Identification of Marker Nucleotides for the Molecular Authentication of Araliae Continentalis Radix Based on the Analysis of Universal DNA Barcode, matK and rbcL, Sequences (범용성 DNA 바코드(matK, rbcL) 분석을 통한 독활(獨活) 유전자 감별용 Marker Nucleotide 발굴)

  • Objectives : Araliae Continentalis Radix and Angelicae Pubescentis Radix have been used as the same medicinal name Korean and Chinese traditional medicines, respectively. The authentic Araliae Continentalis Radix is described only the root of Aralia continentalis in the Korean Pharmarcopoeia. However, the dried root of Angelica biserrata, Levisticum officinale, or Heracleum moellendorffii also has been distributed adulterants of Araliae Continentalis Radix. To develop a reliable method for identifying Araliae Continentalis Radix from adulterants, we carried out the analyses of universal DNA barcode sequences.Methods : Four plants species were collected from different habitate and nucleotide sequences of matK and rbcL were analyzed. The species-specific sequences and phylogenetic relationship were estimated using entire sequences of two DNA barcodes, respectively.Results : In comparative analysis of matK sequences, we were identified 104 positions of marker nucleotide for Ar. continentalis, 3 for An. biserrata, 4 for L. officinale and 8 for H. moellendorffii enough to distinguish individual species, respectively. Furthermore, we obtained marker nucleotides in rbcL at 42 positions for Ar. continentalis, 5 for An. biserrata and 2 for H. moellendorffii, but not for L. officinale. The phylogenetic tree of matK and rbcL were showed that all samples were clustered into four groups constituting homogeneous clades within the species.Conclusions : We confirmed that species-specific marker nucleotides of matK sequence provides distinct genetic information enough to identify four species. Therefore, we suggest that matK gene is useful DNA barcode for discriminating authentic Araliae Continentalis Radix from inauthentic adulterants.

NUMERICAL ANALYSIS OF THE IMPACTING AND SPREADING DYNAMICS OF THE ELLIPSOIDAL DROP ON THE PERFECT NON-WETTING SOLID SURFACE (완전 비습윤 고체 표면 위 타원형 액적의 충돌 및 퍼짐 거동에 대한 수치적 연구)

  • Leidenfrost drops with ellipsoidal shaping can control the bouncing height by adjusting the aspect ratio(AR) of the shape at the moment of impact. In this work, we focus on the effect of the AR and the impact Weber number(We) on the non-axisymmetrical spreading dynamics of the drop, which plays an important role in the control of bouncing. To understand the impact dynamics, the numerical simulation is conducted for the ellipsoidal drop impact upon the perfect non-wetting solid surface by using volume of fluid method, which shows the characteristics of the spreading behavior in each principal axis. As the AR increases, the drop has a high degree of the alignment into one principal axis, which leads to the consequent suppression of bouncing height with shape oscillation. As the We increases, the maximum spreading diameters in the principal axes both increase whereas the contact time on the solid surface rarely depends on the impact velocity at the same AR. The comprehensive understanding of the ellipsoidal drop impact upon non-wetting surface will provide the way to control of drop deposition in applications, such as surface cleaning and spray cooling.

  • One of the important problems in recent AC-PDP technology is high efficiency. In this research, we have been investigated electro-optical characteristics of MgO protective layer after radio frequency(RF) plasma treatment using Ar, $O_2$, and $H_2$ gases. The breakdown voltage order was $O_2$ > Ar > Nontreatment > $H_2$. Also, brightness order was $O_2$ > Ar > Non-treatment > $H_2$. In this experiment, the best result was obtained after $O_2-plasma$ treatment.

  • 최근 저온 대기압 플라즈마 장치의 개발로 대기 및 수질 환경, 바이오 메디컬분야로의 응용 연구가 활발히 진행되어 공기 중 플라즈마의 살균 및 정화효과에 대한 많은 결과가 발표되어 왔다. 본 연구는 면방전 구조의 DBD플라즈마 소스를 제작하여 He과 Ar 기체를 유입하여 미생물인 E.Coli의 변화를 관찰하였다. 면방전 구조의 DBD플라즈마 소스는 1.8 mm 두께의 유리기판위에 포토리소그라피 공정으로 미소전극을 형성하여 고밀도의 방전 셀을 형성하였으며 방전시 발생하는 열 효과를 제어하기 위하여 냉각장치를 제작하여 장착했다. 또한 유리기판과 포토 리소그라피 공정은 방전영역에 제한없이 다양한 크기의 소스제작이 가능하다. 셀 피치가 $400{\mu}m$이며 $cm^2$ 당 200여개의 방전 셀로 구성되어 있어서 기존 메쉬타입의 DBD플라즈마 장치에 비해 균일하게 플라즈마를 조사할 수 있으며 플라즈마 제트 장치에 비해서는 넓은 면적을 동시에 조사할 수 있게 되었다. Ar 과 He기체를 3 L/min의 유량으로 방전공간에 유입하면서 1kV의 구동전압으로 플라즈마를 발생 하였으며, 플라즈마의 조사시간을 20 s, 40 s, 60 s 간격으로 변화를 주어 E.Coli의 변화를 관찰하였다.

Resistance to Anthracnose Caused by Colletotrichum acutatum in Chili Pepper(Capsicum annuum L.)

  • Pepper fruit anthracnose, caused by Colletotrichum acutatum, results in serious yield loss and affects crop quality in many Asian countries, making it a disease of economic consequence. A source resistant to C. acutatum was identified by the AVRDC within the line Capsicum chinense Jacq. PBC932. The resistant breeding line C. annuum AR is the $BC_3F_6$ generation derived from C. chinense Jacq. PBC932. The inheritance of resistance to C. acutatum was analyzed in segregating populations derived from the two crosses HN 11$\times$AR and Daepoong-cho$\times$AR. Detached mature green fruits were inoculated using microinjection method. The disease response was evaluated as the disease incidence at 7 DAI. The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_R$ populations derived from the two crosses fit significantly to a 1:3 Mendelian model. This indicates that the resistance of AR to C. acutatum is controlled by a single recessive gene.

Inhibitory effects of Synurus excelsus and Weigela subsessilis on aldose reductase and HPLC-UV analysis of scopolin, scopoletin, and quercetin

  • The inhibition of aldose reductase (AR) has been shown to prevent the progression of the many complications associated with diabetic hyperglycemia. Several compounds purified from various plant sources have exhibited potent inhibition against AR. In this study, the inhibitory effects of the methanol extracts of the flowers of Synurus excelsus and Weigela subsessilis on AR were determined in vitro. Scopolin and scopoletin are coumarins isolated from the flowers of S. excelsus and W. subsessilis; and quercetin is a known AR inhibitor present in many flowers. To determine and quantify their presence in both plants, HPLC-UV analysis of all three compounds was performed. S. excelsus and W. subsessilis showed potent inhibition against AR having $IC_{50}$ values of 0.17 and $0.14{\mu}g/mL$, respectively. The concentration of scopolin in S. excelsus and W. subsessilis were 34.71 and 174.14 mg/g extract, respectively. Scopoletin was detected in S. excelsus at 3.41 mg/g extract, whereas quercetin was not detected in both plants. This study shows that S. excelsus and W. subsessilis exhibited promising AR inhibitory effects and are both sources of coumarins.

Surface Reactions on the Bi4-xLaxTiO3O12 Thin Films Etched in Inductively Coupled CF4/Ar Plasma (유도결합 CF4/Ar 플라즈마에 의한 Bi4-xLaxTiO3O12 박막의 식각 표면 반응)

  • Etching species in CF$_4$/Ar plasma and the behavior of etching rate of Bi$_4$-$_{x}$L$_{x}$rTi$_3$O$_2$ (BLT) films were investigated in inductively coupled plasma (ICP) reactor in terms of etch parameters. The etching rate as functions of CF$_4$ contents showed the maximum 803 $\AA$/min at 20% CF$_4$ addition in CF$_4$/Ar plasma. The increase of RF power and DC bias voltage caused to an increase of etch rate. The variation of relative volume densities for F and he atoms were measured with the optical emission spectroscopy (OES). The chemical states of BLT were investigated with using X-ray photoelectron spectroscopy (XPS). XPS narrow scan analysis shows that La-fluorides remained on the etched surface. The presence of maximum etch rate at CF$_4$(20%)/Ar(80%) may be explained by the concurrence of two etching mechanisms such as physical sputtering and chemical reaction. The roles of he ion bombardment include destruction of metal (Bi, La, Ti)-O bonds as well as assistant for chemical reaction of metals with fluorine atoms.oms.

  • Mobile IP provides hand-held devices with mobility which allows the user to do work over the network. However, handover time due transfer between access routers causes network delays and data loss. L2Trigger Handover expects this handover to take place, and executes L3 handover before L2 handover takes place, thereby reducing overall handover latency, although it still is an issue since handover latency between AR is not completely eliminated in L2 trigger handover. In this paper took into consideration where MIH is used in MIPv4 and using MIH Table when handover is about to occur in MN(Mobile Node), thereby pre-fetching data needed by Handover. In this way, when the handover is estimated, it improves the init time that L2trigger had. Furthermore we can find that we can execute the handover with shorten init time in smaller and narrow overlap length

  • A statistical approach for the segmentation of speed signals is described in this paper. The main idea of this algorithm is the use of three AR models. Two fixed models are identified at the stationary parts of the signal before and after the spectral change. Changes are detected when the distance between these two models is high. Another model is located between two fixed models and is used to estimate spectral change time. This segmentation algorithm has been tested with connected words and compared to classical methods. The results showed that it can provide more accurate locations of boundaries of segments and can reduce the amount of oversegmentation.

An Economic Feasibility Study of AR CDM project in North Korea (북한 지역을 대상으로 한 조림 CDM 사업의 경제적 타당성 연구)

  • Potentials of AR CDM project in North Korea are assessed and feasible land area for AR CDM project is estimated. According to our estimation, There could be 515,000 hectares of forest lands deforested before 1990 in North Korea and 8,854 hectares at the regional level of Gae-sung City, which are eligible for AR CDM project, based on researches of satellite image analyses conducted from 1980's to 1990's. A baseline scenario assumed 44.73 tones of carbon stored in soil per hectare with no vegetation above ground remained during the project period following the default value of IPCC's Good Practice Guidance for LULUCF considering soil structure, climate and land use of the project area. The scenario also assumes that black rocust (Robinia pseudoacacia) is planted and the CDM project is implemented for 20 years. The costs for producing greenhouse gases CER (certified emission reduction) credits include costs of tree planting and forest management, and costs of project negotiation and transactions for issuing the credits. It is estimated that 376 tones of carbon dioxide per hectare can be accumulated and 503 temporary CER credits per hectare and 265 long-term CER credits per hectare could be produced during the project period. It is estimated to cost US$ 4.04 and US$ 7.67 to provide one unit of temporary credit and long-term credit, respectively. These values can be regarded as the cost of conferring emission commitment of a country or a private entity. However, it is not clear which option is better economically because the replacement periods are different in these two cases.
