EFFECT OF THE CHLORHEXIDINE AND FLUORIDE-CONTAINING VARNISH ON THE LEVEL OF SALIVARY MUTANS STREPTOCOCCI (Chlorhexidine과 fluoride 성분의 varnish가 타액 내 mutans streptococci 수에 미치는 효과)

  • The objective of the present prospective trial was to compare the efficacy of chlorhexidine(Cervitec), Fluoride (Fluorprotector) and combination of chlorhexdine and fluoride varnishes in decreasing the level of salivary mutans streptococci. Forty healthy students of school of dentistry, Chonbuk national university were investigated to evaluate mutans streptococci(MS) counts and randomized into four groups to treat with the experimental varnishes; A) untreated group(n=10), B) chlorhexidine group(n=10), C) fluoride group(n=10), D) chlorhexidine and fluoride group(n=10). Dentocult $SM^{(R)}$ (Orion Diagnostica) strip method was used for measurement of the level of mutans streptococci in saliva. Stimulated saliva were collected at baseline for mutans streptococci counts evaluation (ms1), 12 weeks later the completion of each varnish treatment, mutans streptococci counts were re-evaluated. In varnish group with chlorhexidine, fluoride and combination of chlorhexidine and fluoride, the level of mutans streptococci was lower after 12 weeks than at baseline, but there were no significant differences in saliva(p>0.05), when compared with baseline. After 12 weeks, a remarkable reduction was still found in the subjects with high level of mutans streptococci at baseline, but not different in the low and moderate level of mutans streptococci(p>0.05).

Antimutagenic and Cytotoxicity Effects of Extracts of Eleutherococcus senticosus Maxim fruits (가시오갈피 열매 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was earned out to investigate the antimutagenic and cytotoxic effects of Eleutherococcus senticosus Maxim fruits ethanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using Ames test and SRB assay, respectively. They were extracted with ethanol and then fractionated with hexane, chloroform, ethyl acetate, butanol and water to get active fractions. In the Ames test, most of the extracts had strong antimutagenic effects against the mutagenesis induced by MNNG, 4NQO, B(${\alpha}$)P and Trp-P-l. The ethanol extract (200 $\mu\textrm{g}$/plate) of Eleutherococcus senticosus fruits showed 87.2% inhibitory effect on the mutagenesis induced by MNNG against TA100. And also, The suppression ratio against B(${\alpha}$)P and Trp-P-l in the TA100 showed 96.1% and 95.5%, respectively. In the cytotoxic effects against human cancer cell lines (A549, AGS, MCF-7, Hep3B), the value of inhibition were mostly above 60% for each fraction (1 mg/mL). Hexane fraction (1 mg/mL) against showed the strongest cytotoxic effects of 92.7% compared to those of other fraction and butano fraetion against Hep3B was relatively high growth inhibitory effect of 82%.

Effects of roasting conditions on the physicochemical properties of Coffea arabica beans (배전조건이 아라비카 커피의 이화학적 성질에 미치는 영향)

  • Effects of roasting on physical (weight, volume, density and color) and chemical properties (total phenol, caffeine, chlorogenic acid and antioxidant properties) of three Arabica coffee beans (Brazil Bourbon, Indonesia Mandheling and Kenya Tatu) were investigated. A steady weight loss, volume increase, and bean density decrease were observed during the roasting process. The $L^*$, $a^*$, and $b^*$ values tended to decrease as the roasting temperature and time increased. Caffeine level was approximately 0.73% in green beans, and increased to 40-67% for darker roasts. Green beans contained the highest chlorogenic acid content, which decreased as the roasting temperature and time increased. The light- and medium-roasted coffee showed the highest total phenolol contents. The antioxidant activities were measured using DPPH and ABTS. The light-roasted coffee beans showed the highest antioxidant activities, and an approximately 36-51% loss in antioxidant activity was observed after further roasting. The flavor patterns of roasted coffee beans were analyzed using an electronic nose system based on gas chromatography. This analysis demonstrated that an electronic nose system can be applied to identify the green bean variety and the degree of roasting.

Identification of Antioxidative Components from Ethanol Extract of Rhus verniciflua STOKES (옻나무 에탄올 추출물로부터 항산화 활성 물질의 구조동정)

  • The free phenolic acid fraction of the chloroform extract from 75% ethanol extract of Rhus verniciflua STOKES (RCF) showed stronger antioxidative activity than BHT, BHA and ${\delta}-tocopherol$ at the same concentration. RCF components were isolated and identified by silica gel column chromatography, thin layer chromatography, mass spectrometer and $^1H-NMR\;and\;^{13}C-NMR$. The antioxidative activity was confirmed by electron donating activity, Rancimat method and thiobarbituric acid test in liposome system. RCF-11 could be further separated into three fractions. The antioxidative active compounds were purified and identified as gallic acid, butin and butein. The RCF-13 was purified and identified as sulfuretin.

  • Lipids extracted from two garlic samples (Allium sativum Linnaeus) were studied. Total lipid content of the samples ranged from 310 to 342mg/100g of fresh garlic. These lipids were shown to consist of $36.4{\sim}43.5%$ neutral lipids, $20.3{\sim}24.3%$ glycolipids, and $36.2{\sim}39.3%$ phospholipids. Among the neutral lipids, triglycerides were predominant $(80.5{\sim}83.6%)$ with smaller amounts of free sterols, free fatty acids, and sterol esters being present. 1, 2-Diacylglycerol acetates, 1, 3-diacylglycerol acetates and cholesterol acetates were also tentatively identified. Esterified steryl glycosides and steryl glycosides were major sugar-containing lipids, but monogalactosyl diglycerides, cerebrosides, digalactosyl diglycerides and sulfolipids were relatively minor components. Of the phospholipids, phosphatidyl cholines and serines, and phosphatidyl ethanolamines were the major components, comprising over 85% of this class. Phosphatidic acid and phosphatidyl inositols were also present. The major fatty acids in the total and three lipid classes were linoleic, palmitic, oleic and linolenic acid.

Deposition Characteristics of Lead Titanate Films on $RuO_2$ and Pt Substrates Fabricated by Chemical Vapor Deposition ($RuO_2$ 및 Pt 기판에서 $PbTiO_3$박막의 화학기상 증착특성에 관한 연구)

  • $PbTiO_3$ films were fabricated by electron cyclotron resonance plasma enhanced chemical vapor deposition(ECR-PECVD). Deposition characteristics of $PbTiO_3$films on $RuO_2$ and Pt substrates were investigated with varying the flow rate of metalorganic source and substrate temperature. The residence time of Pb-oxide molecules in much longer on $RuO_2$ than on Pt substrate, while the perovskite nucleation is more difficult on $RuO_2$ than on Pt substrate. Therefore, the process conditions to obtain the single perovskite $PbTiO_3$ phase are more restricted on $RuO_2$ than on Pt substrates. An introduction of Ti-oxide seed layer increases perovskite nucleation density and thus enlarges the process window to obtain the single perovkite phase. The introduction of Ti-oxide seed layer make the PZT film that Ti-components of $PbTiO_3$ are partially substituted with Zr atoms have single perovskite phase for the wide range of Zr/(Zr+Ti) concentration ratios.

Stress Concentration Ratio of GCP Depending on the Mixing Ratio of Crushed Stone and Sand (GCP의 쇄석과 모래의 배합비 별 응력분담비)

  • Gravel compaction pile (GCP) is widely used as it increases the bearing capacity of soft ground and reduces the consolidation settlement. Stress concentration ratio for GCP design is dependent on the area replacement, surcharge pressure and depth. However, a range of stress concentration ratio obtained through field, laboratory experiments and numerical analysis is large. Little study has been done on the stress concentration ratio for the mixing ratio of gravel and sand. The main objective of the study is to evaluate the stress concentration ratio for both area replacement ratio and mixing ratio through literature review and numerical analysis. Numerical analysis using the finite element program ABAQUS 6.12-4 has been performed for the composite ground with GCP. The excess pore water pressure and stress concentration ratio of composite ground have been analyzed for both the area replacement ratio and the mixing ratio. Based on the previous research results, a range of stress concentration ratio obtained from the field tests, laboratory tests, numerical analysis on the GCP studies is found to be 1.7-3.2, 2.0-7.5 and 2.0-6.5, respectively. Based on the numerical analysis results, as the area replacement ratio increases, the stress concentration ratio increases up to 30% and then decreases at 40%. Also, the stress concentration ratio tends to increase up to 70:30 and then to decrease after 60:40.

Bioequivalence of Acer Capsule to Airtal Tablet (Aceclofenac 100 mg) (에어탈 정(아세클로페낙 100 mg)에 대한 에이서 캅셀의 생물학적 동등성)

  • Aceclofenac, 2-[(2',6'-dichlorphenyl)amino]phenylacetoxiacetic acid, is a new nonsteroidal anti-inflammatory drug that belongs to the family of phenylacetic acids. It shows good tolerance and potent analgesic/antiinflammatory properties, and acts on cartilaginous chondriocytes, stimulating their repair mechanism. The purpose of the present study was to evaluate the bioequivalence of two aceclofenac products, $Airtal^{TM}$ tablet (Daewoong Pharmaceutical Co.) and $Acer^{TM}$ capsule (Kyungdong Pharmaceutical Co.), according to the guideliner of Korea Food and Drug Administration (KFDA). The aceclofenac release from the two aceclofenac products in vitro was tested using KP VII Apparatus II method at pH 7.8 dissolution media. Sixteen normal male volunteers, $23.13\pm2.03$ years in age and $66.33\pm7.08$ kg in body weight, were divided into two groups and a randomized $2\times2$ cross-over study was employed. After one tablet or capsule containing 100 mg of aceclofenac was orally administered, blood was taken at predetermined time intervals and the concentrations of aceclofenac in serum were determined using HPLC with UV detector. The dissolution profiles of the two aceclofenac products were very similar at pH 7.8 dissolution media. The pharmacokinetic parameters such as $AUC_t,\;C_{max}\;and\;T_max$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters. The results showed that the differences in $AUC_t,\;C_{max}\;and\;T_{max}$ between two products were $6.50\%,\;-1.06\%\;and\;11.96\%$ respectively, when calculated against the $Airtal^{TM}$ tablet. The powers $(1-\beta)\;for\;AUC_t,\;C_{max}\;were\;89.82\%\;and\;82.84\%$, respectively. Minimum detectable differences $(\Delta)\;at\;\alpha=0.05\;and\;1-\beta=0.8$ were less than $20\%\;(e.g.,\;17.51\%\;and\;19.30\%\;for\;AUC_t,\;C_{max}$, ). The $90\%$ confidence intervals were within $\pm20\%\;(e.g.,\;-3.73\%\sim16.73\%\;and\;-12.34\%\sim10.22\%\;for\;AUC_t,\;C_{max},\;respectively)$. Two parameters met the criteria of KFDA for bioequivalence, indicating that $Acer^{TM}$ capsule is bioequivalent to $Airtal^{TM}$ tablet.

Antimicrobial Effects of Ethanol-Extracted and Sub-Fractionated Materials from Different Parts of Quercus aliena Blume (갈참나무 부위별 에탄을 추출물 및 분획물의 항균효과)

  • This study was to determine the inhibitory effect of ethanol extract of leaf, bark and xylem of Quercus aliena Blume and, consecutively, of organic solvent subfractions against food borne pathogens. The ethanol extracts of leaf, bark and xylem of Quercus aliena Blume were shown to have antimicrobial effect against Gram-positive and Gram-negative bacteria. Listeria monocytogenes and Salmonella Typhimurium were more resistant to ethanol extracts of the Quercus aliena Blume tissues compared with Bacillus cereus and Escherichia coli O157:H7. The ethanol extract of Quercus aliena Blume leaf showed the strongest antimicrobial activity against food borne pathogens, followed by those of xylem and bark. The ethanol extract treatment $(500\~1,000\mu g/mL)$ of Quercus aliena Blume leaf completely reduced the growth of B. cereus and L. monocytogenes within 24 hour whereas $2,000\mu g/mL$ of ethanol extract was needed for complete growth inhibition of E. coli O157:H7 and S. Typhimurium. Among organic solvent subfractions obtained from the ethanol extract of leaf, bark and xylem of Quercus aliena Blume, the ethyl acetate fraction showed the strongest antimicrobial activity, but no antimicrobial activity was observed in chloroform, hexane and water fractions.

Monitoring of Veterinary Drug Residues in Foods Produced in Korea (국내 유통 식품의 잔류동물용의약품 모니터링)

  • This study was conducted to monitor residues of 10 veterinary drugs in food products. Various veterinary drugs were examined including enrofloxacin, ciprofloxacin, norfloxacin, oxolinic acid, amoxicillin, ampicillin, oxytetracycline, tetracycline, chlortetracycline, and doxycycline in beef, pork, egg, chicken, eel, flatfish, and rockfish obtained from 6 different regions (Seoul, Incheon, Daejeon, Gwangju, Daegu, Busan). Residues were detected in 21 (6.5%) samples out of 321 samples. In particular, 2 (1.0%) livestock samples had detected residues among 203 products, and 19 (16.1%) aquaculture samples had residues detected among 118 products. The most frequently detected drug residues in aquaculture products were oxytetracycline and amoxicillin, but the levels were mostly below the MRL (Maximum Residue Limit). In only one flatfish sample, amoxicillin was found at a level higher than the MRL (0.05 mg/kg). In livestock products, residues of most veterinary drugs were not detected. But enrofloxacin was detected in 2 chicken (Korean name: Ogolgae) samples at a higher level than the MRL (0.1 mg/kg as the sum of ciprofloxacin).
