The objectives of this study were to analyze mutant lines of Chinese cabbage ($Brassica$$rapa$ ssp. $pekinensis$) using gene tagging system (plasmid rescue and inverse polymerase chain reaction) and to observe the phenotypic characteristics. Insertional mutants were derived by transferring DNA (T-DNA) of $Agrobacterium$ for functional genomics study in Chinese cabbage. The hypocotyls of Chinese cabbage 'Seoul' were used to obtain transgenic plants with $Agrobacterium$$tumefaciens$ harboring pRCV2 vector. To tag T-DNA from the Chinese cabbage genomic DNA, plasmid rescue and inverse PCR were applied for multiple copies and single copy insertional mutants. These techniques were successfully conducted to Chinese cabbage plant with high efficiency, and as a result, T-DNA of pRCV2 vector showed distinct various integration patterns in the transgenic plant genome. The polyploidy level analysis showed the change in phenotypic characteristics of 13 mutant lines was not due to variation in somatic chromosome number. Compared with wild type, the $T_1$ progenies showed varied phenotypes, such as decreased stamen numbers, larger or smaller flowers, upright growth habit, hairless leaves, chlorosis symptoms, narrow leaves, and deeply serrated leaves. The polyploidy level analysis showed the change in phenotypic characteristics of 13 mutant lines was not due to variation in somatic chromosome number. To tag T-DNA from the Chinese cabbage genomic DNA, plasmid rescue and inverse PCR were applied for multiple copies and single copy insertional mutants. Mutants that showed distinct phenotypic difference compared to wild type with 1 copy of T-DNA by Southern blot analysis, and with 2n = 20 of chromosome number were selected. These selected mutant lines were sequenced flanking DNA, mapped genomic loci, and the genome information of the lines is being recorded in specially developed database.

The quality change of musk melons, divided into ripened (90 days) and over-ripened (92 days) set by the formal day maturing melons, was investigated during marketing period at both 10 and $25^{\circ}C$. The rate of weight loss was increased in all samples as the storage period passed and greater in ripened melons than over-ripened melon. The hardness decreased in both well and over-ripened melon as the storage period passed. Furthermore, changes in hardness were prevented in fruit stored at $10^{\circ}C$ compared to fruit stored at $25^{\circ}C$. Immediately after harvest, the solid solubility of over-ripened melon was 14.6%, while that of ripened fruit was 12.8%. The respiration rate of both well and over-ripened melon increased temporarily when stored at $25^{\circ}C$, which is characteristic of climacteric fruits during the first day of storage; however, no change in respiration rate was observed in fruit stored at $10^{\circ}C$. When sensory evaluation was conducted, there were no differences observed in flavor and taste among samples. However, with the exception of over-ripened melon, the texture of all samples increased significantly with storage time when melon was stored at $25^{\circ}C$. The score of overall acceptability remained high for 12 days in both well and over-ripened melon, while that of ripened melon stored at $10^{\circ}C$ and over-ripened melon stored $25^{\circ}C$ remained high for 7 and 5 days, respectively (p<0.05).

Light with two faces, beneficial and harmful effects is an important signal for every living cell. Optimal adaptation to light environment enhances the fitness of an organism and survival in nature. Understandings of light quality and plant growth provide with the economical guides for artificial light sources like LEDs. Compared with those under white light, the 1 week seedlings of Chinese cabbage (Brassica rapa) under monochromic red and blue light showed normal development and growth. In contrast to extremely long and etiolated hypocotyls of the seedlings under dark, those under far-red etiolated were extremely short. Based on the microarray analysis, blue light induced the vigorous development and growth and two fold changes of transcripts than red light condition. To have insight of gene products under different light qualities conditions, GO term enrichments were calculated and each gene according to their GO terms were categorized. The blue and red lights affected the expressions of genes related to biological process. Especially, the genes related to metabolic process and developmental process and plastid and chloroplast in the cellular component category were induced under blue light. This study provided the molecular biological evidence for various light qualities on the growing process of B. rapa.
Tamjin Dam is built in the upper reaches of the Tamjin River which flows through the Janghung-gun and Gangjin-gun of the Jeollanamdo, Korea. In order to map out a preservation strategy of the fish community from dam construction, We studied the distribution of fish distribution and changes of the habitat environment. we found 49 fish species inhabiting in the downstream and upstream of the Tamjin Dam. Among them, migratory fish were two species sweet smelt, Plecoglossus altivelis and freshwater eel, Anguilla japonica. The Coreoperca kawamebari which designated as a species to be protected by The Ministry of Environment of Korea was also observed. After the dam construction, reservoir would be filled with water and running water system will change to standing water system. Then the habitat and spawning space for mountain torrent fish will be reduced and the migration of migratory fish to upstream will be blocked. Through our study, we proposed several ways to protect fish community. In order to preserve the reduced habitat and spawning area of mountain torrent fish, a fishway has been diagnosed to be built in the shallow reservoir in the entrance of the upriver. The establishment of artificial spawning ground on the riverside has been recommended. In addition, We propose a creation of a shelter for fresh water eel, Anguilla japonica in areas where the depth of the water is about l0m by laying rocks. Since it is difficult for a spawning ground to be formed naturally in the reservoir due to the year-round changes in water level, We suggested a floating spawning facility using an artificial fixture. In the downstream of the dam, a waterway-style habitat and spawning ground in the river and increasing the diversity and abundance of fish fauna in the Tamjin River. A low-cost and highly efficient operational fishway has been recommended so that migratory fish such as Plecoglossus altivelis (sweetfish) can migrate from the lower reaches to the upper reaches of the river.

This study was conducted to test a possibility of water quality improvement using a filter-feeding bivalve (Corbicula leana). In mesocosm scale (width ${\times}$ length ${\times}$ depth: 3 m ${\times}$ 3 m ${\times}$ 0.5 m), we investigated the changes of dissolved nutrient and particulate matter including both abiotic and biotic seston. Short term (16 days) mesocosm experiment was conducted in two stages: the first stage for 8 days and consecutive 8 days of the second stage. Both treatment and control mesocosm were switched over by translocating mussels from the treatment mesocosm to the control, at 8th days since the start of the experiment. This design made it possible to compare mussel effect on the water quality change more clearly. The high mortality of mussel was observed in the treatment of the first stage, but it decreased rapidly and stabilized on the 8th day to less than< 4 ind $day^{-1}$. Chl. a concentration in the treatment mesocosm of the first and second stage decreased to 71 and 88% of initial concentration, respectively, and suspended solids decreased to 70 and 77%. At those times, average filtering rate were 0.46 and 0.61 mL AFDW $mg^{-1}$$hr^{-1}$, respectively. Both $NH_3-N$ and dissolved total phosphorus (DTP) concentrations increased with the mussel mortality. $NH_3-N$ concentration was positively correlated with the mussel mortality, while DTP concentration showed negative correlation with it. After translocating mussel from the treatment to the control, $NH_3-N$ concentration significantly increased compared with that of initial control. Although DTP concentration also increased, there was no significant difference relative to that of initial control. These results suggest that application of this filter-feeding bivalve in a eutrophic reservoir could be a potential tool to improve water quality if mussels could acclimatize successfully in early stage of the introduction.

In the present report, two experiments were carried out with the purposes of knowing the differences of response among forage species to growing period and potassium level in culture solution, and investigating possible relation of the responses with occurence of grass tetany on grazing pasture. The results were as follows; (1) At 25 days after germination, fresh weight of top part as well as the sum of top and root parts of the forages increased rapidly. (2) Italian ryegrass was the highest in potassium (K) content but the lowest in magnesiurn(Mg) content among the three gramineous forages, while tall fescue showed the opposite result to it. And orchardgrass was intermediate of the two forage species (Experiment 1). (3) The K contents of forages generally increased, while Mg content became lower with the increase of K level in culture solution. The highest K contents of Italian ryegrass and orchardgrass were more than 3 times of the lowest values. The K contents of alfalfa and tall fescue increased in the narrower range. The decreases of Mg content of Italian ryegrass and orchardgrass were significant in the ranges of 5ppm to 25 or 50ppm KzO, while the content of the leguminous forages and tall fescue decreased up to 1000 level. (4) Fresh yield, water content and K content of the forages were significantly increased with the increase of K20 application levels up to 25 or 50ppm. (5) The K concentration of forage on a tissue water basis was higher at 50ppm than that at 5ppm $K_20$ level, especially for Italian ryegrass and orchardgrass with the value of 2.6times and 2.5times, respectively. However, the K concentration (tissue water) of leguminous forages increased gradually up to the level of lOOOppm (Experiment 2). It is suggested from the results that rapid changes of water content, Mg content and K concentration (tissue water) may occur to forage on a grazing pasture, when both growing period and K level in the soil affect the changes simultaneously. Under such conditions, plant water especially in Italian reyegrass and orchardgrass can function as toxic material to grazing ruminants.
This experiment was conducted to find out suitable extraction time of available fragrant component in Lilium Oriental Hybrida 'Casa Blanca' based on qualitative and quantitative variation of its fragrant component in its flowering stages. The content of essential oil in its flowering stages increased with the progress of flowering stages except flower bud stage and it had the most oil content in its stage after full bloom. The analysis of the essential oil in Lilium Oriental Hybrida 'Casa Blanca' in its flowering stages shows that its main constituents were farnesol (18.96%), benzyl salicylate (13.81%), butyl-hydroxy toluene (12.87%), geranyl linalool isomer (7.46%), isoeugenol (7.17%) in its each stage. Benzenoids had much content at half bloom stage and full bloom stage while fatty acid derivatives had much content at initial flowering stage and after flowering stage. Most benzenoids such as butyl-hydroxy toluene and isoeugenol, which are some of main constituents of Lilium Oriental Hybrida 'Casa Blanca' had strong antioxidant effect. So they were expected to be used as antioxidant agents for food, feed, vegetable oil. The content of monoterpene compounds like geranyl linalool isomer increased at its later stage. Sesquiterpene such as farnesol, which is main component of lily existed only at full bloom stage. Therefore farnesol and geranyl linalool isomer of Lilium Oriental Hybrida 'Casa Blanca' are expected to be used for important spices with good fragrance. The suitable extraction time for the usable main constituents of Lilium Oriental Hybrida 'Casa Blanca' is as follows; farnesol and benzyl salicylate: at full bloom stage, butyl-hydroxy toluene: at half bloom stage, geranyl linalool isomer and isoeugenol: at stage after full bloom. Finally there was variation in essential oil components and contents of Lilium Oriental Hybrida 'Casa Blanca' in its flowering stages and they are expected to be used usefully for flavor industry, food industry, aromatherapy, when they are extracted at their suitable extraction time.

Black mini tomato 'Hei-G' fruits were harvested at different stages of maturity (immature-mature green and mature-black red) and stored at different temperatures (8, 12, and $20^{\circ}C$) to investigate the quality and lycopene content during storage. Weight loss increased dramatically at higher temperature for both harvesting stages without significant differences. Firmness of immature fruits decreased below the initial level of mature fruit (8.1N) after 5, 8, and 19 days storage, when they were stored at 20, 12, and $8^{\circ}C$, respectively. Soluble solid contents of mature fruit increased at initial storage, and were higher as compared to immature fruits before deterioration at each storage temperature. Decrease in titratable acid of mature fruits depended on storage time and temperature. However, titratable acid of immature fruits showed little change during storage, and so it did not affect flavor. Hunter a value changed greatly in immature fruit stored at high temperature. Unlike ripe tomatoes, there was no significant difference in black tomato Hunter b values of immature and mature fruit at initial and 12 days storage. However, immature fruits stored at $8^{\circ}C$ did not reach full maturity and color development and ripening. High storage temperature increased lycopene production while low storage temperature blocked lycopene development. Shelf life of the immature fruits, which was evaluated by elapsed days to conventional mature stage, was 12 and 15 days when they were stored at 20 and $12^{\circ}C$, respectively. The optimum storage temperature to maintain the quality and lycopene content of mature fruits was $12^{\circ}C$. Moreover, the shelf life of mature fruits stored at $20^{\circ}C$ could reach up to 5 days.
The study was performed to elucidate the effects of ethylene-absorbent on the quality of 'Fuyu' persimmon fruits in the MA package. Five persimmons were packed in a MA package film (low density polyethylene, 0.055 mm film thickness), and stored at $-0.5^{\circ}C$ for 60 days. Two persimmons were repacked in a MA package with or without ethylene absorbent $(1\;M\;KMnO_4+zeolite)$ and stored at $-0.5^{\circ}C$. Ten days later, these packages was moved to $2^{\circ}C$ or $25^{\circ}C$ storage room to examine the effect of the ethylene-absorbent on the quality of the fruits. Ethylene removal by enclosed ethylene absorbent in MA packaging reduced the rate of fruit respiration at $25^{\circ}C$, so that $O_2$ and $CO_2$ concentration in packing were maintained higher and lower, respectively, compared to control. These effects were not observed, however, in $2^{\circ}C$ post-storage. Fruit firmness and sugar composition were also influenced by ethylene absorbent, showing more delayed flesh softening and higher sucrose concentration in ethylene absorbent treated fruits than control. But ethylene-absorbent treatment lowered glucose and fructose concentration. That shows that ethylene could influence on sugar composition by inhibiting sucrose inversion to glucose and fructose. The production of ethanol and acetaldehyde was reduced by ethylene removal, but the effect was not so high as other quality indices.

The measurement of total fresh weight of plants provides an essential indicator of crop growth for monitoring production. To measure fresh weight without damaging the vegetation, image-based methods have been developed, but they have limitations. In addition, the total plant fresh weight is difficult to measure directly in hydroponic cultivation systems because of the amount of nutrient solution. This study aimed to develop a real-time, precise method to measure the total fresh weight of Romaine lettuce (Lactuca sativa L. cv. Asia Heuk Romaine) with growth stage in a plant factory using a nutrient film technique. The total weight of the channel, amount of residual nutrient solution in the channel, and fresh shoot and root weights of the plants were measured every 7 days after transplanting. The initial weight of the channel during nutrient solution supply (Wi) and its weight change per second just after the nutrient solution supply stopped were also measured. When no more draining occurred, the final weight of the channel (Ws) and the amount of residual nutrient solution in the channel were measured. The time constant (${\tau}$) was calculated by considering the transient values of Wi and Ws. The relationship of Wi, Ws, ${\tau}$, and fresh weight was quantitatively analyzed. After the nutrient solution supply stopped, the change in the channel weight exponentially decreased. The nutrient solution in the channel slowly drained as the root weight in the channel increased. Large differences were observed between the actual fresh weight of the plant and the predicted value because the channel included residual nutrient solution. These differences were difficult to predict with growth stage but a model with the time constant showed the highest accuracy. The real-time fresh weight could be calculated from Wi, Ws, and ${\tau}$ with growth stage.
