Bacterial diseases of soybean has been recognized as a limiting factor of soybean production in Korea as it was estimated to cause around 10 percent of yield losses annually. The purpose of the study is to obtain information on the diseases through proving the kinds of pathogens and epidemiology, The wire brush method and multineedle appeared to be the best way of inoculation under all circumstances. Wire brush method, especially, was effective in shortening the incubation period and manifesting the lesion development by introducing more inoculum per unit of area. In case of spray inoculation it was necessary to apply a small amount(1 : 1,000) of wetting agent, twin-20, otherwise it was unabled to produce the diseases under field conditions. Two kinds of bacterial diseases caused by Pseudomonas glycinea and Xanthomonas phaseoli var. sljense were found from surveyed areas in Kore. Wild fire disease on soybean caused by Pseudomonas tabaci had not detectable during the experiment although there were several reports on the disease from other countries. when the pathogens were introduced into sterile soil, bacterial leaf blight pathogen could exsisted until 30 days while bacterial pustule pathogen survived only 4 days under the natural conditions of later June. Both bacteria, however, could produce the disease after more than 10 months period of storage in refrigerator when they were exsisted in infected plant tissues. In warehouse, non-temperature controlled, the bacteria lose their infectability within 6 months period from October to April even though they exsisted in infected tissues. Surface infested seeds with the pathogens could not produced the diseases on seedling stages of soybean plants when the seeds were planted in sterile soil after inoculation by dipping the seeds into bacterial suspensions, although germination was depressed by the pathogens when the seeds were planted on agar media.
The study has been carried out to compare the pathogenicity, physiological characteristics and genetic reliability between rough colony type strain and smooth colony type strains of Pseudomonas mori (Boyer et Lambert) Stevens which were isolated from diseased plant parts in 5 different areas throughout country. The results are summarized as follow. 1. The rough colony type strain showed more agressive reactions to tested host plant varieties than smooth colony type strains though there was no differences in the appearence of lesion types caused by both strains. 2. Both colony types were differentiated morphologically in that the rough colony type strain was having more than 200r long filamentous body without flagella where as the smooth colony type strains have short rods with one or several polar flagella. 3. The colony of smooth type strains was circular, entire, smooth and opaque, while the rough type strain shelved endulated, irregular margin, rough and wrinkled colony on nutrient agar media. 4. There were no differences between both colony types in the physiological and serological test. 5. Both of smooth and rough colony type strains showed genetic reliability through more than 100 succeeding cultures on the media, and were stable to various chemicals such as 1 to 3 percent of NaCl, 5 kinds of organic acid and 4 kinds of antibiotics.
Incidence of soilborne diseases, as a major cause of failure of continuous monocropping becomes severe in recent years. For examples, recent epidemics of club root of chinese cabbage, white rot of garlic, bacterial wilt of potato, pepper phytophthora blight, tomato fusarium wilt and CGMMV of watermelon are the diseases that require urgent control measures. Reasons for the severe incidence of soilborne diseases are the simplified cropping system or continuous monocropping associated with allocation of major production areas of certain crop and year-round cultivation system that results in rapid degradation of soil environment. Neglect of breeding for disease resistance relative to giving much emphasis on high yield and good quality, and cultural methods putting first on the use of chemical fertilizers are thought to be the reason. Counter-measures against soilborne disease epidemics would become most effective when the remedies are seeded for individual causes. As long-term strategies, development of rational cropping system which fits local cropping and economic condition, development and supply of cultivars resistant to multiple diseases, and improvement of soil environment by soil conditioning are suggested. In short-term strategies, simple and economical soil-disinfestation technology, and quick and accurate forecasting methods for soilborne diseases are urgent matter far development. for these, extensive supports are required in governmental level for rearing soilborne disease specialists and activation of collaborating researches to solve encountering problems of soilborne diseases.
Xanthomonas oryzae pv. oryzae, causal agent of bacterial leaf blight (BLB) of rice, had been collected and identified using Biolog and fatty acid analysis. Epidemics of BLB had been occurred all the times at several rice cultivating areas in Korea in 1999-2004. Most X. oryzae pv. oryzae isolated in 1999 and 2002 belonged to Korean race K1, but more than 50% of the pathogen isolated in 2003 belonged to Korean race K3. Especially, most pathogens isolated in Jeonnam and Joenbuk provinces belonged to Korean race K3. Inoculation test of near isogenic lines (NIL) of rice carrying single resistance genes against BLB showed that many isolates belonging to Korean race 1 reacted differently to diverse resistant monogenic lines of rice. Southern blot analysis also showed that the bacterial pathogens belonged to the same race had different numbers of avirulence genes. This results suggested that each Korean race type may respond to many resistance genes of rice. All the K3 races isolated in Jeonnam and Joenbuk provinces were able to cause disease on Xa3 monogenic lines of rice. Since most rice cultivars cultivated in Jeonnam and Jeonbuk were carrying Xa3 resistance genes, the bacterial pathogens isolated in Jeonnam and Jeonbuk were likely to develop to adapt to Xa3 resistance gene. Together with avirulence gene patterns of the bacterial isolates and the results of disease reaction of monogenic lines of rice to them, Korean X. oryzae pv. oryzae was classified into 19 pathotypes. This newly classified pathotypes should help the breeding of new resistance rice cultivars in Korea.
Five hundred sixteen isolates of Botrytis elliptica were isolated from infected leaves of Lilium longiflorum from Kangwon alpine areas in Korea during tile seasons from 1998 to 2000 and resistance of these isolates against some fungicides were examined. The isolation frequency of phenotypes resistant to benomyl, procymidone, and diethofencarb were 90.1, 32.4, and 40.9%, respectively. The isolates were divided into six phenotypic groups; RSS, RRS, SSR, SRR, RSR and RRR, representing sensitive (S) or resistant (R) to benzimidazole, dicarboximide, and N-phenylcarbamate fungicides in order. The percentage of six phenotypes were 40.7, 8.5, 7.2, 2.7, 19.8, and 21.1%, respectively. The RSS phenotype was the most frequently isolated, and tile SRR consisted of the extremely minor populations. In comparison studies on tile overwintering ability of each phenotype in relation to the others, the most frequently isolated RSS and SSR had the higher fitness ability than the less frequently isolated RSR, SRR, and RRR. Recently, population increase of tile RSR and RRR phenotypes may have resulted from the increased applications of the mixture of carbendazim and diethofencarb to control benzimidazole-resistant B. elliptica since 1998. The results of this study indicate that careful application of the fungicides is necessary to achieve effective control of leaf blight on lily in Korea.
This study was conducted to investigate the effect of bulblet size on growth and bulb enlargement of Lilium Oriental hybrids, such as 'Sorbonne', 'Siberia', and 'Casa Blanca', grown in areas of high altitude (780 m), in 2009. The bulblets were oriented from tissue culture and planted to produce virus-free bulblets. All the three cultivars showed high vitality, with more than 93% producing shoots, even from small bulblets with a bulb circumference (BC) of less than 6 cm. Bulblets with BC 9-10 cm showed an increased height and number of leaves rapidly to induce the phase conversation from vegetative growth to reproductive growth. The bulblets of BC 9-10 cm in 'Sorbonne', 'Siberia', and 'Casa Blanca' had 2.0, 2.9, and 2.8 flower bud sets per plant, respectively. However, the flowers from those bulblets were not adequate as standard bulbs to produce cut-flowers for the domestic market. The larger bulblets showed more prosperous characteristics in growth and development of Lilium Oriental hybrids. Small bulblets had a high occurrence of viruses and leaf blight symptoms during cultivation, indicating the aggravated disease symptoms in the previously infected bulbs. 'Sorbonne' cultivars showed a high rate of enlargement of bulbs, and small bulblets under BC 6 cm produced more than 23% of the standard bulbs of BC 14-16 cm, which could produce cut-flowers for export. However, 'Casa Blanca' bulblets with BC 6-9 cm produced low bulbs of BC 14-16 cm at 21% level. 'Casa Blanca' cultivars had more bulb roots, longer root lengths, and higher weights than 'Sorbonne' and 'Siberia' cultivars. 'Casa Blanca' cultivars with BC over 9-10 cm grew rapidly and produced a high quantity of bulbs. In these results, 'Sorbonne' bulblets of BC 9-10, 'Siberia' bulblets of BC 10-12 cm, and 'Casa Blanca' bulblets of BC 10-12 cm produced 89%, 86%, and 93% of the standard bulbs of BC 14-16 cm, respectively. It is recommended that bulblets larger than the above sizes be used to produce cut-flowers for the export market. Experiment results suggest that production of bulbs larger than BC 18 cm requires bulblets that are larger than BC 12-14 cm of the three cultivars for the highest quality cut-flowers.
Potato cv. 'Haryeong' was bred with high solids, resistance to late blight and good culinary quality. It has been registered as new potato variety in 2005. Tuber necrosis symptoms such as severe superficial necrosis, raised surface lesions and ringed necrotic areas were found in tubers of 'Haryeong' during storage of seed potatoes in 2010. Potato virus Y (PVY) was detected from these symptomatic tubers by the analysis of RT-PCR using a primer set specific to coat protein gene of PVY. The nucleotide sequence of RT-PCR product ($PVY^{Hkr}$) was determined and compared to those of other strains, such as $PVY^{Kor}$, $PVY^N$, $PVY^{NTN}$, $PVY^O$, and $PVY^C$ registered in GeneBank. The result showed that $PVY^{Hkr}$ was exactly the same as $PVY^{Kor}$, Korean isolate reported in 2005, except two nucleotides. To verify the PVY was responsible for the tuber necrosis symptoms shown in the tubers of 'Haryeong', a bioassay was done using two viruses (PVY and Potato leafroll virus) and five potato cultivars ('Haryeong', 'Superior', 'Atlantic', 'Dejima', and 'Chubaek'). As expected, the same necrosis symptom appeared in tubers of 'Haryeong' infected with PVY only during the storage period.
This study has the object of deriving schemes for not only securing safety of chestnuts, which is the most important forest product in terms of both production and exportation, to consumers with implementing persistent production of them in harmony with the environment, but also producing environmentally friendly chestnuts that can contribute to the increase of a farm household's income through price differentiation from other chestnuts, which are grown by generally accepted ways. In order to do this, I performed an analysis of the reality and outcome from the management of farm households that produce environmentally friendly chestnuts. The result of the analysis demonstrated some problems such as poor establishment of farming skills in a beginning stage, administrative problems with weeding operations and prevention of blight, difficulty in ensuring distribution channels and high production cost. Therefore, propelling productive expansion of environmentally friendly chestnuts after resolving these problems requires many supports and ideas: establishment and diffusion of farming skill system under the cooperation of civil and government organizations, construction of environmentally friendly INM (Integrated Nutrient Management) and IPM (Integrated Pest Management), creation of production areas for improving productivity, introduction of 'Institution of Income Conservation for Environmentally-Friendly Production' that can help farmers to preserve integrity of income on their switchover to production of environmentally friendly chestnuts from generally-grown chestnuts, and finally strong and persistent promotion of environmentally friendly chestnuts towards existing farmers and consumers.
Three post-harvest fields each in four rice growing areas, Iri, Naju, Jinju and Taegu were randomly selected and surveyed during December 1986 to examine sclerotial density of Rhizoctonia solani overwintering in the field. Surface soil of $0.09m^2$ area was sampled in each field with three replications and sieved to collect sclerotia. Germiability and pathogenicity of collected sclerotia were examined in the laboratory. Number of sclerotia $({\times}10^6)/ha$ in Iri, Naju, Jinju, and Taegu was estimated from the sample as 2.7, 1.2, 0.7 and 0.6, respectively. Based on sample variance with simple random sampling in each area, number of sampling required for estimating average sclerotial density with the precision of 10% apart from a chance of 1 in 20 was calculated to 41, 132, 232, and 395 for Iri, Naju, Jinju and Taegu, respectively. Percentage of germination of sampled sclerotia on potato sucrose agar (PSA) ranged from 42 to 78% depending on the area, and averaged 60%. About 49% of the germinated sclerotia were pathogenic to a rice cultivar Jinheung that was used to test pathogenicity of the sclerotia. Proportion of viable sclerotia that have both germiability and pathogenicity was thus estimated to 0.29 of total sclerotia collected. R. solani cultures obtained from the sclerotia could be distinguished into three groups based on colony morphology on PSA. Size and number of sclerotia formed on PSA differed between group but were not associated with pathogenicity to Jinheung.
Basal rot of freesia caused by a Sclerotium sp. occurred at Incheon areas. Incidence of the disease reached up to 45% and averaged 17.0% in the fields. Typical symptoms consisted of sheath dry and leaf blight due to rots on basal leaves. The causal fungus was identified as Sclerotium sp. based on following mycological characteristics. The fungus formed sclerotia on cultural media and plant tissues, but did not produce asexual spores. On cultural medium, aerial mycelia of the fungus changed color from white to clay with cultural age and smelled musty odor. Numerous irregular and elliptical black microsclerotia of the fungus were formed on potato dextrose agar (PDA) after 5 days of incubation at $25^{\circ}C$ and sized $115{\sim}200{\times}95{\sim}150 (av. 145{\sim}126.5){\mu}m$. The fungus grew at $10{\sim}32^{\circ}C$ and $pH 4.0{\sim}8.5$. However, the optimal temperature and pH for mycelial growth of the fungus were $24^{\circ}C$ and 5.5 respectively. The isolate showed present pathogenicity to not only freesia but gladiolus in the pathogenicity test, and the symptoms were similar to those observed in the fields. Basal rot of freesia caused by Sclerotium sp. is firstly reported in Korea.
