Establishment of Miniaturized Cultivation Method for Large and Rapid Screening of High-yielding Monascus Mutants, and Enhanced Production of Monacolin-K through Statistical Optimization of Production Medium (Monascus 균사체의 소규모 배양을 통한 고생산성 균주의 대규모 선별방법 확립과 통계적 생산배지 최적화를 통한 Monacolin-K 생산성 향상)

  • It is crucial to develop a miniaturized cultivation method for large and rapid screening of high-yielding mutants of monacolin-K, a powerful anti-hypercholesterolemic secondary metabolite biosynthesized by the fungal cells of Monascus ruber. In order to investigate as many strains as possible in a short time, a miniaturized fermentation method especially suitable for the cultivation of the filamentous Monascus mutants was developed using $50m{\ell}$ culture-tube ($7m{\ell}$ of working volume) instead of the traditional $250m{\ell}$ flask ($50m{\ell}$ of working volume). Generally, in filamentous fungal cell fermentations, morphologies in growth and production cultures should be maintained as thick filamentous and compact-pelleted (usually less than 1 mm in diameter) forms, respectively, for enhanced production of secondary metabolites in final production cultures. In this study, we intended to induce the respective optimal morphologies in the miniaturized culture system for the purpose of rapid screening of overproducers. Miniaturized growth culture system was successfully developed due to the mass production of spores in the statistically optimized solid medium. When large amounts of spores were inoculated into the growth cultures, and brown rice flour (20 g/L) was also supplemented to the growth medium, dense filamentous morphologies were successfully induced in the growth cultures performed with the 50 ml culture tubes. It was implied that the amounts of spores inoculated into the growth tube-cultures and the growth medium components should be the key factors for the induction of the filamentous forms in the growth fermentations. Furthermore, in order to statistically optimize production medium, multiple experiments based on Plackett-Burman design and response surface method (RSM) were carried out, resulting in more than 2 fold enhanced production of monacolin-K in the final production cultures with the optimized production medium. Notably, under the production culture conditions with the statistically optimized medium, optimal pellet sizes below 1 mm in diameter were reproducibly induced, in contrast to the thick and viscous filamentous morphologies observed in the previous production cultures.

Effects of Processing Method on Performance and Nutrient Digestibility in Growing-finishing Pigs Fed Lupine Seeds

  • Three trials were conducted to study the effect of processing method on performance, apparent nutrient and amino acid digestibilities in growing-finishing pigs fed lupine seeds. Ground, expanded or flaked lupine seed was prepared for the trials. In trial 1, a total of 72 growing barrows ($Landrace{\times}Large$ $White{\times}Large$, initial BW of $54.22{\pm}5.87$ kg) were allocated to three treatment diets containing 15% percent of differently processed lupine seed for 28 days. Each treatment had four replicates comprising 6 pigs each. In trial 2, a total of 48 finishing barrows ($Landrace{\times}Large$ $White{\times}Large$, initial BW of $84.40{\pm}6.84$ kg) were subjected to three treatments for 21 days. The experimental design was the same as trial 1 except for supplementation with 20% of lupine seed. Each treatment had four replicates comprising 4 pigs each. To evaluate the ileal digestibility of amino acids, a total of 9 barrows ($Landrace{\times}Large$ $White{\times}Large$), with an average initial BW of $41.07{\pm}1.98$ kg, were fed with ground, expanded or flaked lupine for 7 days. Each pig was surgically equipped with a simple T-cannula fitted in the distal ileum. Amino acid composition and presence of anti-nutritional factors (ANFs) in processed lupine sources were also evaluated. The results showed that there was no significant difference (p>0.05) in lupine amino acid composition due to the different processing methods. Expanded and flaked lupine significantly decreased (p<0.05) the concentration of ANFs compared with ground lupine. There was no effect (p>0.05) on the growth performance in growing pigs. However, processing method had a significant effect (p<0.05) on average daily gain (ADG), average daily feed intake (ADFI) and feed conversion ratio (FCR) in finishing pigs. There was no effect (p>0.05) of the different processing methods on the digestibility of dry matter (DM), gross energy (GE), ether extract (EE), Calcium (Ca) and phosphorus (P), but expanded lupine could significantly increase (p<0.05) the apparent digestibility of CP in finishing pigs. Furthermore, expanded lupine had a higher (p<0.05) apparent ileal digestibility for most indispensable and dispensable amino acids compared with ground and flaked lupine. It was concluded that expanded lupine could be beneficial in improving lupine's quality and improve performance and nutrient utilization in growing-finishing pigs.

Effect of the Oriental Medicine Gi-Gong Exercise on the Brain Power, HRV, Live Blood Condition in the Youth (한방기공체조가 두뇌력, 심박변이율, 생혈액형태에 미치는 영향)

  • This study was conducted to identify the effects of the oriental medicine Qigong Exercise on the brain power, HRV, pulsation, live blood condition among young boys and girls. The study was performed with two group(control group and experimental group) in a pre-test/post-test design. The subjects were 44 young boys and girls selected by a some middle school in Busan. The oriental medicine Qigong Exercise program consists of 80-minute sessions three times a week over 5 months. All of the subjects were examined on the congnition assessment tool, stress assessment tool, oriental medicine pulsation 3-D MAC, live blood condition analyzer Prior and post surveys were measured before and after the experiment. In the cognition assessment, the amplitude of ERS were increased afer Qigong Exercise. The Success and the Concentration were significantly increased afer Qigong Exercise, the Error was significantly decreased afer Qigong Exercise. The Cognition strength was significantly increased, but the Reponse time was not significantly decreased afer Qigong Exercise. And the Workload was not significantly decreased, the Total Score was not significantly increased afer Qigong Exercise. Among the stress assessment, RRV tachogram's ‘mean RR’ was significantly increased, ‘mean HRV’ was significantly decreased afer Qigong Exercise. SDNN was not significantly increased, Complexity was not significantly increased afer Qigong Exercise. And TP(RRV power's total power) was not significantly increased, VLF and HF was significantly increased, and LF was significantly decreased afer Qigong Exercise. ANS's norm LF was not significantly decreased, but norm HF was significantly increased afer Qigong Exercise. In the RRV's Phase Plot, RMSSD and SDSD were not significantly increased, pnn50 was not significantly decreased afer Qigong Exercise. On the whole, Parasympathetic Activity and Stress Endurance were significantly increased, but Cardiac Activity and Physical Arousal were not significantly increased afer Qigong Exercise. Cardiac Aging was not significantly decreased afer Qigong Exercise. Sympathetic Activity, Autonomic Nervous System Balance and Heart-load were not significantly decreased afer Qigong Exercise. In the pulsation, press power was increased(15%), and w/t(pressurization time / pulsation time) was decreased(20%) afer Oigong Exercise. And the live blood condition was not changed afer Qigong Exercise. As mentioned above; the oriental medicine Qigong Exercise program was identified the effects of the inspiration of the brain power, heart rate and anti-stress.

Comparative Modeling of Human Tyrosinase - an Important Target for Developing Skin Whitening Agents (피부 미백제의 타겟 단백질인 인간 티로시나제의 3차원 구조 상동 모델링)

  • Human tyrosinase (hTyr) catalyzes the first and rate limiting step in the biosynthesis of a skin color determinant, melanin. Although a number of cosmetic companies have tried to develop hTyr inhibitors for several decades, absence of 3D structure of hTyr make it impossible to design or screen inhibitors by structure-based approach. Therefore, we built a 3D structure by comparative modeling technique based on the crystal structure of tyrosinase from Bacillus megaterium to provide structural information and to search new hit compounds from database. Our model revealed that two copper atoms of active site located deep inside and were coordinated with six strictly conserved histidine residues coming from four-helix-bundle. Substrate binding site had narrow funnel like shape and its entrance was wide and exposed to solvent. In addition, hTyr-tyrosine and hTyr-kojic acid, a well-known inhibitor, complexes were modeled with the guide of solvent accessible surface generated by in-house software. Our model demonstrated that only phenol group or its analogs could fill the binding site near the nuclear copper center, because inside of binding site had narrow shape relatively. In conclusion, the results of this study may provide helpful information for designing and screening new anti-melanogenic agents.

Analysis of Fire Risk Assessment Indicators of Publicly-Used Establishments using Delphi/AHP (Delphi/AHP를 활용한 다중이용업 신종업종의 화재위험평가지표 분석)

  • Through a press release dated July 17, 2018, the Anti-Corruption and Civil Rights Commission recommended that the National Fire Agency develop preventive measures against fire in the "Indoor Archery Ground" and "Room Escape Café" etc., which were originally excluded from the category of "Publicly Used Establishments." This study developed the hierarchy of domains and indicators of measurement for fire risk assessment of the new business of publicly used establishments through the Delphi Method. It analyzed the goodness of fit scores (over 3.00) and secured an average score of 4.25. Using AHP analysis, the ratio of consistency for the domains of measurement of fire risk assessment was found to be 4.0%, which was lower than CR ≤ 0.1 (10%). The consistency of subsequent measurement indicators were distributed in the range of 0.1%~3.6%, and they were identified as being commonly consistent. The indicators of measurement appeared as follows in order of importance and priority: Type of Internal Passage of Establishment and Evacuation Capacity of Exit (0.316), Control of Ignition Source (0.141), Inherent Risk (0.106), Appropriateness and Adaptiveness of Fire Detection System (0.097), Control of Inflammables/Combustibles (0.084), Guides and Facilities helping Evacuation (0.075), Fire Resistant Structure and Finishing Materials (0.060), Compartmentalization and Emergency Exit (0.049), Risk of Fire Expansion (0.046), and Appropriateness and Adaptiveness of Fire Extinguishing Facilities (0.026). The findings of this study are expected to be expansively used as data for future research on the development of fire risk assessment indicators.

Convergence-based analysis on geographical variations of the smoking rates (융복합 기반의 지역간 흡연율의 변이 분석)

  • This study aims to identify geographical variations and factors that affect smoking rates. The data are collected from the Community Health Survey conducted between 2009 and 2011 by Korea Centers for Disease Control and Prevention and other government organizations. Correlation and multiple regression analysis were used to examine the factors influencing smoking rates. For the purpose of investigating regional variations, we employed a decision tree model. The study has found that the significant factors associated with geographical variations in the smoking rates were the rate of hazardous drinking, the completion rate of hypertension education, the experience rate of anti-smoking campaigns, stress awareness rate, hypertension prevalence, health insurance cost, diabetes prevalence, obesity rate, and strength training rate. Convergence-based analysis on geographical variations of the smoking rates is highly important when the regionally customized healthcare programs is implemented. In the future, it is necessary to develop effective program and customized approach for the regions of high smoking rates. Our study is expected to be used as meaningful data for the design of effective health care programs and assessments to lead effective non-smoking program.

Development of Additive to Modify the SDAR (Solvent DeAsphalting Residue) and Laboratory Performance Evaluation of Asphalt Mixture with Modified SDAR (고품위화 정제공정 부산물(SDAR) 활용을 위한 첨가제 개발 및 이를 이용한 아스팔트 혼합물의 실내 공용성능 평가)

  • OBJECTIVES : The objective of this research is to develop additives for the modification of Solvent DeAsphalting Residue (SDAR) to be used as pavement materials, and evaluate the performance of asphalt mixture manufactured using the SDAR modified by developed additives. METHODS : The SDAR generally consists of more asphaltenes and less oil components compared to the conventional asphalt binder, and hence, the chemical/physical properties of SDAR are different from that of conventional asphalt binder. In this research, the additives are developed using the low molecular oil-based plasticizer to improve the properties of SDAR. First, the chemical property of two SDARs is analyzed using SARA (saturate, aromatic, resin, and asphaltene) method. The physical/rheological properties of SDARs and SDARs containing additives are also evaluated based on PG-grade method and dynamic shear-modulus master curve. Second, various laboratory tests are conducted for the asphalt mixture manufactured using the SDAR modified with additives. The laboratory tests conducted in this study include the mix design, compactibility analysis, indirect tensile test for moisture susceptibility, dynamic modulus test for rheological property, wheel-tracking test for rutting performance, and direct tension fatigue test for cracking performance. RESULTS : The PG-grade of SDARs is higher than PG 76 in high temperature grades and immeasurable in low temperature grades. The dynamic shear modulus of SDARs is much higher than that of conventional asphalt, but the modified SDARs with additives show similar modulus compared to that of conventional asphalt. The moisture susceptibility of asphalt mixture with modified SDARs is good if, the anti-stripping agent is included. The performance (dynamic modulus, rutting resistance, and fatigue resistance) of asphalt mixture with modified SDARs is comparable to that of conventional asphalt mixture when appropriate amount of additives is added. CONCLUSIONS : The saturate component of SDARs is much less than that of conventional asphalt, and hence, it is too hard and brittle to be used as pavement materials. However, the modified SDARs with developed additives show comparable or better rheological/physical properties compared to that of conventional asphalt depending on the type of SDAR and the amount of additives used.

Comparative Study on the Effects of Six Species of Microalgae Extracts on Hair Loss Prevention and Scalp Improvement (6종의 미세조류 추출물의 탈모예방 및 두피 개선 효과 비교)

  • This study examines the role of six species of microalgae, including Phaeodacylum tricrnutum (PT), Chaetoceros gracilis (CG), Nanochloris oculata (NO), Pavlova lutheri (PL), Chlorella ellipsoidae (CE), and Scendedemus obliquus (SO), on hair loss prevention and scalp improvement. To determine the effects of microalgae extracts on hair loss prevention and scalp improvement, antioxidant activity, cell proliferation in HaCaT cells and HFDPC cells, and the inhibition level of 5-alpha reductase activity were examined. In the study of antioxidant activity, the $EC_{50}$ values of DPPH anti-radical activities indicated that the SO, CG, and ST9 treatment groups demonstrate significant antioxidant activity. In the study of the hydroxyl radical scavenging activity, CG (6.6~42.1%), ST9 (26.0~44.0%), and SO (7.8~44.3%) demonstrated significant effects. Furthermore, SO promoted the proliferation of HaCaT cells and a human epidermal cell line during a 6-day treatment. In the study of the proliferation of HFDPC cells, a hair follicle dermal papilla cell line, CG, and SO significantly stimulated cell proliferation. Finally, PT, CG, and SO significantly inhibited 5-alpha reductase activity. These results suggest that among the six microalgae used in this study, CG and SO have antioxidant effects, induce cell proliferation, inhibit 5-alpha reductase activity, and can be used for hair loss prevention and scalp improvement.

Effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs

  • Soybean, one of most widely used swine feed component in the world, contains non-starch polysaccharides (NSP). The digestive system of weaned pigs is not yet fully developed, and thus weaned pigs cannot easily digest diets based on corn and soybean meal. Dietary exogenous enzymes supplementation has been intensively investigated to assist digestion of anti-nutritional factors, such as NSP. This experiment was conducted to investigate the effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; mixture of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Growth performance, morphology of ileum, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter, crude protein, and energy of weaned pigs were measured. No significant differences (p > 0.05) were observed for growth performance for the duration of the experimental period, and morphology of ileum, and nutrient digestibility between CON and Cocktail treatment groups. Therefore, the results from the current study indicated that enzyme cocktail supplementation in diets had no influence on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.

  • This study aims at providing useful fundamental information to re-establish the theories of modern fashion by examining the origination and genealogy of street style. The street styles focusing on caucasoid have a variety of genealogies such as western type, beat, teddy boy, hippie, skinhead, punk, neuron-mantic, indie kid, riot grrrl, grunge and techno cyber punk. In the same period, on the contrary, the streets styles focusing on negroid are zootie, hipster, modernist, rude boy, two-tone, rastafarian, funky, B-boy, fly girl, raggamuffine, bhangra, and acid jazz, which are seen as the culture of the large cities formed along Atlantic Ocean and Caribbean sea like England, America and Jamaica. These have root as the main fashion in western society. Ironically, most of the subculture concentrated on the whites were racists. Because of such a reason, the street styles have been formed as resistance culture that was unable to sympathize with their society and characteristics by distinguishing the whites and the colored people. Zootie or hipster that is one of the street fashion styles was formed in the 1940-50s, while the colored people who lived in the west Indies migrated to England or America. As a minimal modernist style called Ivy look in US, in that time, anti-culture formed by teenagers in whitey, teddy boy and mods fashion can be strictly different from the zootie and hipster. The colored people's street styles of the 1960s developed into aggressive and hard forms from the rude boy and two-tone while their resistance toward the whites was stronger. The rastafarian style researched the peak as the colored people's traditional ethnic characteristics or resistance intention for their freedom in the 1970s. With B-boy, fly girl, reggae, rap music, and break dancing in the 1980s, the subculture gradually told on the high fashion as well as the culture between the whites and the colored people. From such aspects, the colored people tried to maintain their unique traditional characteristics. However, their individual values surged by the coming young generation excluded the colored people's characteristic street styles. Focusing on gender, violence and private success among their major concerns, the raga muffin style that represents multi-races and multi-cultures was formed. The jazz style in the 1990s showed cold post-modernistic eclecticism different from that of the 1940s-50s. Simultaneously, the various classes appeared their street styles by emphasizing on each personality. Now that we are living in multi-cultural society, a human race or nationalism concept is getting obscurer. There is no obvious boundary line in the differences between human race and its fashion.
