UV 전처리 유무에 따른 입상활성탄의 세균 생체량 및 군집 구조 비교 (Comparison of Bacterial Biomass and Community of Granular Activated Carbon with or without UV Pre-treatment Process)

  • 생물활성탄 공정은 수처리 과정에서 유기 오염물질을 효과적으로 제거하는 것으로 알려져 있으며, 활성탄에 부착된 세균의 생체량과 종은 오염물질 제거 과정에서 중요한 역할을 한다. 본 연구에서는 입상활성탄 공정에서 활성탄조의 깊이와 가동 기간에 따른 세균 생체량의 변화에 대해 확인해 보았다. 또한 입상활성탄공정 전단에 자외선 (UV) 공정 전처리를 하였을 때 세균 생체량의 변화를 확인하였다. 결과를 살펴보면 활성탄조의 깊이가 깊어질수록 세균 생체량이 감소하는 것을 확인하였다. 그리고 UV 공정 전처리를 한 경우, 공정 기간이 증가할수록 세균 생체량이 감소하는 것을 확인하였다. 그러나, UV 공정 전처리를 하지 않은 경우에는 공정 기간에 따른 세균 생체량의 변화가 나타나지 않았다. 본 연구 결과를 토대로 수처리 실공정에서 생물활성탄 공정 관리에 대한 유용한 정보를 제공할 것이라 여겨진다.

소양호 수중 생태계에서의 세균 생체물질량의 분포 (Distribution of bacterial biomass in the water column of Soyang lake)

  • 소양호에서 측정된 세균 체적, 세균 생물량 및 세균 생산량 등의 미생물학적인 요인의 변화에 미치는 물리화학적 환경요인의 영향을 통계학적 방법으로 분석을 하였다. 상관관계 분석과 중회귀 분석 결과 수온은 대부분의 미생물학적 요인에 폭넓은 영향을 미침을 알 수 있었다. 총 세균수, 세균 체적, 세균 생물 량 및 saprophyte 수외 변화는 엽록소 a와 pheophytin a의 존재와 높은 상관관계를, $^{3}H$-thymidine incorporation rate에 의해 측정된 세균 생산량은 seston의 농도에 큰 영향을 받는 것으로 분석되었다. 소양호 수중생태계에서의 세균 체적 및 세균 생물량의 미생물학적 요인은 미생물 군집에게 탄소와 에너지원으로 작용하는 유기물질의 제공원인 식물성 플랑크톤의 분포와 seston의 농도에 의해 조절되고 있음을 시사하여 준다.

Nitrifying Bacterial Community Structure of a Full-Scale Integrated Fixed-Film Activated Sludge Process as Investigated by Pyrosequencing

  • Nitrifying bacterial community structures of suspended and attached biomasses in a full-scale integrated fixed-film activated sludge process were investigated by analyzing 16S rRNA gene sequences obtained from pyrosequencing. The suspended biomass had a higher number of ammonia-oxidizing bacterial sequences (0.8% of total sequences) than the attached biomass (0.07%), although most of the sequences were within the Nitrosomonas oligotropha lineage in both biomasses. Nitrospira-like nitrite-oxidizing bacterial sequences were retrieved in the suspended biomass (0.06%), not in the attached biomass, whereas the existence of Nitrobacter-like sequences was not evident. The suspended biomass had higher nitrification activity (1.13 mg N/TSS/h) than the attached biomass (0.07 mg N/TSS/h). Overall, the results made it possible to conclude the importance of the suspended biomass, rather than the attached biomass, in nitrification in the wastewater treatment process studied.

Contribution of Marine Microbes to Particulate Organic Matter in the Korea Strait

  • To assess the relative contribution of bacterial and phytoplankton biomasses to particulate organic matter (POM) in the water column, microbial abundance and biomass were from two transects in the western channel of the Korea Strait in 1996. Bacterial abundance had a mean value of $5.9{\times}10^5$ cells/ml and chlorophyll-a averaged 0.14 ${\mu}g/l$. Bacterial abundance in the Korea Strait showed a positive relationship with chlorophyll-a concentration, while the distribution of POM did not covary with chlorophyll-a. Particulate organic carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon and nitrogen biomasses were 7.29 ${\mu}gC/l$ and 1.24 ${\mu}gN/l$, respectively, during the study periods. Bacterial biomass was larger in October than in August due to the autumn phytoplankton bloom. Phytoplankton biomass based on chlorophyll-a was 7.67 ${\mu}gC/l$ for carbon and 1.10${\mu}gN/l$l for nitrogen. The ratio of bacterial carbon (BC) to phytoplankton carbon (Cp) averaged 0.95 in the Korea Strait in 1996. Bacteria may play a more significant role in the dynamics of POM than phytoplankton do in August, with BC/Cp ratio of 1.26. The ratio of BC to Cp increased with a decrease in chlorophyll-a concentration. Averaged over all the samples in both cruises, the contribution of microbial biomass to POC and PON was about 43% and 51%, respectively. Bacterial assemblage constituted a significant fraction of POC (21%) and PON (27%). Phytoplankton accounted for 22% of POC and 24% of PON. Microbial biomass played a more important role in the dynamics of POC and PON in October than in August due to a significant increase in microbial biomass in the southern transect (transect-B) in October by the autumn phytoplankton bloom. This study showed that marine microbes may constitute a significant part in the reservoir of POM in the Korea Strait.

정수장 활성탄 여과지의 생물막과 그 활성도 (Biofilms and their Activity in Granular Activated Carbons Established in a Drinking Water Treatment Plant)

  • Bacterial biomass and its activity were measured in two kinds of granular activated carbon (GAC), the experimental and existing biofiltration system in a drinking water plant. The bacterial biomass was around 210 to 250 nmol P/g WW with phospholipid concentration at acclimation of ozonation treatment. The phospholipid biomass shows more or less a declining gradient along filter depth and no clear seasonality in its values. On the other hand, the microbial activity of [$^3H$]-thymidine and [$^{14}C$]-acetate incorporation within cells increased significantly along the filter depth, showing the difference of three fold between the upper and bottom layer. These factors support the different microbial composition or metabolic activity along the depth of GAC column. Turnover rates, the rate of bacterial biomass and production of biofilm, ranged from 0.26 /hr to 0.37 /hr, indicating a highly rapid recovery itself at amature state. In the non-ozonation treatment, the bacterial biomass was lower than in the ozonation and biological activity also declined towards the filter depth. The biomass levels during cessation of ozonation in the existing GAC filters were 68% of the actively ozonated state.

Variation in Microbial Biomass and Community Structure in Sediments of Peter the Great Bay (Sea of Japan/East Sea), as Estimated from Fatty Acid Biomarkers

  • Variation in the microbial biomass and community structure found in sediment of heavily polluted bays and the adjacent unpolluted areas were examined using phospholipid fatty acid analysis. Total microbial biomass and microbial community structure were responding to environmental determinants, sediment grain size, depth of sediment, and pollution due to petroleum hydrocarbons. The marker fatty acids of microeukaryotes and prokaryotes - aerobic, anaerobic, and sulfate-reducing bacteria - were detected in sediments of the areas studied. Analysis of the fatty acid profiles revealed wide variations in the community structure in sediments, depending on the extent of pollution, sediment depth, and sediment grain size. The abundance of specific bacterial fatty acids points to the dominance of prokaryotic organisms, whose composition differed among the stations. Fatty acid distributions in sediments suggest the high contribution of aerobic bacteria. Sediments of polluted sites were significantly enriched with anaerobic bacteria in comparison with clean areas. The contribution of this bacterial group increased with the depth of sediments. Anaerobic bacteria were predominantly present in muddy sediments, as evidenced from the fatty acid profiles. Relatively high concentrations of marker fatty acids of sulfate-reducing bacteria were associated with organic pollution in this site. Specific fatty acids of microeukaryotes were more abundant in surface sediments than in deeper sediment layers. Among the microeukaryotes, diatoms were an important component. Significant amounts of bacterial biomass, the predominance of bacterial biomarker fatty acids with abundance of anaerobic and sulfate-reducing bacteria are indicative of a prokaryotic consortium responsive to organic pollution.

낙동강하구 연안수역의 세균 생산성과 환경요인의 영향 (Heterotrophic Bacterial Secondary Productivity and Effect of Environmental Parameters is Naktong Embayment Korea)

  • 낙동강 연안 수역에서 세균의 생산성과 생산물량을 측정하여 미생물의 먹이고리에 기여하는 정도를 파악하기 위해 미생물학적 요인과 주요 환경요인을 측정하였다. 낙동강 연안 수계는 겨울철 갈수기에 유입되는 부영양화 담수의 영향을 많이 받았다. 종속영양세균과 총세균의 개체수는 각각$5.4{\times}10^2 ~ 3.2{\times}10^4, 2.2{\times}10^5 ~ 9.8{\times}10^5 $cells/ml이었으며, 세균의 평균 체적과 생물량은 각각 0.023~0.201$\mu$$m^3$/cell, 0.010~0.140$\mu$g-C/ml로 정점별, 계절별 차이는 크지 않았으나 체적과 총세균수가 반비례하였다. Thymidine pool size는 12.93~44.66nM로, 동계에 비해 하계에 세균이 이용하기 쉬운 pool로 이루어져 하계의세균 이차 생산성 측정치가 과소 평가될수 있음을 보여주고 있다. 종속영양세균의 이차생산성은 0.12~22.38$\mu$g-C/l/h의 변화 범위를 나타내었으며, 담수 유출부와 인접한 정점에서 가장 낮은 값을 보였다. 동계에 세균 생산성의 분포는 엽록소 a 및 수온 변화와 밀접한 상관성을 보였으며, 또한 세균 생산량도 많은 영향을 미치는 것으로 나타나 본 조사 수역에서 측정된 세균 생산성의 상당 부분은 식물플랑크톤의 공합성 산물을 이용하는 세균 생물량의 증가임을 알 수 있다.

낙동강 하류 상수원수의 생물활성탄에 의한 수질개선 및 세균분포 특성 (Improving Water Quality and Bacterial Characteristics during Water Treatment Process Using Biological Activated Carbons on Downstream of the Nakdong River)

  • Improvement of water quality and Investigation of bacterial characteristics have been conducted in a pilot plant using biological activated carbon (BAC) in water treatment process at the downstream of the Nakdong River. Most of water control parameters were highly improved after passing through BAC. Approximately 54% of dissolved organic carbon was removed in coal-based BAC process. Bacterial biomass and bacterial production appeared $9.8{\times}10^8 CFU/g and 7.1mg-C/m^3$.hr in coal-based BAC, respectively. Predominant bacteria species grown in BAC were identified as Pseudomonas, Flavobacterium, Alcaligenes, Acinetobacter and Aeromonas species. Particularly Pseudomonas vesicularis was dominant in both coal-based and coconut-based BACs, while Pseudomonas cepacia was dominant in wood-based BAC.

FISH 기법을 이용한 생물활성탄 공정에서의 운전기간별 부착 박테리아 군집변화 분석 (Analysis of Attached Bacterial Community of Biological Activated Carbon Process Using FISH)

  • The concentration of organic compounds was analyzed at each step of BAC process though $BDOC_{total/rapid/slow}$. Further, bacteria communities and biomass concentrations measured FISH and ATP methods were analyzed. The bed volume (BV) of steady state is different from that of based on assessment of organic compounds removal. Bed volumes in DOC, $BDOC_{rapid}$ and $BDOC_{total/slow}$ removal at steady state were around 27,500 (185.8 day), 15,000 (101.4 day) and 32,000 (216.2 day), respectively. A biomass didn't change after the bed volume reached 22,500 (152.0 day) according to analyzing ATP concentration of bacteria. The concentration of ATP was 2.14 ${\mu}g/g$ in BV 22,500 (152.0 day). The total bacterial number was $4.01{\pm}0.4{\times}10^7$ cells/g at the bed volume 1,150 (7.8 day) (the initial operation) and the number of bacteria was $9.27{\pm}0.2{\times}10^9$ at the bed volume 58,560 395.7 day) that increased more than 200 times. Bacterial uptrend was reduced and bacterial communities were stabilized since BV 18,720 (126.5 day). When BV were 1,150 (7.8 day), 8,916 (60.2 day), 18,720 (126.5 day), 31,005 (209.5 day), 49,632 (335.3 day), 58,560 (395.7 day), a proportion of total bacteria for the Eubacteria were 60.1%, 66.0%, 78.4%, 82.0%, 81.3% respectively. ${\gamma}$-Proteobacteria group was the most population throughout the entire range. The correlation coefficient ($r^2$) between Eubacteria biomass and ATP concentration was 0.9448.
