  • We investigated the synergistic apoptotic effects of co-treatments with Chios gum mastic (CGM) and eugenol on G361 human melanoma cells. An MTT assay was conducted to investigate whether this co-treatment efficiently reduces the viability of G361 cells compared with each single treatment. The induction and augmentation of apoptosis were confirmed by DNA electrophoresis, Hoechst staining, and analyses of DNA hypoploidy. Western blot analysis and immunofluorescent staining were also performed to evaluate expression and translocation of apoptosis-related proteins following CGM and eugenol co-treatment. Proteasome activity and mitochondrial membrane potential (MMP) changes were also assayed. The results indicated that the co-treatment of CGM and eugenol induces multiple pathways and processes associated with an apoptotic response in G361 cells. These include nuclear condensation, DNA fragmentation, a reduction in MMP and proteasome activity, an increase of Bax and decrease of Bcl-2, a decreased DNA content, cytochrome c release into the cytosol, the translocation of AIF and DFF40 (CAD) into the nucleus, and the activation of caspase-9, caspase-7, caspase-3, PARP and DFF45 (ICAD). In contrast, separate treatments of $40{\mu}g/ml$ CGM or $300{\mu}M$ eugenol for 24 hours did not induce apoptosis. Our present data thus suggest that a combination therapy of CGM and eugenol is a potential treatment strategy for human melanoma.

  • Cytokine-induced apoptosis inhibitor 1 (CIAPIN1), known as an anti-apoptotic and signal-transduction protein, plays a pivotal role in a variety of biological processes. However, the role of CIAPIN1 in inflammation is unclear. We investigated the protective effects of CIAPIN1 in lipopolysaccharide (LPS)-exposed Raw 264.7 cells and against inflammatory damage induced by 12-O-tetradecanoylphorbol-13-acetate (TPA) in a mouse model using cell-permeable Tat-CIAPIN1. Transduced Tat-CIAPIN1 significantly reduced ROS production and DNA fragmentation in LPS-exposed Raw 264.7 cells. Also, Tat-CIAPIN1 inhibited MAPKs and NF-κB activation, reduced the expression of Bax, and cleaved caspase-3, COX-2, iNOS, IL-6, and TNF-α in LPS-exposed cells. In a TPA-induced animal model, transduced Tat-CIAPIN1 drastically decreased inflammation damage and inhibited COX-2, iNOS, IL-6, and TNF-α expression. Therefore, these findings suggest that Tat-CIAPIN1 might lead to a new strategy for the treatment of inflammatory skin disorders.

  • Long non-coding RNAs (lncRNAs) are involved in multiple cellular events, as well as in tumorigenesis. Colon cance-rassociated transcript-1 (CCAT1) gene encodes an lncRNA whose over-activation was observed in an expanding list of primary human solid tumors and tumor cell lines, however its biological roles in acute myeloid leukaemia (AML) has not been reported yet at present. In this study, the aberrant upregulation of CCAT1 was detected in French-American-British M4 and M5 subtypes of adult AML patients. By gain- and loss-of-function analysis, we determined that CCAT1 repressed monocytic differentiation and promoted cell growth of HL-60 by sequestering tumor suppressive miR-155. Accordingly, a significant decrease in miR-155 level was detected in AML patients. Reintroduction of miR-155 into HL-60 cells restored monocytic maturation and repressed cell proliferation. Furthermore, CCAT1 could up-regulated c-Myc via its competing endogenous RNA (ceRNA) activity on miR-155. In conclusion, these results revealed new mechanism of lncRNA CCAT1 in AML development, and suggested that the manipulation of CCAT1 expression could serve as a potential strategy in AML therapy.

  • To fully utilize scarce spectrum resource, it's necessary to develop and evaluate promising business models prior to making technology R&D plan and industrial promotion policy. The purpose of this paper is to design potential business models, evaluate the propriety of commercializing the models, and discuss promotion policies after exploring promising sectors consuming spectrum resources. The research is based on TV white space, which is vacant TV channels in region or time domain and considered as core spectrum resource along with digital terrestrial television switchover. As the result, four kinds of business models were derived, including broadcasting and telecommunication types. Each model was discussed from four standpoints: customer value proposition, profit formula, key resources, and key processes, and the propriety for commercialization was evaluated by three dimensions: technological evaluations, business-oriented evaluations, and user-oriented evaluations. The promotion policies of government and market participants for the activation of TV White space-based business models were discussed as well.

  • The purpose of this study is to analyze the actual condition of the already established rural theme park, draw the weak and strong points, and come up with an efficient performance plan for Changwon Persimmon Theme Park through the benchmarking of the best overseas case. According to the analysis, it is necessary to introduce a facility and a program for family customers as a plan for promoting Changwon Persimmon Theme Park. It is required to secure safety through public operation and management, improve scalability of the facility through the establishment of multi-functional complex facility, and come up with a plan for operating a special program associated with local resources. Therefore, this study drew three activation factors: the establishment of operation and management system, the development of operation contents by facility, and the establishment of operation in each area. First, to establish the operation and management system, it is necessary to classify the organization of the theme park into the operation business area and into operation & management area by job function. Secondly, it is necessary to establish a special program in consideration of space and facility, suggest a differentiated plan for operation contents, and create a facility operation plan and program for Changwon Persimmon Theme Park. Lastly, to operate and manage Changwon Persimmon Theme Park smoothly, it is necessary to set the theme park space as open collaboration space which the local residents can share and participate in by avoiding the conventional administration-based business performance and operation. This study suggested the plan for establishing an operation and management system for Changwon Persimmon Theme Park and its operation, the establishment of a special program, a business performance strategy, and the use of internal and external spaces of the theme park.

  • The development of IT technology help citizens demand information and public services through internet that they want, and the changes also help the government converge public opinions and become important measures for citizens to participate in deciding governmental policies. So, this thesis makes an overture of a possibility for utilizing PCRM, a way for citizens to participate in Police Administration, by utilizing IT technologies in which everybody can participate without difficulty. The utilization ways are, on a large scale, classified with a macroscopic aspect and microscopic aspect. One of the macroscopic aspects is, first, the consideration of access to services and convenience in utilization. Second, services should have a both-sidedproperty. Ultimately, citizens'participation should have a positive mutual participation level. One of the microscopic aspects is, first, that the police organizations have to establish a strategy to analyze and fractionate customers. Second, the police organizations have to make an effort to develop policy services and P.R strategies. Third, police organizations have to make an effort to manage customers. Fourth, police organizations have to execute customer management in general. So, all of the police organizations have to maintain unity, to some extent, about citizen participation plazas. In addition, at the same time, consciousness aspects have to be accompanied for the reasonable utilization of PCRM. Citizens have to cultivate citizen consciousness, ownership consciousness for society, and positive participation consciousness to solve social problems.
