갈근이 비만 랫드 간조직의 비만관련 유전자 발현에 미치는 영향 (The effects of Pueraria lobata extract on gene expression in liver tissue of rat with estrogen-deficient obesity)

  • Objective : It is known that Pueriaria lobata has an anti-osteoporetic effect, anti-cancer effect, anti-pyretic effect, and anti-diabetic effect. The aim of this study was to evaluate anti-obesity effect of Pueriaria lobata extract (PLE), and elucidate the effect of it on gene expression related to lipid metabolism. Method : The experiments were performed with the use of ovariectomized rats as estrogen-deficient obesity model. They were grouped NC (normal control), OC (estrogen-deficient control), PLH (100mg/kg of PLE), PLL (20mg/kg). PLE was orally administered for 6 weeks. Body weights and serum lipid level were estimated, and real-time PCR was performed to investigate the effect of PLE on gene expression in liver. Results : PLE decreased the body weight and serum cholesterol and triglyceride, but increased HDL-cholesterol. And PLE increased leptin, CYP27, CPT1, CYP8B1, ACAT2, LDLR, and SCD1, but reduced $PPAR{\gamma}$, PGC1A, HMG-CoA-R, ACAT1, SCD1, and APoB gene expression in liver tissue of rat with estrogen-deficient obesity. Conclusion : It is concluded that Pueriaria lobata reduced body weight, and its effect was expressed by regulation of gene expression related to lipid metabolism in rats with estrogen-deficient obesity.

Effects of Purslane Extract on Obesity and Diabetes in High-Fat Diet-Induced Obese Mice

  • The frequency of obesity has risen dramatically in recent years but only few safe and effective drugs are currently available. In addition, obesity can induce type 2 diabetes (T2DM), hyperlipidemia and fatty liver disease. Recently, protective effect of purslane extract (PE) on obesity has been reported, but little is known about the role and mechanism of PE in obesity. This study aimed to evaluate the effect of PE on obesity and diabetes in obese mice. In addition, the effect of PE was compared with anti-obesity and diabetes drugs. High-fat diet (HFD)-induced obese mice were treated for 8 weeks with drugs as follows: PE, orlistat, metformin, voglibose or pioglitazone. While PE mixed with normal diet did not have any effects on BW in non-obese mice, PE mixed with HFD significantly reduced BW gain, insulin resistance, and glucose intolerance, without affecting food intake and appetite in obese mice. The effect was comparable to the effects of anti-obesity and diabetes drugs. Furthermore, PE significantly increased the activity of hepatocellular anti-oxidant enzymes, leading to protection of liver from oxidative stress in obese mice. These results suggest that PE treatment may be a useful tool for preventing obesity and complication of obesity.

In vitro Anti-obesity Effect of 4-hydroxybenzyl Alcohol from Cudrania tricuspidata

  • The present study was investigated on in vitro anti-obesity effect of 4-hydroxybenzyl alcohol from Cudrania tricuspidata. We isolated various compounds from Cudrania tricuspidata. Among these compounds, anti-obesity effects of 4-hydroxybenzyl alcohol was examined by lipase activity assay, cyclic adenosine monophosphate (cAMP)-specific phosphodiesterase type IV (PDE4) activity assay, and citrate synthase activity assay. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts inhibited the enzymatic activities of lipase, PDE4, and citrate synthase. Lipase is known to mediate the hydrolysis of triacylglycerol in adipose tissue and cholesterol esters in other tissue or cells. Also, PDE4 hydrolyses cAMP, a crucial secondary messenger for in metabolic pathways including glucose and lipid metabolism, lipolysis, and thermogenic function. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts induced the inhibitory effect against each enzymatic activity on several specific substrates as observed by detection at 405 or 412 nm. These findings might be attributable to the inhibition of adipogenesis, and partial prevention of obesity. In conclusion, these results show that 4-hydroxybenzyl alcohol and Cudrania tricuspidata may be a critical candidate as a natural anti-obesity source.

고지방식이로 유도된 C57BL/6J 마우스 비만모델에서 참마홍국발효의 비만 억제 및 지질 저하 효과 (Anti-Obesity and Lipid Lowering Effect of Discorea japonica Thunb. Fermented with Monascus in High-Fat Diet Induced Obese C57BL/6J Mice Model)

  • This study was conducted to investigate the anti-obesity effect of Discorea Japonica Thunb. fermented with Monascus After inducing obesity by feeding hight fat diet (diet induced obesity model: DIO) for four weeks, each 8 rats were assigned to normal (Nor), high fat diet (HF), high fat diet containing orlistat (PC), high fat diet containing different concentration of Discorea Japonica Thunb. fermented with Monascus (UPDM_L, UPDM_H) and Discorea Japonica Thunb. (UPD) extract. Although the UPD, UPDM_L (ultrafine pulverized Discorea Japonica Thunb. fermented with Monascus: 400 mg/kg) and UPDM_H (DIO oral administration ultrafine pulverized Discorea Japonica Thunb. fermented with Monascus: 800 mg/kg) showed weight gain inhibition effects, the results of poor obesity inhibition rather than PC were confirmed. However, it showed a more effective weight loss effect in UPDM_H than UPD, and significantly reduced the weight of epididymal fat and subcutaneous fat. Furthermore, the possibility of anti-obesity effects of Discorea Japonica Thunb. fermented with Monascus can be confirmed by observing the effects of reducing serum total cholesterol, triglyceride and LDL concentrations, reducing ALT and AST levels, and inhibiting fat build-up in liver tissue. It is believed that Discorea Japonica Thunb. fermented with Monascus can be expected to utilize as a functional material that is important to improve anti-obesity and metabolic syndrome.

복분자 함유 생약추출물의 체중 조절 효과에 관한 연구 (Effect of Decreasing Body Weight with Plant Extracts Containing Rubi Fructus)

  • Obesity is one of causes of the all adult diseases. We investigated the body weight decrease effect of the selected plants by digestive enzyme activity inhibition test. In a preliminary test, Inonotus obliquus and Rubus coreanus Miq. were found to be effective. Based on this result & previous result, we manufactured the mixture of plant extracts named as Misol$^{TM}$. Misol is applied anti-obesity beverage and rice. When we administered Miso/ sub TM/ to rats, it was found to be effective in body weight decrease. Result that is experimented during 20 weeks, is effective body weight and total cholesterol, triglyceride decrease. In case of human, anti-obesity beverage administered group showed 5.65% body weight decrease, 4% waist length decrease and 6% abdomen length decrease respectively. And anti-obesity rice administered group showed 2.3 kg body weight decrease, 2.9 cm waist length decrease and 3.74 cm abdomen length decrease respectively. This body control effect was supported by plant extracts in the Misol, anti-obesity beverage and rice - administered group. From these results, we suggest that this functional food could be helpful for body weight controlling obesity.

Anti-obesity Effect of Black Vinegar Fermented with Herbal Extracts

  • Vinegar has been widely produced for a variety of industrial and domestic use as well as medicinal use. For sale of the commercial vinegar with herbal extracts, we produced an experimental black vinegar through sequential fermentation of alcohol, followed by acetic acid according to the manufacturer's procedure. To investigate the effect of anti-obesity of black vinegar on biochemical values, we evaluated enzyme activities via acetyl-CoA carboxylase (ACC), which plays a critical role in the lipid metabolism. We found that increased phosphorylated adenosine monophosphate (AMP) activated protein kinase (AMPK) and ACC in L6 mouse muscle cells treated with the manufactured vinegar. Based on the results, supplementation of experimental herbal black vinegar inactivates ACC, enhancing the phosphorylation of AMPK. Thus, the lipid oxidation and inhibitory effect of fatty acid synthesis by the black vinegar expects to facilitate the anti-obesity activity.

고수 에탄올 추출물의 고지방식이 비만 동물모델에서의 항비만효과 (Ant-Obesity Effect of Coriandrum sativum L. Ethanol Extract in High Fat-Induced Obesity Animal Model)

  • This study investigated the anti-obesity effects of Coriandrum sativum L. ethanol extracts in a high fat diet-induced obesity model (DIO). We confirmed the anti-obesity effects by analysing the expression of the related proteins, weight gain, dietary intake, dietary efficiency, blood biochemistry, histological analysis and western blot analysis. After oral administration of Coriandrum sativumL. ethanol extracts at concentrations of 250 and 500 mg/kg, a significant improvement in dietary efficiency, reduction in weight gain, triglycerides, total cholesterol and LDL-cholesterol in blood lipid was observed for 8 weeks. In addition, improvement in blood glucose and metabolism confirmed through glucose tolerance test was observed. Further, the concentration of alanine transaminase (ALT) in blood was significantly decreased, which improved the fatty liver caused by high-fat diet intake as confirmed by liver tissue analysis. This phenomenon was confirmed to decrease the expression of fat accumulation-related PPARγ and FAS protein in the liver tissue. Especially, it is believed that FAS, a liposynthetic enzyme, has a stronger inhibitory effect than PPARγ. Therefore, Coriandrum sativum L. ethanol extract is thought to improve obesity by reducing blood lipids levels, improving glucose metabolism and inhibiting synthesis of the fat that accumulates in the liver in high-fat diet-induced obesity animal models.

의이인탕 구성약물의 비만관련 연구 분석 (Analysis of the Obesity-Related Research for Each Constituent Herb of Euiiin-tang)

  • The purpose of this study is analyzed of exploratory research potential as anti-obesity agents of Euiiin-tang. Four Korean databases and 2 Korean Journals (Riss4U, KISS, OASIS, DBPIA, and Journal of Korean Rehabilitation Medicine, Journal of Korean Medicine of Obesity Research) were searched using search word 'individual herbs' and 'obesity', 'weight loss', 'fat', 'hypertension', 'hyperlipidemia', 'diabetes'. Clinical and Experimental Research published in the journal were analyzed, review research, studies of pharmacopunctures and studies of mixed herbal medicine were excluded. We collected 23 studies. Seven studies of Coicis Semen, 10 stdies of Ephedra Herba, 2 study of Angelica gigas Nakai, 3 studies Atractylodis Rhizoma Alba, 1 studies of Glycyrrhiza inflata Batal. Most studies were experiment researches which were composed of in vivo or in vitro, and clinical trial was 5 studies of Ephedra Herba. Main constituent herb, Coicis Semen, Ephedra Herba were thought to represent an anti-obesity effect. Through the result, we can assume to be likely effect of Euiiin-tang as obesity medicine.

죽엽(竹葉)과 황금(黃芩) 복합물의 항비만 효과 (Synergistic combination effect of anti-obesity in the extracts of Phyllostachys pubescence Mael and Scutellaria baicalensis Georgi)

  • Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and considerable adverse effects affecting tolerability and safety. Therefore, most anti-obesity durgs have been withdrawn. We tried to develop anti-obesity agent by combinations from herbs that are used in food ingredients as well as in traditional medicines. Methods : The 80% (v/v) ethanol extracts from Bamboo (Phyllostachys pubescence) leaf (BL) and Scutellaria baicalensis (SB) and their 1:1 combination (BLSB) was evaluated on high fat diet induced obese mice compared to Omega-3 as a positive control. The mice were divided into six groups (n=5), one group fed a normal diet (ND), and the others fed a high fat diets for eight weeks. Two weeks after starting feeding the diets, the high fat diet groups were orally administered vehicle and Omega-3, BL, SB, and BLSB at dosage of 200 mg/kg/day for six weeks. All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : BLSB group showed significant reductions in body weight gain and fat weights of liver and epididymal adipose tissue compared to BL or SB alone as well as control. Total-cholesterol and LDL-cholesterol levels significantly decreased, and HDL-cholesterol level increased. In liver tissue, macrovesicular steaotisis was remarkably improved and its fat cell size was also significantly decreased. Conclusions : These results suggested that a combination preparation of bamboo leaf and S. baicalensis has anti-obesity effect and have synergistic effect compared to bamboo leaf or S. baicalesis.

Anti-Obesity Effects of Red Beet Extract

  • Obesity is caused by the accumulation of triglycerides in adipocytes by the differentiation and lipid synthesis process of pre-adipocytes, and excessive accumulation of adipocytes by the activated Adipogenesis process within the differentiated cells. Therefore, inhibiting the differentiation of adipocyte cells or controlling the adipogenesis process is known as an effective treatment method for obesity. This study evaluates the inhibition of Red beet root extract on pancreatic lipase and pre-adipocyte cell differentiation. Also it evaluates the Red beet root extract activities on C/EBP-𝛼,𝛽, and PPAR-𝛄. The experiments proved that the Red beet root extract inhibits pancreatic lipase by concentration dependency. Further, in 3T3-L1 inhabitation experiment, it was found Red beet root extract inhibited adipocyte formation. Red beet root extract also inhibits the expression of C/EBP-𝛼, C/EBP-𝛽, and PPAR-𝛾 which effect the process of adipocytic differentiation. We therefore concluded that RBE has a high potential to further studies on anti-obesity effect.
