In order to study the protease from Agaricus bisporus (Lange), the crude protease preparation was separated by fractionation of mushroom extracts with ammonium sulfate. It was found that extracts from Agaricus bisporus (Lange) Sing. contained protease. The optimum pH of the enzyme was 6.0, and the pH range at which the enzyme was stable was 4.0 to 7.0. The optimum temperature at which the enzyme showed the highest proteolytic activity was $50^{\circ}C$, while the enzyme was instantly inactivated at about $60^{\circ}C$. The enzyme activity was inhibited by $Ag^+$, $Hg^{2+}$, $Cu^{2+}$, $Ba^{2+}$, $Fe^{3+}$, $Co^{2+}$, $Ca^{2+}$, $Pb^{2+}$, $Mg^{2+}$ and $Mn^{2+}$. The $K_m$ value was 0.32 mM with Hammarsten casein.
Basidiomycete, Agaricus blazei murill has grown Brazil naturally. It was first cultivated 1997 in Korea. Proteoglycan or polysaccharide which have the effect of immunopotency and anticancer were extracted from it. From the mycelium of cultivated mushroom, were extracted lectin, linoleic acid, FIII-2-b, and simmilar to glucomannan from fruit body of Basidiomycete Agaricus blazei. Fruit body was more studied than mycelium. The experiments were most in vivo study. The effect were shown on S-180, MethA tumor, Ehrlich ascites carcinoma, Shionogi carcinoma 42, Meth A fibrosarcoma by transferation of tumor cell line, specially effectiveness on the S-180. About immunopotenciation, were shown as activation of NK cell, pancreatic T-cell, helper T cell, enhancement of population of cytotoxity T cell. It was effect on the MethA tumor cells in vitro cell cytotoxity and has induction of apoptosis. Forthermore cytotoxity of many other tumor cell line, aneiognensis, cell cyle studies will be needed.

The partial nucleotide sequences of the genomic dsRNA mycoviruses infecting Pleurotus ostreatus (isolates ASI2596, ASI2597, and Bupyungbokhoe) and Agaricus blazei Murrill were determined and compared with those of the other dsRNA mycoviruses. Partial nucleotide sequences of the purified dsRNA from ASI2596 and ASI2597 revealed RNA-dependent RNA polymerase sequences that are closely related to Oyster mushroom isometric virus 2, while nucleotide sequences and the deduced amino acid sequence from dsRNA mycovirus infecting Agaricus blazei did not show any significant homology to the other dsRNA mycoviruses. Specific primers were designed for RT-PCR detection of these dsRNA viruses and were found to specifically detect each dsRNA virus. Northern blot analysis confirmed the homogeneity of RT-PCR products to each purified dsRNA. Altogether, our results suggest that these virus-specific primer sets can be employed for the specific detection of each dsRNA mycovirus in infected mushrooms.
Physiologically active material ${\beta}$-glucan extracted from mushroom has an antitumor effect. Agaricus blazei murill(A blazel) fruiting bodies were reported to contain large amounts of ${\beta}$-glucan. The purpose of this study was to maximize ${\beta}$-glucn extraction from A. blazei by microwave assisted extraction method. Optimal extraction conditions were obtained under various extraction time and applied power which was examined as 200, 500 and 650 watt. The structure and concentrations of the raffinates were analyzed by nuclear magnetic resonance spectroscopy(NMR) and infrared spectroscopy(IR) It was proved that the extraction yield of microwave assisted extraction method was higher than that of conventional solvent extraction method. It was also observed that microwave assisted extraction method requires less amounts of solvents and extraction time compared to the conventional solvent extraction method.

Fish paste containing mushroom, Agaricus bisporus, was prepared for the supplementary value of the product, and it was tested for the color, textural properties, and sensory attributes. With increasing amounts of the mushroom in fish paste, L value decreased and a value increased in the color changes while the b value was not significantly different. As the result of textural properties, folding test in all test samples showed AA that mean good flexibility. The hardness, elasticity, gumminess and brittleness of fish paste with the mushroom were increased but the strength was not significantly different. In overall acceptance of sensory evaluation, fish paste containing 10% of mushroom received the highest score. These results suggest that Agaricus bisporus can be applied to fish paste for the purpose of the varieties of product.
This study were conducted to investigate pin-heading induction and growing temperature of Agaricus brunescens. The temperature for fruit body formation was better at $15^{\circ}C$ than at 18 and $21^{\circ}C$. As the temperature go up to $21^{\circ}C$, the fruit body number was decreased but the fruit body size and weight was increased. Optimal temperature for both pin-heading and growth of Agaricus brunescens was $18^{\circ}C$.
Mushrooms(Pleurotus ostreatus, Agaricus bisporus and Flammulina velutipes) are popular food sources in Korea, and have been reported as therapeutic foods, useful for preventing various diseases. In this study we researched HPLC conditions for the determination of nucleic acids in extracts of the three type of mushrooms. The method for nucleic acids analysis of mushrooms was developed using HPLC with UV detection. To determine the nucleic acids, mushroom extracts were extracted in hot water at $90^{\circ}C$ by reflux extraction for 1 hr. Then, the extracts were hydrolyzed by enzymes RP-1G and 50000G. The HPLC conditions were simple, rapid, and sensitive, and were applicable for the analysis of 4 nucleosides(cytidine, uridine, guanosine and inosine) and 3 mono-nucleotides(5'-CMP, 5'-UMP, and 5'-IMP) in the mushrooms. The nucleic acids in the mushrooms were cytidine, guanosine, inosine, uridine, 5'-CMP, 5'-IMP, and 5'-UMP. The analysis results for total nucleic acids in the mushroom extracts(Pleurotus ostreatus, Agaricus bisporus, and Flammulina velutipes) indicated levels of 25.28, 27.75, and 19.87 mg/g, respectively. In conclusion, this method can be used successfully for qualitative and quantitative analysis of nucleic acids in Pleurotus ostreatus, Agaricus bisporus, and Flammulina velutipes.

Amylases and cellulases are important enzymes that can be utilized for various biological activities. Ten different wild Nigerian mushrooms (Agaricus blazei, Agaricus sp., Corilopsis occidentalis, Coriolus versicolor, Termitomyces clypeatus, Termitomyces globulus, Pleurotus tuber-regium, Podoscypha bolleana, Pogonomyces hydnoides, and Nothopanus hygrophanus) were assayed for production of these secondary metabolites. The results revealed that most of the tested wild fungi demonstrated very good amylase and cellulase activities. With the incorporation of carboxymethyl-cellulose (a carbon source) into the culture medium, Agaricus blazei had the highest amylolytic activity of 0.60 unit/mL (at $25^{\circ}C$, pH 6.8). This was followed in order by P. tuber-regium and Agaricus sp. with 0.42 and 0.39 unit/mL, respectively ($p {\leq} 0.05$). Maltose and sucrose supplementation into the submerged liquid medium made N. hygrophanus and P. hydnoides to exhibit very low amylase activities of 0.09 and 0.11 unit/mL, respectively. Introducing peptone (an organic nitrogen source) into the basal medium enhanced the ability of C. versicolor to produce a cellulase value of 0.74 unit/mL. Other organic nitrogen sources that supported good cellulase activities were yeast extract and urea. Sodium nitrate (inorganic nitrogen source) generally inhibited cellulase production in all mushrooms. The best carbon source was carboxymethyl-cellulose, which promoted very high cellulase activity of 0.67 unit/mL in C. versicolor, which was followed in order by P. tuber-regium, T. chypeatus, and C. occidentalis ($p {\leq} 0.05$). Sucrose was the poorest carbon compound, supporting the lowest values of 0.01, 0.01, and 0.14 unit/mL in P. hydnoides, A. blazei, and Agaricus sp., respectively.
In this study, the α-amylase inhibitory activity, α-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the α-amylase inhibitory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The α-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50~1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5~8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mushrooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as α-amylase, α-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.
The Basidomycetes fungus Agaricus blazei Murill has been well known as a health food for the prevention of cancer, diabetes, hyperlipidemia, arteriosclerosis and chronic hepatitis. This study was concentrated to investigate the characteristics of crude protein-bound polysaccharides(PBP) compositions extracted from Agaricus blazei Murill. In order to produce crude polysaccharides, culture conditions were examined using YMK media. Total sugars and protein contents of PBP were detected by phenol-sulfuric acid method and Bradford -assay. Hexosamine was found to be involved in the linkage, N-linked and O-linked types. To identify helical conformation existence, wavelength was measured using Congo red after the treatment with alkali solution.
