Studies on the Aphid Transmission of Some Cruciferous Viruses (십자화과식물 바이러스의 진딧물 매개에 관한 연구)

  • This is the fist report on detailed aphid transnsmission studies of cruciferous virus in Korea, and experiments aimed to get basic informations for control of vectors. Aphid transmission of turnip mosaic virus prevalent on radish in the field was studied. Results obtained were as follows: 1. Myzus persicae, Lipaphis erysimi, Aphis gossypii and Aphis craccivora were found to transmit turnip mosaic virus. 2. The proper time for turnip mosaic virus transmission by Myzus persicae was 1 hour of fasting, 3 minutes for acquisition, and 1 minute for inoculation: Lipaphis erysimi was 2 hours for fasting, 5 minutes for acquisition, and 3 miuutes for inoculation: while Aphis gossypii needed 1hour for fasting, and 3 minutes for each of the acquisition and inoculation periods. 3. There was Po great difference in probing patterns between nonfasted and fasted aphids for 2 hours. All the fasted aphids began feeding after 4 minutes, 4. When Myzus persicae were transferred artificially at 1-2 minute intervals, the number of probes with aphids fasted for 2 hours was much greater than that of nonfasted aphids. Aphids fasted for 2 hours mainly transmitted the virus before 4 minutes, with an acquisition feeing period of less than 3 minutes

Study of transmission of Candida albicans in denture by dental polishing lathe (의치에서 치과기공용 연마기를 통한 캔디다균의 전염성에 대한 연구)

  • Purpose: The purpose of this study was to evaluate the transmission of candida in denture by dental polishing lathe. Materials and Methods: Maxillary complete dentures made from the same model were infected with Candida albicans. The infected dentures were polished by dental polishing lathe with sterile pumice and distilled water. And then sterile maxillary complete dentures were polished with same method. Polishing surface was wiped with a cotton swab and the sample was checked for Candida albicans. The polishing wheel at room temperature was checked for Candida albicans every 24 hours for 3 days. Results: A considerable number of Candida was found on the polished sterile dentures. And the Candida albicans in polishing wheel was detected at up to two days. Conclusion: An anti-infection measure is urgently needed when dental polishing lathe is used for denture polishing, because of the possibility of fungal infection.

Studies on black rot of rice seedlings (벼 검은빛 모썩음병에 관한 연구)

  • The study was carried on a new disease of rice seedling which named 'Black rot of rice seedling'. None of papers on this disease was reported recently. The symptom of this disease was quite different from other seedling rot of rice plant. Submerged mycelial growth in water was black colored and it was surrounded whole surface of seed. Soil on which diseased seedlings were placed was black colored by this disease and the seedlings, finally stopped to grow within 3 em in height, and appeared with black rot symptom. Sporulation of the pathogen does not occurred under both of natural and artificial conditions. The mycelium appeared in dark color and with distinguished septation under the microscope. With those results it is easily recognized that the pathogen does not belong to those which causal organism:; of other seedling rot of rice plant like as Phycomycetes or Fusarium species. The study should be continued on identification of the causal organism. Favorable environments for the disease did not differ from other seedling rot of rice plant, and varietal resistance also did not significantly differ among the varieties used in this experiment. Dissemination of the disease was carried out not by planted seeds but by infected soil.

Significance of Semame Seedborne Fungi, with special Reference to Corynespora cassiicola (참깨의 종자전염성 진균과 그 병원성 : Corynespora cussiicola를 중심으로)

  • Alternaria sesami, A. sesamicola, A. tenuis, A. longissima, Cercospora sesami, Cephalosporium sp., Corynespora cassiicola, Fusarium equiseti, F. moniliforme, F. oxysporum, F. semitectum, Macrophomina phaseolina and Myrothecium roridum were detected from 40 seed samples of sesame. A sesami, A. sesamicola, A. tenuis and C. cassiicola were the predominant fungi. Except C. cassiicola, all fungi were almost completly reduced and wiped out the infection by pretreatment with chlorine. Plating components also indicate that C. cassiicola was well-established infections. Seedborne infection of C. cussiicola caused heavy seed rot and seedling mortality. Detailed description has been given on the habit character of C. cassiicola under stereoscopic microscope and the variation in colony character and spore morphology have been taken into account. In inoculation experiments, C. cassiicola produced severe leaf and stem spots and blights on sesame plants resulted in ultimate death of the plants. A. sesami, A. sesamicola A. longissima and C. sesami also produced mild to severe leaf spotting and leaf blight when suspension of their conidia were sprayed on to plants. In soil inoculation experiments, F. oxysporum and M. phaseolina were the most pathogenic causing seed rot and seedling blight.

Incidence of Watermelon Mosaic Virus in Cucurbits (박과 작물에 발생하는 Watermelon Mosaic Virus에 관한 연구)

  • Cucurbits including pumpkin (Cucurbita pepo), gourd (Lagenariaa siceraria), cucumber (Cucumis sativus), melon(Cucumis melo) and watermelon(Cucurbita anguria) were diseased with mosaic symptoms. The causal virus was identified as watermelon mosaic virus(WMV). The WMV was transmitted by Myzus persicae Sulzer, and no seed borne virus was found. The virus caused large local lesions on the inoculated leaves of the Chenopodium amaranticolor and mosaic symptom on the upper leaves of Cucumis melo, Cucumis sativus, Lagenaria siceraria, Cucurbita anguria and Cucurbita pepo. There were no symptoms on the inoculated leaves of the Nicotiana tabacum var. Bright yellow, Nicotiana glutinosa, Vigna unguiculata. Petunia hybrida and Datura stramonium. Thermal inactivation point was $55\~65^{\circ}C$, dilution end point was $10^{-4}\;10^{-5}$ and longevity in vitro of the virus was $7\~8$ days. The virus showed positive reaction against watermelon mosaic virus antiserum in microprecipitin tests. The virus particles were flexuous rods in size of 750 nm.

  • Out of 50 seed samples of Sesamum indicum L. tested, Alternaria sesami and Corynespora cassiicola were detected in 29 samples and 38 samples, respectively. Heavy infection of A. sesami and C. cassiicola caused seed rot and seedling blight on water agar as well as in pots. In case of light infection the seed germinated showing elongation of the radicle and hypocotyl, but soon light brown or dark brown lesions appeared at certain parts of the shoot and root where sporulation of the fungi was observed after prolonged incubation in moisture. Four seed disinfectants were compared for the control of seed-borne infection of A. sesami and C. cassiicola. Complete control of seed-borne infection of C. cassiicola was obtained by seed treatment with Benlate T and Homai and seed treatment with Busan 30 was found superior to control seed-borne infection of A. sesami.

Effects of Light on Reproduction of Gibberella zeae and Overwintering of Soil-Borne Conidia (밀붉은곰팡이병균의 분생포자 및 자낭각 형성에 미치는 광선의 영향 및 토양에서의 분생포자월동)

  • 1. Continuous light induced more conidia than alternating light and darkness treatment in isolate Chinju 1. Isolate Suwon 3 produced much more conidia on synthetic medium than Chinju 1 in light. Conidial formation in Suwon 3 increased remarkably with alternate light and darkness in 8 days incubation. 2. Light was essential for perithecial formation in Chinju 1. No matured perithecia were observed in Suwon 3 with any treatment. Abundant perithecia were produced in Chinju 1 but only perithecial initiation occurred in Suwon 3. Suwon 3 produced significantly more conidia than Chinju 1, while perithecial formation was reversed. 3. Conidial numbers in soil decreased significantly through the winter at 10, 30 and 50 per cent soil moisture, with the most striking decrease at 10 per cent levels, but the number recovered again beginning in March, regardless of the soil moisture. The above results provide us a useful clue to support the possbility the conidia may serve as a primary inoculum.

Primary Inoculum of Strawberry Anthracnose in Nursing Field (육묘상에서 딸기탄저병의 1차전염원)

  • This experiment was carried out to investigate the primary inoculum of strawberry anthracnose in nursery field. The pathogen, Colletotrichum gloeosporioides was not detected in soil and weeds of nursery field but symptom of anthracnose was developed in mother plants collected from market after incubation in humid chamber, The symptom of anthracnose was expressed in the strawberry plant that reserved for 17 days in field after inoculation by spore suspension but was not observed there after. When inoculated leaves were observed by SEM, only appressoria were observed 7 days after inoculation. So, it is guessed that dissemination of Colletotrichum sp. into nursery held will be by contamination of mother plants, and diagnosis by naked eyes may be impossible because symptom will be not developed if environment is to be adequate to penetration and in case of imperfect penetration after germination, the pathogen remains appressorium to achieve penetration.

Identication of Bean Yellow Mosaic Virus from Trifolium repens (흰 토끼풀에서 분리한 강낭콩황반모자이크 바이러스의 동정)

  • The virus isolated from white clover, Trifolium repens showing mosaic symptom was identified as bean yellow mosaic virus (BYMV) based on the host range, physical properties, aphid transmission, serology and morphology of the virus particles. Chenopodium amaranticolor and C. quinoa produced local lesions on the inoculated leaves and chlorotic spot on the upper leaves. Broad bean and cowpea produced local lesions on the inoculated leaves and mosaic with vein necrotic symptoms on the upper leaves. French bean showed vein necrosis on the inoculated leaves, yellow mosaic on the upper leaves and bud blight. The average size of virus particles was 740nm in length. The virus was also transmitted by Myzus persicae. The thermal inactivation point of the virus isolate was $60\;to\;65^{\circ}C$, the dilution end point $10^{-3}\;-\;10^{-4}$ and the longevity in vitro was 3 days Serological tests with the virus purified from Trifolium repens were positive to BYMV antiserum.

An Epidemiological Study for on Outbreak of Typhoid Fever in Busan Area (부산 일부지역에 유행한 장티푸스 환자에 대한 역학적인 조사)

  • An epidemiological study was conducted, from April 28 to June 3, 1975, to investigate source and course of infection of typhoid fever occurred in Munhyeon Dong, Nam Gu, Busan. The obtained results were as follows: 1. The entity of unknown febrile disease was identified as Salmonella typhi, D group and it's symptoms were abdominal pain, diarrhea, hepatomegaly, nausea and vomiting, aplenomegaly, rose spots, tenderness of ileocecal region etc., in that order. 2. The average duration from onset to recovery was 25.2 days and incidence rate was 39.5 per 1,000 population. 3. The source of infection was presumed a charity patient. 4. By the duration of report on the cases from onset, 10-19 days group was the highest. 5. The positive rate of initial stool culture was 38.9% and secondary was 5.6%. 6. Distribution of the cases by age and sex showed that 5-9 years old group was the highest as 30.5%, and male was higher than female. 7. Distribution of the cases by education level, under primary school as 66.6% was the highest. By the living standard, the highest was low income earner group as 77.8%. 8. Utilization rate of medical facilites was drug store (41.7%), herbal drug (8.3%), clinic (5.6%) in that order.
