the resolution of adjacent explosive constituents. When 15 explosive constituents were developed in MECC, most constituents were separated successively while RDX/TNB and DNN/DEP were not, and detection limits of separated compounds are in range of 1 to 4 ppm.

  • A borate compound was adopted as new host material for $Eu^{3+}$, $Tb^{3+}$ and $Tm^{3+}$ activators. The phosphor samples, $Gd_{1-x}Eu_xCa_3(GaO)_3(BO_3)_4$, $Gd_{1-x}Tb_xCa_3(GaO)_3(BO3)_4$ and $Gd_{1-x}Tm_xCa_3(GaO)_3(BO_3)_4$ have been synthesized by conventional solid-state reaction. The crystalline phase for the resulting powders was identified using an X-ray diffraction $system^1$. Their photoluminescence properties under the excitation of UV ray were investigated. The Eu, Tb or Tm-doped $GdCa_3(GaO)_3(BO_3)_4$ emits efficient red, green or blue light, respectively. It was observed that the optimum concentration of Eu or Tb activator for the borate host was much higher than other $Eu^{3+}$ or $Tb^{3+}-doped$ phosphors.
