This study was performed to assess airborne fungi concentrations during fall in eight subway stations in Seoul, Korea. The purpose of this study was to investigate appropriate culture media and evaluate factors affecting airborne fungi concentrations. Results indicated that airborne fungi concentrations showed log-normal distribution. Thus, geometric mean (GM) and geometric standard deviation (GSD) were calculated. The GM of airborne fungi concentrations cultured on malt extract agar (MEA) media was 466 $cfu/m^3$ (GSD 3.12; Range 113~4,172 $cfu/m^3$) and the GM of concentrations cultured on DG18 media was 242 $cfu/m^3$ (GSD 4.75; Range 49~6,093 $cfu/m^3$). Both of GM values exceeded 150 $cfu/m^3$, the guideline of World Health Organization (WHO). There was no significant difference between two fungi concentrations cultured on MEA and DG18 media, respectively. Two factors, such as relative humidity and depths of subway stations were significantly related to airborne fungi concentrations. It is recommended that special consideration should be given to deeper subway stations for improvement of indoor air quality.
Objectives: Bioaerosols released by treating organic-waste resources cause a variety of environmental and hygiene problems. The objective of this study was to investigate the distribution characteristics of the airborne bacteria emitted from a pig manure composting plant, a principal site for organic-waste resource facilities. Methods: Three types of pig manure composting plant were selected based on fermentation mode: screw type, rotary type and natural-dry type. Each site was visited and investigated on a monthly basis between September 2009 and August 2010. A total of 36 air samplings were obtained from the pig manure composting plants. The air sampling equipment was a six-stage cascade impactor. Quantification and qualification of airborne bacteria in the air samples was performed by agar culture method and identification technique, respectively. Results: The mean concentrations of airborne bacteria in pig manure composting plant were 7,032 (${\pm}1,496$) CFU $m^{-3}$ for screw type, 3,309 (${\pm}1,320$) CFU $m^{-3}$ for rotary type, and 5,580 (${\pm}1,106$) CFU $m^{-3}$ for natural dry type. The screw type pig manure composting plant showed the highest concentration of airborne bacteria, followed by the natural dry type and the rotary type. The ratio of respirable to total airborne bacteria was approximately 40-60%. The predominant genera of airborne bacteria identified were Micrococcus spp., Staphylococcus spp. and Escherichia spp. Conclusion: Monthly levels of airborne bacteria were highest in August and lowest in November regardless of fermentation mode. There was no significant correlation relationship between airborne bacteria and environmental factors such as temperature, relative humidity and particulate matters in pig manure composting plants.
From the point of view to prevent airborne infection-related diseases such as H1N1, SARS, and MERS, an actual application of air cleaning and purification systems including technologies like UVGI has become increasingly important. Recently, an air purification system using REME (Reflective Electro Magnetic Energy) developed in the U.S. is applied for indoor air purification and sterilization technology to counteract the outbreak of new airborne infections. In this study, an air sterilization performance experiment using REME was carried out. The results verified that air sterilization performance in the case of installing a REME system in a medical center was 31%, namely the number of floating bacteria decreased by 31% after only a five-day operation. In addition, the number of culture collections in the REME operating air conditioning systems using nonpathogenic Geobacillus stearothemophilus as a biological indicator decreased maximally to 67%. A field application of REME technology will be useful to prevent airborne infection-related diseases, especially in response to public health crises due to the advent of emerging diseases.
The objectives of this study are (a) to investigate the distribution patterns and exposure concentrations of biological agents in sawmill industries and (b) to compare sampling methods of biological agents. The representative processes of 5 sawmills were selected to measure total airborne bacteria, fungi, endotoxin as well as dust. Airborne bacteria and fungi were measured with one stage impactor, six stage impactor and gelatin filteration methods. Endotoxin was collected with polycarbonate filters and analysed by kinetic chromogenic Limulus Amebocyte Lysate method. Geometric mean levels of airborne bacteria, fungi, endotoxin and dust were 1,864 CFU/$m^3$, 2,252 CFU/$m^3$, 31.5 EU/$m^3$ and 2.4 mg/$m^3$. The ratios of indoor/outdoor concentrations were 3.7 for bacteria, 4.1 for fungi, 3.3 for endotoxin and 9.7 for dust. The respiratory fractions of bacteria were 68.0, 50.9, 49.2 and 45.1% in band-saw, table-saw, rip-saw process and outdoor air. The respiratory fractions of fungi were 78.7, 90.8, 87.5 and 84.8% in band-saw, table-saw, rip-saw process and outdoor air, respectively. There was no significant differences in bacterial concentrations among single stage, six stage impaction and filteration methods. But, fungal concentrations measured with filtration methods were significantly higher than those with impaction methods. Geometric mean levels of airborne bacteria and fungi were higher than the OSHA guideline values of 1,000 CFU/$m^3$. The respiratory fractions of fungi were above 75%. The concentrations of biological agents were significantly different among culture-based sampling methods. In the exposure assessments of biological agents, further studies are needed for the comparisons of diverse sampling methods and the investigations of environmental factors.
The primary culture of human nasal epithelial cells was performed using the inferior nasal turbinate tissues, and infected with Hantaan virus to examine the hypothesis of airborne transmission of Hantaan virus in humans. The primary culture cells were identified as epithelial cells by morphologic and immunologic analyses. The viral antigens were detected in the primary human nasal epithelial cells infected with Hantaan virus by immunofluorescence staining. The ICAM-1 induction by Hantaan virus or $IFN-{\gamma}$ was examined in the primary human nasal epithelial cells and human lung fibroblasts (WI-38). Hantaan virus induced the surface ICAM-1 in WI-38 cells in a time-dependent manner, and $IFN-{\gamma}$ induced the surface ICAM-1 in a dose-dependent manner in HNEC and WI-38 cells. These results revealed that the human nasal epithelial cells are susceptible to Hantaan viral infection supporting the hypothesis of airborne transmission of Hantaan virus in humans. The human lung fibroblasts also might have an important role in the pathogenesis of Hantaan virus through the induction of ICAM-1.

Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained isolates, 248 isolates were identified as Trichoderma fungi including T. harzianum, T. atroviride, T. citrinoviride, and T. pseudokoningii, by morphological and molecular analysis. T. harzianum was dominant among the four identified species. All the four Trichoderma species grew fast on solid nutrient media tested (potato dextrose agar [PDA], malt extract agar [MEA], Czapek's Dox + yeast extract agar [CYA] and cornmeal dextrose agar). Compact mycelia growth and mass spore production were better on PDA and CYA. In addition, T. harzianum and T. citrinoviride formed greenish and yellowish mycelium and spores on PDA and CYA. Greenish and yellowish pigment was saturated into PDA only by T. pseudokoningii. These four Trichoderma species could produce extracellular enzymes of sawdust substrate degradation such as β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease. Their mycelia inhibited the growth of oak wood mushroom mycelia of two tested cultivars on dual culture assay. Among of eleven antifungal agents tested, benomyl was the best to inhibit the growth of the four Trichoderma species. Our results demonstrate that the airborne Trichoderma fungi need to be properly managed in the cultivation houses for safe mushroom production.
Mold grows more easily when humidity is higher in indoor spaces, and as such is found more often on wetted areas in housing such as walls, toilets, kitchens, and poorly managed spaces. However, there have been few studies that have specifically assessed the level of mold in the indoor spaces of water-damaged housing in the Republic of Korea. We investigated the levels of airborne mold according to the characteristics of water damage types and explored the correlation between the distribution of mold genera and the characteristics of households. Samplings were performed from January 2016 to June 2018 in 97 housing units with water leakage or condensation, or a history of flooding, and in 61 general housing units in the metropolitan and Busan area, respectively. Airborne mold was collected on MEA (Malt extract agar) at flow rate of 100 L/min for 1 min. After collection, the samples were incubated at $25^{\circ}C$ for 120 hours. The cultured samples were counted and corrected using a positive hole conversion table. The samples were then analyzed by single colony culture, DNA extraction, gene amplification, and sequencing. By type of housing, concentrations of airborne mold were highest in flooded housing, followed by water-leaked or highly condensed housings, and then general housing. In more than 50% of water-damaged housing, the level of airborne mold exceeded the guideline of Korea's Ministry of Environment ($500CFU/m^3$). Of particular concern was the fact that the I/O ratio of water-damaged housing was greater than 1, which could indicate that mold damage may occur indoors. The distribution patterns of the fungal species were as follows: Penicillium spp., Cladosporium spp. (14%), Aspergillus spp. (13%) and Alternaria spp. (3%), but significant differences of their levels in indoor spaces were not found. Our findings indicate that high levels of mold damage were found in housing with water damage, and Aspergillus flavus and Penicillium brevicompactum were more dominant in housing with high water activity. Comprehensive management of flooded or water-damaged housing is necessary to reduce fungal exposure.
To evaluate the indoor air quality of food manufacturing plants, the presence of viable bacteria and fungi was assessed in the indoor air of the facilities at which 9 food items were manufactured. Air samples were collected from the general zone, low clean zone and clean zone of each factory with an air sampler, in combination with plate counts agar using for bacteria, and dichloran-glycerol agar for fungi. The samples were incubated at $25^{\circ}C$ for 4 to 7 days. After culture, the colony forming units (CFU) on each plate were counted and corrected with a positive hole conversion table. The average concentration of bacteria was $2.2{\times}10^3\;CFU/m^3$ in the general zone, $1.2{\times}10^3\;CFU/m^3$ in the low clean zone and $7.3{\times}10^2\;CFU/m^3$ in the clean zone. The average concentration of fungal microbes was $2.5{\times}10^3\;CFU/m^3$ in the general zone, $2.6{\times}10^3\;CFU/m^3$ in the low clean zone, and $2.0{\times}10^2\;CFU/m^3$ in the clean zone. No meaningful differences were detected between the general zone and the low clean zone, but the clean zone had significantly lower concentrations than the other zones. Additionally, the identification of the fungi was performed according to morphological method using a giant culture and slide culture. The fungi were identified as belonging to 18 genera, and the genera Cladosporium(33%), Penicillium(29%) and Aspergillus(26%), predominated. Aspergillus isolates were identified to species level, and A. ochraceus, a mycotoxigenic species, was identified. As part of the effort to control the quality of the indoor air of food manufacturing plants, our results show that continued studies are clearly warranted.
본 논문에서는 실외 대기 환경의 바이오에어로졸 혹은 입자상물질의 미생물 메타게놈 특성과 이에 영향을 미치는 기후 및 환경 인자의 영향을 고찰하였다. 시료 채취 지역 및 환경 조건 특성별 대기 중 세균과 곰팡이 농도를 요약 하고, 에어로졸과 PM 시료의 세균과 곰팡이의 메타게놈 특성을 조사하기 위한 비배양법 기반 분석방법과 메타게놈 특성을 정리하였다. 또한, 세균과 곰팡이의 메타게놈 특성과 다양성 및 특성에 미치는 기상 인자와 환경 인자의 영향을 고찰하였다. 대기 중 미생물의 생존, 생장과 분산은 지역 기상 조건 및 대기 오염 물질에 의해 크게 영향을 받았다. 일반적으로 기온이 상승함에 따라 AM 농도는 증가하지만, 여름에는 고온과 강한 자외선의 영향으로 AM 농도가 감소하였다. 습도와 미생물 농도는 양의 상관성을 보이나, 습도가 너무 높으면 AM의 분산이 지연되었다. 이러한 종합적인 고찰 결과는 대기권에서 미생물의 역할과 기능을 이해하고, 이들 미생물에 의해 야기되는 환경 및 공중보건 문제를 해결하기 위한 전략 수립 및 저감 기술 개발에 활용될 수 있다.
The incidence of hospital infection has been seriously increased in the general hospital in recent years. This study was performed on hospital air and materials in a General Hospital in Seoul from June to December in 1980. The results were as follows: 1. Air sampling was done in multiple strategic areas by exposing standard petridishes for 5 minutes. There was a significant difference of airborne microbe between places. ($F._{99}$ = 3.2, p < 0.01). 2. The mean colony count was 8.6$\pm$6.2 colonies / plate / 5 minutes. 3. Gram stains of colony in air sampling were Gram (+) cocci 66.5%. Gram (+) rod 18.4%, Gram (-) cocci 1.3%, Gram (-) rod 8.7% Fungus 4.5%. 4. For the evaluation of sterilization of steam sterilizer and ethylene oxide gas sterilizer, biological monitoring were done by commercial spore strip. Positive culture was obtained in 2 out of 41 tests on 3 steam sterilizers, and in 3 out of 13 tests on ethylene oxide gas sterilizer. 5. Product sampling and culture were done for 2 kinds of disinfectants and 30 sets of various operation package or dressing materials. Positive culture was obtained in one disinfectants.
