Monitoring of Carbamate Pesticide Residues in Agricultural Products Supplied for the Army (군납 농산물 중 카바메이트계 농약의 모니터링)

  • This study was monitored for 14 pesticide residues in agricultural products for the army, such as fruit vegetables(pepper, cucumber, pumpkin, melon and water melon), leafy vegetables(Korean cabbage, spinach, lettuce, crown daisy, cabbage, green onion), mushrooms(agaric, p'yogo), and bean sprouts produced in Kyunggi-do and Inchon-City. From January to December 2005, ten carbamate pesticides in 356 samples were analyzed by HPLC. One kind of pesticide was detected in 8 samples of detection rate (2.53%), and two pesticides were in one sample of detection rate(0.27%). Aldicarb, bendiocarb, fenobucarb, methiocarb, isoprocarb, and propoxur were not found in all samples. Detection rates of pesticides were 0.84% for methomyl, 0.56% for carbofuran, 0.56% for ethiofencarb, and 0.28% for carbayl. Dectection ranges of pesticides were from 0.01 to 2.9 mg/kg for ethiofencarb, from 0.1 to 0.23 mg/kg for methomyl, from 0.20 to 0.24 mg/kg for carbofuran, and 0.01 mg/kg for carbaryl, respectively. Consequently, detection levels of all pesticides in samples were less than the maximum residue limits(MRLs) in Korea representing that all agricultural products for the army were safe.

Development of an Inverted Y-Shaped Strip for the Detection of Organophosphorus and Carbamate Residual Pesticides (유기인계 및 카바메이트계 농약 신속 검출을 위한 역 Y자 스트립의 개발)

  • The inverted Y-shaped strip detection method based on acetylcholinesterase (AChE) was developed for the rapid detection of organophosphorus and carbamate pesticides. The inactivation of AChE by organophosphorus and carbamate pesticides has been well known. The AChE catalyzes acetylthiocholine into thiocholine having (-) and (+) charges, and the (+) charge results in aggregation of gold nanoparticle (GNP). Malaoxon and carbofuran were used as standard organophosphorus and carbamate for the development of the inverted Y-shaped strip, respectively. In order to optimize the method, various angles of the Y-shaped strip, different types of nitrocellulose membrane, and concentration of AChE were tested as key parameters. The detection limit of the method was 10 ng/mL for both malaoxon and carbofuran pesticides. No cross-reaction was observed to other pesticides such as atrazine, cyanazine, simazine, bifenthrin, boscalid, metalaxyl, and chlorobenzilate. Recoveries from lettuce spiked when known concentrations of malaoxon and carbofuran were found ranging from 96.4 to 100.7% and 81 to 112.7%, respectively. This study suggests that the inverted Y-shaped strip method based on AChE may be a useful tool for the sensitive, specific, rapid detection of organophosphorus and carbamate pesticides in agricultural products.

Creating Highly Sensitive and Selective Biochip Sensors for the Detection of Organophosphorus/carbamate Pesticides (고감도 및 고선택성의 유기인계/카바메이트계 농약 검출용 바이오칩 센서)

  • Biochip sensing technologies offering in-situ, fast and real-time measurements in addition to portability can be powerfully utilized in a wide spectrum of research areas including environmental science, food science, medical diagnostics and drug development. In this article, we introduce current research trends and economic aspects of the development of various optical biochip technologies for the analysis of organophosphorus/carbamate pesticides in environmental samples, which is of global importance with serious consequences for both current and future generations. In particular, we will highlight recent efforts made in the creation of highly sensitive and selective optical biochip sensors in conjunction with nanobiotechnologies and microfabrication for the rapid detection of organophosphorus/carbamate pesticides.

Adsorption Characteristics of Organophosphorus and Carbamate Pesticides in Four Soils and the Evaluation for Their Leaching Potential Using Two Screening Models (유기인계 및 카바메이트계 농약의 토양흡착성과 간이선발모형을 이용한 용탈 잠재성 평가)

  • Soil adsorption study was carried out to define the mobility of pesticides and to evaluate leaching potential in soils. Nine pesticides including metolcarb, molinate, fenobucarb, isazofos, diazinon, fenitrothion, dimepiperate, parathion and chlorpyrifos-methyl were subjected to adsorption experiment for four types of soils, such as upland, paddy, forest and volcanic ash soil. Based on Koc values, metolcarb and molinate were classified as mobile, fenobucarb as mobile or moderately mobile isazofos as moderately mobile, diazinon, fenitrothin, dimepiperate, and parathion as slightly or moderately mobile and chlorpyrifos-methyl as slightly mobile. Two evaluation methods, Groundwater Ubiquity Score (GUS) index and standard indices of soil-chemical adsorption and biodegradation (half-life), were used for the estimation of pesticide leaching potential. Leachabilities of metolcarb, molinate and fenobucarb were evaluated as high, and isazofos, dimepiperate and diazinon as a little potential, while fenitrothion, parthion and chlorpyrifos-methyl showed very low leaching potential. The leaching potential of pesticides was determined on the basis of intrinsic properties of the pesticides and the soil properties. Among the soil properties, organic matter gave a great influence on the leachability in soil. Therefore, leachabilities of the pesticides used were expected less in Sineom soil with relatively higher organic matter than Gangseo, Jungdong and Yesan soil with lower organic matter.

Studies on screening test for carbamate insecticides by thin layer chromatography (Thin Layer Chromatography에 의한 유기 카바메이트계 농약의 스크리닝 테스트에 관한 연구(1))

  • 한국에서 사용하고 있는 carbamate계 살충제의 사용율(%)은 1968년경에는 2.1%인데 비해서 현재는 급증되고 있는 실정에 있다. 따라서 이로 인한 자타살 사건, 환경오염, 살포시 급만성 중독 등의 사고가 빈번히 발생하고 있는 동시에 법화학적 분석에도 많은 문제점이 되두되고 있으므로 1차적으로 시판되고 있는 carbamate계 살충제를 thin layer chromatography에 의해서 성분의 분리 확인 시험을 실시한 결과 1) Silica Gel G-60 및 Silica Gel F-254에서 적합하였고 2) 전개용매는 chloroform: benzeneP: carbontetra chloride (60:30:25), cyclohexane: acetone (80:20), cyclohexane:acetone(70:30), ether:hexane(80:20)에서 9종의 살충제가 잘 분리되었으며 3) UV light (2537 ${\AA}$) 및 2.6-dibromoquinone-4-chlorimide는 전개는 Rf치를 육안으로 구분하는데 가장 양호하였고 4) Carbamate계 살충제 중 Bux-2와 Sevin은 Silica Gel G-60에서 UV light (2537 ${\AA}$)에서 특이한 청보라 및 청색형광을 관찰할 수 있었다. 5) 이상의 결과를 보아 법화학적 시료중에 함유된 카바메이트계 농약의 분석은 가능할 것이며 또한 이 방법의 적용이 현재의 실정으로서는 적합한 것으로 사료되는 바이다.

Mass-Production of Acetylcholinesterase Sensitive to Organophosphosphates and Carbamates Insecticides (유기인계 및 카바메이트계 농약의 고감수성 아세틸콜린에스테라이즈의 대량생산)

  • For the simple rapid bioassay of organophosphorus and carbamate pesticide residues, a mass-production system of acetycholinesterase (AChE, EC 3.1.1.7, MAChE) using baculovirus and insect cell culture was constructed. The cDNA for AChE was synthesized from Drosophila melanogaster in Halla Mountain, the lipid anchor tail was removed by PCR and was used for the site-directed mutagenesis of three amino acid residues (E107Y, F368L, L408F). The mutated cDNA was inserted into the baculovirus vector and expressed in insect cells. Maximum cell growth and enzyme activity were reached when the cells $(2{\times}10^6\;cell/ml)$ were infected four times at four-day-intervals. His-tag containing MAChE was purified using Ni-NTA column and used for characterization. The activity was maintained under various pHs (3-10) and temperatures $(20-50^{\circ}C)$ under experimental conditions. As an extraction solution for pesticides, methanol is more effective than ethanol. Against major organophosphate and carbamate pesticides, the MAChE showed better sensitivity than AChE and AChE from housefly (Taiwan).

Diamondback moth (Plutella xylostella L.) resistance to organophosphorus and carbamate insecticides in Kangwon alpine vegetable croplands (강원도 고랭지대 배추경작지 배추좀나방(Plutella xylostella L.)의 유기인계 및 카바메이트계 살충제에 대한 저항성 발달)

  • Diamondback moth (Plutella xylostella L.) recently became a single worst insect which is not controlled effectively by organophosphorus and carbamate insecticides in Kangwon alpine croplands. The objective of this study was to determine if diamondback moth has developed a resistance to organophosphorus and carbamate insecticides. Resistance of diamondback moth, collected at Keichon, Jangpyong Taebaek, Chunchon, and Hongchon, was determined by the concentration required to kill fifty percent of population, $LC_{50}$. Their response of resistance varied to insecticides and locations: Taebaek populations were 35 and 70 times more resistant to chlorpyrifos and fenitrothion, respectively, than susceptible(S) population. Hongchon populations were 94 and 254 times more resistant to chlorpyrifos and fenitrothion, respectively, than S population. In addition, Chunchon populations were 37 and 19 times more resistant to profenofos and benfuracarb, respectively, than S population. However, the field populations did not differ in resistance to diazinon, phenthoate, flupyrazofos, carbofuran, and furathiocarb. This study show that field populations of diamondback moth found in Kangwon alpine vegetable croplands have developed a resistance and/or multiple resistance to some insecticides, implying that farmers are losing organophosphorus and carbamate insecticide options for selective control in vegetable crops.

Development of a Colorimetric Rapid Detection Method for Organophosphorus and Carbamate Pesticides using Gold Nanoparticle Aggregation Principle (금 나노 입자 응집 원리를 이용한 유기인계와 카바메이트계 비색-신속 농약검출법 개발)

  • A colorimetric rapid detection method based on acetylcholinesterase (AChE) was developed for the analysis of organophosphorus (OP) and carbamate (CB) pesticides. The AChE catalyzes acetylthiocholine into thiocholine having (-) and (+) charges, and the (+) charge results in gold nanoparticle (GNP) aggregation. The in-activation of AChE by OP and CB has been well known. In order to optimize the colorimetric method, optimal dilution times of commercial serum containing AChE, diameter of GNP, and concentration of acetylthiocholine were tested as a key parameter. The colorimetric detection limits of the method were 7.5 ng/mL for both dimethyl amine and carbofuran pesticides in 60% ethanol. No cross-reaction to other chemicals, such as aflatoxin B1 and ochratoxin A, which can be contaminated with pesticides in agricultural products, was observed. Recoveries from lettuce, sesame leaf, and cabbage lettuce spiked with known concentrations of dimethyl amine and carbofuran were found to be ranged from 83.85 to 133.16%. These results indicated that the colorimetric rapid method based on AChE can be a useful tool for the sensitive, specific, rapid, and accurate detection of OP and CB pesticides in fresh vegetables.
