Biochemical Methane Potential of Animal Manure and Cultivated Forage Crops at the Reclaimed Tideland (가축분뇨와 간척지 사료작물의 메탄발생량)

  • Anaerobic biodegradability(AB), which can be determined with the ultimate methane yield by the decomposition of organic materials, is one of the important parameters for the design and the operation of anaerobic digestion plant. In this study, Biochemical Methane Potential(BMP) test has been carried out to evaluate the methane yield of animal manures, such as pig and cattle slurries, and different forage crops cultivated at the reclaimed tideland, such as maize, sorghum, barley, rye, Italian ryegrass(IRG), rape, rush and sludge produced from slaughterhouse wastewater treatment plant(SWTP). In the ultimate methane yield of animal manure, that of pig slurry(no used a EM) was 407 $mlCH_4/gVS_{fed}$ higher than 242 $mlCH_4/gVS_{fed}$ of cattle slurry. The ultimate methane yield of spike-crop rye was 442.36 $mlCH_4/gVS_{fed}$ the highest among different forage crops, the other showed the value above a methane yield of 300 $mlCH_4/gVS_{fed}$. The forage crop could be used as a good substrate to improve the methane production in anaerobic co-digestion together with animal manure.

Recovery of Sustainable Renewable Energy from Marine Biomass

  • Marine biomass is considered an important substrate for anaerobic digestion to recovery energy i.e. methane. Nevertheless, marine biomass has attracted little attention by researchers compared to terrestrial feedstock for anaerobic digestion. In this study, biochemical methane potential (BMP) test was used to evaluate generation of renewable energy from starfish. A cumulative biogas yield of $748{\pm}67mL\;g^{-1}VS^{-1}$ was obtained after 60 days of digestion. The cumulative methane yield of $486{\pm}28mL\;CH_4\;g^{-1}VS^{-1}$ was obtained after 60 days of digestion. The methane content of the biogas was approximately 70%. The calculated data applying the modified Gompertz equation for the cumulative $CH_4$ production showed good correlation with the experimental result obtained from this batch study. Since the result obtained from this study is comparable to results with other substrates, marine biomass can be co-digested with food waste or swine wastewater to produce $CH_4$ gas that will help to reduce the gap in global energy demand.

  • The effect of alkaline pre-treatment on the solubilization of waste activated sludge(WAS) was investigated, and the biodegradability of WAS, pretreated WAS, [PWAS], food waste and two types of mixture were estimated by biochemical methane potential [BMP] test at $35^{\circ}C$. The biodegradability of PWAS and mixture waste were significantly improved due to the effect of alkaline hydrolysis of WAS. An alkaline pre-treatment was identified to be one of the useful pre-treatment for improving biodegradability of WAS and mixture waste. In high-rate anaerobic co-digestion system coordinate with an alkaline pre-treatment in process, the digesters were operated at the HRT of 5, 7, 10 and 13 days with a mixture of FW $50\%\;and\;PWAS\;50\%,\;$In term of $CH_4$ content, VS removal and specific methane production [SMP] which are the parameters in the performance of digester, the optimum operating condition was found to be a HRT of 7 days and a OLR of 4.20g/L-day with the highest SMP of 0.340 L $CH_4/g$ VS.

  • Bladder exstrophy is a rare congenital condition of the pelvis, bladder, and lower abdomen that opens the bladder against the abdominal wall, produces aberrant growth, short penis, upward curvature during erection, wide penis, and undescended testes. Exstrophy affects 1/30,000 newborns. The bladder opens against the abdominal wall in bladder exstrophy, a rare genitourinary condition. This study is vital to provide appropriate therapy choices as a basis to improve patient outcomes. This study may explain bladder exstrophy and provide treatment. Epispadias, secretory placenta, cloacal exstrophy, and other embryonic abnormalities comprise the exstrophy-spades complex. The mesenchymal layer does not migrate from the ectoderm and endoderm layers in the first trimester, affecting the cloacal membrane. Embryological problems define the exstrophy-aspidistra complex, which resembles epimedium, classic bladder, cloacal exstrophy, and other diseases. Urogenital ventral body wall anomalies expose the bladder mucosa, causing bladder exstrophy. Genetic mutations in the Hedgehog cascade pathway, Wnt signal, FGF, BMP4, Alx4, Gli3, and ISL1 cause ventral body wall closure and urinary bladder failure. External factors such as high maternal age, smoking moms, and high maternal body mass index have also been associated to bladder exstrophy. Valproic acid increases bladder exstrophy risk; chemicals and pollutants during pregnancy may increase bladder exstrophy risk. Bladder exstrophy has no identified cause despite these risk factors. Exstrophy reconstruction seals the bladder, improves bowel function, reconstructs the vaginal region, and restores urination.

Lifecycle cost assessment of best management practices for diffuse pollution control in Han River Basin (한강수계 비점오염원 저감시설의 생애주기비용 평가)

  • Diffuse pollution management in Korea initiated by the Ministry of Environment (MOE) resulted to the construction of pilot facilities termed Best Management Practices (BMPs). Twelve BMPs installed for the diffuse pollution management in the Kyung-An Stream were monitored since 2006. Data on the mass loading, removal efficiency, maintenance activities, etc. were gathered and utilized to conduct the evaluation of long-term performance of BMPs. The financial data such as actual construction, design and maintenance cost were also collected to evaluate the lifecycle cost (LCC) of BMPs. In this study, most of the maintenance activity was focused in the aesthetic maintenance that resulted to the annual maintenance cost of the four BMP types was closely similar ranging from 8,483 $/yr for retention pond to 8,888 $/yr infiltration system. The highest LCC were observed in constructed wetland ($418,324) while vegetated system had the lowest LCC ($210,418). LCC of BMPs was not so high as compared with the conventional treatment facility and sewage treatment plant. On the other hand, the relationship of removal efficiency on unit cost for TSS and TN was significant. This study will be used to design the cost effective BMP for diffuse pollution management and become models for LCC analysis.

Optimization of Methane Yield in Anaerobic Digestion of Sewage Sludge with Microwave Pretreatment (극초단파 전처리를 적용한 하수슬러지 혐기성소화에서 메탄수율 최적화)

  • The objective of this study was to find an optimum methane yield condition in anaerobic digestion of sewage sludge with microwave pretreatment. The pretreatment process was carried out using a lab scale industrial microwave unit (2,450 MHz frequency). The digestion efficiency of pretreated sludge was evaluated by biochemical methane potential (BMP) test. Box-Behnken design and Response Surface Analysis (RSA) were applied to determine the optimal combination of sludge mixing ratio (0 to 100%), power (400 to 1600 W), holding time (0 to 10 min) and pretreatment temperature (60 to 100℃). BMP test results showed that Volatile Solid (VS) removal efficiency was up to 48% at a condition of 0% for mixing ratio, 1600 W for power, 5 min for holding time, and 80℃ for pretreatment temperature. Methane production was up to 832.3 mL/g VSremoved at a condition of 50% for mixing ratio, 1000 W for power, 5 min for holding time, and 80℃ for pretreatment temperature. The results of the variance analysis (ANOVA) showed that the p-value of the power and pretreatment temperature among the independent variables were significant (p<0.05), and in particular, the pretreatment temperature significantly affected on the solubilization and methane production. The optimum condition for the maximum methane yield (847 mL/g VSremoved) was consist of 38.4% of mixing ratio, 909.1 W of power, 4.1 min of holding time, and 80℃ of temperature within the design boundaries.

First flush modeling of the radial type surface runoff and a placement strategy for stormwater inlets to improve the effectiveness of the first flush treatment in a small impervious catchment (방사형 강우 유출의 초기세척 모의 및 소규모 불투수 배수구역에서의 초기우수 처리효과 상승을 위한 집수시설 배치 방안)

  • In this study, general characteristics of dynamic behavior of stormwater runoff from a small impervious catchment was investigated from a series of simulations for a radial type surface runoff. Based on the simulation results, a better placement strategy for stormwater inlets to improve performance of a structural best management practice (BMP) was suggested. The degree of pollutant first flush from an ideal radial type impervious catchment was simulated using a 1-D diffusion wave equation coupled with a pollutant transport equation. The results showed that the first flush of the chemical oxygen demand was the strongest when the catchment length ranged 30-50m at a bed slope of 0.02. This result suggested that a required degree of the first flush can be intentionally obtained by just changing the locations and numbers of stormwater inlets, and thereby adjusting the catchment length. Particularly, the overall performance of a structural BMP in reducing pollutant load can be improved by placing the stormwater inlets at locations for obtaining a required first flush strength.

  • There is no genetic activity information with the functions of dental pulp and periodontal ligament in human. The purpose of this study was to identify the gene-expression profiles of, and the molecular biological differences between periodontal ligament and dental pulp obtained from human permanent teeth. cDNA microarray analysis identified 347 genes with a fourfold or greater difference in expression level between the two tissue types 83 and 264, of which were more plentiful in periodontal ligament and dental pulp, respectively. Periodontal ligament exhibited strong expression of genes related to collagen synthesis (FAP), collagen degradation (MMP3, MMP9, and MMP13), and bone development and remodeling (SSP1, BMP3, ACP5, CTSK, and PTHLH). Pulp exhibited strong expression of genes associated with calcium ions (CALB1, SCIN, and CDH12) and the mineralization and formation of enamel and dentin (SPARC/SPOCK3, PHEX, AMBN, and DSPP). Among these genes, SPP1, SPARC/SPOCK3, AMBN, and DSPP were well known in dental research. However, the other genes are the newly found and it may help to find a good source of regenerative therapy if further study is performed.

A Study on Characteristics of Solubilization and Biogas Production for Sewage Sludge using Thermal Pretreatment (열적가용화공정을 이용한 하수슬러지의 가용화 특성 및 바이오가스 생산량 평가)

  • In this study, an applicability of thermal pretreatment for primary and secondary sludge, which are generated in a sewage treatment plant, was evaluated. The efficiency and charateristics was investigated with each sludge after pretreatment under the condition of $100{\sim}220^{\circ}C$ for 30 minutes. As the result, it was found that $SCOD_{Cr}$, $NH_4{^+}$, VFAs concentrations increased as the pre-treatment temperature increased. For COD solubilization, it was also highly dependent on an increase of temperature resulting in acceleration on hydrolysis and acid fermentation. In the BMP (Biochemical Methane Potential) experiment, for the primary sludge, it showed the higher biogas production rate at a temperature of $220^{\circ}C$, however, the effect was insignificant (5.6%). Whereas, for the secondary sludge, the increase on biogass production rate was 38.8% ($180^{\circ}C$) and this means that the secondary sludge is more suitable for an applicability of thermal pretreatment.

Simvastatin Induces Osteogenic Differentiation and Suppresses Adipogenic Differentiation in Primarily Cultured Human Adipose-Derived Stem Cells

  • Recent in vitro and in vivo animal studies have reported that statin, a cholesterol-lowering drug, stimulate osteogenic differentiation. In the present study, we investigated the effect of simvastatin on osteogenic and adipogenic differentiation in primarily cultured human adipose-derived stem cells (hADSCs). The simvastatin treatment significantly increased the positive cell numbers in alkaline phosphatase and von Kossa staining, and enhanced the expression levels of bone morphogenic protein (BMP)-2, core binding factor alpha 1 (cbfa1), collgen type I and osteonectin mRNAs. Lastly, hADSCs were cultured in the adipogenic media with or without simvastatin to examine the effect of simvastatin on adipogenic differentiation. In the RT-PCR analysis, there were notable decreases in mRNA expression of aP1, C/EBP-$\alpha$ and PPAR-$\gamma$ in hADSCs cultivated in simvastatin-added medium, compared to those in simvastatin-free medium. It suggests that the adipogenic differentiation was significantly inhibited by simvastatin treatment. These observations indicate that simvastatin induces osteogenic differentiation and suppresses adipogenic differentiation in hADSCs.
