To develop an easy and rapid method of quantifying lipid contents of marine dinoflagellates, we quantified lipid contents of common dinoflagellate species using a colorimetric method based on the sulpho-phospho-vanillin reaction. In this method, the optical density measured using a spectrophotometer was significantly positively correlated with the known lipid content of a standard oil (Canola oil). When using this method, the lipid content of each of the dinoflagellates Alexandrium minutum, Prorocentrum micans, P. minimum, and Lingulodinium polyedrum was also significantly positively correlated with the optical density and equivalent intensity of color. Thus, when comparing the color intensity or the optical density of a sample of a microalgal species with known color intensities or optical density, the lipid content of the target species could be rapidly quantified. Furthermore, the results of the sensitivity tests showed that only $1-3{\times}10^5cells$ of P. minimum and A. minutum, $10^4cells$ of P. micans, and $10^3cells$ of L. polyedrum (approximately 1-5 mL of dense cultures) were needed to determine the lipid content per cell. When the lipid content per cell of 9 dinoflagellates, a diatom, and a chlorophyte was analyzed using this method, the lipid content per cell of these microalgae, with the exception of the diatom, were significantly positively correlated with cell size, however, volume specific lipid content per cell was negatively correlated with cell size. Thus, this sulpho-phospho-vanillin method is an easy and rapid method of quantifying the lipid content of autotrophic, mixotrophic, and heterotrophic dinoflagellate species.
Reactive oxygen species (ROS) generated by harmful algal blooms (HABs) exert detrimental effects on aquaculture systems. Fish gill cells deteriorate upon exposure to HABs, suggesting that internally generated ROS in HABs influences the external environment. Therefore, we investigated the internal and external changes in ROS concentrations during growth using fluorescence staining of four representative HABs: Alexandrium affine, Chattonella marina, Karenia mikimotoi, and Margalefidinium polykrikoides. The concentrations of H2O2 and O2- produced by A. affine were low; H2O2 from M. polykrikoides was primarily detected internally throughout the experiments, and O2- was not detected. High H2O2 and O2- concentrations were observed in K. mikimotoi during the death phase, with weak external O2- concentrations. Regarding C. marina, which produces large amounts of ROS, H2O2 was observed internally during the exponential phase, whereas weak O2- concentrations were measured externally in the stationary phases. Collectively, our results highlight that ROS concentrations and internal/external distributions are functions of HABs and growth stage. These differences indicate the potential allelopathic mechanisms of proliferating HABs and suggest a possible impact of ROS on aquaculture organisms.
한국 연안에서 출현하는 주요 적조생물의 적조발생 특성을 알아보기 위해 편모조류 10종을 대상으로 하여 수온과 염분, 광도에 따른 종의 성장률을 조사하였고, 또한 최근 13년간의 적조발생자료와 1999년도 및 2000년도 남해안연안에서 조사한 적조생물의 출현밀도를 비교, 분석하였다. 수온에 따른 적조생물의 성장은 Heterocapsa triquetra, Eutreptiella gymnastica, Alexandrium tamarense의 경우 대체로 $16{\sim}22^{\circ}C$에서 최대성장률을 보였으나, $10{\sim}16^{\circ}C$의 저수온에서도 비교적 높은 성장률을 보였고 $22^{\circ}C$이상의 고수온에서는 오히려 성장률이 급격히 감소되는 것으로 나타나 저온성종의 특성을 보였다. Prorocentrum micans와 Pyramimonas sp.는 $19^{\circ}C$ 이하의 저수온에서는 낮은 성장률을 보였으나 $22{\sim}25^{\circ}C$에서는 높은 성장률을 보여 고온종의 특성을 나타냈으며, Akashiwo sanguinea, Hetemsigma akashiwo Prorocentrum minimum, Scrippsiella tnchoidea는 $16{\sim}25^{\circ}C$의 광범위한 수온 범위에서 비교적 높은 성장률을 보여 광온성종의 특성을 보였다. 또한 이러한 결과는 이 종들이 자연상태에서 적조를 일으키는 수온 범위와도 대체로 일치하였다 염분에 따른 성장은 대부분의 종이 염분 $30{\sim}35$ psu에서 높은 성장률을 보였는데, E. gymnastica는 35 psu 이상의 고염분보다는 $10{\sim}30$ psu의 저염분에서 더 높은 성장률을 나타내 저염성을 보였으며, H. akashiwo, P. minimum, H. triquetra는 $15{\sim}40$ psu의 넓은 염분 범위에서 양호한 성장률을 보여 광염성종의 특성을 보였다. 이종들은 한국 연안에서 강우 직후 염분이 25 psu이하로 하강하는 저염분 상태에서 빈번히 적조를 일으키고 있는데, 이것은 이종들의 저염 및 광염성 특성과 밀접한 관련이 있는 것으로 판단되었다. 광도별 성장은 H. akashiwo, P. minimum, Pyramimonas sp.는 $150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$의 고광도에서 비교적 높은 성장률을 보였다. A. sanguinea, A. tamarense, H. triquetra는 $150{\sim}100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$에서 가장 높은 성장률을 보였고 $100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ 이상의 고광도에서 는 오히려 성장률이 급격히 감소되는 것으로 나타났다. 전반적으로 수온과 일조량이 높은 시기인 하계에 적조를 일으키는 고온성 종은 고광도에서 성장이 좋은 것으로 나타났고, 수온과 일조량이 낮은 동, 춘계에 적조를 일으키는 저온성 종은 저 광도에서 성장이 양호한 것으로 나타났다.
광양만의 와편모조류 시스트를 연구하기 위하여 2001년 8월 24일 광양만의 20개 정점에서 TFO 채니기를 이용하여 표층 퇴적물과 식물플랑크톤 조사를 위한 표층 해수를 채수하였다. 퇴적물을 분석한 결과, 와편모조류 시스트는 gonyaulacoid 그룹 11종, protoperidinioid 그룹 13종, diplopsalid그룹 2종, gymnodinioid 그룹 2종, 그리고 tuberculodinioid그룹 1종, calciodinellid 그룹 1종으로 총 IT속 30종이 동정되었다. 와편모조류 시스트는 115∼2,188cysts/g의 농도로 분포하고 있었으며 , 전반적으로 조사해역의 서쪽으로 갈수록 증가하였다. 시스트 농도가 가장 높게 나타난 지역은 북서쪽에 위치한 정점 11로서, 11속 18종(2,188 cysts/g)의 시스트가 발견되었으며, 반면에 가장 낮게 나타난 지역은 조사해역의 중앙부에 위치한 정점 3으로, 6속 9종(115 cysts/g)의 시스트가 발견되었다. 가장 많이 발견된 와편모조류 시스트는 SPiniferites bulloideus였으며, Alexandrium sp., Brigantedinium simplex, S. delicatus 등이 우점하였다. 퇴적물 시료에 기록된 8종의 와편모조류 시스트의 유영세포가 해수 시료에서도 발견되었다: Polykrikos swartzii/kofoidii complex, Scripssiella trochoidea, Protoperidinium claudicans(cyst name: Votadinium spinosum), P. pentagonum(: Trinovantedinium capitatum capitatum), P. conicum(: Selenopemphix quanta), P. leonis(: Quinquecuspis concretum), P. conicoides(: Brigantedinium simplex), Gonyaulax spp.(: Spiniferites spp.). 본 연구에서 퇴적물 시료의 타가영양 와편모조류 시스트 농도가 가장 높은 정점과 표층 해수 시료의 규조류 현존량이 가장 높은 정점이 일치하였다. 이는 부영양화로 인해 영양염류가 증가한 해역에서 규조류가 많이 증식하고, 뒤이어 이를 먹이로 하는 타가영양 와편모조류도 증식함에 따라, 많은 양의 타가영양 와편모조류 시스트가 퇴적물에 집적되는 현상이 장기간에 걸쳐 진행된 결과로 생각된다 앞으로 해역의 부영양화와 퇴적물의 타가영양 와편모조류 시스트와의 관계를 보다 명확하게 규명하기 위해서는,표층수의 식물플랑크톤과 퇴적물의 와편모조류 시스트에 대한 장기간 모니터링에 따른 연속적 인 연구가 필요할 것으로 사료된다.
The annually outbreak of paralytic shellfish poisoning (PSP) were caused by toxic dinolagellate A. tamarense and A. catenella in Korea. The purpose of this study were to investigate the distribution of PSP-causative organisms, A. tamarense and A. catenella and their species classification. Sediment (Saemangeum, the south open sea) and water samples (southeastern coast) were sampled to establish clonal isolates in 2003. After isolation and purification, strains were cultured under $17^{\circ}C$, f/2 media, 14:10=L:D cycle. PST analysis and species identification were performed by HPLC-FD method and specific DNA probe, respectively. Thirty-ons strains were isolated from the Saemangeum reclamation, southeastern coast including Jinhae Bay and south open sea. PSTs were detected in all cultured strains. In eight strains from south offshore, major toxin components are GTX5, C1/2 and minors are GTX3/4, dcGTX3, neoSTX. Sixteen strains from south coastal area have GTX1/4, neoSTX, C1/2 as major toxin components and GTX2/3 as minors. Seven strains from the Saemangeum reclamation have GTX5, C1/2 as major toxin components and GTX1/2/3/4 as minors. Thus, among eight south offshore isolates, four A. tamarense have more toxic (38.31~l19.16 fmol.$cell^{-1}$) than A. catenella (3.78~13.13 fmol.$cell^{-1}$). With the previous results of different toxin composition, toxin components and toxin contents, .it is toxin profile that could used to diagnosis of regional toxic population and geographical distribution of both A. tamarense and A. catenella and their toxigenic strains.

Twenty-three cultures of harmful algal bloom (HAB)-(causing dinoflagellates were isolated from the coastal waters of Korea. For each of the 14 morphospecies, the nuclearencoded small subunit (SSU) rDNA was analyzed to determine the phylogenetic relatedness of the species. Despite temporal and spatial isolation, 3-4 clonal cultures of Alexandrium catenella, Cochlodinium polykrikoides, and Gymnodinium catenatum had 100% identical SSU rDNA sequences. In contrast, heterogeneities in the SSU rDNA sequences were observed in Akashiwo sanguinea and Lingulodinium polyedrum strains. Extreme sequence polymorphism was shown within the SSU rRNA genes of an Al. tamarense clonal culture. A homology search in GenBank revealed that 11 dinoflagellate species were located in clusters corresponding to their morphological classification. The SSU rDNA sequences of C. polykrikoides, Gyrodinium instriatum, and Pheopolykrikos hartmannii, which were determined for the first time in this study, showed the following phylogenetic relationships: C. polykrikoides formed an independent branch separated from other dinoflagellates; Gyr. instriatum was placed in a monophyletic group with Gyr. dorsum and Gyr. uncatenum; and Ph. hartmanii, which forms a distinct two-celled pseudocolony, belonged to Gymnodinium sensu Hansen and Moestrup.
The nucleotide sequences of the internal transcribed spacer regions (ITS1 and ITS2) of ribosomal DNA (rDNA), and the 5.85 rRNA gene, have been determined for 13 strains of dinoflagellates in order to analyze the phylo-genetic relationship. The DNA sequences contained considerable variation in the ITS regions, but little in the 5.85 rDNA. In addition, the ITS1 was more variable than the ITS2 in all species examined. The nucleotide length of this region varied from 519 bp to 596 bp depending on the taxa. The investigated taxa were divided into three large groups based on the ITS length, i. e., a group with short ITS region (A. fraterculus and Alexandrium sp.), a with ITS region group (P. micans, P. minimum and P. triestinum) and a with ITS region group (G. impudicum, C. polykrikoides, G. sanguineum, G. catenatum and H. triquetra). The relationship between nucleotide length of ITS1 and that of ITS2 was negative, whereas G+C content and nucleotide length showed positive correlation. In phylogenetic analyses producing NJ trees, the topology was similar cluster and clearly divided the taxa into three groups based on 5.8S rDNA that were similar to those based on morphological characteristics. In particular, G. impudicum was more closely related to G. catenatum than to C. polykrikoides using phylogenetic analysis. From this study, we chew that the length of ITS region contributes to discriminate Korean harmful algal species and ITS analysis is a useful method for resolving the systematic relationships of dinoflagellates.

Occurrence of Cochlodinium polykrikoides red tides have resulted in considerable economic losses in the aquaculture industry in many countries, and thus predicting the process of C. polykrikoides red tides is a critical step toward minimizing those losses. Models predicting red tide dynamics define mortality due to predation as one of the most important parameters. To investigate the roles of heterotrophic protists in red tide dynamics in the South Sea of Korea, the abundances of heterotrophic dinoflagellates (HTDs), tintinnid ciliates (TCs), and naked ciliates (NCs) were measured over one- or two-week intervals from May to Nov 2014. In addition, the grazing impacts of dominant heterotrophic protists on each red tide species were estimated by combining field data on red tide species abundances and dominant heterotrophic protist grazers with data obtained from the literature concerning ingestion rates of the grazers on red tide species. The abundances of HTDs, TCs, and NCs over the course of this study were high during or after red tides, with maximum abundances of 82, 49, and $35cells\;mL^{-1}$, respectively. In general, the dominant heterotrophic protists differed when different species caused red tides. The HTDs Polykrikos spp. and NCs were abundant during or after C. polykrikoides red tides. The mean and maximum calculated grazing coefficients of Polykrikos spp. and NCs on populations of co-occurring C. polykrikoides were $1.63d^{-1}$ and $12.92d^{-1}$, respectively. Moreover, during or after red tides dominated by the phototrophic dinoflagellates Prorocentrum donghaiense, Ceratium furca, and Alexandrium fraterculus, which formed serial red tides prior to the occurrence of C. polykrikoides red tides, the HTDs Gyrodinium spp., Polykrikos spp., and Gyrodinium spp., respectively were abundant. The maximum calculated grazing coefficients attributable to dominant heterotrophic protists on co-occurring P. donghaiense, C. furca, and A. fraterculus were 13.12, 4.13, and $2.00d^{-1}$, respectively. Thus, heterotrophic protists may sometimes have considerable potential grazing impacts on populations of these four red tide species in the study area.
Ceratium 적조생물을 메타놀이나 수용성 물질을 분리하거나 cell-free medium 추출물을 Cochlodinium polykrikoides 성장에 미치는 영향을 조사했다. cell-free medium을 25% 이상 첨가한 시험구에서 가장 우수한 C. polykrikoides 성장을 가져왔다. 그러나 메타놀이나 수용성으로 처리한 추출물에서는 고농도로 배지에 첨가시켜도 C. polykrikoides 성장에는 큰 영향이 없었다. 특히 cell-free medium은 Gyredinium impudicum이나 Chaetoceros 종에서도 C. polykrikoides와 비슷한 양상을 보였으나, Alxendrium 적조생물에 대해서는 현저히 성장을 저해시키는 결과를 나타내었고, Prorocentrium minimum에서도 성장을 촉진시키지는 못했다. 이러한 결과로 보와서 Ceratium 추출물은 C. polykrikoides성장을 촉진시킬 수 있는 주요한 역할을 하는 것으로 보여서 두 적조생물이 공존하게 되면 상호간에 영향을 미칠 수 있을 것으로 판단된다.

Microalgae have been utilized in diverse industries including aquaculture. Among the microalgae, dinoflagellates are known to have various bioactive compounds, and thus the interest in their application to industry has increased. In order to test their potential as food materials for aquaculture animals, the crude protein contents and compositions of amino acids of six dinoflagellates Heterocapsa rotundata (family Heterocapsaceae), Ansanella granifera (Suessiaceae), Alexandrium andersonii (Ostreopsidaceae), Takayama tasmanica (Brachidiniaceae), Takayama helix, and Gymnodinium smaydae (Gymnodiniaceae) belonging to diverse families were analyzed. The percentage of the amount of the crude protein relative to dry weight of T. tasmanica was the highest (65%) and that of A. andersonii was the lowest (26%). However, the highest percentage of total detected amino acids in crude protein was found in A. andersonii (98.2%). In all six dinoflagellates, glutamic acid was the most dominant amino acid in crude protein. However, the second main amino acid was aspartic acid for H. rotundata, A. granifera, T. helix, and G. smaydae, but were arginine and leucine for A. andersonii and T. tasmanica, respectively. Furthermore, T. tasmanica and T. helix did not have taurine and gamma-aminobutyric acid, whereas the other dinoflagellates possessed them. The percentages of essential amino acid contents of the dinoflagellates met the requirement levels for juvenile shrimps. In addition, the dinoflagellates were not toxic to the brine shrimp Artemia salina. Compared with the other microalgae reported so far, H. rotundata and A. andersonii can be used for arginine-rich diets, T. tasmanica for valine and leucine-rich diets, A. granifera for histidine-rich diets, T. helix for threonine-rich diets, and G. smaydae for lysine-rich diets. Therefore, based on their biochemical composition and toxicity to Artemia, the dinoflagellates could be used as essential amino acid sources for cultivating animals in the aquaculture industry.

저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
