Cloning of Chondroitinase ABC from Bacteroides stercoris HJ-15, a Human Intestinal Anaerobic Bacterium (사람 장내세균군집 유래 Bacteorides stericoris HJ-15의 Chondroitinase ABC의 클로닝)

  • The gene encoding chondroitinase ABC from a genomic library of Bacteroides stercoris HJ-15, which was isolated from human feces, was cloned. The cloned gene consisted of 3,090 bp and was predicted to encode a 1,029−amino-acid protein. The B. stercoris chondroitinase ABC gene was not homologous to other chondroitinase ABC genes; however, its amino acid sequence showed 71% homology to that of Bacteroides thetaiotaomicron. The gene was cloned in the pET-26b+ expression vector and expressed under the T7 promoter in Escherichia coli BL21(DE3). The purified recombinant chondroitinase ABC degraded chondroitin sulfates A, B, and C.

Isolation and Characterization of the Colletotrichum acutatum ABC Transporter CaABC1

  • Fungi tolerate exposure to various abiotic stresses, including cytotoxic compounds and fungicides, via their ATP-driven efflux pumps belonging to ATP-binding cassette (ABC) transporters. To clarify the molecular basis of interaction between the fungus and various abiotic stresses including fungicides, we constructed a cDNA library from germinated conidia of Colletotrichum acutatum, a major anthracnose pathogen of pepper (Capsicum annum L.). Over 1,000 cDNA clones were sequenced, of which single clone exhibited significant nucleotide sequence homology to ABC transporter genes. We isolated three fosmid clones containing the C. acutatum ABC1 (CaABC1) gene in full-length from genomic DNA library screening. The CaABC1 gene consists of 4,059 bp transcript, predicting a 1,353-aa protein. The gene contains the typical ABC signature and Walker A and B motifs. The 5'-flanking region contains a CAAT motif, a TATA box, and a Kozak region. Phylogenetic and structural analysis suggested that the CaABC1 is a typical ABC transporter gene highly conserved in various fungal species, as well as in Chromista, Metazoans, and Viridiplantae. We also found that CaABC1 was up-regulated during conidiation and a minimal medium condition. Moreover, CaABC1 was induced in iprobenfos, kresoxim-methyl, thiophanate-methyl, and hygromycin B. These results demonstrate that CaABC1 is necessary for conidiation, abiotic stress, and various fungicide resistances. These results will provide the basis for further study on the function of ABC transporter genes in C. acutatum.

Study on the Effect of the ABC Dry Chemical on Wood in Pyrolysis (ABC 분말소화약제의 열분해 시 목재에 미치는 영향에 관한 연구)

  • This study the effect of pyrolysis products ABC dry chemical and of monoammonium phosphate on wood surface. When the pyrolysis product was removed from the wood surface, monoammonium phosphate was removed due to the high viscosity of the transparent pyrolysis product, but the ABC dry chemical was removed in a lump form. Thermal analysis showed that the pyrolysis characteristics of each sample were similar but the weight of pyrolysis residue was 55.9% for ABC dry chemical and 25.2% for monoammonium phosphate. The additives added to the ABC dry chemical also affect the weight of the pyrolysis residue and the fire protection effect of metaphosphoric acid.

Isolation and Characterization of Novel Chlorella Species with Cold Resistance and High Lipid Accumulation for Biodiesel Production

  • Chlorella spp. are green algae that are found across wide-ranging habitats from deserts to arctic regions, with various strains having adapted to survive under diverse environmental conditions. In this study, two novel Chlorella strains (ABC-002, ABC-008) were isolated from a freshwater lake in South Korea during the winter season and examined for possible use in the biofuel production process. The comparison of ABC-002 and ABC-008 strains with Chlorella vulgaris UTEX265 under two different temperatures ($10^{\circ}C$, $25^{\circ}C$) revealed their cold-tolerant phenotypes as well as high biomass yields. The maximum quantum yields of UTEX25, ABC-002, and ABC-008 at $10^{\circ}C$ were 0.5594, 0.6747, and 0.7150, respectively, providing evidence of the relatively higher cold-resistance capabilities of these two strains. Furthermore, both the biomass yields and lipid content of the two novel strains were found to be higher than those of UTEX265; the overall lipid productivities of ABC-002 and ABC-008 were 1.7 ~ 2.8 fold and 1.6 ~ 4.2 fold higher compared to that of UTEX265, respectively. Thus, the high biomass and lipid productivity over a wide range of temperatures indicate that C. vulgaris ABC-002 and ABC-008 are promising candidates for applications in biofuel productions via outdoor biomass cultivation.

  • This study tried to identify the key success factors of ABC by identifying the influence of ABC's success factors on project performance and analyzing the moderating effect of communication. For this purpose, factors applicable to construction projects were extracted through case studies related to the success factors of ABC, an activity-based financial management technique. The survey method was conducted as an online survey method using the Delphi method. For statistical analysis, frequency analysis and factor analysis were performed with SPSS Statistic 20, and hypothesis testing was performed with SmartPLS 2.0. As a result of the analysis, it was found that linkage with quality initiatives affects not only ABC's success factors on project performance, but also communication moderation effects. It was confirmed that linkage and communication with quality initiatives are the most important key success factors for ABC's success. Based on the results of this study, it is expected that if ABC and quality management are well linked, it will be effective in improving project performance.

Prediction and Annotation of ABC Transporter Genes from Magnaporthe oryzae Genome Sequence (벼도열병균 게놈서열로부터 ABC transporter 유전자군의 예측 및 특성 분석)

  • Magnaporthe oryzae is destructive plant-pathogenic fungus and causes rice blast. The pathogen uses several mechanisms to circumvent the inhibitory actions of fungicides. ATP-binding cassette (ABC) transporters are known to provide protection against toxic compounds in the environment. PC facilitated bioinformatic analysis, particularly with respect to accessing and extracting database information and domain identification. We predicted ABC transporter genes from the M. oryzae genome sequence with computation and bioinformatics tools. A total of thirty three genes were predicted to encode ABC transporters. Three of thirty three putative genes corresponded to three known ABC transporter genes (ABC1, ABC2 and ABC3). Copy numbers of the ABC transporter genes were proven by Southern blot analysis, which revealed that twenty genes tested exist as a single copy. We amplified the DNA complementary to RNA corresponding to eleven of these by reverse transcriptase polymerase chain reaction.

  • As a short lived group, ABC had an important effect on the design of Modern architecture. Because it played an important role in the foundation of CIAM, and affected the style of Internationalism and Neue Sachlichkeit in Modern architecture. ABC group was the result of Elementarism and Constructivist International. And El Lissitzky was a central figure in the birth of ABC group as a Constructivist. ABC's design resembled the design of ASNOVA, but it modified it as a more real and functional design. This study amins at the analysis of the effects of ABC group on the design of Modern architecture.

  • The characteristic of this study is to compare and analyze costing information from ABC cost accounting system after ABC cost accounting system is designed and implemented from the manufacturing company's traditional cost accounting system through field researches. This is different from previous researches which investigate surveys or adoption procedures and contents of some companies already adopting ABC cost accounting system. Statistically, differences between costing information are not really similar. However, the advantage of ABC cost accounting system is to reduce some activities which do not well contribute creating value after analyzing productions and supporting activities of the company, and dividing into two groups, activities creating value and non-creating value. Therefore, the cost accounting information of ABC cost accounting system is very useful system to offer valuable information for the management of the company. The accuracy of information of ABC cost accounting system depends on setting up the central point of activities and implementing cost drivers. In particular, when it comes to supporting activities, choosing cost accounting grivers which are highly relevant to producing products and collecting data for cost accounting drivers is really difficult. This is the limitation of this study. In other words, when implementing cost accounting drivers of ABC cost accounting system, aconceptual framework is different from applying to the real producing place. Therefore, when implementing on ABC cost accounting system, future researches are needed to find cost accounting drivers which are more close to the theoretical model, and implement ABC cost accounting system which can collects relevant data.

  • In this study it is intended to develop the ontology which can express the relation between objects with emphasis on the structural representation of semantics. Its interoperability with other kinds of previous ontology and metadata was also considered so that the developed ontology may be applicable to the real situation. The ABC Ontology can get extended into another field where the application of the concept of event is Possible, for ABC Ontology Provides the fundamental framework on the axis of event. In this study it is Music where ABC Ontology can be applied properly, which results in creating Music Ontology. Music Ontology Provides the infrastructure of knowledge for reasoning of Potential meaning as well as the simple semantic connection of terms. The extended model of ABC Ontology has been developed by applying Music Ontology, which is the domain ontology and conveys meaning, to ABC Ontology that represents the whole framework. The representation of conceptual relation in ABC Ontology turns into the association of the framework and meaning in the extended model of ABC Ontology, with reasoning rules which are typical in ontology Also, interoperability of the extended model of ABC Ontology is examined in consideration of co-operating with metadata different from those in it.
