초음파 유도 난포란 채란에 의한 젖소 및 한우의 체외수정란과 송아지 생산 (Production of In vitro Fertilized Bovine Embryos and Calves by Ultrasound-guided Ovum Pick-up in Holstein and Hanwoo)

  • The objective of this study was to produce calves by transfer of embryos derived from slaughter house(SHD) and ultrasound-guided ovum pick-up (OPU). At 60 hrs after injection of 400 mg FSH dissolved in 25% polyvinylpyrrolidone(PVP) by single dose, ultrasound-guided follicular oocyte aspiration was ferformed. Day-7 and day-8 blastocysts produced by in vitro maturation (IVM), fertilization (IVF) and culture(IVC) of the oocytes derived from SHD and OPU were nonsurgically transferred into recipients. The results obtained were as follows. The cleavage rate and the development rate to blastocysts were not significantly (P<0.05) different between the oocytes obtained by SHD (72.9% vs. 34.1%) and OPU (75.9% vs. 38.4%). The oocyte recovery rate from the number of follicles by ultrasound-guided aspiration were not significantly (P<0.05) different between Holstein (61.7%) and Hanwoo (60.1%), but the rate of oocytes useful for IVF was significantly (P<0.05) higher in Hanwoo (69.3%) than Holstein (59.6%). The cleavage rate and the development rate to blastocysts was not significantly (P<0.05) different between Holstein (74.9% vs. 39.2%) and recipients on day 8 of estrus cycle resulted in 13 pregnancies (34.2%). One of them was sacrificed during gestation period due to mastitis and another was aborted spontaneous. The resulting 14 calves were morphologically normal at birth. Seventy eight fresh OPU-IVF embryos were transferred into 21 recipients on day 8 of estrus cycles, resulting in pregnancy of 12 recipients (41.4%). Two of them were sacrificed during gestation period due to mastitis and the other two were aborted. Nevertheless, the 11 OPU-calves have been born normally.

송아지 난소에서 초음파 유도에 의한 한우의 미성숙난자 채취시에 bST-FSH 처리효과에 관한 연구 (Effect of bST Co-Treatment with FSH on Transvaginal Ultrasound-Guided Oocyte Retrieval(TVR) in Calves)

  • The aim of this study was to evaluate the effect of recombinant bovine somatotropin (bST; Boostin-S, LG Chem) treatment with FSH (Super OV) or PMSG on superovulatory response for transvaginal ultrasound-guided oocyte retrieval (TVR) in calves. Eight Korean Native Cattle(KNC) heifer calves; 150 to 240 days old; were randomly assigned to four treatment groups: 1) FSH(75 mg); 2) FSH (75 mg) + bST(500mg) 3) PMSG(1;000 IU); 4) PMSG(1, 000 IU) + bST(500 mg). Experimental calves in group 1 (n=2) and 2(n=2) were weekly superovulated for 4 consecutive weeks with daily injection of FSH for 3days and the next day subjected to TVR session. Animals in group 3 (n=2) and 4(n=2) were weekly stimulated for 4 consecutive weeks with a single dose of 1, 000 IU PMSG. TVR was performed on 72 hours after PMSG injection. Calves in group 2 and 4 was received injection of 500 mg of bST every 10 days. At each TVR session, follicle number and size were recorded; the oocytes collected and graded according to cumulus and cytoplasm investment. Collected oocyte were determined viable oocyte according to morphological quality with granulation of oocyte and number and status of cumlulus cells. IVM and IVF were performed and assessed cleavage rate on day 3 after fertilization. A Sonovet 600(Medison, Co., Ltd) realtime ultrasound scanner with a 6.5 MHz convex transducer, fixed at the tip of 500 mm estended handle equipped with a needle guide was used in collecting oocyte. Differences between groups were analysed by chi-square test. The population of large follicle ($\geq$5 nun) and aspiration rate were not significant different among the 4 groups. But, the number of small follicles (<5 mm) and aspirated oocyte in the KNC calves treated with bST were 1.3~1.6 times higher than in KNC calves treat with FSH or PMSG alone. In conclusion, the administration of bST with FSH or PMSG at superovulation for TVR in calves was increase the nurnber of small follicle which was influenced the number of aspiratable follicle.

한우 송아지에서의 요도하열(hypospadias) 발생 증례 (A Case of Hypospadias in Korean Native Calf)

  • Hypospadias is a malformation in which the urethra open on the ventral side of the penis and its frequency is about from 0.3% to 0.5% male birth in human. It is considered a disorder both genetic and environmental factors involves in pathogenesis. A KNC(Korean native calf) showing symptoms of fusion defect of the male ventral urethra, penile defect, bifid scrotum and low body weigh was born. To prevent an ascending urinary tract infection, dermatitis and loss of hair, surgical operation was performed to make a genitals like a female. After the operation, the KNC got a regenerative hair and normal urination. However the KNC became anorexia, loss in weight and weak. Seventy days after birth, the KNC died. The view of autopsy was a yellowish hepatomegaly, hydronephrosis, closed ureter and complex disorder in interanl organ. The symptoms of hepatomegaly and hydronephrosis could be due to a fluid therapy as a consequence of ureter obstruction. The normal urination of the KNC before it got sick, suggested that ureter obstruction in the case of this KNC was formed at its postnatal growth stage.

한우 송아지의 성장에 따른 혈액화학치에 관한 연구 (Blood Chemistry Profiles in Indigenous Korean Calves According to Age)

  • Blood chemistry values according to ages have rarely been investigated in indigenous Korean calves. The present study aimed to clarify the changes in blood chemistry values of indigenous Korean calves with age. Blood samples were collected from 29 calves (11 females and 18 males) reared at the Gyeongsangbukdo Livestock Research Institute and the levels of various chemical components in the serum were analyzed. The total protein, albumin and globulin concentration in the calves increased gradually over time. The glucose concentration increased significantly at 2 weeks after birth, and then decreased slightly 4 weeks after birth. The blood urea nitrogen concentration increased from 2 weeks after birth, and the levels in females were higher than those in males at 2 and 4 weeks after birth. The creatinine concentration decreased significantly from 2 weeks after birth, and the levels in females were higher than those in males at 2 weeks and 6 months after birth. The total bilirubin concentration decreased gradually from 2 weeks after birth. The glutamic oxaloacetic transaminase and glutamic pyruvic transaminase concentration increased gradually from 2 weeks after birth. The triglyceride concentration tended to increase from 2 weeks after birth. The total cholesterol concentration increased significantly at 2 weeks after birth, and then decreased slightly 4 weeks after birth. The total high density lipoprotein cholesterol concentration increased significantly from 2 weeks after birth. The calcium concentration was maintained between $9.8{\pm}0.8mg/dL$ and $11.6{\pm}0.9mg/dL$ throughout the investigation period, although some levels fluctuated at 4 weeks and 6 months after birth. The phosphorus concentration increased significantly at 2 weeks after birth, and then decreased slightly 4 weeks after birth. In conclusion, the data obtained from this study may be valuable for use as a standard for interpreting results of blood biochemical analyses in indigenous Korean calves.

소피부사상균이 한우 송아지 경매가격에 미치는 영향 (The effect of bovine dermatophytosis on auction price in Hanwoo calves)

  • This study examined the dermatophytes of calves aged between 6 and 12 months in the Hanwoo calf auction market. Moreover, the research analyzed how dermatophytosis affected the auction price of Hanwoo calves based on their sex and age. The incidence rate of dermatophytosis was found to be 85 cases out of 1,955 calves (4.3%). The major location of dermatophytosis lesions were in the head region. Specifically, the highest prevalence was observed in the forehead (42.4%), followed by the eyes (30.1%), and the ears (18.8%). The auction prices of Hanwoo calves were observed that the average price for normal calves was 2,936,428 won, while calves with dermatophytosis were sold at 2,767,059 won. Comparing auction prices according to gender and age, it was observed that male calves and calves aged between 8 and 12 months had significantly lower auction prices compared to normal calves. The results provided valuable insights into the current situation of dermatophytosis in Hanwoo calves. Moreover, analyzing the impact of dermatophytosis on the auction prices of these calves, it has generated essential data that can serve as a foundation for implementing and enhancing ongoing management and prevention measures for dermatophytosis in cattle.

체외생산된 한우 배반포기배로부터 송아지 생산을 위한 체계 II. 한우 배반포기배를 간편하면서 효율적이고 성공적으로 유리화 동결하는 방법 (Systems for Production of Calves from Hanwoo(Korean Cattle) IVM/IVF/IVC Blastocyst II. Simple, Efficient and Successful Vitrification of Hanwoo Blastocyst)

  • 본 실험은 체외 생산된 한우 배반포기배에 적합한 동결 / 융해 방법을 찾고자 실시하였다. 체외배양 7일째에 생산된 배반포기배는 동해제 EFS40(40% ethylene glycol, 18% ficoll, 0.3 M sucrose 그리고 10% FBS가 첨가된 m-DPBS)과 embryo container인 EM grid (V-G) 또는 straw(V-S)를 이용해서 초자화 동결하였다. 동결과 융해는 두 방법 모두 2 단계로 실시하였으며, 처리시간은 V-G 방법이 2 분과 3분, V-S 방법이 3.5분과 10분 각각 소요되었다. 체외 생존능 평가는 융해 후 24시간째의 재팽창율과 48 시간째의 부화율로 조사하였다. 본 실험에서 얻어진 결과는 다음과 같다. 팽창 배반포기배를 이용하여 동결액 노출과 동결과정의 냉해가 배의 생존에 미치는 영향을 조사하였던 바, 융해 후 24시간째, 동결액 노출군 (100.0%)의 결과는 대조군 (100.0%)과 차이가 없었으며, 두 동결군 (V-G: 87.8%, V-S: 77.8%)의 생존율과 비교해 볼 때 유의하게 높았다 (P<0.00l). 그러나, 융해 후 48시간째 각 처리군의 부화율을 조사하였던 바, V-G 군 (67.8%)은 V-S 군 (53.3%)보다 유의하게 높게 나타났으며 (P<0.05), 동결액 노출군 (73.3%)과도 유의한 차이를 나타내지 않았다. 또한, 배발달단계 (초기, 팽창, 부화초기 배반포)와 동결에 사용된 embryo container(EM grid, straw)가 체외 생존율에 미치는 영향을 동시에 비교하였던 바, embryo container 에 상관없이 빠르게 발달된 배반포기배가 느리게 발달하는 난자군보다 유의하게 높은 생존율을 나타내었다 (초기 : 57.1, 24.4%; 팽창 : 84.7, 60.6%; 부화초기 : 91.7, 80.0%)(P<0.001). 특히, 팽창 배반포기배와 부화초기 배반포기배에서, 융해 후 48시간째, V-G군(67.8, 95.0%)의 부화율이 V-S군(53.0, 65.0%)보다 유의하게 높게 나타나 동결시 EM grid 의 유용성을 확인할 수 있었다 (P<0.05, P<0.001). 따라서, 한우 배반포기배는 EM grid를 사용하는 초자화 동결방법으로 간편하면서도 효율적이고 성공적으로 동결보존 할 수 있다는 것을 알았다.

  • 본 실험은 체외 생산된 한우 배반포기배가 초자화 동결과 1- 단계 융해 후 체외 / 체내에서 성공적으로 생존할 수 있는지의 여부를 확인하고자 실시하였다. 초자화 동결은 glycerol (G) 또는 / 과 ethylene glycol(EG), 그리고 10% FBS가 들어있는 D- PBS 동결액을 이용하여 배반포기배를 10% G 에서 5분, l0%G와 20% EG 혼합액에서 5분 그리고 2S%G와 2S% EG 혼합액에서 30 초간 노출시켰으며, straw에 넣은 후 질소 증기에 3분간 쐬고 액화질소에 침지함으로써 완성하였다. 1-단계 융해는 straw 자체를 $25^{\circ}C$와 36$^{\circ}C$에 각각 5분과 3분간 처리함으로써 이루어졌으며, 융해 후 회수된 난자는 단층배양이 유도된 난구세포 소적에서 48시간 배양하거나 대리모인 소에 이식하였다. 체외 생존능 평가를 융해 후 24시간째의 재팽창율과 48시간째의 부화율로 조사하였을 때, 대조군의 정상 발달 (100.0, 63.3%)보다는 유의하게 낮았지만 초자화 동결군의 결과 (85.4, 43.8%)는 높게 나타났다 (P<0.001, P<0.05). 발달단계에 따른 체외생존능을 조사하였을 때, 빠르게 발달된 배반포기배가 느리게 발달하는 난자군보다 유의하게 높은 생존율을 나타내었다 (부화초기 : 88.0, 48.0%; 팽윤 : 81.1, 45.3%; 초기 : 66.7, 14.3%) (P<0.05). 또한, 배반포기배가 생산된 나이(체외수정 후 7, 8, 9 일) 에 따른 초자화동결군의 체외생존능을 조사하였을 때, 배의 나이가 어릴수록 체외생존능은 유의하게 높은 경향을 나타내었다 (7 일 : 67.3, 34.5%; 8 일 : 76.9, 40.7%; 9 일 : 60.9, 23.9%) (P<0.05). 초자화동결된 한우배반포기배의 1-단계 융해 후 체내생존 가능성을 확인하고자 8마리의 소에 이식하였을 때, 4마리가 임신된 것을 확인하였다. 따라서, 이상의 높은 생존율을 고려하여 볼 때, 본 연구에서 사용된 초자화동결과 1-단계 융해는 기존의 동결란을 융해 이식할 때 필요로 했던 실험장비나 고도의 숙련된 기술 없이도 실험현장에서 쉽게 이용할 수 있으면서 효율적인 결과를 얻을 수 있는 방법이라는 것을 알 수 있었다.

韓牛 牝牛의 Body Condition Score가 繁殖形質에 미치는 影響 (Body Condition Score of Hanwoo Cows and Reproductive Performances Performances)

  • 본 연구는 산차, 계절, 영양상태, 번식주기에 따라 변화하는 한우 빈우의 몸 상태(Body Condition)를 주기적으로 등급화 하여 조사한 값과 동일한 시기에 조사된 한우 번식우의 발육 및 번식형질과의 상관관계를 추정하고, 한우 빈우에서 측정된 몽상태 점수(Body Condition Score, BCS)를 번식 및 사양관리의 수단으로 적용할 수 있는 범위의 기초자료로 제공하고자 실시하였다. 각 형질별 최소자승평균을 추정하였는 바, 분만간격은 406.7일, 임신기간은 287.7일, 분만후 첫종부 일수는 66.2일, 수태당 종부횟수는 1.78회였다. 송아지 생시체중은 23.3kg, 이유시 체중은 70.7kg 이었다. 종부시 BCS와 분만시 BCS의 평균은 4.55, 5.29 이었으며, 수태당 종부횟수를 BCS 점수에 따라 추정한 바, 종부시 BCS가 5 이하에서는 1.50${\sim}$1.74회였으나, 6 이상에서는 2.00${\sim}$3.00회 로 나타났다. 종부시와 분만시 BCS의 분포는 종부시에는 BCS 4${\sim}$5에 서 57%를, 분만시 BCS 5${\sim}$6에 서 46%로 나타내었다. 주요 번식형질의 표현형 상관은 종부시 BCS와 분만시 BCS, 분만간격, 임신기간, 수태당 종부횟수 사이에는 0.16, 0.26, 0.08, 0.06으로 정의 상관을 보였고, 분만시 BCS와 분만간격, 임신기간, 수태당 종부횟수 사이에는 0.10, 0.13, 0.10으로 역시 정의 상관관계를 보였으나, 분만간격과 임신기간, 임신기간과 수태당 종부횟수간에는 -0.11, - 0.13으로 부의 상관을 보였다.

육성비육 거세한우에 대한 황토 급여가 성장 및 도체특성에 미치는 효과 (Effect of Red clay(Hwangto) on Growth Performance and Carcass Characteristics in Growing-Fattening Hanwoo Steers)

  • 육성비육우에 대한 황토급여 효과 구명을 위하여 거세한우 송아지 10두(평균체중 : 144.7kg)를 대상으로 황토급여 유무에 따른 2개 처리(T1 : 황토 무급여, T2 : 황토 자유채식) 와 황토급여 수준에 따른 4개 처리(T3 : 황토 무급여, T4 : 황토 2% 첨가, T5 : 황토 5% 첨가, T6 : 황토 자유채식)를 두어 6개월령부터 24개월령까지 540일간 사양시험을 실시한 결과를 요약하면 다음과 같다. 시험 1에서, 일당증체량은 육성기 및 비육후기에는 황토 무급여구가, 비육전기에는 황토 자유채식구가 우수하였으나 전기간 동안에는 황토급여 유무에 따른 차이가 없었고, 1kg 증체당 소요된 농후사료 및 TDN량도 황토급여 유무에 따른 뚜렷한 차이는 없었다. 도체조사 결과, 도체율, 정육율 및 거래정육율은 황토급여 유무에 따른 뚜렷한 차이가 없었으나 육량지수, 전단력 및 관능검사의 각 항목 등은 황토급여시 개선되는 경향이었다. 그 밖에 경제성분석결과 처리구별 소득은 처리구간에 따른 뚜렷한 차이는 없었다. 시험 2에서, 일당증체량은 육성기에는 황토 무급여구가, 비육전기에는 황토 5%구가 그리고 비육후기에는 황토 자유채식구가 우수하였으나 전기간에는 황토급여 수준에 따른 뚜렷한 차이가 없었으나, 1kg 증체당 소요된 농후사료 및 TDN량 관행사육구에 비해 황토 급여구에서 각각 4.5-8.3% 및 1.2-1.6% 적게 소요되었다. 도체조사 결과 등지방두께, 배최장근단면적 및 육량지수는 황토급여 수준에 따른 뚜렷한 차이가 없었지만, 근내지방도는 황토 5% 및 자유채식구에서 높은 것으로 나타났고, 황토를 자유채식시 관행사육에 비해 소득이 15.5% 증가하는 것으로 나타났다. 이상과 같은 결과를 요약해 볼 때 거세한우 비육우에 대해 가공하지 않은 황토 급여시 육량 및 사료이용성에는 뚜렷한 효과가 없는 반면 육질개선에는 다소 영향을 미치는 것으로 나타났다. 따라서 황토급여시에는 비육후기에 자유채식의 형태로 급여하되, 이용성 증대를 위해 미세입자로 하거나 지장수 형태로 급여하는 것이 바람직 할 것으로 판단된다.

초유를 섭취한 한우 송아지의 출생후 12주 동안의 혈청 면역글로불린과 각종 바이러스 항체가의 변화 (Changes in the serum immunoglobulin levels and viral antibody titers of colostrum-conferred Korean native calves during the first 12 weeks postpartum)

  • The changes in serum total protein and immunoglobulin levels, and BVD, IBR and PI-3 viral neutralizing antibody titers in colostrum-conferred Korean native calves during the first 12 weeks postpartum were studied, and the results obtained were summerized as follows: The Mean concentration of total protein, total immunoglobulin, IgG, IgM and IgA in sera of 9 calves at birth were $3.8{\pm}0.5g/dl$, $0.27{\pm}0.15mg/ml$, $0.06{\pm}0.08mg/ml$, $0.21{\pm}0.11mg/ml$, and extremely low concentration, respectively. Serum total protein level reached a maximum at 20 hours after birth, total immunoglobulin, IgG, and IgM levels at 24 hours, and IgA level at 28 hours, respectively. Serum IgA level reached a minimum at 4 weeks old, IgM level at 5 weeks, total immunoglobulin level at 8 weeks, and IgG level at 10 weeks, respectively. After then those levels had begun to increase, but total protein level was still decreasing at 12 weeks old. The half-lives of IgG, IgM, and IgA were 21.1 days, 4.0 days, and 2.6 days-respectively. In 10 Korean native cows immediately after parturition, serum neutralizing antibody titers specific to BVD, IBR and PI-3 virus were $8.7{\pm}1.5{\log}_2$, $5.7{\pm}1.2{\log}_2$, and $6.8{\pm}1.01{\log}_2$, respectively. And colostral neutralizing antibody titers against BVD, IBR, and PI-3 virus were $10.1{\pm}1.4{\log}_2$, $6.8{\pm}1.3{\log}_2$ and $7.8{\pm}1.7{\log}_2$, respectively. Before suckling the colostrum, SN antibody titers against BVD, IBR, and PI-3 virus were undetectable from all of 9 Korean native calves. Nevertheless SN antibody titer against BVD virus reached a maximum level ($9.2{\pm}0.6{\log}_2$) at 24 hours after birth, that against IBR virus ($6.1{\pm}1.0{\log}_2$) at 20 hours after birth, and that against PI-3 virus ($6.8{\pm}0.9{\log}_2$) at 32 hours after birth, respectively. In 12 weeks old calves, the SN antibodies against BVD and IBR virus were still decreasing, but that against PI-3 virus reached a minimum at 10 weeks, and increased after 12 weeks of age. The half-lives of SN antibodies against BVD, PI-3 and IBR, virus were 16.0 days, 22.6 days, and 25.5 days, respectively.
