  • Drought is one of the environmental factors inhibiting plant productivity and growth, leading to oxidative damage. This study aims to identify the role of sodium hydrosulfide (NaHS) as a hydrogen sulfide (H2S) donor in drought stress tolerance in Brassica napus. Drought-induced stress symptoms appeared eight days after treatment, showing wilted leaves and a significant reduction of leaf water potential. Drought-induced increase of lipid peroxidation was significantly reduced by NaHS application. NaHS-treated plants mitigated stress symptoms under drought conditions by reducing hydrogen peroxide (H2O2) content, confirmed with H2O2 localization in situ. Furthermore, NaHS promotes photosynthetic activity by maintaining chlorophyll and carotenoid content, thereby supporting plant growth under drought conditions. Pyrroline-5-carboxylate and proline contents were significantly increased by drought but further enhanced by NaHS treatment, indicating the important roles of proline accumulation in drought stress tolerance. In conclusion, this study provides valuable insight into the roles of NaHS in alleviating drought stress by reducing oxidative stress and promoting proline accumulation. Therefore, NaHS may serve as an effective strategy to enhance crop production under drought-stress conditions.

  • Thermal stress of pigs causes decreased feed consumption and weight gain rate, immunosuppression, reproductive disorders, and increased mortality. The concept of the temperature-humidity index (THI) has been widely used to evaluate the degree of thermal stress of pigs. However, use of this concept is strongly restricted for animals living in the enclosed facilities. In this study, Building Energy Simulation (BES) technique was used to realize the energy flow among outside weather conditions, building materials, and animals. Especially, mechanisms of sensible and latent heat generation from pigs according to surrounding air temperature and their weight were designed to accurately evaluate the THI values inside the pig house. The THI values computed by the BES model were compared to those calculated by method of the report (NIAS, 2016), the model of this study predicted the start date of heat stress about 9~76 days earlier compared to the NIAS model. Results of the BES model also showed higher frequencies of the THI above the THI threshold for pigs, indicating that conventional model has a possibility of underestimating the degree of heat stress of pigs.

  • Oxidative stress is a cascade reaction characterized by a significant increase in the amount of oxidized components. Free radicals produced by oxidative stress are one of the common features in several experimental models of disease, and contribute to wide range of neurodegenerative diseases, including Alzheimer's disease. Iron (II) species can participate in the Fenton, and Fenton-like reactions, to react with hydrogen peroxide and generate hydroxyl radical. As iron accumulation and oxidative stress are associated with the pathological progression of neurodegenerative diseases, iron chelation and antioxidant therapies have become strategies to combat these diseases. Due to the complexity of the redox system in vivo, a multifaceted approach may be an attractive therapeutic strategy. Further investigations are highly expected for the prevention and treatment of neurodegenerative diseases in future.

  • The decrease in sperm quality under heat stress causes a great loss in animal husbandry production. In order to reveal the mechanism underlying the sperm quality decrease caused by heat stress, we first established a mild heat-treated mouse model. Then, the sperm quality was identified. Further, the testicular proteome profile was mapped and compared with the control using 2D electrophoresis and mass spectrometry. Finally, the differential expressed proteins involved in the heat stress response were identified by real-time PCR and Western blotting. The results showed that heat stress caused a significant reduction in mouse sperm quality (P<0.05). Further, 52 protein spots on the 2D gel were found to differ between the heat-shocked tissues and the control. Of these spots, some repair proteins which might provide some explanation for the influence on sperm quality were found. We then focused on Bag-1, Hsp40, Hsp60 and Hsp70, which were found to be differently expressed after heat shock (P<0.05). Further analysis in this heat-shocked model suggests numerous potential mechanisms for heat shock-induced spermatogenic disorders.

  • Phytochemicals which exist in various plants and fungi are non-nutritive compounds that exert numerous beneficial bioactive actions for animals. In recent years following the restriction of antibiotics, phytochemicals have been regarded as a primal selection when dealing with the challenges during the producing process in the poultry industry. The selected fast-growing broiler breed was more fragile when confronting the stressors in their growing environments. The disruption of oxidative balance that impairs the production performance in birds may somehow be linked to the immune system since oxidative stress and inflammatory damage are multi-stage processes. This review firstly discusses the individual influence of oxidative stress and inflammation on the poultry industry. Next, studies related to the application of phytochemicals or botanical compounds with the significance of their antioxidant and immunomodulatory abilities are reviewed. Furthermore, we bring up nuclear factor (erythroid-derived 2)-like 2 (Nrf2) and nuclear factor kappa B ($NF-{\kappa}B$) for they are respectively the key transcription factors involved in oxidative stress and inflammation for elucidating the underlying signal transduction pathways. Finally, by the discussion about several reports using phytochemicals to regulate these transcription factors leading to the improvement of oxidative status, heme oxygenase-1 gene is found crucial for Nrf2-mediated $NF-{\kappa}B$ inhibition.

  • Purified myofibrils were prepared from ante-mortem stress and control lots of Taiwan country chicken breast and thigh muscles at death and afler storage at $4^{\circ}C$ for 0, 1, 2, 3, and 7 days post-mortem. Sodium dodecyl sulfate polycrylamide gel electrophoresis (3.2%) and densitometer were used to examine the effect of ante-mortem stress and control storage of muscle on titin and nebulin. Results indicated that titin and nebulin were more rapidly degraded in the control and the ante-mortem stress light muscles than in the control and ante-mortem dark muscles of Taiwan country chicken. In contrast, nebulin was shown to be more resistance to degradation in the ante-mortem stress dark muscle than in the control light muscle.

  • The objective of present study was to investigate the effect of seasons on reproductive performance of Hanwoo and Holstein heifers. Heat stress in summer or cold stress in winter stress to Hanwoo and Holstein heifers may bring reproduction failure, which would pose an important economic loss, even around Daegwallyeong region located in high mountainous area. Seasonal differences in the serum levels of LH, FSH and progesterone ($P_4$) in response to environmental factors (hot and cold) out of 20 pubertal Hanwoo heifers in Daegwallyeong, Gangwon Province and 20 non-lactating Holstein heifers in Chonan city of Republic of Korea at 2-3 years of age were compared. Blood samples for hormonal analysis were from jugular vein after detection of estrus repeatedly over four seasons within four-week intervals (Spring: May to June, Summer: July to August, Autumn: October to November and Winter: January to February). In Hanwoo heifer population, averages of LH and FSH concentration in spring and in summer were greater compared to those in winter (p<0.05). LH or FSH levels tended to be greater (p=0.06) in spring and less (p=0.09) in winter compared to the levels in autumn. Only in summer, cattle seemed to show lower LH or FSH secretion (p<0.05). Similar to the results in Hanwoo heifers, the serum concentrations of LH and FSH in Holstein heifers decreased further by heat stress in summer when P 4 levels were high during luteal phase. The results demonstrate significant effect of summer heat on reproduction of Hanwoo or Holstein heifers. Although parameters indicating the extent of heat stress were not measured in this study, we suggest that serum hormone levels could be considered as successful indicators of summer heat stress condition for Hanwoo and Holstein heifers even under rather cool summer climate.

  • Objective: The oxidative stress status and changes of chicken ovary tissue after shading were studied, to determine the mechanism of the effect of shading on follicular development. Methods: Twenty healthy laying hens (40 weeks old) with uniform body weight and the same laying rate were randomly divided into two groups (the shading group and normal light group). In the shading group, the cage was covered to reduce the light intensity inside the cage to 0 without affecting ventilation or food intake. The normal lighting group received no additional treatment. After 7 days of shading, oxidative stress related indicators and gene expression were detected. Results: Analysis of paraffin and ultrathin sections showed that apoptosis of ovarian granulosa cells (GCs) increased significantly after light shading. Enzyme linked immunosorbent assay results revealed that the levels of total antioxidant capacity, malondialdehyde, superoxide dismutase (SOD), glutathione, catalase (CAT), and other substances in the sera, livers, ovaries, and follicular GCs of laying hens increased significantly after shading for 7 days; and reactive oxygen species (ROS) levels in the livers of laying hens also increased significantly. ROS in the serum, ovarian and GCs also increased. After shading for 7 days, the levels of 8-hydroxy-2 deoxyguanosine in the sera and ovarian tissues of laying hens increased significantly. Cell counting kit-8 detection showed that the proliferation activity of GCs in layer follicles decreased after shading for 7 days; the expression level of the anti-apoptotic gene B-cell lymphoma-2 in ovarian tissue and follicular GCs was significantly reduced, and the expression levels of pro-apoptotic caspase 3 (casp3), and SOD, glutathione peroxidase 2 (GPX2), and CAT were all significantly increased. Conclusion: Oxidative stress induced by shading light has a serious inhibitory effect on follicular development during reproduction in laying hens.

  • Objective: Considering the importance of dairy farming and the negative effects of heat stress, more tolerant genotypes need to be identified. The objective of this study was to investigate the effect of heat stress via temperature-humidity index (THI) and diurnal temperature variation (DTV) in the genetic evaluations for daily milk yield of Holstein dairy cattle, using random regression models. Methods: The data comprised 94,549 test-day records of 11,294 first parity Holstein cows from Brazil, collected from 1997 to 2013, and bioclimatic data (THI and DTV) from 18 weather stations. Least square linear regression models were used to determine the THI and DTV thresholds for milk yield losses caused by heat stress. In addition to the standard model (SM, without bioclimatic variables), THI and DTV were combined in various ways and tested for different days, totaling 41 models. Results: The THI and DTV thresholds for milk yield losses was THI = 74 (-0.106 kg/d/THI) and DTV = 13 (-0.045 kg/d/DTV). The model that included THI and DTV as fixed effects, considering the two-day average, presented better fit (-2logL, Akaike information criterion, and Bayesian information criterion). The estimated breeding values (EBVs) and the reliabilities of the EBVs improved when using this model. Conclusion: Sires are re-ranking when heat stress indicators are included in the model. Genetic evaluation using the mean of two days of THI and DTV as fixed effect, improved EBVs and EBVs reliability.

  • Objective: This study was conducted to assess effects of heat stress on growth performance, physiological parameters, and blood profiles of Hanwoo steers during early-fattening period in climate chambers. Methods: Four Hanwoo steers (body weight, 454.3±10.9 kg; age, 14±0.1 month) were allocated into four levels of temperature-humidity index (THI) in a 4×4 Latin square design for 21 days (pre-adaptation, 7 d; heat stress, 7 d; post-adaptation, 7 d) per period. Experimental treatments were assigned according to THI chart based on National Institute Animal Science (NIAS, 2022): Comfort (25.5℃ to 26.5℃, 60%; THI 73 to 75), Mild (28℃ to 29℃, 60%; THI 77 to 79), Moderate (29.5℃ to 30.5℃, 80%; THI 82 to 84), and Severe (31℃ to 32℃, 80%; THI 85 to 86) in separate climatic controlled chambers. Results: The dry matter intake (DMI) of the formula feed was lower in Severe compared to Mild and Comfort (p<0.05). The DMI of rice straw was the lowest in Severe and lower in Moderate than Comfort and Mild (p<0.05). Both average daily gain and feed conversion ratio of Severe and Moderate were lower than those of Mild and Comfort (p<0.05). Water intake was the highest in Severe and lower in Moderate compared with Comfort and Mild (p<0.05). Heart rate and rectal temperature increased as THI level increased (p<0.05). Glucose was the lowest in Severe and lower in Moderate compared to Comfort (p<0.05). On the contrary, non-esterified fatty acid was the highest in Severe and lower in Moderate compared with Comfort (p<0.05). Blood urea nitrogen of Moderate and Severe were higher than those of Comfort and Mild (p<0.05). Cortisol increased as THI increased (p<0.05). Conclusion: This study demonstrated the negative effects of heat stress on the performance and physiological responses of Hanwoo steers during the early-fattening period. In addition, it is judged that the THI chart for Hanwoo steers of National Institute of Animal Science (2022) was properly calculated.
