Antithrombosis and Antioxidant Activity of Methanol Extract from Different Brands of Rice (시판 쌀 메탄을 추출물의 항혈전 및 항산화 활성)

  • Antithrombosis and antioxidant activity of the methanol extracts, which were prepared from 51 different brands of rice, were evaluated to investigate bioactive-substances. Among them, the extract of sample No. 2, 25, 40 and 41 showed strong thrombin inhibition activities ($401\~764\%$ inhibition at 5 mg/mL), and the extract of sample No. 5, 9, 23, 36, 44 and 48 showed strong DPPH scavenging activities ($40.8\~64.1\%$ at 0.4 mg/mL), respectively. The average contents of total flavonoid and polyphenol with 50 brands of rice were 51.89 mg/kg and 195.23 mg/kg, respectively. However, the activities were not linked with the contents of flavonoid and polyphenol. By the treatment at $100^{\circ}C$ for 10 min, the activities of the selected rices were severely decreased. Maximal remaining activities of antithrombosis and antioxidant were $208\%$ of No. 40 and $23.2\%$ of No. 48, respectively. These results support a functional superiority of rice-based livelihood, and suggest that the development of healthy food using functional ingredients of rice is possible.

Relationship between Plant Protection Rate and Coefficient of Variation of Microbial Products for Ginseng Cultivation (인삼재배용 미생물 제품의 식물보호율과 변이계수와의 관계)

  • The plant protection rate of the marketed microbial products for ginseng cultivation was investigated against Rhizoctoina solani and Pythium sp. in a seedling pot experiment. A significant difference was found among the mean plant protection rates (Pm) of the microbial products, including Tolclofos-methyl (Rhizolex). The best microbial product, C-ISR2, showed a 33% and 33.6% net Pm (total Pm-control Pm) in the two tests against Rhizoctonia solani. In one test with a 58.6% control plot Pm, the total pm was 91.6%, indicating that plant protection can be done only with a microbial product in a well-conditioned field. The net Pm of C-ISR2 against Pythium sp. was 26.4%. The net Pm of a microbial product against a pathogen seems to be fixed. A significant negative linear correlation was found between the Pm and the coefficient of variation (CV) of the protection rate in all the three experiments. This indicates that the protection processes of control, microbial products, and chemical pesticides are in the same system. Pm was only dependent on CV, probably due to each seedling's microenvironment. In the linear correlation equation between the Pm and the CV of the microbial products and the control plot, the intercept of the vertical axis will be the theoretical Pm when CV is zero.

Classification of Korean Polygonatum Collections Based on Cluster Analysis (군집분석에 의한 한국 자생 둥글레속 수집종의 분류)

  • Morphological characteristics and growth patterns of 20 Polygonatum collections indigenous to Korea were examined and the collections were classified to obtain the basic data for practical use of Polygonatum genetic resources. Based on the cluster analysisi 20 collections were distinctly classified into seven groups with average distance greater than 0.6 between groups. Group I was p. sibiricum Delar, and group II included p. odoratum var. pluriforum Ohwi, P. odoratum var. pturiforum Ohwi 'Variegata' and P. odoratum var. maximowiczii Koidz.. Group III was P. odoratum var. thunbergii Hara, group IV included P. lasianthum var. coreanum Nakai, and group V was P. involucratum Maxim. and P. desoulavyi Komarov. group Ⅵ was P. inflatum Komarov and group Ⅶ was P. humile Fischer ex. Maxim. Morphologically, group I was larger than the other groups, group II and III were medium, and group IV to Ⅶ were small. In the classification of genus Polyognatum, stem length, stem habit, phyllotaxis, stem angularity, petiole, inflorescence, perianth, bract and rhizome were particularly important characters. Group I to III were thought to be useful as the edible and medicinal resources plants, and group IV to Ⅶ were thought to be useful as ornamental plants.

Initial Development of Forest Structure and Understory Vegetation after Clear-cut in Pinus densiflora Forest in Southern Gangwon-do Province (강원도 남부 지역에서 소나무림 개벌 후 초기 임분 구조 및 하층식생 발달)

  • Open- to closed canopy stage and it's ecological characteristics in vegetation succession are commonly described, but poorly understood in Korea. Vegetation development on structure, environment and understory abundance were studied for 16 yr in post-clearcut Pinus densiflora forests in the southern Gangwon-do province by applying space-for-time approach. We sampled 210 plots (10 for structure and 200 for understory) for four seral stages (1yr, 3yr, 10yr and 16yr). After clear-cut, mean stem density increased gradually to $5,714{\pm}645$ stems/ha after 16 years and mean basal area was also from $5.5{\pm}0.7m^2/ha$ after 10 years and doubled at $10.0{\pm}1.6m^2/ha$ in 16 years. Woody debris and bared soil on the forest floor peaked at 11--- after 10 years and at 10.3--- after 3 years, respectively. In understory mean cover declined with all growth form groups following succession, but in richness, forb specie increased with structural development during 16 years. Our study suggested that overstory development did not suppressed whole understory properties especially in richness, thus appeared to act as a filter selectively constraining the understory characteristics. However only long-term studies are essential for elucidating patterns and processes that cannot be inferred form short-term or space-for-time researches. Strong negative relationship between overstory and understory characteristics in conventional models surely reexamined.

Screening of $\alpha$-Amylase and $\alpha$-Glucosidase Inhibitor from Nepalese Plant Extracts (100종 네팔 식물 추출물로부터 $\alpha$-Amylase 및 $\alpha$-Glucosidase저해제의 선별)

  • In the course of screening for anti-acidosis and anti-diabetes agent from natural products, the inhibitory activities of Nepales plant extracts against microbial $\alpha$-amylase and $\alpha$-glucosidase were evaluated. Among the 100 different kinds of ethanol extracts, Cedrus deodara (Roxb.) G. Don and Myrica nagi Thunb showed strong inhibition activities against $\alpha$-amylase. The $IC_{50}$ values of C. deodara (Roxb.) G. Don, M. nagi Thunb and acarose, a commercial available anti-diabetes agent, were 44.5, 47.5 and $50.5\;{\mu}g/mL$, respectively. Considering the crude extract of C. deodara (Roxb.) G. Don, and M. nagi Thunb, these extracts have strong potentials as anti-acidosis or anti-diabates agent. In a while, Cleistocalyx operculatus (Roxb.) extract showed a good inhibition activity to $\alpha$-amylase and $\alpha$-glucosidase, even it was recently reported. The selected three extracts did not show any hemolysis activity against human red blood cell up to 1 mg/mL, and the inhibition activities were maintained by heat or acid treatment. Moreover, treatment of HCl (0.01N) for 1 h into C. operculatus (Roxb.) and M. nagi Thunb increased the inhibition activity from 50% to 70%. Our results suggest that C. deodara (Roxb.) G. Don, M. nagi Thunb, and C. operculatus (Roxb.) are potential as anti-acidosis and anti-diabetes agents.

Design of a Prototype System for Graft-Taking Enhancement of Grafted Seedlings Using Artificial Lighting - Effect of air current speed on the distribution of air temperature and relative humidity in a graft-taking enhancement system (인공광을 이용한 접목표 활착촉진 시스템의 시작품 설계 - 활착촉진 시스템 내의 기온과 상대습도 분포에 미치는 기류속도의 효과)

  • Grafting of fruit-bearing vegetables has been widely used to increase the resistance to soil-borne diseases, to increase the tolerance to low temperature or to soil salinity, to increase the plant vigor, and to extend the duration of economic harvest time. After grafting, it is important to control the environment around grafted seedlings for the robust joining of a scion and rootstock. Usually the shading materials and plastic films are used to keep the high relative humidity and low light intensity in greenhouse or tunnel. It is quite difficult to optimally control the environment for healing and acclimation of grafted seedlings under natural light. So the farmers or growers rely on their experience for the production of grafted seedling with high quality. If artificial light is used as a lighting source for graft-taking of grafted seedlings, the light intensity and photoperiod can be easily controlled. The purpose of this study was to develop a prototype system for the graft-taking enhancement of grafted seedlings using artificial lighting and to investigate the effect of air current speed on the distribution of air temperature and relative humidity in a graft-taking enhancement system. A prototype graft-taking system was consisted by polyurethane panels, air-conditioning unit, system controller and lighting unit. Three band fluorescent lamps (FL20SEX-D/18, Kumho Electric, Inc.) were used as a lighting source. Anemometer (Climomaster 6521, KANOMAX), T-type thermocouples and humidity sensors (CHS-UPS, TDK) were used to measure the air current speed, air temperature and relative humidity in a graft-taking system. In this system, air flow acted as a driving force for the diffusion of heat and water vapor. Air current speed, air temperature and relative humidity controlled by a programmable logic controller (UP750, Yokogawa Electric Co) and an inverter (MOSCON-G3, SAMSUNG) had an even distribution. Distribution of air temperature and relative humidity in a graft-taking enhancement system was fairly affected by air current speed. Air current speed higher than 0.1m/s was required to obtain the even distribution of environmental factors in this system. At low air current speed of 0.1m/s, the evapotranspiration rate of grafted seedlings would be suppressed and thus graft-taking would be enhanced. This system could be used to investigate the effects of air temperature, relative humidity, air current speed and light intensity on the evaportranspiration rate of grafted seedlings.

Analysis of Temperature Gradients in Greenhouse Equipped with Fan and Pad System by CFD Method (CFD 기법을 이용한 팬 앤 패드 냉방 온실의 온도경사 분석)

  • Evaporative cooling pad system is one of the main cooling methods in greenhouses and its efficiency is very high. However, it has some disadvantages such as greenhouse temperature distributions are not uniform and installation cost is expensive. In this study, a CFD simulation model f3r predicting the air temperature distribution in the fan and fad cooling greenhouse was developed. The model was calibrated and validated against experimental data and a good fit was obtained. The influence of different outside wind, fan and pad height, ventilation rate, shading, and greenhouse length, were then examined. In order to reduce the internal temperature gradients, it is desired that the prevail wind direction and the fan and pad heights are considered. The simulation indicates that high ventilation rates and shading contribute to reduce the temperature gradients in the fan and pad cooling greenhouse. In order to maintain the desired greenhouse temperature, the pad-to-fan distance should be restricted according to the design climate conditions, shading and ventilation rates. The developed CFD model can be a useful tool to evaluate and design the fan and pad systems in the greenhouses with various configurations.

Enhancement of Tomato Tolerance to Biotic and Abiotic Stresses by Variovorax sp. PMC12 (Variovorax sp. PMC12 균주에 의한 토마토의 생물학 및 비생물학적 스트레스 저항성 증진)

  • Rhizobacteria play important roles in plant growth and health enhancement and render them resistant to not only biotic stresses but also abiotic stresses, such as low/high temperature, drought, and salinity. This study aimed to select plant growth promoting rhizobacteria (PGPR) with the capability to mitigate biotic and abiotic stress effects on tomato plants. We isolated a novel PGPR strain, Variovorax sp. PMC12 from tomato rhizosphere. An in vitro assay indicated that strain PMC12 produced ammonia, indole-3-acetic acid (IAA), siderophore, and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase, which are well-known traits of PGPR. The aboveground fresh weight was significantly higher in tomato plants treated with strain PMC12 than in non-treated tomato plants under various abiotic stress conditions including salinity, low temperature, and drought. Furthermore, strain PMC12 also enhanced the resistance to bacterial wilt disease caused by Ralstonia solanacearum. Taken together, these results indicated that strain PMC12 is a promising biocontrol agent and a biostimulant to reduce the susceptibility of plants to both abiotic and biotic stresses.

Effect of Polysaccharides from Astragalus membranaceus on Exercise-Induced Fatigue and Oxidative Damage in Skeletal Muscle in Exhaustive Exercise Animal Models (과도 운동에 의해 유발되는 피로 및 골격근 산화적 손상에 대한 황기 다당체의 효과)

  • Background: Astragalus membranaceus is a well known oriental medicinal herb. The polysaccharides of the aboveground parts (AMA) and the radix (AMR) of A. membranaceus are the most important functional constituents. Methods and Results: The aim of this study was to determine the effects of AMA and AMR on the oxidative damage induced in the skeletal muscle of rats subjected to exhaustive exercise. Sprague-Dawley rats were randomly divided into exercise and non-exercise groups; in the groups receiving the test compounds, AMA and AMR were administered orally for 30 days. Skeletal muscle samples were collected from each rat after running to exhaustion on a treadmill to determine the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) and the concentation of malondialdehyde (MDA). The antioxidant enzyme activities of SOD and GSH-Px of skeletal muscle of AMA- and AMR-treated groups were significantly higher than those of the control and commercial sports drink (SPD)-treated groups in exhaustive exercise rats. In addition, MDA concentrations in the skeletal muscle of the AMA- and AMR-treated groups were significantly lower than those of the control and SPD-treated groups. In the present study, the effects of AMA and AMR on exercise endurance capacity were also evaluated in mice subjected to a swimming exercise test. AMA and AMR supplementation prolonged the swimming time of mice and enhanced exercise endurance capacity. AMA and AMR possess the ability to retard and lower the production of blood lactate, and prevent the decrease of serum blood glucose. Conclusions: These results showed that, AMR and AMA exerted beneficial effect in mice, increasing the activity of the antioxidant systems and inhibiting oxidative stress induced by exhaustive exercise. The compounds improved exercise performance and showed anti-fatigue effects against exhaustive exercise.

  • Erwinia pyrifoliae is a Gram-negative bacterium causing black shoot blight in apple and Asian pear trees. E. pyrifoliae strain EpK1/15 was isolated in 2014 from an apple twig from the Pocheon, Gyeonggi-do, South Korea. In this study, we report the draft genome sequence of E. pyrifoliae EpK1/15 using PacBio RS II platform. The draft genome is comprised of a circular chromosome with 4,027,225 bp and 53.4% G + C content and a plasmid with 48,456 bp and 50.3% G + C content. The draft genome includes 3,798 protein-coding genes, 22 rRNA genes, 77 tRNA genes, 13 non-coding RNA genes, and 231 pseudo genes.
