This study was conducted to develop a method for direct determination of phagocytic activities in human circulatic systems and to measure the phagocytic activities in human leukocytes from the alcoholics, since phagocytic activity was considered to be very important in human immune mechanism at early stage for the health care of the alcoholics. The subjects for this study were 130 among which 95 males and 3 females were diagnosed as alcoholism and 32 was healthy blood donors. A thin layer of heat-killed Staphylococcus aureus Cowan I was placed on a plastic dish and reacted with whole blood to measure the phagocytic plaque formation by human leukocytes. In order to determine the health conditions of the subjects, some clinical laboratory tests, such as white blood cell counts, hemoglobin contents (Hgb), mean corpuscular volume of red blood cells(MCV), serum electrophoresis, B and T-lymphocytes, T-lymphocyte subtypes and phytohemagglutination test were also implemented. Compared to the non-alcoholism, new and old alcoholic inpatients showed statistically significant differences on levels of Hgb and MCV (p<0.05), but showed that T and B-lymphocyte numbers decreased and Helper T cell/Suppressor T cell ratio ($1.6{\pm}0.8$%) increased. Compared to non-alcoholism, phagocytic plaque activities of leukocytes from alcoholic patients decreased significantly and an unusual pattern in phagocytic plaque was observed, showing a strange body and chain shaped phagocytosis. Based upon these results, it is concluded that a phagocytic-plaques of Staphylococcus aureus Cowan I by leukocytes was very simple and useful method for the early immunological determination of phagocytic activities in alcoholic patients without requiring any special equipments.
A new baekji(Angelica dahurica Bentham et Hooker) variety, Baekji l, was developed through a pure line selection at the Gyeongbug Provincial RDA during the period of 1990 to 1995. The variety was characterized to have high plant height. Long root length and large root diameter as compared with a check varicty of Bonghwa baekji but emergence date, flowering date and leaf number of Baekji l was similar to that of the check variety, and it was also more tolerant to nematode and heat stress. Peeled root color of Backji l was yellowbrown but number of the lateral root of the variety was greater. The dried-root yield of Baekji l in yield trial, regional adaptation trial and farmer's field trial was always $21\sim31%$ higher than that of the check variety.

Next, continuous advanced the ${\beta}-globurin$ fractions at first month as primary crisis in a diagram, and indicated the maximum ratio at 8th months as the second crieir on the its diagram of after vaccinati on, however, few changed in non-vaccinated bovine serum. (Figs 4.) Especially, ${\gamma}-globurin$ fractions are advanced the maximum ratio as 41. 45% 4.48% anp advanced to be widely range much as about 22.55% more than control serum at 5th months of after the vaccination. That is one of the most considerable evalution in Black-leg No. 2 vaccination to Korean calevs as great presence of the maximum immune antibodies at for 5th month after the vaccination. (Table 4. and Fig 4.) 5. In the relationship between vaccinated rabbit and Korean calves, serum protein fraractions were to be changed within the 3 weeks, so that albumin fractions of vaccinated immune bovine serum are increased as directly ratio, while vaccinated rabbit immunized serum showed the decrease as to reciprocal ratio. Although, conclude that ${\gamma}-globurin$ fractions are increased gradually by and large on the vaccinated immune bovine serum(B. P. S.) and vaccinated immune rabbit serum(R. P. S.) together. (Table 6).
Mercury known as quicksilver, is the most common cause of heavy metal toxicity. Toxicity caused by excessive mercury exposure is now being recognized as a widespread environmental problem and is continuing to attract a great deal of public concerns. The mercury genotoxicity could be its effect on DNA repair mechanisms, which constitute the defense system designated to protect genome integrity. The objective of this study is to confirm that mercuric chloride inhibits the repair of gamma ray-induced DNA damage. The earthworm of Eisenia fetida was chosen for this study because it is an internationally accepted model species for toxicity testing with a cosmopolitan distribution. Experiments were done to identify the levels of DNA damage and the repair kinetics in the coelomocytes of E. fetida irradiated with 20 Gy gamma rays alone or with gamma rays after 40 mg $kg^{-1}$$HgCl_2$ treatment by means of the single cell gel electrophoresis assay. The Olive tail moments were measured during 0~96 hours after irradiation. The repair time in the animals treated with the combination of $HgCl_2$ and ionizing radiation was nearly five times longer than that in the animals treated with ionizing radiation alone. Also, E. fetida exposed to mercury showed a statistically lower repair efficiency of gamma ray-induced DNA damage. The results suggest that the mercury could even have deleterious effects on the DNA repair system. Influence of mercury on the DNA repair mechanisms has been confirmed by this study.
The experiment was carried out to investigate Insecticide susceptibility of the spiraea aphid (Aphis citricola) in apple orchards In Korea, using dipping method. Although insecticIde susceptibility vaned with local and seasonal populations, the susceptibility to demeLon S-rnethyl and phosphamidons was different from that Lo the other insecticides. The LCso values were 10 ppm for deltamethrin and chloropyrifos, 103 ~ 629 ppm for demeton S-methy1, acephate, phosphamidon, monocrotophos and vamldothion, 12,200 ppm for EPN, and 1,745 ppm for pirimicarb. 'When Insecticide susceptibility was compared between the S-clone selected from a population on the host plant spiraea and the R-clone selected from a population on the apple tree, the resistance ratio as expressed by RIS ratio of $LC_{50}$ was 78 for phosphamidon and 546 for pirimicarb. Esterase zymogram determined by the agar gel electrophoresIs revealed a significant difference between the clones. The activity of the E2, E5, E6 and E7 of the R-clone was higher than that of the S-clone, It is suggesLed that the Increased esterase activity may be involved in the mechanism of insecticide resistance in the spiraea aphid, although the involvement of other factor(s) may not be ruled out.
This study was conducted to evaluate the susceptibility of rice(Oryza sativa L.) cultivars to herbicide thiobencarb through the determination of protein patterns using SDS-PAGE. In both greenhouse and laboratory screening tests, IR 10198-66-2 and IR 9660-50-3-1 showed relatively tolerant response to thiobencarb, while IR 22, IR 31802-48-2-2-2 and IR 20656-R-R-R-6-1 were susceptible to it. The total protein content of susceptible cultivars markedly decreased as the thiobencarb concentrations increased, but tolerant cultivars such as IR 10198-66-2 and IR 9660-50-3-1 showed very slight changes suggesting that protein synthesis of susceptible cultivars may be inhibited by thiobencarb. The protein profiles of the tolerant cultivars were not much affected by thiobencarb treatment. However, the protein spots with molecular weights of 14.4 kD and 55 kD in the susceptible cultivar of IR 22 disappeared with the treatment of 3 ppm thiobencarb, indicating that differential susceptibilities of rice cultivars against thiobencarb can be attributed to their difference in protein metabolism affected by thiobencarb.
Heterogeneity in antigenic composition of Aspergillus fumigatus isolates from clinical specimens and in antibody response of patients infected with this fungus was investigated by immunoblotting. A considerable quantitative and qualitative difference was found in composition of the culture filtrate antigens derived from a reference strain (ATCC 13073) and 8 clinical isolates of A. fumigatus on SDS-PAGE and immunoblots. The crude CF antigen of a strain AFG7 was selected to identify the serologically reactive and specific components by immunoblotting. Out of more than 36 components separated by electrophoresis, transblotted to nitrocellulose sheet, and reacted with sera that showed a positive reaction to A. fumigatus or other fungal antigens on immunodiffusion tests, merely four or so were found useful to serodiagnosis of aspergillosis. An antigen of 82KD was found most reactive and specific component so as to be contained in the standard preparation. Several other components, for example 11KD, 26KD, 30KD and 31KD, also possessed relatively high reactivity and specificity and seemed to be worth while purifying and characterizing. Antibody binding activity (reactivity) of the antigenic components was clearly shown on immunoblots because some were faintly stained with Coomassie blue but darkly stained on immunoblots, while some others behaved contrary to them. A number of components seemed to carry not only species specific but cross reactive antigenic determinants. Immunoblotting proved very useful to identify serologically reactive and specific components that should be present in the antigen to be employed to the serodiagnosis of aspergillosis.

Conformational changes of allivin, a major allergen protein in garlic, by gamma irradiation were observed. Allium solution (3.0 mg/mL in 0.01 M PBS, pH 7.4) and raw garlic were irradiated in a gamma irradiation facility to be absorbed with doses of 2.5, 5.0, 7.5 and 10.0 kGy. After irradiation, a half of irradiated allivin solution was filtered through 0.22 ${\mu}{\textrm}{m}$ filtration kit. UV spectrum and SDS-PAGE were used for observing the conformational changes of allivin by gamma irradiation. Turbidity of allivin solution was increased by optical observation, and the result was validated by the increase of absorbance at 660 m. Turbidity of irradiated and filtered allivin solution significantly (p<0.05) decreased, and the concentration of soluble protein in the solution decreased by increase of the dose. Aggregation among allivin molecules was observed by SDS-PAGE, but irradiated and filtered samples showed decrease of allivin with reducing aggregation trend. SDS-PAGE profiles of irradiated raw garlic were not different. Results indicate that gamma irradiation can decrease the allivin in solution state which may impact on allergenic reaction of garlic.

A vehicle fuel economy is very important issue in the view of fuel cost and environmental regulation. The fuel economy is much improved according to the development of electric, electronic and mechanical technology, but up to now the measurement of it tests the given mode(LA-4, FTP-75, etc) within computer simulation program and engine dynamometer. This fuel economy is different with it of real road. The one of main reason is not considered the gradient of the road. To estimate the effects of fuel economy at highway with gradient in this paper, we measure the amount of fuel consumption and calculate the fuel economy of it with running the Youngdong highway with high gradient. Also this paper analysis and compares the fuel economy with gradient and without gradient when the vehicle runs the same driveway. Then we calculate the total energy created the difference of fuel consumption amount of the two cases and calculate the consumpted energy by tire driving force from the torque and power of engine in the simulation. This paper verifies the relation of the driving force and the total energy by creating the difference of fuel consumption amount. This paper also proposes the method of fuel economy improvement despite of gradient at the result.
It has been known that the silk degumming treated by hot alkali solution is easy to handle but is liable to yield poor-quality silk due to the degree of degumming loss, incomplete-degumming or over-degumming. Therefore, many studies have been carried out on the silk degumming by enzyme in order to improve the quality of silk. However, no attention has been paid to the physicochemical analysis of enzymatic degummed silk. In this paper, two different degumming methods, soap and enzymatic, are compared in aqueous solution state of silk fibroin. The results can be summarized as follows: There was no significant difference between two solutions on the bases of polarizing microscopy, TEM observation and SDS-PAGE. Spherulite of silk fibroin was not observed in polarizing microscopy, however the leaf-shape fibril structure was developed upon solidification. The size of spherulites of silk fibroin in TEM observation were 30~120nm with a wide range of size distribution. The intrinsic viscosity of enzymatic degummed fibroin solution was lower than that of soap degummed solution. This can be explained that the silk fibroin was more degraded by enzymatic degumming method compared with the soap degumming method. SDS-polyacrylamide gel electrophoresis showed that the fibroin molecule was composed of large component of molecule weight above 50 kd and small component of molecule weight about 20 kd. There was no difference in crystallinity between two degumming methods on the bases of results of DSC thermograms and IR spectra.
