A General Strategy for the Synthesis of Amino-Substituted 2-Pyridones Using a Palladium-Catalyzed Amination Reaction

  • A novel library of amino-substituted 2-pyridones has been constructed through a two-step sequence of microwave-promoted Buchwald-Hartwig amination of 2-benzyloxy halopyridines followed by debenzylation. Microwave-promoted amination of 3- or 4-halopyridine in the presence of a suitable palladium catalyst and ligand system provided amino-substituted 2-benzyloxypyridines in excellent yields. Then, debenzylation of 2- benzyloxypyridines afforded the corresponding 2-pyridones with high efficiency.

  • A fast, efficient, and high yielding method for the preparation of amines by reductive amination of aldehydes and ketones using sodium borohydride in the presence of cellulose sulfuric acid in EtOH and under solvent-free conditions at room temperature is described.

  • A rapid and selective copper-catalyzed amination of aryl halides with sodium azide was established by using 10 mol % of CuI, and 20 mol % of N,N'-dimethylethylenediamine in DMSO under microwave irradiation for 10 min. The catalytic system with 4-substituted aryl iodides was found to be the most effective leading to a nearly complete conversion.

  • $\beta$-Glucans, which were isolated from sangwhang (Phellinus linteus), were subjected to reductive amination and the biological properties of the derivative were investigated. The degree of substitution of the aminated sangwhang $\beta$-glucan was calculated by elemental analysis to be 1.13. Bronchoalveolar lavage (BAL) experiments showed that the aminated derivative increased nitric oxide production. In addition, the amination enhanced in vitro cytotoxic activities against HT1080 and SNU-C2A cell lines. Thus, the aminated derivative is shown to enhance immune systems by the incorporation of amino groups into the polymer structure.
