Therapeutic Effect of Ozone Gas on Bovine Mastitis (젖소 유방염에 대한 오존가스의 치료효과)

  • The potential therapeutic effect of ozone gas on bovine mastitis was investigated. Eighteen quarters from 18 lactating cows with chronic mastitis were included. The 18 quarters were assigned to the control group (treatment with antibiotics for 3 days), experimental group I (0.1 ppm ozone treatment, for 7 days) and experimental group II (1ppm ozone treatment, for 3days). In experimental group I, milk somatic cell counts were lower on day 7 after ozone treatment, compared to the pretreated counts, but were higher than the control counts. In experimental group II, somatic cell counts were significantly decreased (p<0.05) on day 7 compared to the pretreatment counts, and they were lower than the control counts. There were no changes in leukocyte, neutrophil, or lymphocyte numbers, N/L ratios, N/L ratios, or serum total protein in the control and experimental groups. We concluded that ozone gas treatment(1ppm for 3days) might be effective for treatment of bovine mastitis.

A cytotaxonomic study of Galium (Rubiaceae) in Korea (한국산 갈퀴덩굴속(Galium L.)의 세포분류학적 연구)

  • In this study, the somatic chromosome of 14 taxa of Korean Galium L. were investigated. Among them were a few taxa for which the somatic chromosome number was determined for the first time. The somatic chromosome numbers of Korean Galium L. were 2n = 22, 24, 44, 48, 66, 72, 77, 88 and so basic chromosome numbers were x = 11 or 12. Those taxa having the basic chromosome number x = 11 showed polyploidy, including diploid, tetraploid, heptaploid, and octoploid. Tetraploid and hexaploid can be observed in those taxa with the basic number x = 12. The eleven taxa reported 11 for the first time are G. spurium var. echinospermon (Wallr.) Hayek (2n = 44), G. gracilens (A. Gray) Makino (2n = 22), G. pogonanthum Franch. & Sav. (2n = 22, 44), G. trachyspermum A. Gray (2n = 22, 44), G. japonicum (Maxim.) Makino & Nakai (2n = 77), G. trifloriforme Kom. (2n = 44), G. dahuricum Turcz. var. dahuricum (2n = 48, 72), G. dahuricum var. tokyoense (Makino) Cufod. (2n = 22), G. kinuta Nakai & Hara (2n=66), G. verum var. trachycarpum for. nikkoense (Nakai) Ohwi (2n = 44), G. verum var. asiaticum for. pusillum (Nakai) M. Park (2n = 44). The taxa with the same chromosome numbers as previously reported ones were G. boreale L. (2n=22) and G. verum var. asiaticum Nakai for. asiaticum (2n = 44). The chromosome number of G. trifidum L. (2n = 22) was different from the previous report. Two infraspecific taxa of G. dahuricum showed differences in their basic chromosome numbers (x = 11 for G. dahuricum Turcz. var. dahuricum and x = 12 for var. tokyoense (Makino) Cufod. The somatic chromosome number for G. dahuricum Turcz. var. dahuricum was found to be 2n = 48 (tetraploid) or 72 (hexaploid), while that of G. dahuricum var. tokyoense (Makino) Cufod. was found to be 2n = 22 (diploid). Therefore, basic chromosome numbers for members of the genus Galium can be used as valuable characters in delimiting infrageneric sections and investigating interspecific relationships.

Anatomical Observation of Somatic Embryogenesis in Oenanthe javanica ($B^{L}.$) DC. (미나리 체세포 배발생과정의 해부학적 관찰)

  • This experiment was carried out to observe the origin and developmental pattern of somatic embryos of Oenanthe javanica ($B^{L}.$) DC. The experiment included observation of embryogenic cells and their development stages by light microscope, transmission electron microscope and scanning electron microscope. The embryogenic cells, which were smaller than non-embryogenic cells in size with expanded nucleus and dense cytoplasm. When stained with hematoxylin, the embryogenic cells were readily distinguished from the non-embryogenic cells of which cell walls were stained with safranin. It was observed at somatic embryos developed from single cells on the epidermis of developing embryos or in the surface or inside of embryogenic clumps by segmentation pattern. Observation with a transmission electron microscope revealed that the embryogenic cells had dense cytoplasm expanded nucleus, small vacuoles, large amyloplasts containing starch grains, and abundant organelles including lipid bodies. Under a scanning electron microscope, embryogenic callus was shown to consist of very smaller cells than non-embryogenic cells in an orderly arrangement and covered with a net-like structure, while the non-embryogenic callus consisted of large cells, irregular in size and arrangement, and covered with a gelatin-like material.

Effect of Artificial Soils and Aqueous Solutions for Plantlet Acclimatization of Somatic Embryos of Aralia elata (두릅나무 체세포배 유래 소식물체의 순화에 미치는 배양토 및 공급액의 효과)

  • In order to develop effective acclimatization methods for Aralia elata plantlets regenerated from somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. The plantlets were transplanted into plastic boxes containing artificial soil in the presence of either several levels of MS liquid media, distilled water, 2% sucrose or 0.1% hyponex solution. They were then cultured by spraying of distilled water twice a week and maintained in the normal tissue culture room. Perlite was proved to be better than vermiculite on survival rate and growth of the plantlets. As the size of perlite (larger than 0.2 cm in diameter) increased, both the survival rate and growth of the plantlets improved. Among the various MS liquid media and different aqueous solutions tested, distilled water appeared to result in the best survival rate and growth. MS media were also effective in increasing survival rate and supporting growth when diluted to 1/4 and/or 1/8. The acclimatized plantlets could be transplanted directly onto the nursery bed and grown normally. The above results suggest that plantlets regenerated from somatic embryos of Aralia elata be effectively acclimatized using a plastic box containing perlite with distilled water treatment.

  • Somatic embryos or adventitious buds were formed from the segments of zygotic embryo of Lycium chinense Mill. on Murashige and Skoog (MS) medium supplemented with auxins (2,4-D, NAA, IAA) and / or cytokinins (zeatin, kinetin, BAP). Embryogenic callus formed on MS medium containing 0.5 mg/L 2,4-D and then differentiated into somatic embryos without transfer to hormone free medium. On the other hand, adventitious buds were formed on medium with 0.01 mg/L 2,4-D and 0.5 mg/L zeatin. Among various parts of zygotic embryos, the morphogenic potential was higher in the cotyledonary region, and the most organogenic potential was found in cotyledon followed by radicle, hypocotyl, and whole embryo. Histologically, bipolar structure of the heart-shaped embryos were confirmed and in adventitious buds only shoot apical meristems were found to exist.

Somatic Embryogenesis and Plant Regeneration in Immature Flower Bud Cultures of Carnation (카네이션의 미숙화뢰 배양을 통한 체세포배 발생 및 식물체 재분화)

  • Immature flower buds of 'Desio' carnation were cultured on MS agar medium supplemented with 1 ㎎/L 2,L-D. Embryogenic calli were formed from 5-10% of the buds less than 20 ㎜ in length, but only non-embryogenic calli were produced from explants of shoot apex leaf, internode, and flowere buds larger than 20 ㎜. The same method was applied to 16 cultivars of cut Sower carnation and embryogenic calli were obtained in 7 cultivars. Several embryogenic callus lines were selected and maintained through subcultures over 120 weeks without loss of embryogenic competence. The embryogenic cultures were also proliferated rapidly in liquid agitation cultures using MS medium supplemented with 1mg/L 2,4-D. Numerous embryos were formed on the periphery of the cell aggregates upon transfer to auxin-free MS agar medium. Plantlets were transplanted in potting soil and grown to bloom in six months.

Effect of Abscisic Acid on the Number of Somatic Embryo Cotyledons in Tissue Cultures of Aralia cordata Thunb. (땅두릅(Aralia cordata Thunb.)의 조직배양에서 체세포배의 자엽 수 변화에 미치는 ABA의 영향)

  • In order to elucidate the effect of abscisic acid (ABA) on the abnormality of somatic embryos, somatic embryos were induced from embryogenic cell clumps derived from cotyledon segment of Aralia cordata. When embryogenic cell clumps were pretreated medium containing 0.2 mg/L ABA for 3 weeks before transferring to MS basal medium, the frequency of embryos with normal cotyledons enhanced 68% as compared with control. However when clumps pretreated in medium containing 0.2 mg/L ABA were transferred to medium containing 0.1 mg/L ABA, the Sequency decreased to about 29%. In the case of globular embryos cultures in medium containing various concentrations of ABA (0.01 to 1.0mg/L), the frequency of dicotyledonary embryo formation decreased propotionally to ABA concentration. Also, when somatic embryos at various stages were cultured in medium containing ABA, those with polycotyledons appeared at higher frequency.

Effects of Feeding TMR Feed with Dietary Citrus Byproducts on Chemical Properties of Raw Milk from Holstein Cow's Raw Milk (원유의 이화학적 특성에 미치는 감귤 부산물 TMR 사료 급여의 영향)

  • In this study, the effects of feeding providing Holstein cows with with TMR feed including amended with undried citrus byproducts on the properties of their raw milk were investigated. Two samples were used for the experiment: T0 (raw milk produced by dairy cows fed with TMR feed not including citrus byproducts) and T1 (raw milk produced by cows fed with TMR feed including citrus byproducts). The All experiments were conducted with Holstein cows at a dairy farm in the on Jeju island, and were repeated three times, in each, after of which raw milk obtained from 7 cows of each samplein each group was analyzed[ED highlight - please ensure this is correct]. The daily milk yield and somatic cell numbers of T0 and T1 were 24.16 kg and 25.97 kg, and 660,000 thousands and 445,000 thousands, respectively, which means that feeding citrus byproducts to cows increases daily milk yield and reduces somatic cell numbers. There was were no significant differences between T0 and T1 in terms of the raw milk's total plate count, specific gravity, titration acidity, and or the amount of milk protein, lactose, nonfat solids, free amino acids and volatile compounds, while raw milk in T1 showed significantly lower levels of milk fat in raw milk. The antioxidant activity of raw milk was improved by citrus byproducts TMR feed containing citrus byproducts.

Development of Transgenic Plant (Codonopsis lanceolata Trautv.) Harboring a Bialaphos Resistance Gene, bar (Bialaphos 저항성 유전자 bar를 이용한 형질전환 더덕개발)

  • Codonopsis lanceolata ("Deoduck" in Korea) is a perennial herb, and belongs to family, Campanulaceae. Its taproot is used a good source of a wild vegetable as well as an herbaceous medicine. In this study, to develop a bialaphos-resistant transgenic Codonopsis, seed germination mechanism and somatic embryogenesis of the plant were investigated, and Agrobacterium-mediated transformation with bar gene encoding phosphinothricin acetyltransferase (PAT) was performed. Attempt were made to regenerate plant from cells via somatic embryogenesis. When the cotyledons, nodes and leaf disks were cultured on MS medium containing 2,4-D and zeatin, embryogenic calli were induced. Upon transferring the somatic embryos to N6 solid medium without plant growth regulators, they developed into plantlets under continuous illumination. All plants were dead on MS basal medium containing 10 mg/L phosphinothricin (PPT) and Basta, respectively. The explants did not produce calli in the medium containing 200 mg/L kanamycin. The explants were cocultured with Agrobacterium tumefaciens for 2 days, and transformants were selected in MS basal medium containing 1.0 mg/L 2,4-D, 100 mg/L kanamycin and 500 mg/L carbenicillin. After the selection, embryogenic calli were induced and then somatic embryos were produced by subsequent subculturing. The somatic embryos were germiated on N6 basal medium containing 200 mg/L kanamycin and 500 mg/L carbenicillin. PCR analysis showed that nptII and bar genes were introduced in the Deoduck transformants. After the confirmation of bar gene expression in RNA and protein level, the transgenic Deoduck will be used to study the genetics of filial generation with the herbicide control gene, bar.gene, bar.

Induction of Embryogenic Callus and Plant Regeneration by Mature Embryo Culture of Onion (Allium cepa L.) (양파의 성숙배 배양을 통한 체세포배발생 캘러스 유기 및 식물체 재분화)

  • To obtain regeneration system of onion, we analyzed the effects of 2,4-D and BA concentration on the embryogenic callus induction from mature embryos. The highest embryogenic callus induction ratio was shown on MS medium (Murashie and Skoog 1962) containing $2.5\;\cal{mg/L}\;or\;5\;\cal{mg/L}$ picloram after mature embryos were placed on medium. When induced callus were cultured on half strength of MS medium containing $1\;\cal{mg/L}$ Kinetin, the highest shoot formation ratio was observed on MS medium containing $1\;{mg/L}$ 2,4-D and $1\;{mg/L}$ BA. Embryogenic callus were cultured in MS liquid medium containing $1\;\ccal{mg/L}$ of 2,4-D and $1\;\cal{mg/L}$ BA. The suspension cultured cell clumps could be mass propagated. Embryogenic callus were friable, but non-embryogenic callus included a lot of moisture, hence the identification between embryogenic and non-embryogenic callus as easily achieved. When embryogenic callus as cultured on half strength of MS medium containing $1\;\cal{mg/L}$ Kinetin, shoots were induced. The whole plantlet was obtained on rooting medium containing $0.5\;\cal{mg/}$ of NAA.
