Combinations of plant growth regulators, darkness treatments, and the order of expanding leaves for explants were evaluated for optimizing in vitro shoot regeneration rate of 'Whangkeumbae' pear. In a MS medium, supplemented with $8.88{\mu}M$ 6-benzylaminopurine (BA) and $0.49{\mu}M$ indole-3-butyric acid (IBA), green foci were observed on the surface of the callus 8 days after culture initiation. Some adventitious buds were later induced from those green foci, resulting in the appearance of normal shoots. In a medium containing $22.20{\mu}M\;BA$, the surface of the callus became compact and greenish, and many adventitious buds were formed over the entire area of the callus surface. When comparing BA concentration via histological observation, the section which had been treated with $22.20{\mu}M\;BA$ exhibited closer cell aggregation than those with $8.88{\mu}M\;BA$. The darkness treatment enhanced the formation of adventitious shoots for up to 3 weeks. The youngest two expanding leaves, proximal to the shoot apex, were proved to be the most regenerative, and yielded the highest shoot number per regenerating leaf. A fourth strength MS medium, which was supplemented with $0.54{\mu}M\;NAA$, yielded good quality plantlets, with regard to root number and root length.
인삼 조직배양에 있어 multi-shoot 대량번식을 통한 이차대사산물의 이용가능성을 조사코자 강제통기방식에 의해 $CO_2$를 기내에 공급하여 배배양하였던 바 multi-shoot 분화특성 및 사포닌 함량에 미치는 $CO_2$의 효과는 다음과 같다. 부정아 수와 부정아 유래의 신초수는 무처리와 $CO_2$ 2처리구간에 차이가 적었고 분화율은 50%내외이었고 부정아에서 분화한 신초는 경엽의 전개가 빠르고 생장량이 높은것이 특징이었다. 한 개의 자엽에서 분화되는 shoot primordium의 수는 대체로 15.4개-23.9개로 $CO_2$ 2, 500ppm처리구가 가장 양호하였고 $CO_2$ 10, 000ppm의 고농도는 오히려 shoot primordium수 뿐만 아니라 신초 분화율도 저하시켰다. 무처리의 건물율은 7,50%이었던데 반하여 $CO_2$ 2, 500ppm처리구는 건물율이 8.59%로 여타 $CO_2$ 처리구에 비하여 가장 높았다. 기내개화율은 무처리가 7.6%이었으나 $CO_2$, 2, 500-5, 000ppm처리구는 16.3-15.7%로 무처리에 비하여 2배 가량 높았다. 그러나 1개의 치상 배에서 얻어지는 개화수는 1.2-1.3개로 $CO_2$ 처리농도에 따른 차이가 없었다. 조사포닌 함량은 $CO_2$ 2, 500ppm처리구의 경우, 캘러스와 신초가 혼재되어 있는 소식물체는 5.80%이었던데 비하여 multi-shoot가 8.32%로 높았고 ginsenoside함량은 multi-shoot의 경우, ginsenosid Rd, Re 그리고$Rg_1$이 특히 많았다.
This study describes culture conditions for a plant regeneration system via a combined pathway of somatic embryogenesis and organogenesis in root explant cultures of the commercial rose cultivar 'Charming'. Root explants formed white calluses at a frequency of 30% after 6 weeks of culture on Schenk and Hildebrandt (SH) medium supplemented with $11mg\;1^{-1}$ 2,4-dichlorophenoxyacetic acid. After 6 weeks of transfer to SH medium without growth regulators, initial white calluses gave rise to globular somatic embryos at a frequency of 2.8%, which were subsequently dedifferentiated to embryonic tissues. Somatic embryos or embryonic tissues initially derived from root explants did not undergo development beyond cotyledonary stage. To produce adventitious shoots, embryonic tissues were sliced and cultured on SH medium with $0.5mg\;1^{-1}$ 6-benzyladenine. After 4 weeks of culture, 28% of embryonic tissue explants formed adventitious shoots. Regenerated shoots were rooted on half strength SH medium with $0.1mg\;1^{-1}$${\alpha}-naphthalaneacetic$ acid and subsequently grown to maturity. Root-derived embryonic tissues were proliferated by subculture, while retaining the capacity for shoot production for a few years.
An efficient protocol for in vitro organogenesis was achieved from callus-derived immature and mature leaf explants of Momordica charantia, a very important vegetable and medicinal plant. Calluses were induced from immature leaf explants excised from in vitro (15-day-old seedlings) mature leaf explants of vivo plants (45 days old). The explants were grown on Murashige and Skoog (MS) medium with Gamborg (B5) vitamins containing 30 g $1^{-1}$ sucrose, 2.2 g $1^{-1}$ Gelrite, and 7.7 lM naphthalene acetic acid (NAA) with 2.2 ${\mu}M$ thidiazuron (TDZ). Regeneration of adventitious shoots from callus (30-40 shoots per explant) was achieved on MS medium containing 5.5 ${\mu}M$ TDZ, 2.2 ${\mu}M$ NAA, and 3.3 ${\mu}M$ silver nitrate ($AgNO_3$). The shoots (1.0 cm length) were excised from callus and elongated in MS medium fortified with 3.5 ${\mu}M$ gibberellic acid ($GA_3$). The elongated shoots were rooted in MS medium supplemented with 4.0 ${\mu}M$ indole 3-butyric acid (IBA). Rooted plants were acclimatized in the greenhouse and subsequently established in soil with a survival rate of 90%. This protocol yielded an average of 40 plants per leaf explant with a culture period of 98 days.
쇠무릎(A. japonica) 다량증식 방법의 일환으로 액아를 이용한 multiple shoot유도를 위한 최적 조건을 조사하였다. 기내에서 증식하고 있는 식물체로부터 액아를 적출하여 NAA, 2,4-D 및 BA가 여러 농도로. 첨가된 MS 배지(3% sucrose, 0.2% gelrite)에서 6주 간 배양한 결과, shoot의 발생은 1mg/L NAA와 2mg/L BA가 첨가된 처리구에서 액아 당 평균 25.8개로 가장 많았다. 신초의 발생 빈도는 조금 낮지만 발생된 신초가 완전한 식물체로 되기 위한 크기를 고려할 때의 조합은 0.5 mg/L NAA와 1 mg/L BA가 첨가된 처리구에서 액아 당 19.7개의 신초가 발생하여 가장 좋은 결과를 얻었다. 한편 발생된 신초는 절취하여 0.1mg/L IBA가 첨가된 1/2 MS 배지에서 배양했을 때 뿌리의 발생과 신장이 가장 양호하였으며, 식물체의 생육도 왕성하였다. 발근된 식물체를 기외로 이식 후 활착시켜 쇠무릎의 액아 배양을 통한 다량증식의 가능성을 확인하였다.
Calystegia soldanella L. is a perennial herbaceous halophyte belonging to the convolvulaceae family, which mainly grows in coastal sand dunes in Korea. Shoots and rhizomes are edible, and roots called 'Hyoseon Chogeun' are known to have medicinal effects such as antipyretic, sterilization, and diuretic. In addition, physiological activities of antioxidant, anti-inflammatory, antiviral, antifungal and PTP-1B (protein tyrosine phosphate-1B) inhibition have been reported. In this study, in vitro induction cell lines of C. soldanella L. collected from the coastal sand dunes in Jeju island was redifferentiated into adventitious roots that can be used as medicinal resources. Also the biomass of mass-proliferated adventitious roots using a bioreactor were evaluated. Plants of C. soldanella L. were collected from the crevice of the seashore in the coastal area of Taeheung 2-ri, Namwon-eup, Seogwipo-si. Then, it was separated into leaves, stems, rhizomes, and roots, and surface sterilized with 70% ethyl alcohol and 2% NaOCl (sodium hypochlorite). After washing with sterilized water, each organ section was cultured in Hormone-free MS medium (Murashige & Skoog Medium). As a result, the induction response rates were evaluated at 85% and 55%, respectively, in terms of callus formation and shoot generation in the rhizome segment. In the case of the adventitious roots morphological characteristics induced by single-use treatment of auxin-based plant growth regulators IBA and NAA from redifferentiated shoots were compared. Most efficient adventitious root culture method as a rooting rate, number, length, and biomass proliferation in the bioreactor system was confirmed when treated by culturing in MS salts, Sucrose 30 g·L-1, and IBA 1mg·L-1 for 4 weeks. In this study, the medium composition and culture period were confirmed using a bioreactor system to mass-proliferate adventitious roots derived from C. soldanella L. in Jeju island. Also this adventitious root line developed a new medicinal material could increase value of the bio-industry ingredient through quantitative and qualitative screening of phyto-bioactive compounds.

Petiole explants from 20 cultivars of Catharanthus roseus were cultured on various shoot-inducing media to assess their competence for adventitious shoot formation. After eight weeks of culture on Murashige and Skoog' s medium supplemented with 4.4 $\mu\textrm{m}$6-benzyladenine and 0.5 $\mu\textrm{m}$$\alpha$-naphthaleneacetic acid, petiole explants from 'Cooler Icy Pink' exhibited the greatest frequency of adventitious shoot formation at 40%, which was followed by 'Little Bright Eye'. By comparing with a previous study on assessment of competence for adventitious shoot formation in hypocotyl explant cultures of various cultures, it is indicated that the relative degree of their competence among cultivars varies to the organ used for the source of explant. Excised adventitious shoots were readily rooted on half-strength MS basal medium. Regenerated plantlets were successfully transplanted to potting soil and grown to maturity in a greenhouse.
미류나무 절간조직(2년생)을 이용하여 식물생장조절물질이 함유된 MS배지에서 대량의 식물체를 증식시켰다. 대부분의 식물체는 BA 1.0 + TDZ 0.1 mg/L가 함유한 배지에서 줄기의 형성층 부분에서 절편당 $24.6\;{\pm}\;4.6$개의 줄기가 유기되었으며, 몇몇 줄기는 피목 부분의 조직에서 유기되었다. 신장된 줄기는 NAA 0.01 mg/L가 포함된 배지에서 $94.6\%$의 발근이 이루어졌다. 신장된 줄기의 발근은 식물생장조절물질에 따라 차이를 보여 NAA와 IBA에서 $90\%$ 이상의 발근이 촉진되었으나, cytokinin류를 처리한 시료에서는 $50\%$ 이하의 낮은 발근율을 나타냈다. 발근된 유식 물체는 Cell-tray의 인공상토(peatmoss : vermiculite : perlite = 1 : 1 : 1)에 이식하여 8주간 생육후 $98\%$의 높은 생존율을 나타냈다. 본 연구결과, 2년생 미루나무류의 줄기에서 직접 대량의 줄기를 유도하여 대량증식과정을 단순화 시켰을 뿐만 아니라 조직배양묘 경화시 multi-cap을 이용하여 생존율을 향상시킬 수 있었다.
부정아 유도를 통한 E. pellita의 재분화 체계를 확립하고자 기내 발아묘를 재료로 부정아 유도에 미치는 BA 및 NAA 효과를 시험하였다. 기내 부정아 유도는 재료의 노화정도, 절편 및 생장조절제에 따른 차이가 큰 것으로 나타났다. 발아 후 7일된 식물체가 발아 후 14일 된 것 보다 양호한 반응을 보였고, 절편은 자엽 > 배축 > 뿌리 순으로 부정아 유도가 이루어졌다. 뿌리에서는 부정아가 전혀 유도되지 못했다. 적정배지는 MS 배지에 0.5 mg/L BA, 0.2 mg/L NAA로 나타났으며, 유도된 부정아는 DKW + BA 0.2mg/L + NAA 0.01 mg/L 배지에서 정상적인 줄기로 재생되었고, 다경으로 생장되고 자발적인 발근도 이루어 졌다. 이 상의 결과는 어린 자엽과 배축에서 부정아 유도를 통해 E. pellita의 증식이 가능함을 보여주었다.
