Effects of Garlic on Uncoupling Protein 2 (UCP2) Transcriptional Regulation in Metabolic Tissues of UCP2 Transgenic Mice Fed on a High-Fat Diet (마늘이 고지방 식이를 섭취한 UCP2 형질전환 마우스의 대사성 조직에서 UCP2 전사 조절에 미치는 영향)

  • This study was performed to investigate the effects of garlic on uncoupling protein 2 (UCP2) transcriptional regulation of UCP2-luciferase transgenic mice fed on a high fat diet to induce obesity. To examine the transcriptional regulation of UCP2, we generated transgenic mice with a UCP2 promoter (-1,830/+30 bp) containing luciferase as a reporter gene. UCP2-luciferase transgenic mice were fed a 45% high-fat diet for 8 weeks to induce obesity. Subsequently, mice were maintained on either a high-fat control diet (TG-CON), or high-fat diets supplemented with 2% (TG-GL2) or 5% (TG-GL5) garlic for a further 8 weeks. Dietary garlic reduced body weight and energy efficiency ratio in the TG-GL5 group, compared to the TG-CON group. Furthermore, garlic supplementation significantly decreased white adipose tissue fat mass and plasma levels of triglycerides, total cholesterol, and leptin in the TG-GL2 and TG-GL5 groups, compared to the TG-CON group. Specifically, UCP2 promoter activity in metabolic tissues such as liver, white adipose tissue, brown adipose tissue, and skeletal muscle was increased by garlic supplementation. These results suggest that dietary garlic was partially associated with an increase of UCP2 transcriptional activity in metabolic tissues for decreasing obesity.

  • The effects of adding goldenrod leaf (GL) and stem powder (GS) (1 and 2%) to sausage meat were evaluated for quality characteristics. The compositional properties, pH, cooking yield, instrumental color, emulsion stability, viscosity, texture, and sensory properties were investigated. Adding GL and GS increased the moisture and ash concentration in sausages. Furthermore, increasing the amount of added GL and GS helped to develop (p<0.05) cooking yield, emulsion stability, and viscosity in the meat batters. Sausages with 2% GL and GS had significantly higher springiness and hardness than those of the control. No significant differences in cohesiveness were observed among the treatments. CIE L*and CIE a*values were highest (p<0.05) in the control sausage. Treatment with 2% GL resulted in the highest CIE b* values (p<0.05) in batter and sausages. No significant difference was observed between the control and other treatments for the overall acceptability. Overall, this study indicated that GL and GS could be used as dietary fiber in sausages to improve quality characteristics.

Glehnia littoralis Root Extract Induces G0/G1 Phase Cell Cycle Arrest in the MCF-7 Human Breast Cancer Cell Line

  • Glehnia littoralis (GL) is widely used as an oriental medicine for cough, fever, stroke and other disease conditions. However, the anti-cancer properties of GL on MCF-7 human breast cancer cells have not been investigated. In order to elucidate anti-cancer properties and underlying cell death mechanisms, MCF-7cells ($5{\times}10^4/well$) were treated with Glehnia littoralis root extract at 0-400 ug/ml. A hot water extract of GL root inhibited the proliferation of MCF-7 cells in a dose-dependent manner. Analysis of the cell cycle after treatment of MCF-7 cells with increasing concentrations of GL root extract for 24 hours showed significant cell cycle arrest in the G1 phase. RT-PCR and Western blot analysis both revealed that GL root extract significantly increased the expression of p21 and p27 with an accompanying decrease in both CDK4 and cyclin D1. Our reuslts indicated that GL root extract arrested the proliferation of MCF-7 cells in G1 phase through inhibition of CDK4 and cyclin D1 via increased induction of p21 and p27. In summary, the current study showed that GL could serve as a potential source of chemotherapeutic or chemopreventative agents against human breast cancer.

  • Fix ℤ/2 is the prime field of two elements and write 𝒜2 for the mod 2 Steenrod algebra. Denote by GLd := GL(d, ℤ/2) the general linear group of rank d over ℤ/2 and by ${\mathfrak{P}}_d$ the polynomial algebra ℤ/2[x1, x2, …, xd] as a connected unstable 𝒜2-module on d generators of degree one. We study the Peterson "hit problem" of finding the minimal set of 𝒜2-generators for ${\mathfrak{P}}_d$. Equivalently, we need to determine a basis for the ℤ/2-vector space $$Q{\mathfrak{P}}_d:={\mathbb{Z}}/2{\otimes}_{\mathcal{A}_2}\;{\mathfrak{P}}_d{\sim_=}{\mathfrak{P}}_d/{\mathcal{A}}^+_2{\mathfrak{P}}_d$$ in each degree n ≥ 1. Note that this space is a representation of GLd over ℤ/2. The problem for d = 5 is not yet completely solved, and unknown in general. In this work, we give an explicit solution to the hit problem of five variables in the generic degree n = r(2t - 1) + 2ts with r = d = 5, s = 8 and t an arbitrary non-negative integer. An application of this study to the cases t = 0 and t = 1 shows that the Singer algebraic transfer of rank 5 is an isomorphism in the bidegrees (5, 5 + (13.20 - 5)) and (5, 5 + (13.21 - 5)). Moreover, the result when t ≥ 2 was also discussed. Here, the Singer transfer of rank d is a ℤ/2-algebra homomorphism from GLd-coinvariants of certain subspaces of $Q{\mathfrak{P}}_d$ to the cohomology groups of the Steenrod algebra, $Ext^{d,d+*}_{\mathcal{A}_2}$ (ℤ/2, ℤ/2). It is one of the useful tools for studying these mysterious Ext groups.

The Effects of Glechoma longituba on Rats Fed High Diet (금전초(金錢草) 추출물(抽出物)이 고지방식이(高脂肪食餌)로 유발한 비만(肥滿) 흰쥐에 미치는 영향(影響))

  • This present study was carried out to investigate the body weight-regulatory effects of Glechoma longituba in high fat diet-induced obese rats. Control group rats were fed with high fat diet and administered normal saline for 8weeks. Experimental groups rats were fed with high fat diet and administered extract of Glechoma longituba for 8 weeks. And observed that, body weight of rats and total cholesterol, triglyceride, free fatty acid, phospholipid in serum of rats. The results were as follows: 1. There were decrease of body weight of rats in GL group, but these results showed no significant efficacy. 2. There were significant decrease of serum total cholesterol level in GL group. 3. There were decrease of serum triglyceride level in GL group, but these results showed no significant efficacy. 4. There were decrease of serum free fatty acid level in GL group. but these results showed no significant efficacy. 5. There were significant decrease of serum phospholipid level in GL group. According to above mentioned results, Glechoma longituba is expected to be applied to the prevention or treatment of obesity and its complications.

  • In this paper, we implement the ski jump visualization tool using the OpenGL graphics library. Among the evaluation elements of jump, the distance of jump is most important. Also, the distance of jump depends on Lift-to-Drag Ratio. In the previous studies, the visualization too according to the consequential changing of variety factors is insufficient. This study provides an easy style flying posture visualization tool to the user who using Lift-to-Drag Ratio. Furthermore, there is to implement an international standard jumping hill by virtue of entered five main data. The results of this study can be applied with reference data for better ski jump.

Psychotropic Effects of Ginseng Saponine on Agonistic Behavior in Male and Female Mice

  • Psychotropic actions of crude ginseng saponins(CGS), pure ginsenoslue Rbl(GS-bl) and gin- senoside Rgl(GS-gl) isolated from the root of Panax ginseng, were evaluated by determining their effects on agonistic behavior in male(Experiment 1) and female(Experiment 2) mice, using a biologically relevant method. The results of experiment 1 demonstrated that CGS and GS-bl significantly suppressed aggressive episodes (offensive sideways posture and attack bite) in a dose-dependent manner when the resident was drugged, whereas G5-gl was ineffective. However, when the intruder was treated with one of three ginseng saponins, agonistic behavior between resident and intruder males was not altered. In experiment 2, acute administration of CGS and G5-bl significantly suppressed maternal aggression, whereas GS-gl was ineffective. As compared with the vehicle-treated group, chronic treatment with CGS and GS-bl significantly suppressed maternal aggression, while GS-gl showed a tendency to increase the frequency of attack bite by females. These findings clearly indicate that the root of Panax ginseng contains psychoactive ingredient, which can suppress both intermale and maternal aggression in mice. We suggest that the present results have important implications for the clinical usefulness of ginseng saponins in psychiatric medicine.

  • An immobilized Trigonopsis variabilis cells having an high activity of D-amino acid oxidase(DAO) was used to convert CPC into GL-7-ACA. The optimal pH of the reaction system was 8.0-8.5, and the optimal temperature was 40$\circ$C. When immobilized cell was used repeatedly in semi-batchwise reaction, the system retained 80% of the initial activity after used of 12 times for over 12 hours. The storage stability of the immobilized cell was maintained for 30 days at 4$\circ$C. The CPC concentration for the maximal reaction rate was about 30 mM and 40 mM for free and immobilized cells, respectively. Substrate inhibition of CPC concentration more than 50 mM was overcomed by 20~25% by immobilization. Pure oxygen supply into reaction system was most efficient in D-amino acid oxidase reaction. Continuous conversion to GL-7-ACA from CPC has been developed with an bioreactor system containing immobilized T variabilis cells. By opera- tion of the reactor for 5 hours, the average conversion yield of >80% and GL-7-ACA production of 40~45 mM per hour could be obtained.

Production of Poly-$\beta$-hydroxybutyrate from Methanol by Fed-batch Cultivation of methylobacterium sp. GL-10 (Methylobacterium sp. GL-10의 유가식 배양에 의한 Methanol로 부터 Poly-$\beta$-hydroxybutyrate의 생산)

  • The production of poly-$\beta$-hydroxybutyrate(PHB) from methanol by batch and fed-batch cultivations of Methylobacterium sp. GL-10 was studied. PHB accumulation was stimulated by the nutrients deficiency including, NH4+, SO42-, and K+. The nitrogen deficiency was the most critical factor for PHB accumulation. In batch cultivation, the maximum cell concentration and PHB content were 1.86g/l and 0.62g/l, respectively, with 1.0%(v/v) of methanol and 0.5g/1 of ammonium sulfate. The mass doubling time of Methylobacterum sp. GL-10 was in the range of 4-5 hrs. The cell growth and PHB accumulation were severely inhibited at the methanol concentration over than 2% (v/v). To overcome methanol Inhibition, constant feeding and intermittent feedillg fed-batch cultivations were adopted, using C/N molar ratio as a control factor. In constant feeding fed-batch process, cell concentration was increased up to 2.67g/1, and PHB yield was enhanced from 0.33 of batch culture to 0.53. The relatively low cell concentration was caused by methanol accumulated in culture broth at late growth phase. To prevent methanol accumulation and to maximize PHB production, DO-state intermittent fed-batch cultivation was attempted. The cell and PHB concentration was reached up to 4.55g/1 and 1.80g/1, respectively. It was possible to maintain methanol concentration low and also to feed nutrient of desired C/N molar ratio.
