  • 본 연구는 피토케미칼과 펩타이드의 융합된 신소재의 항노화 효능에 관한 연구이다. 대표적인 피토케미칼인 갈릭산 및 시키믹산에 식물유래 Pentapeptide인 Exorphin을 융합시켜 만든 Galloyl Exorphin 및 Shikimoyl Exorphin은 콜라겐 합성능 및 항염증 효능 등을 보여주었다. 이러한 피토케미칼 융합 펩타이드 신소재는 피토케미칼이 가진 Stability 및 Solubility를 향상시키며, 피토케미칼 및 펩타이드 소재가 가진 각각의 효능을 유지시켰다. 이러한 결과는 피토케미칼 융합 펩타이드 소재가 항노화 소재로서 화장품 포물레이션에 널리 활용될 수 있음을 의미한다.

Effect as antimicobial agents of pufied antipeptide from acid-extracts, Halocynthia roretzi (멍게 추출물에서 정제한 항균 펩타이드의 항균제재로서의 효과)

  • 참멍게의 체액세포로부터 산추출 후, 조 추출물에서 천연항균소재를 개발하기 위해 먼저 멍게 조 추출액을 직접 Tricine-SDS PAGE를 통하여 주요 펩타이드들의 분자량의 범위를 살펴본 결과 6kDa 이하의 분자량의 펩타이드들이 다량 존재함을 알수 있었다. 펩타이드들의 size별 항균활성을 알아보기 위해 여러 사이즈의(100, 50, 30, 10 kDa)의 한외여과만으로 여과하여 그 여과액들의 specific 활성을 알아본 결과 여과막의 cut-off size에 상관없이 거의 인정한 specific activity를 가짐을 알 수 있었다. 멍게 조 추출액의 여러 미생물에 대한 항균 스펙트럼을 알아보기 위해 E.coli, P. aeruginosa, S. typhi, V. parahaemolyticus, L. monocytogenes, B. sutillus, S. aureus, S. mutans 균주들을 $10^5 CFU/ml$로 부터 4log 감소시키는 농도를 측정한 결과 각각 200, 50, 60, 10, 25, 30, 100, 100ppm 농도였으며, 대표적 상용화 항균 펩타이드인 Nisin과의 항균활성 비교 결과 비슷하거나 월등히 뛰어난 결과를 보여주었다. 또한 추출액의 열안정성을 측정하기 위해 $100^{\circ}C$에서 10분간 가열한 후 원액과의 항균력의 차이를 Radial diffusion assay로 알아본 결과 항균력의 차이가 거의 없음을 알 수 있었다.

Hypolipidemic Effects of Peptide Fractions of Casein on Serum Lipids in Rats Fed Normal or High Fat Diet (정상 또는 고지방식을 섭취한 흰쥐에서 Casein 펩타이드 분획물이 혈청 지질농도에 미치는 영향)

  • In the recent studies, many researchers are interested in foods as functional components rather than nutrient sources. Cow's milk is considered as an excellent food sources because of its many nutrients. Casein is a major milk protein and has been reported to have hyperlipidemic and hypercholesterolemic effects. But several reporters have suggested that peptide fractions and hydrolysate of casein have hypolipidemic effects differing from intact protein, casein. Therefore, the objective of the study was to investigate how the casein peptide fractions affect lipid metabolism in rats fed normal or high fat diets. The peptide fractions and hydrolysate of casein were obtained by casein hydrolysis with trypsin. The male rats (Sprague-Dawley), weighing approximately 150 g, were fed each experimental diet containing casein (CAS), casein hydrolysate (CH), casein hydrolysate precipitate (Cpt) and two kinds of peptide fractions (CL & CB) for three weeks, respectively. In the exit I, the male rats were fed normal fat diets (7% soybean oil & cholesterol-free; Expt. I), and in the expt II, fed high fat diets (18% beef tallow & 1% cholesterol; Expt. II). Crude protein contents were calculated from nitrogen contents. Amino acid composition of each fraction was also analyzed. The concentration of total lipid, total cholesterol and triglyceride in serum, liver and feces were measured. As the results of study, tole rats fed peptide fractions with normal fat diets (Expt. I) had no effects on total lipid, total cholesterol and triglyceride concentration in serum and liver and fecal excretion. However, in the rats fed hydrophobic casein peptide fractions (CB) with high fat diet, fecal lipids excretion were significantly increased and the lipids concentration of serum and those of liver tended to decrease, numerically.

Intermacromolecular Complex Formation between Helix Strilctilral Polypeptides through Hydrogen Bonding (수소 결합을 통한 Helix 폴리 펩타이드사이의 복합체 형성)

  • Polypeptide has been used broadly as an active ingredient in cosmetics We thought it is very important to investigate the adsorption behavior of polypeptide in order to pre-estimate the effect of these polypeptides. For the study of polypeptide adsorption, we have investigated complex formation of basic homopolypeptides, poly(L-proline) Form I [PLP(I)], Form II [PLP(II)] and poly(4-hydroxy-L-proline) (PHLP) with acidic homopolypeptides, poly(L-glutamic acid) (PLGA), poly(D-glutamic acid) (PDGA) and poly(L-aspartic acid) (PLAA) through hydrogen bonding in a hydroalcoholic medium with viscometer, 1ight scatter, pH meter and circular dicroism (CD). The polypeptides used in this study have helical structure in some conditions. The result exhibited that al 1 the complexes were formed as the composition of basic/acidic homopolypeptide : L:2 irrespective of the complex systems used. A more favorable complex is formed in the PLP(II)-PLGA system than PHLP-PLGA because PLP(II) has a more flexible helical conformation, whereas PHLP has a more rigid helical conformation. The right-handed helix PLGA formed the complex favorably and quickly with the left-handed helix PLP(II), whereas the left-handed helix PDGA formed the complex favorably with the right-handed helix PLP(I). The effect of side chain of the acidic homopolypeptides on the complexation was also studied. The result showed that more favorable condition for the complexation was PLGA-PLP(II) system which has longer side chain at acidic homopolypeptide than PLAA - PLP(II). All the above facts were well supported by CD measurement for the complex systems. By the CD spectra for the complexes we could deduce the conformational change of each homopolypeptide in the complexes On the basis of the above results, we performed the adsorption test of PLP(I, II) and PHLP on the hair having a left-handed helix. The adsorption amount of each polypeptide was analyzed by HPLC. The result showed that PLP(I) was adsorbed more than PLP(II), PLP(II) was adsorbed more than PHLP on the hair. On adsorbing polypeptides having a helical structure on the hair through hydrogen bonding, it could be concluded that the helical polypeptides having the opposite directional structure to the hair are adsorbed more than those having the same directional structure with the hair and also the polypeptides having a flexible conformation are adsorbed more than those having a rigid conformation.

  • Microorganism, including some bacteria isolated from soil, were found to secrete an extracellular soymilk-clotting enzyme. Using this bacterial enzyme experiments were carried out to optimize the hydrolyzing conditions for the production of soy peptides. The soy peptides produced by hydrolyzing 11S globulin with enzyme treatment at $65^{\circ}C$, pH 6.1, for 1hr were found to have a accessible possibility. The obtained coagulum by enzymatic reaction was very flocculation with fine structural formation. Properties of peptide Y and W of the enzyme hydrolysates at pH 6.1 were superior to that of isoelectric precipitation because these peptides were miscible with water in all proportions.

Molecular Cloning of a Gene Encoding a Putative Antibacterial Peptide from Bombyx mori (누에에서 새로운 항세균성 펩타이드 유사 유전자의 분리와 염기서열 결정)

  • To isolate a novel gene for antibacterial peptide, an inducible clone(BmInc8) was selected by differential screening strategy from Bombyx mori cDNA library prepared from lavae injected with Escherichia coli. This clone contained a cDNA insert of 564 nucleotides and encoded 59 amino acids with an apparent molecular mass of 6.3 kDa. The cDNA sequence of BmInc8 had 61.2% identity compared to that of the bactericidin from Manduca sexta and also the deduced amino acids sequences from this insert had 65% identity compared to that of the cecropin D peptide Hyalophora cecropia. The transient expression assay of this insert using prokaryotic expression vector system revealed that the expressed peptide displayed the antibacterial activity. The cDNA sequence was deposited in GenBank under the accession number U30289.

Design of Short Indolicidin Analogs with Enhanced Prokaryotic Selectivity (증가된 원핵세포선택성을 가진 짧은 인돌리시딘 유사체의 설계)

  • Indolicidin (ID) is a 13-residue Trp-rich antimicrobial peptide (AMP) isolated from bovine neutrophils. In addition to having a high antimicrobial potency, it is also toxic to mammalian cells. To develop novel ID-derived AMPs with shorter lengths and enhanced prokaryotic selectivities (meaning potent antimicrobial activity against bacterial cells without toxicity against mammalian cells) over the parental ID, several ID analogs were designed and synthesized. Finally, 10-residue ID analogs (SI, SI-PA, SI-WF and SI-WL) with much higher prokaryotic selectivity than the parental ID were developed. Our results suggest that the hydrophobic and aromatic amino acids at the central position of the analog SI with the highest prokaryotic selectivity are important for potent antimicrobial activity, but two Pro residues do not affect antimicrobial activity. The order of prokaryotic selectivity for ID and its designed analogs was SI > SI-PA > SI-WF > SI-WL > ID > SI-WA. Taken together, our designed short ID analogs could be developed as therapeutic agents for treating bacterial infections.

Characterizing and Analysis of the Peptide in Extract from Camellia sinensis (녹차에 함유된 펩타이드 분리 방법 및 추출물의 특성 분석)

  • 본 연구는 알려진 녹차의 다양한 항산화, 항염 효능이 있는 Polyphenol 류 이외의 생물학적 활성을 가지는 펩타이드 및 당단백질 소재에 대한 특성과 분석을 위하여 연구되었다. 녹차에서 수용성 성분을 추출 후 알콜 침전방법을 사용하여 단백질과 펩타이드류를 분리하여 실험에 사용하였다. 전체 추출물 중에는 Glutamic acid가 14073.9ug/g, (39.11%)로 가장 높게 분석되었으며, GPC 분석결과 Mn 886, Mw 25218, Mp 890, Mw/Mn 28.46으로 분석이 되었으며, 전체적으로 3개의 피크로 분석되었다. 함유된 펩타이드를 분석하기 위하여 HPLC를 이용하여 RT 16.148min의 Fraction을 분리하여 펩타이드 분석결과 녹차에서 유래한 TNTLSN이라는 hexapeptide를 동정하였다. 이러한 펩타이드는 친수성이며, 안정도가 높은 특징을 가지고 있고, 분자량이 1000이하로 피부 흡수도가 높을 것으로 기대되며, Biodegradable하며, Renewable 한 자연 친화적인 화장품 소재로서 널리 활용될 수 있을 것으로 판단된다.

Peptide isolated from Hermetia illucens larvae inhibits mice from Klebsiella pneumoniae infection in the kidney (동애등애유충에서 분리된 펩타이드의 신장에서의 폐렴간균 감염 억제 효능)

  • Overuse of antibiotics has significantly contributed to an increase in microbial antibiotic resistance, causing difficulties in the suppression of microbe-borne infectious diseases. In this study, we determined the anti-Klebsiella pneumoniae effect in the kidneys of mice induced by peptides isolated from H. illucens larvae. Mice were intranasally infected with a high dose of K. pneumoniae and 1 day later, peptides were introduced through the intramuscular route. Mice were sacrificed on day 10 upon K. pneumoniae infection to determine the bacterial loads in the kidneys. Mice receiving peptide treatment demonstrated significantly reduced bacterial loads, reduced bodyweight loss, and higher survival in a dose-dependent manner compared to control. These results indicate that peptide isolated from H. illucens larva inhibits K. pneumoniae infection in the kidney. The peptide from H. illucens larva could be a potential candidate for the development of an effective antibacterial drug.
