The Difference of Ginsenoside Compositions According to the Conditions of Extraction and Fractionation of Crude Ginseng Saponins (추출 및 분획조건에 따른 인삼 조사포닌 중 ginsenoside 조성 차이)

  • This study was carried out to investigate the difference of ginsenoside compositions in crude ginseng saponins prepared by five different methods including three new methods. Two known methods are hot methanol(MeOH) extraction/n-butanol(n-BuOH) fractionation and hot MeOH extraction/Diaion HP-20 adsorption/MeOH elution. Three new methods are hot MeOH extraction/cation AG 50W $absorption/H_2O$ elution/n-BuOH extraction, cool MeOH extraction/Diaion HP-20 adsorption/MeOH elution and direct extraction with ethyl acetate(EtOAc)/n-BuOH. Analysis of ginsenoside composition in the crude saponins by conventional HPLC/RI(Refractive Index) did not show great difference between methods except EtOAc/n-BuOH method. However, HPLC/ELSD (evaporative light scattering detector) employing gradient mobile phase afforded fine resolution of ginsenoside Rf, $Rg_1$ and $Rh_1$, and great difference of ginsenoside compositions between methods. LC/MS revealed that large amount of prosapogenins were produced during the pass through the cation exchange (AG 50W) column being strongly acidic. Six major ginsenosides such as $Rb_1,w;Rb_2,$ Rc, Rd, Re and $Rg_1$, 5 prosapogenins and one chikusetsusaponin were identified by LC/MS. A newly established HPLC method employing ODS column and gradient mobile phase of $KH_2PO_4/CH_3CN$ revealed that malonyl ginsenosides were detected only in the crude saponin obtained from cool MeOH extraction.

  • Parameter estimations were made for the reversed-phase adsorption of perillyl alcohol (POH), a potent anti-cancer agent, on octadecylsilyl-silica gel (ODS). The average particle diameter of ODS was about $15\;{\mu}m$, and the particles were packed in the column $(3.9\;\times\;300mm)$. The mobile phase used was a mixture of acetonitrile and water, in which the acetonitrile ranged between 50 and $70\;(v/v\;\%)$. The first absolute moment and the second central moment were determined from the chromatographic elution curves by moment analysis. Experiments were carried out using POH solutions within the linear adsorption range. The fluid-to-particle mass transfer coefficient was estimated using the Wilson-Geankoplis equation. The axial dispersion coefficient and the intra particle diffusivity were determined from the slope and intercept of a plot of H vs $1/u_0$, respectively. The contributions of each mass-transfer step were axial dispersion, fluid-to-particle mass transfer, and intraparticle diffusion.

  • Perchlorate ($ClO{_4}^-$) is the material that is used as propellants of rockets and material of explosive as a form of ammonium perchlorate salts. Ammonium perchlorate solution of high concentration is recovered from expired rocket through demilitarization process by the water-jet method. If people take perchlorate in food and water, it interferes with adsorption of iodide which is the substance needed to synthesize thyroid hormone in the thyroid gland. It has an bad influence upon disturbing pregnancy and synthesis of growth hormone. So the effective method is necessary to remove perchlorate anion in water. By considering economic aspect, we studied effective desorption (regeneration) of perchlorate anion from adsorbent with studies on removal and adsorption of perchlorate anion. Desorption experiment was conducted as batch type. Depending on various conditions (concentration, pH, cation anion form) elution, we evaluated amount, efficiency of desorption(amount of adsorption/desorption ${\times}$ 100). Also, research confirmed the efficiency of mixed resins between anion exchange resin and activated carbon and expected synergic effect from advantages of both adsorbents.

A Study on Phosphate Removal Efficiency by Pre-Treatment Conditioning of Oyster Shells (굴 패각의 전처리 조건에 따른 인산염 제거효율에 관한 연구)

  • In this study, we investigated phosphate removal efficiency according to pretreatment (pyrolysis temperature, pyrolysis time, particle size) of oyster shells as a basic study for their recycling. And XAFS analysis and isothermal adsorption experiments were performed to investigate the phosphate removal characteristics of oyster shells. As a result, the removal efficiency was good at $600^{\circ}C$ pyrolysis temperature with 6 hour pyrolysis time and 0.355 ~ 0.075 mm particle size. Isothermal adsorption experiments showed that the Langmuir model is suitable for adsorption of oyster shells. XAFS analysis showed that calcium phosphate formed on the oyster shell pyrolyzed at $600^{\circ}C$. In other words, it was confirmed that the formation of calcium phosphate by the calcium ion elution of the oyster shell contributes to the decrease of phosphate concentration.

Separation and Adsorption-Desorption Characteristics of Heavy Rare Earth Elements (Gd, Tb, Dy) using P507 Resin (P507 추출수지를 이용한 중희토류 원소(Gd, Tb, Dy)의 흡탈착 분리특성에 관한 연구)

  • This study was conducted to establish the adsorption-desorption mechanism and the optimum condition of chromatographic operation for separations of heavy rare earth elements (Gd, Tb, Dy) using a p507-containing resin. By employing Langmuir and Freundlich isotherm together with pseudo first and second order kinetics, absorption-desorption reaction mechanism was investigated. Langmuir and Freundlich isotherm was applied under assumption that adsorption reaction occurs in form of monolayer, and because the result was identical to the assumption, now we know adsorption of heavy rare earth elements occurs in form of monolayer. Concerning the pseudo first and second order kinetic, the pseudo second order seemed to be more suitable to represent heavy rare earth element adsorption mechanism. By using the extraction chromatography to separate heavy rare earth elements, ${\alpha}^{Tb}_{Gd}=1.24$, and ${\alpha}^{Dy}_{Tb}=1.03$ were confirmed in eluent HCl 0.25 M which indicates almost perfect separations of three elements. Furthermore, as concentrations of eluent became higher, the resolution value decreased and the elution area got shortened.

Synthesis of CHBM-DVB Ion Exchanger and It's Ion Exchange Behavior (CHBM-DVB 이온교환체의 합성과 그의 이온교환작용)

  • The salicylic acid-formaldehyde condensed copolymer (SAF) and the 3-carboxy-4-hydroxy benzyl methacrylate divinylbenzene copolymer (CHBM-DVB) were synthesized. The capacities of SAF and CHBM-DVB ion exchangers were 4.6meq/g and 3.9 meq/g, respectively. The adsorption of several heavy metal ions on these weakly acidic ion exchangers and the property of ion exchanger were discussed. The distribution coefficients in hydrochloric acid solution and the elution behavior of several heavy metal ions were also discussed.

A study on the separation and recovery of uranium (우라늄의 분리 및 회수에 관한 연구)

  • The anionic exchange resins with the Dowex-1 and Amberlite CG-400 form were transformed into resin of sulfate and acetate acid form, respectively. The uranyl complex ions with SO$_{4}$$^{2-}$ and CH$_{3}$COO$^{-}$ were adsorbed on the anion exchange resion mentioned above, and these complex ions were eluted as mixture eluents of 0.7M HNO$_{3}$ - 0.5M NH$_{4}$NO$_{3}$ by anion exchange chromatography. The optimum adsorption conditions of uranyl anion complex ions adsorbed on the upper of the resin colmun were 1.5-2.0 ml/min of flow rates at pH 2.0 and adsorptive power of uranyl complex ion of sulfuric acid type were nearly consistent with the Caussion normal distribution curve, whereas the elution state of UO$_{2}$(Ac)$_{2}$$^{4-}$ with acetic acid type was departed. The weighing form obtained from resin of sulfuric acid and aceric acid type was U$_{3}$O$_{8}$ whose recovery was 91.7%. The possibility of recovering uranium from the monazite sulfate solution using a strong base anion resin, Amberlite CG-400(sulfate form), was successfully recovered more than 90%.

  • Activated carbons (ACs) are widely used in adsorption for the removal of gaseous and aqueous pollutants[1,2]. The microbicidal property of ACs is also very important, in order to decrease the risk of water and air contamination with microorganisms. For this purpose, ACs have impregnated with silver or with metallic oxides[2]. However, in the case of Ag supported ACs prepared by impregnation, there are some problem, such as, heat-treatment, highly decrease in specific surface area and pore volume by deposited Ag, and rapidly elution of Ag at the initial stage of usage[3]. Characteristics of Protein Chromatography by Affinity Membrane Mudule (친화성 막모듈에 의한 단백질 크로마토그래픽 특성)

  • Protein affinity membrane was prepared via the coating of chitosan gel on the porous flat polysulfone membrane surface, followed by the immobilization f the reactive dye (Cibacron Blue 3GA) to the chitonsan gel. The maximum protein binding capacity of affinity membrane was about 70${\mu}g/cm^2$ determined by the batch adsorption experiments of human serum albumin (HSA). Using module of this membrane, the characteristics of protein chromatography were investigated through the experiments of elution and frontal chromatography of HSA. This membrane module promises as a chromatography column, since it represented a lower pressure drop and a greater reproducibility. The protein separation ratio was significantly influenced by the flow rate of mobile phase and the injection quantity of HSA. The dynamic protein binding capacity of module decreased from the equilibrium binding capacity with increasing flow rate and approached the value of 15 - 20 ${\mu}g/cm^2$ for flow rates above 6 mL/min.

Characterization of Adsorbents for the Solid-Phase Extraction of Trace Diphenylhydantoin from Biological Samples (생체시료로 부터 미량 디페닐히단토인의 고상추출에 사용될 흡착제의 특성)

  • Ths sorption and desorption properties of four different solid adsorbents were evaluated for the trace enrichment of diphenylhydantoin from biological samples. Graphitized carbon black(GCB) gave the highest adsorption coefficient. And among the organic solvents examined, methanol gave the highest desorption coefficient. Using the GCB column, the optimum elution volume of the eluting solvent was evaluated from the breakthrough curve of diphenylhydantoin. The usefulness of GCB as the solid adsorbent was examined for the solid-phase extraction of diphenylhydantoin from serum in the concentration range of $20-50\;{\mu}g/ml$.
