Growth and Survival Rates of Flat Oyster, Ostrea denselamellosa, by Condition of Larval Cultivation (벗굴 (Ostrea denselamellosa) 유생의 생육조건에 따른 성장과 생존)

  • For the effective seedling production of flat oyster, Ostrea denselamellosa, dietary value of live food, densities, water temperature and salinity on growth and survival rate of the larvae were examined. In rearing larvae by feeding them phytoplankton diets, the optimal survival rate and growth rate of larvae were found using a mixed phytoplankton diet which was mixed with Isochrysis galbana, Chaetoceros calcitrans and Chlorella sp. The highest growth and survival rates of the larvae were 208.4% and 38.8% with the phytoplankton diet. In growth and survival rates of larvae with various rearing densities, the highest survival and growth rates were 228.1% and 29.0% at the density of 2 individuals/ml. In observing rearing experiments of the flat oyster larvae under various temperature conditions, average growth rates of the larvae in respect to shell length were 202.2%, 240.4%, 250.6% and 121.3% in natural water temperatures (18-22$^{\circ}C$), 24$^{\circ}C$, 28$^{\circ}C$ and 32 $^{\circ}C$, respectively. And average survival rates of the larvae were 16.0%, 32.0%, 13.0% and 0% in natural water temperatures (18-22$^{\circ}C$), 24$^{\circ}C$, 28$^{\circ}C$ and 32$^{\circ}C$, respectively. In rearing at various salinities, the highest growth rates of the larvae in shell length was 240.0% at 30.0 psu and the highest survival rate was 31.0% at 25 psu.

Development of Transgenic Crops and Research Projects for Biotechnology Application (유전자 전환작물 개발 연구 현황과 과제)

  • The main objective of this topic is to establish strategies and to plan biotechnology researches which are related to the agricultural improvements especially focusing on the crop breeding in Korea. From 1960's to 1980's government policy had been emphasized to develope high yielding cultivars for the self sufficient supply of the staple food crops. As a result, considerable increase of rice production has been made with accumulating technology and man's powers. Recently genetically modified crops harboring useful characteristics have been developed using biotechnology and released in the developed countries. National research institutes and private companies have been developed biotechnology researches to establish competitive capabilities, however they have not been successfully used in commercialization. Therefore it is necessary to promote the practical. application by connecting molecular technology with conventional breeding. Proposed research projects are; (1) basic researches including plant genome studies, (2) developing new cultivars through gene transformation, (3) screening and producing antioxidants, secondary metabolite substances and edible vaccines. To set a government policy, both domestic and international research trends were reviewed and possibility of success based on the economic view point were discussed. The intellectual property and preservation of environment play a key role to decide the research priority. It is also necessary for us to make one step system for the distribution of research resources such as microorganisms, genes cloned, plant seeds and research informations for promoting research activities.

Effect of Vermicast Application of Earthworm Feeding Food Waste in Vegetable Plants (Radish and Chinese Cabbage) Growth (음식물 쓰레기를 먹이로 한 지렁이분립이 채소류(열무와 엇갈이배추) 생육에 미치는 영향)

  • This study was performed to make the method of food waste manufacturing for feeding to earthworm and different ratio of earthworm cast was investigated for applying vegetable plants that cultivated in a family or roof garden. We made conventional treatment of gardening soil and prepared soils with earthworm cast (the only fertilizer) at 30, 50, 70 and 100% for planting vegetable plants and then we studied its growth feature. The treatment was repeated three times with 12 weeks for each time. The cultivation of radish and chinese cabbage were managed at the same cultivation level for RDA standard cultivation method, and each sample of soils and plants was analyzed by appropriate analysis method. The growth investigation was performed 12 week after planting, and as for growth characteristics, height, leaf diameter, leaf number, leaf length, fresh weight and dry weight were investigated. The chemical characteristics, soils and plants, were surveyed. As for the growth characteristics investigated, radish and chinese cabbage showed the biggest growth in 70% and 50% treatment with average growth of all characteristics, height, leaf diameter, leaf number, leaf length, fresh weight and dry weight, respectively. Applying these results, a new equation for vermicast recommendation for radish and Chinese cabbage in summer (June-August) was presented. The earthworm cast 50% level with the gardening soil showed the high production in chinese cabbage. In case of radish, earthworm cast 70% with gardening soil level showed great growth, significantly.

  • Mycorrhiza refers to the association between a plant and a fungus colonizing the cortical tissue of the plant's roots during periods of active plant growth. The benefits afforded by plants from mycorrhizal symbioses can be characterized either agronomically, based on increased growth and yield, or ecologically, based on improved fitness (i.e., reproductive ability). In either case, the benefit accrues primarily because mycorrhizal fungi form a critical linkage between plant roots and the soil. The soilborne or extramatrical hyphae take up nutrients from the soil solution and transport them to the root. This mycorrhizae-mediated mechanism increases the effective absorptive surface area of the plant. There are seven major types of mycorrhizae along with mycoheterotrophy: endomycorrhizae (arbuscular mycorrhizae, AM), ectomycorrhizae (EM), ectendomycorrhizae, monotropoid, arbutoid, orchid, and ericoid. Endomycorrhizal fungi form arbuscules or highly branched structures within root cortical cells, giving rise to arbuscular mycorrhiza, which may produce extensive extramatrical hyphae and significantly increase phosphorus inflow rates in the plants they colonize. Ectomycorrhizal fungi may produce large quantities of hyphae on the root and in the soil; these hyphae play a role in absorption and translocation of inorganic nutrients and water, and also release nutrients from litter layers by producing enzymes involved in mineralization of organic matters. Over 4,000 fungal species, primarily belonging to Basidiomycotina and to a lesser extent Ascomycotina, are able to form ectomycorrhizae. Many of these fungi produce various mushrooms on the forest floor that are traded at a high price. In this paper, we discuss the benefits, nutrient cycles, and artificial cultivation of mycorrhizae in Korea.

Quality Changes in Tomato Fruits Caused by Genotype and Environment Interactions (재배환경과 유전형의 상호작용에 따른 토마토 과실 품질 변화)

  • Bred and grown around the world, tomato (Solanum spp.) has highly valuable fruits containings various anti-oxidants such as lycopene, flavonoids, glutamine, and ${\beta}-carotene$. Several studies have explored, way in which to enhance the growth, management and quality of tomato, we focus on the management of growth for yield rather than quality. The expression of superior agronomic traits depends on where cultivars are grown. We evaluated 10 cultivars grown in three environment for their lycopene. HTL3137 ($70.48mg{\cdot}kg^{-1}$), which was grown in Yoeju in spring/summer, contained the highest lycopene content, while HTL10256 ($20.9mg{\cdot}kg^{-1}$), which was grown in Suwon in spring/summer, contain the least lycopene.Correlations between color components and lycopene content varied according to growing location and season. In spring/summer-grown tomatoes from Suwon, no significant correlation was observed between any color component (redness [R], greenness [G], blueness [B], luminosity, $L^*$, $a^*$, $b^*$, hue and chroma) and lycopene content. A correlation was observed between B and lycopene content in tomatoes grown in Yeoju during the same season. In tomatoes grown in Yeoju in fall/winter, significant correlations were found between lycopene content and G, luminosity, $L^*$, and hue. Variance in interactions between genotype, environment, and genotype ${\times}$ environment (G ${\times}$ E) using Minimum Norm Quadratic Unbiased Estimate (MINQUE) analysis indicated that lycopene content depends on genotype (51.33%), environment (49.13%), and G ${\times}$ E (21.43%). However, when the Additive Main Effects and Multiplicative Interaction (AMMI) was used, the G ${\times}$ E value was highest.

Properties of Pepper growth and Yield, Cost Down with No-Tillage Organic Cultivation in Vinyl Greenhouse (시설고추 무경운 유기 재배의 생육 및 수량 특성과 생산비 절감효과)

  • To investigate the possibility of sustainable agriculture in no-tillage pepper this study was carried out in vinyl greenhouse with organic cultivation having no pesticide certification. 1. Growth and yield in pepper cultivation General growth in pepper was suppressed with decreasing hill spacing, primary branch length, and stem width. Fruit diameter and fruit weight in no-tillage increased significantly, and yield of pepper increased by 10% compared with conventional tillage. From results organic cultivation in no-tillage improved a quality of pepper compared with conventional tillage. 2. Production cost of conventional tillage and no-tillage Production cost of conventional tillage and no-tillage was not different in seed cost, inorganic fertilizer cost, pesticide cost, repair cost, light agricultural tool cost, agriculture facilities depreciation cost and so on. Intermediary goods cost in no-tillage was decreased by 11% for organic fertilizer cost, light and heat expenses and power rate, heavy agricultural tool cost, and repairing expenses compare with conventional tillage. Employment effort cost and work effort cost were decreased, and farm income and farm income rate were increased by 11% and 5%, respectively, in no-tillage. In this work, yield and gross income were increased by 10% and 25%, respectively, in no-tillage. Therefore material cost, intermediary goods cost, working expensive, farm income, and income rate were increased by 34%, 3%, 2%, 52% and 22%, respectively.

Urban aquaculture of catfish, Silurus asotus, using biofloc and aquaponics systems (바이오플락과 아쿠아포닉스를 이용한 도심형 양식시스템에서의 메기양식)

  • This study was conducted to determine whether the water in which nitrate accumulated during long-term fish culture in an aquaponics system without water exchange could be removed and reused as catfish-culturing water. The catfish (Silurus asotus) were cultured in the urban aquaculture system using BFT (Biofloc Technology) aquaculture and an aquaponics system (two rearing tanks, 3 tons each) without exchanging the rearing water. After 151 days (from March to August) of rearing, 2.8 g of fry had grown to an average weight of 171.3 g (total weight, 56.53 kg) and 235.5 g (total weight 71.1 kg), respectively. The overall survival rate was 65% in the urban aquaculture system. However, the survival rate was 77.7% before separation into the two tanks. The survival rates after the separation were 92.9% and 78.0%. In the early biofloc watermaking process, there was a high mortality rate. After water stabilization, the mortality rate decreased and some mortality occurred during the period when the total amount of suspended solids (TSS) increased. The results of monthly blood analysis of the catfish showed that the AST concentration was significantly higher in April. Blood ALT levels and triglycerides showed no difference in the rearing period and the glucose, cholesterol, and total protein levels were significantly higher in July. There was no difference in the other periods. The plants produced by the aquaponics system using catfish-rearing water were lettuce, basil, chard, and red chicory. These showed smooth growth and a total of 148.85 kg of plants were harvested in five months. It was possible to remove nitric acid from the aquaponics system and reuse it as catfish-rearing water. Maintaining proper plant quantity according to the capacity of the catfish showed that the combination of agricultural and aquatic products was possible.

Allelism and Molecular Marker Tests for Genic Male Sterility in Paprika Cultivars (파프리카 시판 품종에 대한 유전자적 웅성불임성의 대립성 및 분자표지의 유용성 검정)

  • Paprika (Capsicum annuum L.), a colored bell-type sweet pepper, is one of the most important money making vegetable crops in Korea. The cultivation area, total production, and exports of paprika are gradually getting increased, but the paprika cultivars used in Korea are all imported. It was well-known that the genic male sterility (GMS) is the main way to produce paprika hybrid seeds. However, it is little known that how many and what kinds of ms genes are used for breeding of paprika $F_1$ varieties. In this study, eight paprika cultivars ('Special', 'Debla', 'Plenty', 'Fiero', 'Boogie', 'Fiesta', 'Derby', and 'Minibell'), popularly cultivated in Korea and three different genic male sterile lines ('GMSP', 'GMS3', and 'GMSK') were used. For allelism test among the $F_1$ cultivars, half diallel crosses were performed. The result demonstrated that the most of the GMS in paprika cultivars except for 'Minibell' were same allele. To identify which GMS gene(s) were used for paprika $F_1$ cultivars, top crosses between previously known GMS lines and the $F_1$ cultivars were performed. As a result, we found that the $ms_k$ and the $ms_p$ genes were alleles for the GMS of 'Minibell' and for the other cultivars, respectively. We also confirmed that the GMS gene identification using GMSK-CAPS marker linked to the $ms_k$ gene and the PmsM1-CAPS marker linked to the $ms_p$ gene in $F_2$ progenies of 'Minibell' and 'Fiesta' and 'Derby' cultivars, respectively. In addition, we developed the PmsM2-CAPS marker for 'Plenty', 'Fiero', and 'Boogie' cultivars. We expect that these markers will be very useful for breeding new maternal (male sterile) line of paprika.

Virulence of Vibriosis in Larva Stage of Pacific oyster, Crassostrea gigas (초기 유생단계 참굴, Crassostrea gigas에 대한 Vibrio의 병원성)

  • The potential pathogenicity of Vibrio splendidus biovar II, which was isolated from triploid larvae of pacific oyster with bacillary necrosis and fish pathogenic V. anguillarum were investigated. The 5-day-old larvae infected with V. splendidus biovar II at the dose of $1.81{\times}10^{4}$ CFU/$m\ell$ started to die within 8 hours after exposure and the mortality were reached to 100% in 16 hours. However, $1.13{\times}10^{4-5}$ CFU/$m\ell$ of V. anguillarum caused 5.5-20% mortality of the larvae after 24 hours. The 10-day-old larvae infected with V. splendidus biovar II at the dose of $5.0{\times}10^{5}$ CFU/$m\ell$ showed mortality from 8 hours after challenge and led to a marked mortality of 90.47% after 24 hours. But V. anguillarum at doses of $5.08{\times}10^{3-6}$ CFU/$m\ell$ did not show mortality in the 10-day-old larvae. Therefore V. splendidus biovar II exhibited stronger virulence in 5-day-old larvae than 10-day-old and young oyster. Changes in the concentration of Vibrio in sea water showed that V. anguillarum decreased from $1.13{\times}10^{5}$ CFU/$m\ell$ to 1.7${\times}$105 CFU/$m\ell$ and V. splendidus biovar II increased from $1.81{\times}10^{4}$ CFU/$m\ell$ to $1.7{\times}10^{7}$ CFU/$m\ell$. This strong survival ability of V. splendidus biovar II in seawater was thought as one of the virulence factors against oyster larvae. The mortalities of 5-day-old and 10-day-old oyster larvae were decreased by addition of 30 $\mu{g}/m\ell$ of antibacterial agent(oxytetracycline or streptomycin). These results suggest that bacillary necrosis by V. splendidus biovar II can be occurred in oyster larvae in Korea. And virulence of V. splendidus biovar II is stronger than that of V. anguillarum in oyster larvae causing significant mortality at the density of $10^{4}$ CFU/$m\ell$.

Combination Culture of Rotifer Brachionus rotundiformis and Copepod Apocylops sp. (로티퍼 Brachionus rotundiformis와 코페포다 Apocyclops sp.의 혼합 배양)

  • The small size food organism(under the size 150${\mu}m$) is needed as food for early stage of marine fish larvae of small mouse (e.g the group of grouper). This study was investigated to develop a method for copepod Apocyclops sp. culture in combination with the rotifer B. rachionus for stable culture of copepod species and harvest of various size food organisms. The culture conditions as temperature, salinity, culture volume, photo period, culture preiod and observation interval were 25${\circ}C$, 22ppt, 40ml, all dark except to observation time, 16 days and every two day during the experimental period, respectively. The Tetraselmis suecica was used as the food for the two testing orgtanisms. After every two day counting, theses two organisms were transferred to fresh culture tanks with Tetraselmis suecica of $7{\times}10^5$cells/ml. In the mixed culture of B. rotundiformis and A. sp., growth of rotifer was suppressed by mixed culture with A. sp. whereas the growth of copepod Apocyclops was promoted in the mixed culture with rotifer B. rotundiformis (the maximum density was 22 individuals/ml through the 16 culture days). Moreover, the number of copepod nauplius were promoted about 2 times in the mixed culture compared to the numbers in single species culture. With this combination culture, the havested two food organisms of variable sizes. This size variation of food organisms was useful tools for larval rearing of small mouse marine fish larvae and next step food organism size of post hatched larvae.
