민긴뿌리버섯(Oudemansiella radicata)의 자실체로부터 추출한 조다당류의 항암 및 면역 활성 효과에 관한 연구 (Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Fruiting Body of Oudemansiella radicata)

  • 민긴뿌리버섯은 송이버섯과에 속하는 버섯으로 예로부터 식용은 물론 항암, 고혈압 및 진균감염증의 치료에 널리 이용해온 식의약용 버섯이다. 민긴뿌리버섯의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하여 Sarcoma 180에 접종된 ICR mice에 주사하여 수명연장 및 항암효과를 조사하였다. 세포독성 실험결과, 각각의 세포는 $10{\sim}1000\;{\mu}g/ml$ 추출물 농도에서 70% 내외의 생존율을 보여 세포독성을 나타내지 않았다. 각각의 조다당류가 투여된 실험군이 대조군에 비해 평균수명이 각각 $42.9{\sim}66.7%$ 연장되었다. 중성염용액 추출물은 B 임파구의 alkaline phosphatase 활성을 대조군과 LPS군에 비해 약 $1.4{\sim}3$배 내외의 증가율을 보였다. 총 복강 세포수도 대조군에 비하여 최고 3.5배 정도 증가하였으며, 혈액 중 백혈구의 수도 대조군에 비하여 약 2.5배 증가하였다. 그리고 면역에 관련된 장기인 간, 비장 및 흉선의 체중이 대조군에 비하여 증가된 것을 확인하였다.

  • 뽕나무버섯부치는 주름버섯목, 송이버섯과에 속하는 버섯으로 예로부터 만성간염, 담낭염 및 암의 치료에 널리 이용해온 맛이 좋은 식의약용 버섯이다. 뽕나무버섯부치의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하여 Sarcoma 180가 접종된 ICR mice에 주사하여 항암 및 면역증강 효과를 조사하였다. 세포독성 실험결과, 각각의 세포는 $10{\sim}2000\;{\mu}g/ml$ 추출물 농도에서 세포독성을 나타내지 않았다. 각각의 조다당류가 투여된 실험군은 대조군에 비해 수명이 각각 $28.8{\sim}46.5%$ 연장되었다. 중성염용액으로 추출한 조다당류는 B 임파구의 alkaline phosphatase 활성을 대조군에 비해 약 $1.8{\sim}2.1$배 내외의 증가율을 나타냈다. 중성염추출 조다당류를 투여한 생쥐의 총 복강 세포 수는 대조군에 비하여 최고 9배 정도 증가하였으며, 혈액 중 백혈구의 수도 대조군에 비하여 약 1.9배 증가하였다. 또한 면역에 관련된 장기인 간, 비장 및 흥선의 체중이 대조군에 비하여 증가된 것을 확인하였다.

야생 비늘 버섯의 항암 성분에 관한 연구 (Studies on Antitumor Components of Wild Pholiota squarrosa (Fr.) Quel.)

  • In order to investigate the antitumor components of Korean wild higher fungi, the carpo-phores of Pholiota squarrosa collected in Kaung Nung area were extracted with water by refluxing for eight hours at 80-90.deg. C. The extracts were dialized for six days against distilled water and lyophilized to produce crude polysaccharide powders. They were found to have the antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Pholiota squarrosa was 78.7% in the dose of 20mg/kg/day for the period of ten days. The tumor in two of the ten mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The total polysaccharide was 42% by Anthrone method and the protein was 55% by Lowry-Folin method in the antitumor component of Pholiota squarrosa.

  • A series of 4-(2-chloroethyl)nitrosocarbamoyl- and 4-methyl nitrosocarbamoyl analogs of 4-Aza-$5{\alpha}$-cholestane (5a and 5b), 4-Aza-5-cholestene (6a and 6b) were synthesized and evaluated for their inhibitory activity against the Sarcoma 180 cell. The steroidal nitrosoureas, 5a, 5b, 6a and 6b displayed modest activity in vitro.

Antitumor and Immunostimulating Activities of $Elfvingia$ $applanata$ Hot Water Extract on Sarcoma 180 Tumor-bearing ICR Mice

  • $Elfvingia$ $applanata$, a medicinal mushroom belonging to Basidiomycota, has been used in the effort to cure cancers of the esophagus and stomach, and is also known to have inhibitory effects on hepatitis B virus infection. The hot water soluble fraction (as Fr. HW) was extracted from fruiting bodies of the mushroom. $In$ $vitro$ cytotoxicity tests showed that hot water extract was not cytotoxic against cancer cell lines such as Sarcoma 180, HT-29, HepG2, and TR at concentrations of 10-2,000 ${\mu}g/mL$. Intraperitoneal injection with Fr. HW resulted in a life prolongation effect of 45.2% in mice previously inoculated with Sarcoma 180. Treatment of Fr. HW resulted in a 2.53-fold increase in the numbers of murine spleen cells at a concentration of 50 ${\mu}g/mL$, compared with control. Incubation of murine spleen cells with Fr. HW at a concentration of 500 ${\mu}g/mL$ resulted in improved immune-potwntiating activity of B lymphocytes through an 8.3-folds increase in alkaline phosphatase activity, compared with control. Fr. HW generated 12.5 ${\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7, a mouse macrophage cell line, at the concentration of 50 ${\mu}g/mL$, while lipopolysaccharide, a positive control, produced 15.2 ${\mu}M$ of NO. Therefore, the results suggested that antitumor activities of Fr. HW from $E.$ $applanata$ might, in part, be due to host mediated immunostimulating activity.

  • The present study has been undertaken to detect edible medicinal plants with antineoplastic property on the basic of a number of traditional Korean medical literatures, besides studies on development of anti-cancer medical wild plants growing in Korea and to prove experimentally their efficacy by in vitro and in vivo tests.235 species from 45 family 79 genus were screened primarily as edible sources of antitumor effect. Among those the crude. extracts of 40 spp. showed considerable cytotoxicity in vitro and especially Pegangkuen(Patrinia scabiosaefolia), Deod-eog(Codonopsis lanceolata), Okssusu(Zea may), and Geureong(Eragrositis ferru-ginea) exhibited significant antitumor activity against sarcoma 180 asites mice. However, additional researches should be mode for the confirmation of their availability as antitumor plants.

韓國과 日本産 抗腫瘍性 資源의 Screening에 대하여 (Screening for Antitumor Efficacy from the medical plants in Korea and Japan)

  • 21plants, which collected from Korea and Japan, were applied to antitumor and cytotoxic screening tests against sarcoma 180 a ascitec in mice. The results are summariged as follows : 1) The total packed cell volume method has been used for the antimeoplastic screening for from natural higher plants in Korea. By this method, we have found out that Selaginella involves, Patrinia hispida, Archyranthes japonica and Solanum nigrum having significant activity and also Cydonia sinensis and Rubia akane showed slight activity to antitumor 2) The total packed cell volune method has been used for the antineoplastic screening for from natural higher plants in Japan. Among the 21 tested plants, lsodon japonicus having strong antitumor activity and also Torilis japonica, Aralia elata, Leonurus sibiricus and Rubia cordifolia showed significant activity to anticancer tumor while Forsythia spp and Solanum nigrum showed slight activity to antitumor. 3) Among the 21 tested Korea plants, lsodon excisus and Forsythia Koreana showed strong antitumor activity by the V79 cytotoxic cell screening test.

Antitumor Activity of Protein-Plysacharides Produced form Vibrio anguillarum

  • 해수로부터 분리 확인한 비브리오균에서 생산하는 단백 다당체의 항종양 활성에 대하여 조사하였다. 비브리오균을 0.5%-peptone와 0.5% yeast extract가 함유된 해수를 기질로 배양하여 단백 다당체를 얻었다. 단백다당체의 단당류의 함량은 xylose, mannose, galactose, glucose 및 fructose의 순서이고, 주요 아미노산 glutamic acid, serine 및 aspartic acid였다. 단백 다당체의 항종양 활성을 Sarcoma-180에 대하여 0.5mg/kg/day 및 5mg/kg/day을 쥐에 주사했던 바 20.9% 및 43.9%의 억제 효과가 있었다.

턱수염 버섯 배양(培養) 균사(菌絲)의 항종양(抗腫瘍) 성분(成分) (An Antitumor Constituent of the Cultured Mycelia of Hydnum repandum)

  • 한국산 담자균류의 항종양성 성분을 탐색하기 위하여 턱수염버섯의 균사를 분리하여 인공배지에서 진탕배 양후, 그 배양균사체로부터 수용셩 단백성 다당류를 추출하였다. 이 성분이 ICR마우스에 이식한 육종 180에 대하여 54.3%의 종양억제율을 나타내었다. 이 성분은 다당류 42%와 단백질 28%로 구성되어 있었다. 그 중 다당류는 glucose (57.8%), mannose(19.3%), galactose(10.8%), xylose (6.8%), 및 fucose (5.7%)를 함유하고 있었다.

밀버섯의 항암성분에 관한 연구 (Studies on Antitumor Components of Collybia confluens)

  • To find antitumor components from higher fungi, the mycelia of Collybia confluens (Pers. ex Fr.) Kummer were cultured in artificial media. For efficient production of the mycelia, the influences of various modifications of culture conditions were examined. A water-soluble protein-bound polysaccharide fraction, Fr. A, was obtained from the mycelia by hot water extraction. When Fr. A was purified and fractionated by DEAE-cellulose and Sepbadex G-200 gel filtration chromatographies into four fractions which were designated B, C, C-I and C-II. The tumor inhibition ratios of these fractions ranged from 46% to 75% against the solid forms of sarcoma 180 in ICR mice at doses of 20 and 50 mg/kg/day when given intraperitoneally. Especially, Fr. C which was named Collyban(CB) exhibited a marked life-prolonging effect of the mice against ascitic forms of sarcoma 180 at a dose of 50 mg via i.p. administration. To extend spectra of the antitumor activities and eliminate the effects of allograft rejection, the characterization of antitumor effects of CB was performed in syngeneic host-tumor systems. It did not show any antitumor activity against L1210 murine leukemia in $CD_2Fl$ mice but prolonged their life span against ascitic forms of $MM_{46}$ carcinoma in $C_3H/He$ mice. Also it exhibited antitumor activity against human cervical cancer HeLa cells that were xenografted into nude mice having BALB/c genetic backgrounds by the i.p. injection at a dose of 100 mg/kg/day. In order to characterize the antitumor components, CB was examined by chemical analysis. It was acidic protein-bound polysaccharides composed of 31% polysaccharide, 27% protein and 3% hexosamine. CB was fractionated into two fractions, Fr. C-I(M.W.: 500 Kd) and Fr. C-II(M.W.:30 and 8 Kd) by Sephadex G-200 gel filtration chromatography.
