Enhancement of the resistance of Korean catfish(Silurus asotus) to experimental Edwardsiella ictaluri and Aeromonas hydrophila infection by $\beta$-glucan administration (글루칸 투여에 의한 한국산 메기(Silurus asotus)의 Edwardsiella ictaluri와 Aeromonas hydrophila감염증에 대한 저항성의 증가)

  • The effect of $\beta$-glucan administration on the resistance of Korean catfish(Silurus asotus) to experimental Edwardsiella ictaluri and Aeromonas hydrophila infection was evaluated. Fish were either intraperitoneally received $\beta$-glucan($200\sim1000{\mu}g$/100g body weight) dissolved in physiological saline once or twice at an intervals of 3 days, or placed in $\beta$-glucan bath($100{\mu}g/m\ell$) prepared with filtered tap water for 30 or 60 min. Bacterial challenge was performed by intraperitoneal injection of 0.1 ml of bacteria suspension($2{\times}10^7 CFU/m\ell$) 3 days after $\beta$-glucan administration. The $\beta$-glucan injected fish showed an significantly enhanced resistance against experimental infections. The resistance was much higher in the twice-injected fish than in the once-injected fish. But glucan bath did not affect survival rate after the challenge. The protective effect in the $\beta$-glucan injected fish was higher to A. hydrophila than to E. ictaluri. These results indicate that $\beta$-glucan injection can increase the resistance of Korean catfish against experimental E. ictaluri and A. hydrophila infection and that twice injection of $\beta$-glucan at an 3 day-intervals is more effective to the resistance.

Factors Affecting on Pregnancy Rate of Recipients Following Transfer of Hanwoo Embryos Produced In Vivo (한우 체내 수정란 이식 후 수태율에 미치는 요인)

  • This study was performed to improve the pregnancy rates of recipients following transfer of bovine embryos produced in vivo. Superovulation response didn't showe significant differences between each season (4.18 in spring; 4.36 in summer; 5.50 in fall; 4.38 in winter). Pregnancy rate was significantly different (p<0.05) between fresh (43.4%) and frozen embryos (17.2%). In administration of hCG to recipients, the pregnancy rate of fresh embryos (45.7%) was slightly higher than that of control (35.3%), but the pregnancy rates of frozen embryos in control group (25.0%) was higher than that of hCG group (16.0%). When synchrony of recipient and embryo was -2, -1, 0 and 1, the pregnancy rates were 20.0, 45.0, 30.3 and 26.3%, respectively. The pregnancy rates of recipients synchronized by naturally or $PGF_{2{\alpha}}$, CIDR/$PGF_{2{\alpha}}$ and E/P/CIDR/$PGF_{2{\alpha}}$/E treatments were 35.3, 48.0, 29.0 and 40.0%, respectively. Gestation lengths and birth weights of female and male calf were 288 and 290.5 days, 28.3 and 30.0 kg, respectively. The results were showed that the superovulation response was not affected by seasons, and also pregnancy rate didn't increase by administration of hCG, synchrony of embryo and recipients, synchrony methods. Further study and concern should be focused on improving the embryo freezing and pregnancy rate for commercial embryo transfer.

Vitamin B12 Deficiency after a Total Gastrectomy in Patients with Gastric Cancer (위암으로 위 전절제술 후 비타민 Bl2 결핍과 치료)

  • Purpose: The most common metabolic defect appearing following a gastrectomy is anemia. Two types have been identified: One is related to a deficiency in iron and the other is related to an impairment in vitamin B12 metabolism. The purpose of this study is to evaluate the incidence and treatment of vitamin B12 deficiency after a total gastrectomy. Materials and Methods: Serum vitamin B12 concentrations were measured in 128 patients who had undergone a total gastrectomy. The group with a serum concentration under 200 pg/ml was supplemented at one-month intervals with Actinamide and five-six months later, serum concentrations of the vitamin B12 were rechecked. Results: The group with vitamin B12 under 200 pg/ml was 61 (47.6%) of the 128 patients who had undergone a total gastrectomy. In this group, the cumulative incidences of vitamin B12 deficiency were 7.0, 23.4, 33.6, 39.1, 41.4, and 47.7% at 6 months, 1, 2, 3, 4, and 5 or more years, respectively. The treated group with vitamin B12 under 200 pg/ml had 36 (28.17%) of the 128 patients. The 16 of those cases with vitamin B12 levels of $100{\sim}150\;pg/ml$ were supplemented 5.21 times and the vitamine B12 was elevated to above 650 pg/ml. The other 20 cases with an average of vitamin B12 levels of $150{\sim}200pg/ml$ were supplemented an average of 4.75 times, and the vitamin B12 was elevated to above 780 pg/ml. Conclusion: It is necessary to supplement vitamin B12 even 1 year later after a total gastrectomy. The group with vitamin B12 under 200 pg/ml was supplemented $5{\sim}6$ times at one-month intervals with Actinamide $1,000\;{\mu}g$ IM injections and reached normal levels.   • By this study, on-line real-time radiometric system was developed using a 64 channels linear array of solid state detectors to measure wall thickness of insulated piping system. This system uses an Ir-192 as a gamma ray source and detector is composed of BGO scintillator and photodiode. Ir-192 gamma ray source and linear detector array mounted on a computer controlled robotic crawler. The Ir-192 gamma ray source is located on one side of the piping components and the detector array on the other side. The individual detectors of the detector array measure the intensity of the gamma rays after passing through the walls and the insulation of the piping component under measurement. The output of the detector array is amplified by amplifier and transmitted to the computer through cable. This system collects and analyses the data from the detector array in real-time as the crawler travels over the piping system. The maximum measurable length of pipe is 120cm/min. in the case of 1mm scanning interval.

  • Bisphosphonate가 백서 발치와의 치유과정에 미치는 영향에 대해 관찰하고자 생후 2개월 된 Sprague-Dawley계의 백서 18마리를 사용하여, 실험군과 대조군 모두 발치를 용이하게 하고 외상을 최소화 하기 위해 발치 전에 하루 ${\beta}-Aminoproprionitrile(APN)$ 0.2g/㎏을 5일간 투여한 후 발치하고, 감염방지를 위해 매일 1.5㎎/㎏ ampicillin을 3일 동안 근육주사 하였다. 실험군에서는 Biosphosphonate 1㎎/㎏을, 대조군에서는 vehicle(0.9%saline)을 1주에 3회씩 2주 동안 피하 주사하였으며, 각 군은 발치 후 1주, 2주, 4주 간격으로 3마리씩 희생시켜 H&E 염색에 의한 조직학적인 관찰, TRAP 염색에 의한 파골 세포의 활성, 경도 측정기에 의한 경도 측정을 통해 다음과 같은 결과를 얻었다. 1. 각 주간의 전반적인 치유양상은 시간이 경과함에 따라 실험군과 대조군 공히 유사한 치조골 회복 양상을 보였다. 2. 파골 세포의 활성은 1주, 4주 군에서는 실험군과 대조군에서 유사한 양상을 나타내었으나, 2주 군에서는 실험군에서 대조군에 비해 감소된 파골 세포 활성 양상을 관찰 할 수 있었다. 3. 경도측정에서는 모든 군에서 대조군에 비해 실험군에서 유의성 있는 증가 양상을 보였으며, 시간경과에 따른 경도의 차이에서는 1주군과 4주군 간에서 대조군에 비해 실험군에서 통계학적으로 유의성 있는 증가 양상을 보였다.(p<0.05) 이상의 연구를 통해 Bisphosphonate가 백서 발치와의 치유과정과 파골 세포의 활성에 영향을 미치며, 치유된 발치와의 경도를 증가시키는 것으로 나타났다.

Effects of Parity and Season on Production of Embryos in Superovulated Hanwoo (과배란 처리된 한우의 수정란 생산에 미치는 산차와 계절의 효과)

  • This study was performed to investigate the effects of parity and season on the embryo production in superovulated Hanwoo cows. Superovulation was performed from 1 to 8 times by repeated superovulation treatment of Hanwoo cows (n = 22). Irrespective of estrous cycle, donor cows were received a CIDR, progesterone (50 mg) and estradiol benzoate (2.5 mg). After 4.5 days, the donor cows were superovulated with total 28AU FSH (Antorin R-10) administrated twice daily in a decreasing dose for 4 days. On $6^{th}$ and $7^{th}$ of FSH injection, 2.5 mg and 15 mg $PGF_2{\alpha}$ were injected i.m, respectively. CIDR was removed at the $7^{th}$ FSH injection. The donor cows received $200{\mu}g$GnRH at 48 hrs after $1^{st}$ $PGF_2{\alpha}$ injection. The donor cows were artificially inseminated three times after estrous detection at 12 hr intervals and embryos were recovered 7 days after estrous detection. The mean number of total ova, transferrable embryos, degenerated embryos and unfertilized oocytes were $11.6{\pm}7.9$, $5.5{\pm}4.4$, $3.0{\pm}3.3$ and $2.6{\pm}4.1$ per donor cows, respectively. A higher number of total ova were recovered in parity 3~5 ($14.3{\pm}1.3$) than 1~2 ($8.9{\pm}1.9$, P<0.05). The number of recovered normal embryos is significantly higher in parity 3~5 ($7.3{\pm}0.8$) than that of over 6 ($3.7{\pm}1.5$). Significantly higher number of total ova and normal embryos were recovered in summer ($16.4{\pm}2.3$, $8.1{\pm}1.4$) than in autumn ($10.1{\pm}1.8$, $4.5{\pm}1.1$) and winter ($6.3{\pm}1.8$, $3.3{\pm}1.1$), respectively (P<0.05). Transferable embryos were significantly higher in summer ($7.6{\pm}1.3$) than in winter ($3.0{\pm}1.0$, P< 0.05). The results were showed that parity and season affecting on the production of embryos in superovulated Hanwoo.

Effects of garlic Allium sativum on the immune responses of olive flounder Paralichthys olivaceus (마늘, Allium sativum이 넙치, Paralichthys olivaceus의 면역반응에 미치는 영향)

  • This study was aimed to investigate the effects of injection of garlic, Allium sativum, extract and immersion in garlic juice on the nonspecific immunity and the resistance against the artificial infection of Streptococcus iniae and Edwardsiella tarda of the olive flounder, Paralichthys olivaceus. The nonspecific immune mechanisms were assessed in terms of lysozyme activity, nitroblue-tetrazolium (NBT) assay and superoxide dismutase (SOD) activity etc. Relative percent survival (RPS) was assessed by the challenge with S. iniae BS10 or E. tarda KE-1. Almost of the garlic extract injected groups showed the enhanced level of the tested nonspecific immune factors. In the challenge test with S. iniae and E. tarda, RPS of 5% garlic extract pre-injected group was much higher than that of any other tested groups, respectively. Almost of the garlic juice immersion tested groups exhibited strengthened nonspecific immune defence factors, lysozyme activity, the number of lymphocytes and neutrophils, NBT reduction and SOD activity in kidney. In the challenge with S. iniae and E. tarda, RPS in the 0.25 g/L of garlic juice immersed group was much higher than any other tested groups, respectively. The results suggest that the garlic extract and juice would be effective to enhance the nonspecific immunity and protective ability of olive flounder against fish disease such as S. iniae and E. tarda.

In Vivo Embryo Production and Embryo Transfer in Hanwoo and Jeju Black Cattle Using CIDR (CIDR를 이용한 제주 한우 및 흑우의 체내 수정란 생산과 이식)

  • The objectives of this study was to produce valuable offsprings of Hanwoo and Jeju black cattle using in vivo embryo production and embryo transfer techniques during 5 years ($2001{\sim}2005$) in Jeju. Two hundred and eighty six Hanwoo and sixty nine Jeju black cattles, at random stages of the estrous cycle, received a CIDR. Seven days later, the animals were superovulated with a total of 400 mg pFSH ($Folltropin^{(R)}-V$) administered twice daily in constant doses (each 50 mg) over 4 days. On the 6th administration of FSH, CIDR was withdrawn and 25 mg $PGF_{2{\alpha}}$ was administered. Cows were artificially inseminated thrice after estrous detection at 12 hr intervals. The cows received $250{\mu}g$ GnRH at the time of 2nd insemination. Embryos were recovered 7 or 8 days after the 1st insemination. Recipients were synchronized with donors by insertion of a CIDR for 7 days and administration of 25 mg $PGF_{2{\alpha}}$ at the time of CIDR removal. The collected embryos were transferred to 1,219 recipients by 6 transfer persons. The mean numbers of total ova and transferable embryos from Hanwoo and Jeju black cattle donors were 7.4 and 4.7, respectively The number of transferable embryos differed between Hanwoo (5.0) and Jeju black cattle (3.5, p<0.05), while that of total ova did not differ. Repeated superovulation treatments decreased (p<0.05) the ratio of numbers of the flushed animals vs. superovulated animals in Jeju black cattle, and the numbers of total ova and transferable embryos in Hanwoo. More transferable embryos were collected at summer (5.6) than winter (2.9, p<0.01). The mean pregnancy rate was 40%. The pregnancy.ate was affected by transfer year (2001<2004, p<0.05) and transfer person ($33.0{\sim}41.9%$, p<0.01), while not by donor (embryo) breed. These results showed that in vivo embryo preduction and embryo transfer techniques using CIDR for Hanwoo and Jeju black cattle donors as well as recipient, regardless of their estrous cycle, may enable a stable embryo production and recipient preparation.

  • Groundwater flow in a fractured rock mass is related to the geometric characteristics of the fracture system. The objective of this study aims to analyze the probabilistic density function of fracture properties md their relations to the hydraulic conductivity in volcanic rocks of the northern Yosu area. Fracture characteristics were investigated by core logging and acoustic televiewer logging in four boreholes and the hydraulic conductivity was obtained from the constant pressure injection and fall-off tests. The 303 fractures were grouped into three sets by their orientation and three fracture types by the degree of opening in aperture. As a result of the study, the hydraulic conductivity in the test section intersected by open and semi-open fractures, conductive fractures, and set 1 fractures was very high, while closed fractures did rarely affect the hydraulic conductivity. It was recognized that the hydraulic conductivity in a fractured rock mass was preferentially affected by the aperture size of conductive fractures and fracture intersection frequency and size, secondly.

  • In this study, the characteristics of ITO thin film was investigated to finding a low cost and highly transparent electrodes for display of mobile communication devices. The ITO film was deposited by DC magnetron sputter. The experimental conditions were changed as follows: 1. ambient pressure changed 1 to 3 mTorr with 1mTorr step, 2. bias electric voltage changed with 10V step. The chamber was pumped out by rotary pump until 10-3Torr then the diffusion pump was used to lower the pressure of 10-6Torr. The results shows us the film growth was obvious when the bias voltage was larger than 300V, but the overall thickness tendency was existed: the more voltage is the thicker thickness. At 330V bias voltage condition, the deposition rate was the largest and apparent grain was showed.
