The Studies on In Vivo Embryo Production using Sex-Sorted Sperm in Korean Brindle Cattle (성 감별 정자를 이용한 칡소 체내수정란 생산에 관한 연구)

  • The objective of this study was to investigate the result of in vivo embryo collection and pregnancy rate after embryo transfer using sex-sorted sperm of Korean brindle cattle. Donor Korean brindle cattle superovulation treated by decreasing dose of FSH injection. Embryos were recovered on 7 days after the third artificial insemination. Control group semen straw used artificial insemination contained 20 million sperm. Sex-sorted semen straws contained 4 million sperm or 10 million sperm. As for the result of the recovery of the in vivo embryos derived from sex-sorted sperm, the number of transferable embryos was significantly highly recovered to be $6.20{\pm}2.28/donor$ from the control group and was significantly lowly recovered to be $1.57{\pm}1.72/donor$ from the group treated at a sperm concentration of $10{\times}10^6$ (p<0.05). The number of unfertilized embryo was $0.8{\pm}1.30/donor$ in control group which was significantly lower than the group treated at a sperm concentration of $4{\times}10^6$ (p<0.05). However, there was no significant difference in the number of undeveloped ova between control and treatment groups. Pregnancy rate after embryo transfer was shown to be 35.00% in control group and 12.50% in treatment group. The karyotype analysis of the calf derived from sex-sorted sperm resulted in a similar chromosomal distribution pattern (2n=60, XX) compared to those of common Korean native cattle.

Effects of Pregnancy Stage on Eating Behaviors of Hanwoo Heifers (Bos taurus coreanae) (한우 미경산우 임신 단계가 채식행동에 미치는 영향)

  • The objective of this study was to determine the effect of different pregnancy stages on the eating and ruminating behavior of Hanwoo heifers (Bos taurus coreanae). A total of twelve Hanwoo heifers were divided into four groups depending on pregnancy stage: C: before pregnancy, 9 months old, T1: 3 months pregnant, 16 months old, T2: 6 months pregnant, 19 months old, T3: 9 months pregnant, 22 months old (3 replicates per group). Concentrate of 1% of the body weight was fed to them. Roughage was provided at ad libitum to all treatments. Our results revealed that the intake of roughage was the highest in the T3 group (at 3.68 kg) and the lowest in the C group (at 1.02 kg, p<0.05). Total intake in the order from high to low was: T3 > T2 > T1 > C (p<0.05). The rate of dry matter intake compared to body weight (DMI/BW, %) from high to low order was: T3 > T2 > T1 > C (p<0.05). Eating time, ruminating time, and chewing time were the highest in T3 group (p<0.05). Resting time was the highest in C group (p<0.05). The number of bolus and the number of total chews were significantly higher in T3 compared to those in other groups. However, ruminating time per bolus was not significantly difference among groups. The number of chews per bolus from high to low order was: C > T1 > T3 > T2 (p<0.05). The number of bolus per minute was not significantly different among groups. The number of drinking and the number of defecating were significantly higher in C and T1, respectively (p<0.05). However, the number of urinating had no effect according to pregnancy stage. Eating rate and chewing efficiency were significantly higher (p<0.05) in accordance with pregnancy stage (T3 > T2 > T1 > C). Ruminating efficiency was significantly (p<0.05) higher T1 than that in other groups.

  • Estimate breeding value can be used as single trait animal model was developed directly using the Fortran language program. The program is based on data computed by using the indirect method repeatedly. The program develops a common algorithm and imprves efficiency. Algorithm efficiency was compared between the two programs. Estimated using the solution is easy to farm and brand the service, pedigree data base was associated with the development of an improved system. The existing program that uses the single trait animal model and the comparative analysis of efficiency is weak because the estimation of the solution and the conventional algorithm programmed through regular formulation involve many repetition; therefore, the newly developed algorithm was conducted to improve speed by reducing the repetition. Single trait animal model was used to analyze Gauss-Seidel iteration method, and the aforesaid two algorithms were compared thorough the mixed model equation which is used the most commonly in estimating the current breeding value by applying the procedures such as the preparation of information necessary for modelling, removal of duplicative data, verifying the parent information of based population in the pedigree data, and assigning sequential numbers, etc. The existing conventional algorithm is the method for reading and recording the data by utilizing the successive repetitive sentences, while new algorithm is the method for directly generating the left hand side for estimation based on effect. Two programs were developed to ensure the accurate evaluation. BLUPF90 and MTDFREML were compared using the estimated solution. In relation to the pearson and spearman correlation, the estimated breeding value correlation coefficients were highest among all traits over 99.5%. Depending on the breeding value of the high correlation in Model I and Model II, accurate evaluation can be found. The number of iteration to convergence was 2,568 in Model I and 1,038 in Model II. The speed of solving was 256.008 seconds in Model I and 235.729 seconds in Model II. Model II had a speed of approximately 10% more than Model I. Therefore, it is considered to be much more effective to analyze large data through the improved algorithm than the existing method. If the corresponding program is systemized and utilized for the consulting of farm and industrial services, it would make contribution to the early selection of individual, shorten the generation, and cultivation of superior groups, and help develop the Hanwoo industry further through the improvement of breeding value based enhancement, ultimately paving the way for the country to evolve into an advanced livestock country.

Effects of Increasing Inclusion Levels of Rumen Cellulolytic Bacteria Culture on In vivo Ruminal Fermentation Patterns in Hanwoo Heifers (반추위 섬유소분해 박테리아 배양액의 투여 수준에 따른 한우 반추위 발효에 미치는 영향)

  • This experiment was conducted to observe the effects of anaerobic cellulolytic bacteria culture (Ruminococcus flavefaciens H-20 and Fibrobactor succinogenes H-23) on in vivo ruminal fermentation characteristics in Hanwoo heifers. Four ruminally cannulated Hanwoo heifers ($221\pm7.5kg$) receiving a basal diet containing 3 kg of mixture hay (tall fescue and ochardgrass) and 2 kg of concentrate per day were in a $4\times4$ Latin square with 21-day periods. Treatments were the basal diet without the culture additive (control), the basal diet plus 50 ml/day of bacteria culture of H-20 and H-23 (1%), 150 ml/day of H-20 and H-23 (3%), and 250 ml/day of H-20 and H-23 (5%). In the whole experimental periods, ruminal pH did not differ between treatments. However, the concentration of ruminal ammonia-N was increased in the 3% treatment relative to control and the 1% treatment at 1 hr post-feeding (p<0.05). Avicelase and CMCase (carboxymethyl cellulase) activities in rumen fluid showed no significant difference among treatments. However, xylanase activity was higher in the 5% (119.49, xylose ${\mu}mol$/ml/min) than the 3% treatment (71.02, xylose ${\mu}mol$/ml/min) at 0 hr post-feeding (p<0.05). Concentrations of ruminal total VFA, acetate, propionate and valerate were unaffected by treatments, while butyrate was higher in the 3% treatment (24.48 mM) than control (15.71 mM) at 1 hr post-feeding (p<0.05). Results indicate that minimum 3% inclusion of cellulolytic bacteria cultures improved ruminal fermentation, especially ammonia-N concentration and butyric acid production.

Effects of Brewery Meal-Based Fermented Feedstuff Supplemented with Aspergillus Oryzae or Saccharomyces Cerevisiae on Ruminal Microorganism (Aspergilus Oyzae 혹은 Saccharomyces Cerevisiae를 첨가하여 제조한 맥주박 위주 발효사료가 반추위 미생물에 미치는 영향)

  • This study was conducted to examine effects of brewery meal-based fermented feedstuff supplemented with Aspergillus oryzae(AO) or Saccharomyces cerevisiae(SC) on luminal micro-organism of Korean native cattle. Two cows equipped with luminal cannulas were used as experimental animals. Experiment was done with three treatment groups: $71.5\%$ of commercial feed and $28.5\%$ of com silage(control): $45.0\%$ of commercial feed, $26.5\%$ of fermented feedstuff supplemented with AO and $28.5\%$ of corn silage(TAO): $45.0\%$ of commercial feed, $26.5\%$ of fermented ffedstuff supplemented with SC and $28.5\%$ of corn silage(TSC). The number of total viable bacteria (p<0.05), anaerobic fungi and protozoa(p<0.05) was higher in TAO and TSC than in control. The number of proteolytic bacteria(p<0.05), cellulolytic bacteria and xylan fermenters tended to be higher in TAO and TSC than in control. The dry matter recovery (DMR) of protozoa was higher in TAO and TSC than in control(p<0.05). The crude protein (CP) content of total microbes and protozoa was higher in TSC than in control and TAO (p<0.05). The CP content of bacteria was higher in TAO and TSC than in control(p<0.05). The ether extract(EE) content of the total microbes was higher in TAO than in control and TSC(p<0.05), and the EE of protozoa and bacteria were higher in TSC than in control and TAO(p<0.05). The ratio of essential amino acids of total microbe was higher in control than in TAO and TSC(p<0.05). The ratio of methionine and alanine of bacteria was higher in TAO and TSC than in control(p<0.05). The results suggested that the feeding of fermented feedstuff supplemented with AO or SC had an influence on the numbers of ruminal microorganism and the changes of microbial body composition.

A Study on the Changes of Taste Components in brisket and shank Gom-Kuk by Cooking Conditions (조리조건에 따른 양지머리와 사골곰국의 맛성분 변화에 대한 연구)

  • The purpose of this study was to investigate the changes of taste components in the boiled beef brisket soup stock and shank soup stock by varying pretreatment, boiling temperature and time. Free amino acids and nucleotides color and sensory evaluation in each samples were analyzed. The results were obtained as follows : 1. The amount of free amino acids in the brisket soup stock pretreated by soaking and blanching showed a tendency to increase in proportion to boiling time. The amount of glutamic acid in the brisket soup stock was much in order of soaking > blanching > roasting pretreatment. While the amount of glutamic acid in the boiled soup stock samples pretreated by soaking and blanching was much more at low temperature than at high temperature, the glutamic acid contents in the boiled soup stock pretreated by roasting were large at high temperature. The amount of glutamic acid in pretreated by soaked soup stock showed the highest and recorded 8.73 mg% at 6 hour-low temperature-boiling. 2. The amount of free amino acids in the shank soup stock did not show any regular tendency and had few changes in quantity by the methods of pretreatment. Each amount of glutamic acid in the shank soup stock pretreated by soaking and blanching was the highest, when boiled for 3 hours at high temperature. The samples pretreated by roasting showed the highest record 2.49 mg%, when boiled for 6 hours at high temperature, but could not recognize any regular tendency in the case of boiling at low temperature. 3. The amount of nucleotides in the brisket soup stock generally showed increase in proportion to boiling time. The amount of 5'-IMP extracted from the brisket soup stock was much in order of blanching > soaking > roaking pretreatment, but few differences between blanching and soaking soup stock samples. The amount of 5'-IMP extracted from soup stock samples pretreated by soaking and blanching was high at low-boiling and by roasting at high-boiling. Each amount of 5'-IMP extracted from soup stocks pretreated by soaking(BSL) and blanching(BBL) was the highest at 6 hour-low-boiling(37.06 mg%), and 5 hours(38.37 mg%) respectively. The amount of 5'in the soup stock pretreated by roasting(BRH) showed the highest records at 6 hour-high-boiling(10.85 mg%). 4. The amount of 5'-IMP extracted from the shank soup stock preteated by soaking and blanching showed a tendency to decrease after 3 hours boiling irrelative of boiling temperature. The amount of 5'in the shank soup stock was much in order of soaking > blanching > roasting pretreatment and showed high at the boiling of high temperature. In the sample pretreated by roasting it showed the highst records when boiled for 6 hours at high temperature(1.55 mg%). 5. The L Value of the brisket soup stock pretreared by roasting at high temperature(BRH) was the lowest and the b value of it was the highest of all the brisket samples boiled for 6 hours. No differences were found in the Value of L, a, and b in shank soup stock by the methods of pretreatment and boiling temperature. 6. The sensory scores in color and flavor of the brisket soup stock showd that BRH was higher than the other samples, and the preference in taste and overall was the highest in BSH while it was the lowest in BRH. The preference in the all sensory characteristics of SSH was higher than any other shank soup stock, but did not show any significant difference statistically.

Effects of Terpenoids-Rich Plant Extracts on Ruminal-fermentation and Methane Production (Terpenoid 함유 식물 추출물의 첨가가 반추위 발효와 메탄 발생에 미치는 영향)

  • This study was conducted to investigate effects of terpenoids-rich plant extracts (TRPE) on the in vitro ruminal fermentation characteristics and methane production. The ruminal fluid was collected from a cannulated Hanwoo cow fed concentrate and timothy in the ratio of 6 to 4. The TRPE as Mint (Mentha arvensis var. piperascens), Pine (Pinus densiflora), Japan cedar (Cryptomeria japonica), Sichuan pepper (Zanthoxylum piperitum), Hinoki cypress (Chamaecyparis obtuse) and Japanese black pine (Pinus thunbergii) were used in this study. The 15 mL of mixture, contains McDougall buffer and rumen fluid in the ratio of 2 to 1. The mixture was dispensed anaerobically 50 mL serum bottles and it is contained 0.3 g timothy substrate and 5% TRPE. The bottles were incubated at $39^{\circ}C$ for 3, 6, 9, 12, 24, 48 and 72 hours. The pH value decrease by increased incubation times and the pH values at all times were significantly (p<0.05) higher in treatments than in control. The digestibility of dry matter at 3 hours was significantly (p<0.05) higher in mint treatment than in control. Productions of total gas and carbon dioxide at before 12 hours was significantly lower (p<0.05) in treatments than in control. The methane production at 24 hours was significantly (p<0.05) lower in treatments than in control. The concentrations of acetic acid and propionic acid at 24 hours were significantly higher (p<0.05) in mint and pine treatments than in control. In conclusion, the terpenoid-rich plant extracts were shown to decreased methane emission and without adversely affected ruminal fermentation. Therefore, the terpenoid-rich plant extracts as mint and pine were shown to decreased methane production and it has potential possibility for ruminal fermentations.

Effects of Applying Cattle Manure on Carrying Capacity of Organic Livestock per Unit Area of Summer Forage Crops (우분뇨 시용이 하계사료작물의 단위면적당 유기가축 사육능력에 미치는 영향)

  • This study was carried out to select a proper forage crop, and to estimate the proper level of application of cattle manure and carrying capacity of organic livestock per unit area. Corns and forage sorghum hybrids were cultivated with different types of livestock manures and different amount of them to produce organic forage. For both corns and forage sorghum hybrids, no fertilizer plots had significantly (p<0.05) lower annual dry matter (DM), crude protein (CP) and total digestible nutrients (TDN) yields than those of other plots, whereas the N-P-K (nitrogen-phosphorous-kalium) plots ranked the highest yields, followed by 150% cattle manure plots and 100% cattle manure plots. DM, CP and TDN yields of in cattle manure plots were significantly (p<0.05) higher than those of no fertilizer and P-K (phosphorous-kalium) plots. The yields of in cattle slurry plots tended to be a little higher than those of in composted cattle manure plots. Assuming that corn and forage sorghum hybrids produced from this trial were fed at 70% level to 450kg of Hanwoo heifer for 400g of average daily gain, the carrying capacity (head/year/ha) of livestock ranked the highest in 150% cattle slurry plots (mean 6.0 heads), followed by 100% cattle slurry plots (mean 5.3 heads), 150% composted cattle manure plots (mean 4.7 heads), 100% composted cattle manure plots (mean 4.4 heads), and no fertilizer plots (mean 2.8 heads) in corns (or the cultivation of corns). Meanwhile, in the case of forage sorghum hybrids, 150% cattle slurry plots (mean 6.4 heads) ranked the highest carrying capacity, followed by 150% composted cattle manure plots (mean 4.8 heads), 100% cattle slurry plots (mean 4.4 heads), 100% composted cattle manure plots (mean 4.1 heads), and no fertilizer plots (mean 2.8 heads). The results indicated that the application of livestock manure to cultivated soil could enhance not only DM and TDN yields, but also the carrying capacity of organic livestock as compared with the effect of chemical fertilizers. In conclusion, the production of organic forage with reutilized livestock manure will facilitate the reduction of environmental pollution and the production of environmentally friendly agricultural products by resource circulating system.

Effects of Nitrate-rich Plant Extracts on the in vitro Ruminal Fermentation and Methane Production (질산염 화합물 함유 식물 추출물이 in vitro 반추위 발효성상과 메탄 발생에 미치는 영향)

  • This study was conducted to evaluate the effects of nitrate-rich plants extracts on the in vitro rumen fermentation characteristics and rumen methane production. The extracts of nitrate-rich plants, as potato, carrot, chinese cabbage, lettuce and spinach were used in this study. The ruminal fluid was collected from a cannulated Hanwoo cow fed concentrate and timothy in the ratio of 6 to 4. The 20mL of mixture, comparing McDougall's buffer and rumen fluid in the ratio 2 to 1, was dispensed anaerobically 50mL serum bottles containing 0.3g of timothy substrate and extracts of nitrogen-rich plants. The serum bottles were incubated 39℃ for 9, 12, 24, 48 hours. The pH value was decreased by increased incubation times and normal range to 6.31 to 6.96. The dry matter digestibility was significantly(p<0.05) lower in chinese cabbage than in control at 9h incubation time. Ammonia concentration was significantly(p<0.05) lower in potato, chinese cabbage, lettuce than in control and the rumen microbial growth rate was significantly(p<0.05) higher in carrot than in control at 24h incubation time. The concentrations of acetate and propionate was significantly(p<0.05) lower in treatment than in control. The concentration of butyrate was showed a different pattern depending on treatments. Total gas emissions was significantly(p<0.05) lower in chinese cabbage, lettuce, spinach than in control at 12h, 24h incubation time. Methane production was significantly(p<0.05) lower in potato, chinese cabbage, spinach than in control, carbon dioxide production was significantly(p<0.05) lower in treatment than in control. In conclusion, supplementation of the nitrate-rich plant extracts in ruminal fermentation in vitro resulted in decreasing the methane production without adversely affecting the fermentation characteristics. Particularly the chinese cabbage extract was regard as a potential candidate for reducing the methane emission in ruminants.

Effects of Applying Livestock Manure on Productivity and Organic Stock Carrying Capacity of Summer Forage Crops (가축분뇨시용이 하계사료작물의 생산성 및 유기가축 사육능력에 미치는 영향)

Meanwhile, in case of sorghum $\times$ sorghum hybrids, N+P+K plots (mean 5.7 heads) ranked the highest carrying capacity, followed by $100{\sim}150%$ cattle slurry plots (mean $4.8{\sim}5.2$ heads), 150% composted cattle manure plots (mean 4.7 heads), 100 % composted cattle manure plots (mean 4.3 heads), P+K plots (mean 3.8 heads), and no fertilizer plots (mean 3.4 heads). The results indicated that replacing chemical fertilizer by livestock manure application to cultivation soil for forage crops could enhance not only DM and TDN yields, but also organic stock carrying capacity. In conclusion, it was conceived that organic forage production by reutilizing livestock manure might contribute to reduced environmental pollution and the production of environment friendly agricultural products through resources recycling.
