Cigarette smoking (SM) is considered to be well known environmental toxin which contributes to the onset of various diseases. SM cause direct lungs damage, activate lungs inflammatory responses, and in some cases leads to the development of lung cancer. Cytokines in injured starfish (Asterina pectinifera) is the potential changes in its expression during the regeneration process. Especially, expression of TGF-${\beta}1$ has increased in arm cut starfish extract after eight days. Also, starfish including saponin like the ginseng. Saponin is widely used in the world because of some effective pharmacological activities. Therefore, the current study was designed to elucidate the pharmacological activities of starfish extract against cigarette smoking induced damage in cell line and pulmonary tissue. We investigate that the effect of eight days starfish extract after arm cut (8d) and intact starfish extract on cell line and mouse lung injury by SM. In cell proliferation analysis, although cigarette smoking extract (CSE) was co-treated, the higher proliferation ability is shown in 8d treatment than intact starfish extract. 8d and intact starfish extract was directly transported to pulmonary cells through respiratory organ by nebulizer inhalation. In this case of cigarette smoking, the pulmonary structure was damaged and functions become abnormal. However, 8d treated groups showed similar with the control group compared with SM group. Among them, 8d was proved to be more effective than intact starfish extract. These results demonstrate that 8d could more protect pulmonary structure and function than intact starfish extract against cigarette smoking by ginseng like saponin and regulation of inflammatory cytokines.

In this study, we investigated the effect of the extracts of Cyrtomium fortunei J.Sm. (CFJ) on lipopolysaccharide (LPS) induced inflammation in mouse BV-2 microglial cells. Nitric oxide (NO) production and cell viability were measured using the Griess reagent and the (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) (MTT) assay. Inflammatory cytokines were detected by quantitative polymerase chain reaction (qPCR) in BV-2 microglial cells with and without CFJ extracts. Subsequently, mitogen-activated protein kinases (MAPKs) and antioxidant markers were assessed by western blot analysis. It was found that the CFJ extract significantly decreased the production of pro-inflammatory cytokines (interleukin [IL]-6, tumor necrosis factor [TNF]-α, and IL-1β) and NO in BV-2 microglial cells that were stimulated with LPS. In addition, the expression levels of the phosphorylation of the MAPK family (p38, c-Jun N-terminal kinases [JNK], and extracellular-signal regulated kinase [ERK]) were reduced by CFJ. Also, treatment with CFJ significantly increased the activities of superoxide dismutase type 1(SOD1) and Catalase in BV-2 microglial cells. Our results indicate that CFJ has a potent suppressive effect on the pro-inflammatory responses of activated BV-2 microglia. Therefore, CFJ has the potential to be an effective treatment for neurodegenerative diseases, as it can inhibit the production of inflammatory mediators in activated BV-2 microglial cells.

Background: Despite its anticancer activity, cisplatin exhibits severe testicular toxicity when used in chemotherapy. Owing to its wide application in cancer therapy, the reduction of damage to normal tissue is of imminent clinical need. In this study, we evaluated the effects of catechin hydrate, a natural flavon-3-ol phytochemical, on cisplatin-induced testicular injury. Methods: Type 2 mouse spermatogonia (GC-1 spg cells) were treated with 0-100 μM catechin and cisplatin. Cell survival was estimated using a cell proliferation assay and Ki-67 immunostaining. Apoptosis was assessed via flow cytometry with the Dead Cell Apoptosis assay. To determine the antioxidant effects of catechin hydrate, Nrf2 expression was measured using qPCR and CellROX staining. The anti-inflammatory effects were evaluated by analyzing the gene and protein expression levels of iNOS and COX2 using qPCR and immunoblotting. Results: The 100 μM catechin hydrate treatment did not affect healthy GC-1 spg cells but, prevented cisplatin-induced GC-1 spg cell death via the regulation of anti-oxidants and inflammation-related molecules. In addition, the number of apoptotic cells, cleaved-caspase 3 level, and BAX gene expression levels were significantly reduced by catechin hydrate treatment in a cisplatin-induced GC-1 spg cell death model. In addition, antioxidant and anti-inflammatory marker genes, including Nrf2, iNOS, and COX2 were significantly downregulated by catechin hydrate treatment in cisplatintreated GC-1 cells. Conclusions: Our study contributes to the opportunity to reintroduce cisplatin into systemic anticancer treatment, with reduced testicular toxicity and restored fertility.
Objectives: This study investigated the anti-osteoarthritic effects of Kyejiinsam-tang (hereinafter referred to KIT) on the monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods: Anti-oxidative effects of KIT were measured by scavenging activities of DPPH, reactive oxygen species (ROS) and nitric oxide (NO). Scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were also measured for inhibitory effects against the production of inflammatory mediators (tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, interleukin-6). Osteoarthritis was induced in rats by injecting MIA in the knee joint. Rats were divided into a total of 4 groups (n=6). The normal group were not treated at all without inducing osteoarthritis whereas the control group were induced for osteoarthritis by MIA and oral medicated physiological saline per day. The positive comparison group was injected with MIA and after 7 days, 2 mg/kg of Indomethacin. The experimental group was injected with MIA and after 7 days was medicated with 34 mg/kg of KIT. Indomethacin and KIT were orally-medicated for each substance a total of 4 weeks, once per day. Weight-bearing on hind legs was measured every week after MIA injection. At the end of the experiment (5 weeks after MIA injection), micro CT (computed tomography)-arthrography and histopathological examinations on the articular structures of knee joint were performed. The effect on inflammatory cytokines and immunological cells in synovial fluid was measured. Volume of cartilage was measured by micro CT-arthrography. Injury to synovial tissue was measured by H & E (hematoxylin and eosin), Safranin-O immunofluorescence. Results: 1. Cytotoxicity against hFCs was insignificant. 2. KIT showed the potent full term for DPPH. 1. NO was significantly reduced by KIT (at 100, $200{\mu}g/m{\ell}$) and ROS was also reduced, but not significantly, by KIT (at $200{\mu}g/m{\ell}$). 2. IL-6 and IL-$1{\beta}$ were significantly reduced by KIT (at 100, $200{\mu}g/m{\ell}$) and TNF-${\alpha}$ was also reduced, but not significantly, by KIT (at $200{\mu}g/m{\ell}$). 1. In hind legs weight-bearing measurement, level of weight increased. 2. Functions of liver and kidney were not affected. 3. IL-$1{\beta}$ was significantly reduced and TNF-${\alpha}$, IL-6 were also reduced but not significantly. 4. PGE2 (prostaglandin E2), LTB4 (leukotriene B4) were significantly reduced in the KIT group. 5. MMP-9 (matrix metalloproteinase-9), TIMP-1 (tissue inhibitor of metalloproteinases-1) and Osteocalcin were significantly reduced in the KIT group. 6. Destruction of cartilage on micro CT arthrography was reduced but had no significant differences. 7. Histopathologically, injury to synovial membrane of the KIT group was decreased and proteoglycan content of KIT group was increased. Conclusions: According to this study, Kyejiinsam-tang has inhibiting effect on the progression of arthritis in MIA-induced osteoarthritis rat. Kyejiinsam-tang has anti-oxidants and anti-inflammation effects, and is related to inhibiting the activity of inflammatory cytokine and injury of volume in cartilage.

We studied anti-microbial and anti-oxidant activities of bio-active components in stem of Juniperus chinensis L. and leaf of Abies koreana Wilson. Those plants of needle leaf tree family were reported to contain anti-cancer compounds. The anti-bacterial activity was tested by Broth dilution method against Escherichia coli and Staphylococcus aureus. As results, Juniperus chinensis L. and Abies koreana Wilson extracts showed 17.0% and 8.5% higher anti-bacterial activity than methyl paraben, respectively. The free radical scavenging activity of Juniperus chinensis L. and Abies koreana Wilson extracts showed 45 % and 44 % at 5,000 ppm. We measured polyphenol (catechin equivalent) and flavonoids quantity. The Juniperus chinensis L. extract contained 312 mg/g of polyphenol and 105 mg/g of flavonoids. The Abies koreana Wilson extract contained 280 mg/g of polyphenol and 103.8 mg/g of flavonoids. The cytotoxicity of extracts was measured by neutral red assay. Extracts did not affect the viability of CCK-986sk cells up to a concentration of 1,250 ppm. In conclusion, these data suggest that extracts of needle leaf trees would be usefull as antiseptic agents and anti-oxidants for cosmetic products.

The Fabaceae family are being used as traditional medicine. The aim of this study was to compare the antioxidant effects as well as the cell protecting effects of extracts of 5 species (Astragalus membranaceus, Caesalpinia sappan L., Glycyrrhiza uralensis F., Pueraria lobate O., Pterocarpus santalinus L.) in Fabaceae family. The extracts from 5 species were tested by radical scavenging activity test, total phenolic contents and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on human liver carcinoma (HepG2) cell line. Anti-oxidant effects of the extracts (5 mg/mL) from C. sappan was 93.49% by radical scavenging activity test. In addition, A. membranaceus extracts showed a weak radical scavenging activity. Anti-oxidant effects of the extracts (5 mg/mL) from A. membranaceus was 7.83% by radical scavenging activity test. Total phenolic contents of the extracts from C. sappan and A. membranaceus were 310.93 mg GAE/g extract, 15.33 mg GAE/g extract, respectively. Cell protecting effects against H2O2 treatment were observed at 100 ㎍/mL concentration of C. sappan and P. santalinus extracts. These results suggest that C. sappan and P. santalinus might be best anti-oxidant in Fabaceae family.
This study was carried out to search for natural anti-oxidants and anti-inflammatory compounds from 5 medicinal plants (Carthamus tinctorius seed, Cyperus rotundus, Schizonepeta tenuifolia, Polygonatum odoratum var. pluriflorum, and Paeonia lactiflora). These plants were extracted with 70% ethanol. In order to measure total antioxidant activity of flavonoids, polyphenol content was measured. Radical scavenging activities of extracts were examined using a-a-Diphenyl-${\beta}$-picrylhydrazyl ($DPPH{\cdot}$), 2,2-azino-bis 3-ethylbenzthiazoline-6-sulfonic acid ($ABTS{\cdot}$), ferric reducing antioxidant power (FRAP) and superoxide anion radical assays. C. tinctorius seed extracts showed the highest polyphenol and flavonoid contents as well as strong $DPPH{\cdot}$, $ABTS{\cdot}$, FRAP, and superoxide anion radical scavenging activity. Also, C. tinctorius seed extracts showed the highest nitric oxide (NO) production inhibitory effect. Theses results indicate that the C. tinctorius seed extracts can be used as a functional material due to their effective anti-oxidative and antiinflammatory activities.
In modern society, interest in antioxidants is increasing as the stress caused by oxidants increases. However, the demand for synthetic antioxidants is decreasing because some studies have confirmed that they are harmful when consumed in large quantities; thus, studies on antioxidants derived from natural substances are actively being conducted to replace synthetic antioxidants. Blueberry, known as one of the world's top ten long-lived foods, is a plant of the Vaccinium (Ericaceae) family, and various pharmacological activities of blueberry including antioxidant activity have been studied. Vaccinium oldhamii (VO) is a deciduous broad-leaved shrub in the same genus as blueberries, and in this paper, we summarize the studies on the efficacy analysis of VO extracts and purified products. The content of phenolic compounds in VO fruits was proportional to antioxidant and anti-influenza activity such as the inhibition of NO production, and the total content of polyphenols and anthocyanin was higher than that in blueberries. VO fruit extracts showed anti-inflammatory activity and anti-cancer activity against human acute leukemia; in contrast, VO branch extracts showed anti-inflammatory activity, activity to inhibit osteoclast differentiation and bone resorption due to inflammatory response, and anti-cancer activity against several human cancer cell lines. Compared to blueberries, VO showed higher phenolic compound content, antioxidant activity, and various physiological activities. In addition, VO is considered to have sufficient value as an alternative crop to blueberries, such as it can be grown natively in Korea, with simple mass cultivation and no need to pay royalties for commercialization.

Baicalin, a biologically active flavonoid form the roots of Scutallaria baicalensis (Skullcap), have been reported to not only function as anti-oxidants but also cause anticancer effect. We investigated the mechanism of baicalin-induced cytotoxicity and the macro scale gene expression analysis in leukemia cell line, HL-60 cells. Baicalin (10 μM) were used to treat the cells for 6h, 12h, 24h, 48h and 72h. In a human cDNAchip study of 65,000 genes evaluated 6, 12, 24, 48. 72 hours after treated with Baicalin in HL-60 cells. Hierarchical cluster against the genes which showed expression changes by more than two fold. One hundred one genes were grouped into 6 clusters according to their profile of expression by a hierarchical clustering algorithm. For genes differentially expressed in response to baicalin treatment, we tested functional classes based on Gene Ontology (GO) terms. This study provides the most comprehensive available survey of gene expression changes in response to baicalin treatment in HL-60 cell line.
The present study was designed to investigate the effects of genistein, daidzein, and quercetin on the antioxidative systems of normal rats. Male Sprague-Dawley rats were divided randomly into seven groups and treated with flavonoids at either 2 or 20 mg/day or through vehicle for four weeks. Lipid peroxidation in the liver was inhibited significantly following administration of quercetin. Genistein and daidzein did not have significant effects except in rats treated with 20mg daidzein/day. Genistein and daidzein treatment did not affect the content of $\alpha$-tocopherol in the serum and liver, while quercetin caused a slight increase. In hepatic glutathione and its related enzymes, genistein and daidzein treatment tended to cause a decrease in $\alpha$-tocopherol content, although no significant difference was found. However, quercetin treatment significantly decreased the content of glutathione together with the activity of glutathione reductase in all doses in the liver but there was no significant difference in the brain. Interestingly, daidzein treatment in the brain at 2mg/day significantly increased glutathione (27.1% p<0.05) compared with the control group, while at 20mg/day glutathione decreased significantly (26.6%, p<0.05). In conclusion, genistein has not antioxidant effects. Daidzein quercetin may have the capacity to produce not only antioxidants but also have adverse effects including the production of pro-oxidants. Therefore, people should consider consumption at a high dosage.
