Cross-Infection of Flacherie and Densonucleosis Virus of the Silkworm, Bombyx mori, to Mulberry Pests. (누에 무름병 및 농핵병 바이러스의 주요 뽕나무 해충에 대한 교차감염)

  • Flacherie virus (FV) and Densonucleosis virus (DNV) of the silkworm, Bombyx mori, which give the most severest damage to the silk production in korea, were fed on the mulberry wild silkworm, Bombyx mori mandarina, the mulberry pyralid, Gryphodes phyloalis, and the American fall webworm, Hypantria cunea, to investigate cross infectivity by serological and histopathological at observation. By the Ouchterlony's double difusion test the mulberry wild silkworm was infected with both FV and DNV type 1 (DNV-1) and the mulberry pyralid with DNV-1, so those were confirmed the cross infection. But the American fall webworm was not recognized the cross infection by the same method. The infection and multiplication of the FV in the mulberry wild silkworm was observed in the cytoplasm of the goblet cell with the appearance of the virus-specific vesicle. In DNV-1 infection to the mulberry wild silkworm and the mulberry pyralid, the nuclei of columnar cell in the midgut of both insects was hypertrophied and the nuclei of midgut cell of the mulberry pyralid positively stained with the feulgen stain. Multiplication of DNV-1 in the midgut cell of the mulberry wild silkworm was replicated in two different patterens as linear arrays and large masses, while that of DNV-1 in the muberry pyralid was multiplied as virus masses in several portion of the nuclei of the midgut cell.

A Geophysical Survey of an Iron Mine Site (철광산 지역에서의 물리탐사 기술 적용 연구)

  • DC electrical and electromagnetic survey was applied to evaluate the reserve of an iron mine site. We analyzed the borehole cores and the cores sampled from outcrops in order to decide which geophysical method was efficient for the evaluation of iron mine site and to understand the geological setting around the target area. Based on the core tests for specific weight, density, porosity, resistivity and P-wave velocity, showing that the magnetite could be distinguishable by the electrical property, we decided to conduct the electrical survey to investigate the irone mine site. According to previous studies, the DC electrical survey was known to have various arrays with high resolutions effective to the survey of the iron mine site. However it was also known that the skin depth is too shallow to grasp the deep structure of iron mine. To compensate the weakness of the DC electrical method, we applied the MagnetoTelluric (MT) survey. In addition, a Controlled Source MT (CSMT) method was also applied to make up the shortcoming of MT method which is weak for shallow targets. From the DC electrical and MT survey, we found a new low resistivity zone, which is believed to be a magnetite reserve beneath the old abandoned mine. Therefore, this study was confirmed for additional utility value.

An Efficient Periodic-Request-Grouping Technique for Reduced Seek Time in Disk Array-based Video-on-Demand Server (디스크 배열-기반 주문형 비디오 서버에서의 탐색 시간 단축을 위한 효율적인 주기적 요청 묶음 기법)

  • In Video-on-Demand (VoD) servers, disk throughput is an important system design parameter because it is directly related to the number of user requests that can be served simultaneously. In this paper, we propose an efficient periodic request grouping scheme for disk array-based VoD servers that reduces the disk seek time, thus improving the disk throughput of VoD disk arrays. To reduce the disk seek time, the proposed scheme groups the periodic requests that access data blocks stored in adjacent regions into one, and arranges these groups in a pre-determined order (e.g., in left-symmetric or right-symmetric fashion). Our simulation result shows that the proposed scheme reduces the average disk bandwidth required by a single video stream and can serve more user requests than existing schemes. For a data block size of 192KB, the number of simultaneously served user requests is increased by 8% while the average waiting time for a user request is decreased by 20%. We also propose an adaptation technique that conforms the proposed scheme to the user preference changes for video streams.

Numerical study on the pressure drop and heat transfer enhancement in a flat-plate solar collector (평판형 태양열 집열기의 압력강하 및 열전달 성능 향상에 관한 수치해석적 연구)

  • The use of artificial roughness in various forms of shapes and sizes is the most common and effective way to improve the performance of a flat-plate solar collector. In the present study, numerical analysis on heat transfer and pressure drop was performed in a rectangular channel with various rib arrays. The uniform heat flux is applied to the channel from the upper side. The forms of ribs considered in this study were rib $90^{\circ}$, groove $90^{\circ}$, groove $60^{\circ}$, baffle $90^{\circ}$, baffle $60^{\circ}$, wave $90^{\circ}$ and wave $60^{\circ}$. Air is the working fluid, and the Reynolds number ranges from 3200 to 17800. Nusselt number and friction factor were investigated to predict the performance of the system with various type of ribs. The average Nusselt number and pressure drop were increased with the increase of velocity in all types of ribs. The highest heat transfer and pressure drop occurred for the baffle $90^{\circ}$, but highest performance factor considering heat transfer and pressure drop together occurred for the groove $60^{\circ}$. Therefore, heat transfer and pressure drop should be considered together when a flat plate solar collector is designed.

Measurement and Evaluation of Scatter Fractions for Digital Radiography with a Beam-Stop Array (Beam-Stop Array를 이용한 DR에서의 Scatter Fraction 측정 및 효용성 평가)

  • Scatter radiation considerably affects radiographic image quality by reducing image contrast and contributing to a non-uniform background. Images containing a large portion of scatter radiation may result in an incorrect diagnosis. In the past few years, many efforts have been made to reduce the effects of scatter radiation on radiographic images. The purpose of this study is to accurately measure scatter fractions and evaluate the effectiveness of beam-stop arrays. To measure scatter fraction accurately, a beam-stop array and the SFC (Scatter Fraction Calculator) program were developed. Images were obtained using the beam-stop array for both an anti-scatter technique with an anti-scatter grid and an air gap technique. The scatter fractions of the images were measured using the SFC program. Scatter fractions obtained with an anti-scatter grid were evaluated and compared to scatter fractions obtained without an anti-scatter grid. Scatter fractions were also quantitatively measured and evaluated with an air gap technique. The effectiveness of the beam-stop array was demonstrated by quantifying scatter fractions under various conditions. The results showed that a beam-stop array and the SFC program can be used to accurately measure scatter fractions in radiographic images and can be applied for both developing scatter correction methods as well as systems.

Three-Dimensional Microstructures Fabricated by Multi-Step Electrochemical Aluminum-Foil Etching (알루미늄 박판의 다단 전해식각 공정을 이용한 3 차원 마이크로 구조물의 제작)

  • We present a simple, cost-effective, and fast fabrication process for three-dimensional (3D) microstructures; this process is based on multi-step electrochemical etching of metal foils which facilitates the mass production of 3D microstructures. Compared to electroplating, this process maintains uniform and well-controlled material properties of the microstructure. In the experimental study, we perform single-step electrochemical etching of aluminum foils for the fabrication of 2D cantilever arrays. In the single-step etching, the depth etch rate and bias etch rate are measured as $1.50{\pm}0.10 {\mu}m/min$ and $0.77{\pm}0.03 {\mu}m/min$, respectively. Using the results of single-step etching, we perform two-step electrochemical etching for 3D microstructures with probe tips on cantilevers. The errors in height and lateral fabrication in the case of the fabricated structures are $15.5{\pm}5.8% $ and $3.3{\pm}0.9%$, respectively; the surface roughness is $37.4{\pm}9.6nm$.

Distribution of DArT Markers in a Genetic Linkage Map of Tomato (토마토 유전자연관지도 상의 DarT 마커 분포)

  • A genetic linkage map was constructed using 188 $F_9$ RILs derived from a cross between $Solanum$ $lycopersicum$ H7996 (resistant to bacterial wilt) and $S.$ $pimpinellifolium$ WVa700 (highly susceptible to bacterial wilt). The map consisted of 361 markers including 260 DArTs, 74 AFLPs, 4 RFLPs, 1 SNP, and 22 SSRs. The resulting linkage map was comprised of 13 linkage groups covering 2042.7 cM. The genetic linkage map had an average map distance between markers of 5.7 cM, with an average DArT marker density of 1/7.9 cM. Based on the distribution of anchor SSR markers, 11 linkage groups were assigned to 10 chromosomes of tomato except chromosomes 5 and 12. The DArT markers were distributed across the genome in a similar way as other markers and showed the highest frequency of clustering (38.8%) at ${\leq}$ 0.5 cM intervals between adjacent markers, which is 3 times higher than AFLPs (13.5%). The present study is the first utilization of DArT markers in tomato linkage map construction.

Fabrication of a Partial Genome Microarray of the Methylotrophic Yeast Hansenula polymorpha: Optimization and Evaluation of Transcript Profiling

  • The methylotrophic yeast Hansenula polymorpha has been extensively studied as a model organism for methanol metabolism and peroxisome biogenesis. Recently, this yeast has also attracted attention as a promising host organism for recombinant protein production. Here, we describe the fabrication and evaluation of a DNA chip spotted with 382 open reading frames (ORFs) of H. polymorpha. Each ORF was PCR-amplified using gene-specific primer sets, of which the forward primers had 5'-aminolink. The PCR products were printed in duplicate onto the aldehyde-coated slide glasses to link only the coding strands to the surface of the slide via covalent coupling between amine and aldehyde groups. With the partial genome DNA chip, we compared efficiency of direct and indirect cDNA target labeling methods, and found that the indirect method, using fluorescent-labeled dendrimers, generated a higher hybridization signal-to-noise ratio than the direct method, using cDNA targets labeled by incorporation of fluorescence-labeled nucIeotides during reverse transcription. In addition, to assess the quality of this DNA chip, we analyzed the expression profiles of H. polymorpha cells grown on different carbon sources, such as glucose and methanol, and also those of cells treated with the superoxide­generating drug, menadione. The profiles obtained showed a high-level induction of a set of ORFs involved in methanol metabolism and oxidative stress response in the presence of methanol and menadione, respectively. The results demonstrate the sensitivity and reliability of our arrays to analyze global gene expression changes of H. polymorpha under defined environmental conditions.

Identifying Genes Related with Radiation Resistance in Oral Squamous Cell Carcinoma Cell Lines (두경부 편평상피세포암주에서 방사선 내성 관련 유전인자의 확인)

  • Background and Objective : Radiation resistance(RR) is one of main determinants of treatment outcome in oral squamous cell carcinoma(OSCC), but accurate prediction of RR is difficult. We aim to establish RR OSCC cell lines and identify genes related with RR by a measurement of altered gene expression after inducing RR. Material and Methods : OSCC cell lines, SCC15, SCC25 and QLL1, were treated with 2Gy radiation per session, and parts of them were alive in finally accumulated dosage of 60Gy through 30 times repetition of radiotherapy for inducing RR cell lines. We compared results of cDNA array and proteomics in non-radiated cell lines and RR cell lines to detect changes of gene expression. Western blot was used for the validation of results. Results : cDNA array revealed 265 commonly up-regulated genes and 268 commonly down-regulated genes in 3 RR cell lines comparing their non-radiated counterpart. Among them, 30 cancer related genes were obtained. Proteomics showed 51 commonly altered protein expressions in 3 RR cell lines and 18 cancer related proteins were obtained. Among the detected genes, we found NM23-H1 and PA2G4 were over-expressed in both cDNA array and proteomics. Western blot showed increased expression of NME1 in RR cell lines but not in PA2G4. Conclusion: We concluded that NM23-H1 may be a candidate of RR related gene and over-expression of NM23-H1 could be a biomarker to predict RR in OSCC.

Cell Biological Function of Secretome of Adipose-Derived Stem Cells on Human Dermal Fibroblasts and Keratinocytes (인체 섬유아세포 및 케라티노사이트에 대한 지방줄기세포 분비물의 세포생물학적 기능)

  • The beneficial effects of adipose-derived stem cell conditioned media (ADSC-CM) for skin regeneration have previously been reported, despite the precise mechanism of how ADSC-CM promotes skin regeneration remaining unclear. ADSC-CM contains various secretomes and this may be a factor in it being a good resource for the treatment of skin conditions. It is also known that ADSC-CM produced in hypoxia conditions, in other words Advanced Adipose-Derived Stem cell Protein Extract (AAPE), has excellent skin regenerative properties. In this study, a human primary skin cell was devised to examine how AAPE affects human dermal fibroblast (HDF) and human keratinocyte (HK), which both play fundamental roles in skin regeneration. The promotion of collagen formation by HDFs was observed at 0.32 mg/ml of AAPE. AAPE treatment significantly stimulated stress fiber formation. DNA gene chips demonstrated that AAPE in HKs (p<0.05) affected the expression of 133 identifiable transcripts, which were associated with cell proliferation, migration, cell adhesion, and response to wounding. Twenty five identified proteins, including MMP, growth factor and cytokines such as CD54, FGF-2, GM-CSF, IL-4, IL-6, VEGF, TGF-${\beta}2$, TGF-${\beta}3$, MMP-1, MMP-10, and MMP-19, were contained in AAPE via antibody arrays. Thus, AAPE might activate the HK biological function and induce the collagen synthesis of HDF. These results demonstrate that AAPE has the potential to be used for clinic applications aimed at skin regeneration.
